Glycosaminoglycans in lung carcinoma.

In order to better understand the chemical composition of carcinomas of the lung, glycosaminoglycans (GAGs) of lung tumors from 34 patients were studied histochemically and quantitated spectrophotometrically. When the GAG quantity in neoplastic tissue was compared with that in normal lung tissue, total GAG contents in various carcinomas of the lung ranged from 1.3 to 4 times the control. There were highly significant differences in the fractions of GAGs between the Clara cell type and the poorly differentiated types of adenocarcinomas. Mucus-producing adenocarcinoma of the goblet cell and the bronchial gland cell types, which histochemically showed abundant glycoprotein in epithelial cells, contained 74% of GAGs as hyaluronic acid. These results suggest that there seems to be a relationship between the composition of GAGs and the subtype and degree of differentiation of various lung carcinomas.

[Long-term follow-up after tumorectomy and irradiation for breast cancer].

From 1968 to 1976, 31 patients underwent breast conservation surgery (tumorectomy) and irradiation. A review of the available pathologic slides has revealed that 11 of that number had definite breast cancer. Of these 11 patients, three experienced a relapse, one with local recurrence only, and two with distant metastasis, though one of these two also had a local recurrence. The patient manifesting a local recurrence only was salvaged by a mastectomy, while the other two with distant metastases have died. Survival and disease-free survival in these 11 patients were at least equal to the results of other Japanese patients of the same ages and stages who had undergone a halsted operation during the same period. As for the other 20 patients, either their pathologic slides were not available, or their diagnosis was equivocal or benign. None suffered a relapse. Cosmetic results were excellent in most patients, and there were no second malignancies in the treatment volume.

Effect of hyperthermia on spermidine/spermine N1-acetyltransferase activity in Ehrlich ascites cells.

When Ehrlich ascites tumor cells were incubated at 42 degrees C, the activity of spermidine/spermine N1-acetyltransferase, a rate-limiting enzyme of polyamine biodegradation, decreased to 20% of the control level within 1 hr. Protein synthesis, judged from the incorporation of [3H]leucine into the acid-insoluble fraction, was less affected by heat exposure. The decrease in the enzyme activity caused by heat treatment could be reversed by switching the incubation temperature back to 37 degrees C. The reversion of enzyme activity was suppressed by cycloheximide but not by actinomycin D.

Diffuse interstitial pulmonary fibrosis and lung cancer.

Forty-two cases of lung cancer complicated with diffuse interstitial pulmonary fibrosis (DIPF) were selected from 13,056 autopsy cases. They were divided into primary (Group I and II) and secondary (Group III) DIPF, and histopathologic and clinicopathologic studies were made. The prevalence of lung cancer with primary DIPF was 17% (8/47) which was significantly (p less than 0.01) higher than that without DIPF (7% or 886/13,009). All eight cases of lung cancer in Group I and II were male, and 7 had cigarette-smoking history. The anatomical sites of lung cancer and DIPF were all peripheral in origin, and in 7 cases located primarily in the lower lobes (left:right = 6:1). Adenocarcinoma was the most common type. The study suggests that male DIPF patients with long clinical history should be followed thoroughly for lung cancer.

Effects of methyl mercuric chloride and sulfhydryl inhibitors on phospholipid synthetic activity of lymphocytes.

The effect of methyl mercuric chloride on the activity of phospholipid synthesis of rat lymph node lymphocytes was compared with that of sulfhydryl inhibitors. Measurement of the radioactivity of [14C]oleic acid and [14C]acetate incorporated into lecithin of cells during short-term incubation showed that all the inhibitors tested similarly reduced the incorporation. However, methyl mercuric chloride (MMC) was the strongest inhibitor, being effective at 4 microM and causing more than 80% decrease at 20 microM. Inhibition by the sulfhydryl inhibitors, at less than 40 microM, ranked as follows: N-ethylmaleimide greater than alpha-chloroacetophenone greater than hydroquinone greater than iodoacetamide. MMC also obstructed the enhancement by phytohemagglutinin of [14C]oleic acid incorporation into lecithin. MMC was effective at 2 microM, while the other agents had little or no effect at this concentration. Further investigation suggested that inhibition of phospholipid synthesis did not depend on reduced incorporation of oleic acid into the cellular membrane but on decreased turnover of the fatty acid into phospholipids after the incorporation. The viability of lymphocytes incubated with the agents was measured by trypan blue dye-exclusion test. More than 90% of the cells treated with MMC at a concentration as low as 20-40 microM died, but the SH inhibitors, including NEM which greatly inhibited the phospholipid synthesis, produced few cell deaths at these concentrations. These observations show that the SH inhibitors affect enzymes in phospholipid synthesis, whereas MMC not only inhibits the enzymes but kills cells.

An increase in water content of mouse erythrocytes infected with Plasmodium yoelii.

Mean cell volume and mean cell water content were examined in Plasmodium yoelii-infected mouse erythrocytes by gas chromatography and 3H-sucrose. Mean cell volume increased by 16% in infected erythrocytes with late trophozoites and schizonts. Mean cell water content further increased by 23% in the infected erythrocytes. Measurement of the erythrocyte potassium and sodium concentrations by an atomic absorption spectrophotometer revealed that the infected erythrocytes contained highly elevated sodium and slightly reduced potassium levels when calculated per single erythrocyte. It is suggested that membrane transport processes of sodium and potassium are changed in P. yoelii-infected erythrocytes and that a passive inflow of sodium takes place, which results in an increase in intra-erythrocytic water content.

Histogenesis of alveolar soft part sarcoma. An immunohistochemical and biochemical study.

In order to clarify the histogenesis of alveolar soft part sarcoma (ASPS), an immunohistochemical and biochemical study was performed on three cases. The immunohistochemical study indicated the presence of actin, desmin, vimentin, and Z-protein in all cases. On the other hand, intermediate filaments other than desmin and vimentin were not detected immunohistochemically. The presence of desmin and Z-protein strongly suggests the myogenic character of this tumor. As to whether ASPS shows striated muscle differentiation or smooth muscle differentiation, the immunohistochemical absence of myoglobin in the three cases suggests that the tumor does not differentiate in the direction of striated muscle. However, biochemical assay of subunits of enolase revealed significantly high amounts of beta-enolase, which is known as a marker for striated muscle, in all three cases. The determined values--735, 426, and 584 ng/mg of protein --are indicative of striated muscle differentiation. In addition, the immunohistochemical study of all cases revealed the presence of beta-enolase in tumor cells. These data definitely show the myogenic character and rhabdomyoblastic differentiation of ASPS.

Neurofilaments of Kultschitsky cells in human lung.

The immunohistochemical localization of three triplet proteins of neurofilaments in normal Kultschitsky cells and tumourlets of the human lung has been studied using avidin-biotin-peroxidase complex (ABC) method. Kultschitsky cells and tumourlets have been stained with antisera against 68 K, 150 K and 200 K dalton components of the neurofilaments, respectively. Ultrastructural observations of human Kultschitsky cells have revealed the presence of bundles of intermediate filaments as well as microtubules and neurosecretory-type granules. In the tumourlets, similarly sized filaments were found, but were relatively scarce. Since intermediate filaments are thought to be specific to differentiated cells and neurofilament proteins are restricted to the neuronal tissues, we conclude that Kultschitsky cells of the lung are of neuronal nature.

Decreased water and potassium content in erythrocytes in essential hypertension.

The water content in erythrocytes of subjects with borderline or established essential hypertension was measured by using gas-liquid chromatography and was found to be lower than that in normotensive controls (p less than 0.01). The water content in erythrocytes of normal controls (n = 14), borderline hypertensive subjects (n = 18), and established essential hypertensive subjects (n = 23) was (mean +/- SE) 71.0 +/- 0.2%, 69.9 +/- 0.2%, and 69.3 +/- 0.1% (vol/vol), respectively. A definite negative correlation was found between water content of erythrocytes and mean arterial pressure in normotensive and hypertensive subjects (n = 60, r = -0.59, p less than 0.001). Although there was no statistically significant between-group difference in the sodium content, the potassium content of erythrocytes from subjects with essential hypertension was significantly lower than that of normotensive controls (0.205 +/- 0.003 vs 0.222 +/- 0.004 mumol/mg dry red blood cells; p less than 0.01). There was no between-group correlation of sodium and water content in erythrocytes, but the potassium content correlated with the water content (n = 46, r = 0.49, p less than 0.001).

Growth of Plasmodium falciparum in sodium-enriched human erythrocytes.

Transport of nutrients into animal cells is driven by transmembrane gradient of Na+ across the plasma membrane. The protozoan malaria parasite, Plasmodium falciparum, however, grows within the host human erythrocytes, in which the cytoplasmic concentration of Na+ is maintained low by the membrane Na+, K+-ATPase. Our experiments show that human erythrocytes enriched with Na+ by treatment with ouabain (an inhibitor of the ATPase) will support the growth of P. falciparum in culture.

Expression of neurofilament triplet proteins in human neural tumors. An immunohistochemical study of paraganglioma, ganglioneuroma, ganglioneuroblastoma, and neuroblastoma.

Intermediate filaments which are specific to neural cells, ie, neurofilaments, consist of three subcomponents--68, 150, and 200 kd. Thirty human neural tumors were examined for the presence of these three subcomponents by means of their monospecific antisera. All 8 paragangliomas contained cells that were positive for the 68-kd component, but only 5 of them had cells positive for the 150-kd and 200-kd components. All 4 ganglioneuromas and 11 ganglioneuroblastomas contained cells that reacted with antibodies to all three components. All 7 neuroblastomas had cells reacting with antibody to 68 kd, but only 3 of them had cells that reacted with antibodies to 150 kd and 200 kd. In each case, the number of positive cells depended on the antibody used. The largest number reacting with antibody to 68 kd and the smallest with antibody to 200 kd. Furthermore, it was possible to detect tumor cells in which the 68-kd subcomponent existed by itself, but no tumor cells in which the 150-kd or 200-kd subcomponent existed alone could be detected. These results seem to indicate that antibody to the 68-kd component is sufficiently discriminating to be applied diagnostically.

Infantile digital fibromatosis. An electron microscopic and immunohistochemical study.

Three cases of infantile digital fibromatosis were studied by electron microscopy and immunohistochemistry. The tumor was made up equally of myofibroblasts containing long narrow bundles of microfilaments with dense bodies. Another striking feature was the intracytoplasmic electron-dense inclusion. The inclusions were constituted of packed fibrils similar to those of the long bundles. At the periphery of these inclusions were seen prominent felt-like filamentous structures; the diameter varied from 5-12 nm. The 10 nm filaments were seen most abundantly in the cytoplasm surrounding the inclusions, but sometimes they were also found within the inclusions themselves, mainly at their periphery. These findings suggest that the inclusions may represent an abnormal accumulation of cytoskeletal proteins. On the other hand, various kinds of immunohistochemical study showed that actin, actomyosin, myosin, and vimentin were distinctly observed in cytoplasms of tumor cells. They were located fringing the intracytoplasmic inclusions, but the inclusion themselves showed negative findings, thus indicating a hollow-like staining pattern. These data can be synthesized as follows. The inclusion results from an accumulation of cytoskeletal proteins (actin-myosin complex and vimentin, at least), and antigenicity seems to be lost by for a certain mechanism or in the degenerative process.

Glycosaminoglycans in malignant diffuse mesothelioma.

Because of frequently encountered diagnostic difficulty due to a morphologic similarity between diffuse pleural mesothelioma and peripheral pulmonary adenocarcinoma, glycosaminoglycans (GAG) of human malignant diffuse mesothelioma were histochemically stained and chemically quantitated, and were compared with GAG of papillary adenocarcinoma of the lung. In all seven patients, the diagnosis of diffuse mesothelioma was confirmed morphologically by such findings as abundant bushy microvilli on cell surface and intermediate filaments in cytoplasm. The total GAG in mesothelioma obtained from fresh materials (5 cases) was significantly increased over that in pleural connective tissue (P less than 0.01) and lung adenocarcinoma (P less than 0.02). Two dimensional electrophoretic separation of GAG of mesothelioma and lung cancer showed hyaluronic acid, heparan sulfate, heparin, dermatan sulfate and chondroitin sulfate; among them, the two predominant fractions were hyaluronic acid and chondroitin sulfate. In the quantitative analysis, the hyaluronic acid content of mesothelioma averaged 57% of the total GAG, but that of lung adenocarcinoma, 38%. The results suggest that chemical analysis of GAG may be useful as supplementary diagnostic procedure to morphologic examination in the differentiation of diffuse mesothelioma from papillary adenocarcinoma of the lung.

Cellular differentiation of epithelioid sarcoma. An electron-microscopic, enzyme-histochemical, and immunohistochemical study.

For the purpose of clarifying cellular differentiation of epithelioid sarcoma, studies based on various methods were performed. Enzyme histochemical studies showed that epithelioid sarcoma tumor cells have characteristics intermediate between epithelial cells and the large plump cells of synovial sarcoma-incomplete epithelial differentiation. For alkaline phosphatase and adenosine triphosphatase particularly, positive cells and negative cells coexisted, as in the large plump cells of synovial sarcoma. Immunohistochemical studies for alpha 1-antitrypsin, alpha 1-antichymotrypsin, vimentin, and keratin also showed that epithelioid sarcoma tumor cells are very similar to the large plump cells of synovial sarcoma and have incomplete epithelial differentiation. For example, the examinations of serial sections and double staining methods revealed that keratin-positive cells are always vimentin-positive in epithelioid sarcoma and in the monophasic area of synovial sarcoma. Electron-microscopically, bundles of intermediate filaments and filopodia toward the intercellular lumen were observed, as in the monophasic area of synovial sarcoma. The results of enzyme-histochemical and immunohistochemical studies of non-neoplastic synovial lining cells, performed here for the first time, are also discussed.

[Does weak local gamma irradiation modify the effect of CDDP on mouse mastocytoma?].

The possibility that an irradiation with small doses of gamma rays given shortly before might enhance the cure effect of cis-diammine-dichloroplatinum (II) (CDDP) was tested in tumor bearing mice. Tumor cells of FMA3, a cloned cell line of murine mastocytoma, were transplanted in the muscle of a hind leg in one experiment and in the abdominal cavity in another experiment. In both cases, a single injection of CDDP at 4 mg/kg was effective to lengthen the mean survival time of mice. gamma rays at the dose of 1, 2 and 4 Gy given 3 hours before did not modify the effect of CDDP.

[Combined effects of gamma rays, cis-diamminedichloroplatinum (CDDP) and systemic hyperthermia mastocytoma transplanted into muscle in the mouse].

Comparison was made between the effects of local irradiation of gamma rays, s. c. injection of cis-diamminedichloroplatinum (II) (CDDP), systemic hyperthermia and their combinations on the i. m. transplanted murine mastocytoma. Increase of the mean survival time (M. S. T.) by a factor of 1.72 and of 1.68 was achieved by a single irradiation at 20 Gy, given on day 5 after transplantation, and by injections of CDDP at 2 mg/kg, given s. c. on days 5 and 12 after transplantation, respectively. Increase of M. S. T. by a factor of 1.10 which was achieved with systemic hyperthermia of 41.8 degrees C of the core body temperature for 5 min, given twice, on days 5 and 12 after transplantation, was not statistically significant. The most effective one among all possible combinations within the 3 modalities was that of radiation and CDDP. Increase of M. S. T. was by a factor of 4.01.
