Leukemogenicity and cell transformation mechanisms in vitro by Gross murine leukemia virus: analysis of virus subpopulations.

The leukemogenic activity of Gross murine leukemia virus adapted to rats was tested in W/Fu rats and NIH/Swiss mice. All animals infected with this virus developed thymic and nonthymic T-cell leukemia with a short latency period. It was observed that cell-free extracts from thymic lymphoma tissue of mice and rats, induced by either Gross murine leukemia virus or Gross murine leukemia virus adapted to rats, consisted of both small-plaque-forming and large-plaque-forming viruses, as determined by the XC plaque test. MCF-type virus was found in these virus complexes. Transformed cell foci were induced in SC-1 cell layers by double infection of the cloned MCF-type virus and an ecotropic virus. SC-1 cells containing transformed cell foci were shown to be tumorigenic upon inoculation into nude mice. The formation of transformed cell foci in mink lung cells was also observed after double infection with the cloned MCF-type virus and a xenotropic virus. The possible mechanism of leukemogenesis by endogenous viruses is discussed.

A new broad-spectrum aminoglycoside antibiotic complex, sporaricin. IV. Sporaricins C and D.

Two new aminoglycoside antibiotics, sporaricins C and D have been isolated from the culture broth of Saccharopolyspora hirsuta subsp. kobensis, which produced sporaricins A and B. The structures of sporaricins C and D have been determined to be 4-N-carbamoyl-glycylsporaricin B and 4-N-formylglycylsporaricin B, respectively. Sporaricins C and D are active against Gram-negative and Gram-negative bacteria including aminoglycoside-resistant strains.

Cortical structure of the lymph node. I. Effect of blockage of the afferent lymph flow to mouse popliteal nodes for protracted periods.

Changes in the lymphoid organization of the popliteal lymph nodes in mice, under conditions where the afferent lymph flow to the nodes was kept blocked for a protracted period, were the subject of this study. Surgical operations to interrupt the afferent lymphatics of the popliteal nodes were performed and the afferent lymph flow remained blocked for 15 weeks after the operation. A striking accumulation of small lymphocytes occurred within the nodes and the cellular density of the parenchyma and lumina of the sinuses greatly increased. These changes persisted throughout the 15 weeks period of observation. Germinal centres gradually diminished in size and ultimately disappeared completely from most of the operated nodes. Lymphatic nodules, which were aggregations of B lymphocytes, decreased in size to some extent but the number of these structures per whole node was not significantly different from that of the unoperated node. The deep cortex remained highly populated with small lymphocytes and showed no indication of fading out or of being replaced by the medullary structures.

Studies on familial amyloid polyneuropathy in Ogawa Village, Japan.

A newly discovered large concentration of familial amyloid polyneuropathy (FAP) in Japan is clinically outlined. The importance of the urinary secretion of IgA in these cases is stressed. Dimethyl sulfoxide administration to these patients induced clinical improvements accompanied by increased excretion of low molecular weight proteins. Amino acid composition of amyloid protein of the Ogawa village-type FAP cases was similar to that of primary amyloidosis.

A new aminoglycoside antibiotic, sannamycin C and its 4-N-glycyl derivative.

Streptomyces sannanensis KC-7038 used for the production of sannamycins A and B has also produced another antibiotic sannamycin C, in the culture broth. Physico-chemical characterization revealed that sannamycin C is a new aminoglycoside antibiotic having 6-N-methylpurpurosamine C and 2-deoxy-3-epi-fortamine. Its 4-N-glycyl derivative indicated inhibitory activity against Gram-positive and Gram-negative bacteria containing aminoglycoside resistant strains.
