Molecular alteration of the 140-megadalton plasmid associated with loss of virulence and Congo red binding activity in Shigella flexneri.

A plasmid of about 140 megadaltons has been associated with the invasiveness of Shigella flexneri. Upon subculturing in liquid media of fully virulent isolates of Shigella flexneri 2a YSH6000, which contains only a 230-kilobase-pair (kbp) plasmid in addition to 3.3- and 4.2-kbp cryptic plasmids characteristic to all S. flexneri strains, loss of invasiveness, loss of Congo red binding activity (Pcr), and complete loss of, or a deletion, or even a single-site IS insertion in the plasmid occurred simultaneously. This was ascribed to the fact that, once a noninvasive Pcr- cell has emerged, it overgrows the wild type as a consequence of its selective advantage in artificial media. A deletion map of the 230-kbp plasmid was made by analyzing SalI digests of 39 deletion derivatives plus 1 formed by insertion of an IS1-like element in independently isolated, noninvasive Pcr- mutants. Of 39 deletion derivatives, 16 belonged to a single type, and 6 belonged to another, suggesting deletion hot spots. The deletion map was confirmed and extended by analyzing 359 SalI-generated partial digests of the wild-type plasmid cloned into pBR322. Three copies of IS1-like elements were found on three different SalI fragments by Southern hybridization. Segments required for the Pcr+ phenotype seemed to occur at several different locations in the plasmid. Each of 28 representative Pcr- mutants were negative by the Sereny test. Hence, many, or possibly all, Pcr determinants were required for full virulence.

Molecular cloning of a genetic determinant for Congo red binding ability which is essential for the virulence of Shigella flexneri.

A DNA sequence of about 1.0 kilobase (kb) derived from a 230-kb (140-megadalton) plasmid in a fully virulent Shigella flexneri 2a strain, YSH6000, was cloned into Escherichia coli K-12 by selecting for the ability to bind Congo red (Pcr+ phenotype). It was mapped and localized within the SalI restriction fragment F on the plasmid. This clone converted an S. flexneri strain which is avirulent as a result of a small deletion in the plasmid to full virulence but did not convert those without the 230-kb plasmid or with a plasmid bearing a more extensive deletion. This indicates that there are more than two virulence determinants on the plasmid. Thus, this sequence contains a genetic determinant which is essential but not sufficient for full virulence and expression of the Pcr+ phenotype in S. flexneri but is essential and sufficient for expression of the Pcr+ phenotype in E. coli K-12. We noted that there exist some other regions on the 230-kb plasmid which express the Pcr+ phenotype in E. coli K-12. Although these regions express the Pcr+ phenotype less markedly than the region cloned in the present study, they do hybridize with it.

Isolation, characterization, and mapping of Tn5 insertions into the 140-megadalton invasion plasmid defective in the mouse Sereny test in Shigella flexneri 2a.

Using Shigella flexneri 2a YSH6000, we isolated 304 independent Tn5 insertion mutants in the 230-kilobase invasion plasmid, pMYSH6000. The site of each Tn5 insertion was assigned to 23 SalI fragments on the previously made SalI cleavage map of pMYSH6000. Among the 304 insertions, 150 were negative in expression of four phenotypes examined (mouse Sereny test [Ser], invasion into epithelial cells [Inv], Congo red binding [Pcr], and inhibition of bacterial growth [Igr] ): 12 were Ser- Inv+ Pcr+ Igr+, and 142 were positive in all four phenotypes. Tn5 insertions in the avirulent mutants were distributed in two separate SalI fragments, F and G, and in four contiguous SalI fragments, B, P, H, and D. Fragment G contains a novel class of determinant(s) which is required only for Ser+ but not for Inv+, Pcr+, and Igr+. Fragment F contains the previously characterized virF locus. B, P, H, and D each contained both virulent and avirulent Tn5 insertions. This indicates that more than two gene clusters exist within this region. Both are required for expression of all four virulence phenotypes.

Effects of apomorphine on morphine analgesia during the state of dopaminergic supersensitivity after chronic treatment with haloperidol.

Morphine induced-reduction in response to a repetitive electrical stimulation of the tail was measured in acute rats and rats chronically treated with haloperidol (1 mg/kg/d, for 7 d) following pretreatment with apomorphine. In acute experiments, a significant enhancement of the anti-struggling action of morphine was produced by haloperidol (1 mg/kg, i.p.). Low doses of apomorphine (30-480 micrograms/kg, i.p.) had no influence on the suppressing action of morphine on the struggling response induced by the tail stimulation. Following chronic treatment with haloperidol, the inhibitory action of morphine on the tail stimulation-induced struggling response was dose-dependently inhibited by very low doses of apomorphine (60-480 micrograms/kg, i.p.). A significant increase in 3,4-dihydroxyphenylacetic acid (DOPAC) levels was observed after administration of haloperidol or morphine in acute rats, whereas no change in DOPAC levels was found after administration of morphine or apomorphine in chronically haloperidol-treated rats. In these animals, basal striatal DOPAC levels were significantly decreased compared with those of vehicle-treated animals controls. The present results suggest that in rats treated chronically with haloperidol, the suppressive action of a low dose of apomorphine on morphine analgesia is due to an increased sensitivity of postsynaptic dopaminergic receptors to apomorphine.

[In vitro culture of mouse embryos for toxicological evaluation. (3): Effect of carcinogens on development of the embryos].

Mouse embryos were collected at the 2 cell or 8 cell stage. The embryos of each stage were exposed to 1 pM-10 nM 4-nitroquinoline-1-oxide (4-NQO) or 1 nM-10 microM N-methyl-nitro-nitrosoguanidine (MNNG) for 24 hr, and cultured to develop to blastocysts within clean medium. Since after exposure to 4-NQO, the appearance of early blastocysts and blastocysts were increased in exposure groups compared with controls, the growth to the 8 cell stage embryo (8-E) appeared to be late in development. Frequency of sister chromatid exchange (SCE) and mitotic index were increased with doses in the blastocysts (2-B) which derived from 2-E. There was little change in the SCE and mitotic index for blastocysts (8-B) which derived from the 8 cell stage embryo (8-E). Cessation of development in the 2 cell stage embryo (2-E) appeared in the 10 microM group during the period of exposure to MNNG. Development to the 8-E stage appeared to be slightly late in the other exposed groups. Thereafter at 48 hr after the initiation of culture, the exposed groups appeared to be more advanced in development than the controls. After this period, developmental rates to blastocysts were increased in the 10-100 nM groups. It appeared that the development of these groups was more advanced. In 2-B after exposure to MNNG, cell counts were dose-responsively increased in the 1 and 10 nM groups. The mitotic index in the 1 to 100 nM groups was higher than the controls.(ABSTRACT TRUNCATED AT 250 WORDS)

[Toxicological evaluation of an in vitro culture of mouse embryos; effect of non-carcinogenic mutagens for the development of embryos].

Short term drug toxicities were investigated using cultures of mouse embryos in the early stage of development. These embryos were collected at the two- or eight-cell stage. They were exposed to bleomycin (B1) or 6-mercaptopurine (MP) for 24 hr, thereafter, they were grown in BMOC-3 medium without these agents until the blastocyst stage. Total culturing period was 72 hr for the two-cell embryos and 48 hr for the eight-cell embryos. At the end of the culture periods, the number of cells, mitotic index and frequencies of sister chromatid exchange in these embryos after these exposures were unaltered. However, the death rate of embryos was elevated by the exposure to either B1 or MP. These agents are regarded as non-carcinogenic mutagens; therefore, it is suggested that these compounds are lethal to the embryos through an induction of mutation.

In vivo voltammetric study of dopamine release in the striatum following microinjection of apomorphine into the substantia nigra zona reticulata.

Effect of microinjection of apomorphine into the substantia nigra zona reticulata (SNR) on the dopamine (DA) release in the ipsilateral striatum was examined in the present study. To measure the DA release, we determined DA and/or DOPAC levels in the striatum by using in vivo voltammetry. The microinjection of apomorphine (10 micrograms/2 microliters) into the SNR produced a rapid and prolonged increase in the DA and/or DOPAC peak in the ipsilateral striatum. These results suggest that release of DA from dendrites of DA cells normally plays a physiological role in the SNR neurons.

Further dopaminergic supersensitivity to dopamine agonists by subchronic administration of haloperidol in rats treated with colchicine.

Effects of subchronic administration of haloperidol on the circling behavior induced by dopamine agonists were examined in rats treated with colchicine. Methamphetamine and apomorphine produced contralateral or ipsilateral rotation in colchicine-treated rats, and these circling behaviors were significantly increased by subchronic treatment with haloperidol. These results suggest that development of supersensitivity following colchicine and subchronic haloperidol may occur through different mechanisms.

[Chemoembolization therapy with cisplatin.lipiodol (CDDP.lipiodol) in primary liver cancer--with special reference to hepatocellular carcinoma].

From Jan., 1985 to Mar., 1986 thirty-six patients with primary liver cancer received transcatheter arterial chemoembolization therapy with Cisplatin (100 mg) blended into Lipiodol (5 ml) and simple embolization therapy with Gelfoam particles. Thirty-three cases out of 36 had hepatocellular carcinoma, one had hepatoblastoma and one had adenocarcinoma. Ten (31%) out of 32 had hepatocellular carcinoma, and showed objective tumor reduction greater than 50% (partial response) regarding the main tumor. Of the 33 there was one sudden death due to intracerebral hemorrhage. Only two out of 25 cases with daughter nodules showed slight reduction. Almost all cases with daughter nodules showed no response to chemoembolization therapy. Five patients died after chemoembolization therapy during the fifteen-month study period. Two patients died of liver abscess or cholecystitis and surrounding abscess, one died of intracerebral hemorrhage, one died of hepatic failure and the remaining case was one of tumor death.

Qualitative structure-activity relationships (QSAR) by the 1H NMR spectra of methylbenz[c]acridines.

The 1H NMR spectra of benz[c]acridine and its methyl derivatives (BAc) in CDCl3 have been measured. The total assignment of 1H NMR spectra and spin coupling constants of BAc were discussed. The good relationship between the delta delta (delta 2--delta 1) and epithelioma index (EI) have been gained. It was suggested that the utilization of exact NMR method in the screening of some chemical carcinogens can be adaptable.
