Cardiovascular effects of Gardenia florida L. (Gardeniae Fructus) extract.

Intravenous injection of Gardeniae Fructus extract in rats significantly lowered the systemic arterial pressure which was related to a decreased cardiac output with decreased stroke volume. Gareniae Fructus extract decreased the myocardial contractility of perfused isolated rat heart. Electrocardiogram revealed evidence of myocardial damage and atrioventricular block after a large dose of the extract.

The terminal hepatic microcirculation in the rat.

The hepatic microcirculation was observed microscopically in the transilluminated liver of the rat. The portal and hepatic venous microvessels were classified into four orders according to their branching hierarchy, and the hepatic sinusoids into branching, direct and interconnecting types according to their topographic arrangements. The diameters of the various orders of microvessels and types of sinusoids were measured by serial photomicrography, and the velocity of the erythrocytes in these various microvessels and sinusoids by the dual-slit photometric technique. The microvascular volume flows were calculated from these data. In both portal and hepatic venous systems, the erythrocyte velocity and the volume flow significantly decreased in successive orders of the microvessels in apparent relation to the cross-sectional areas. The diameters of the three types of sinusoids did not significantly differ, but the velocity of the erythrocytes in the direct sinusoid was significantly faster than that in the branching sinusoid while that in the inter-connecting sinusoid fluctuated widely.

Adrenaline-induced acute massive pulmonary oedema in the dog.

Pericardiotomy followed by intravenous injection of adrenaline (1 mg/kg body weight) provides a reliable method for the production of acute massive lung oedema in the dog. The lung wet weight: heart wet weight ratio is found to be the best assessment of the degree of lung oedema. A ratio greater than two represents massive lung oedema.

Hepatic sinusoidal responses to intraportal injections of phenylephrine and isoprenaline in the rat.

1. Specific alpha- and beta-adrenoreceptor agonists, phenylephrine and isoprenaline, were injected intraportally into the intact rat liver under direct microscopic observation by an in vivo transillumination technique. 2. The diameter of a hepatic sinusoid and the intra-sinusoidal erythrocyte velocity were quantitatively measured, and the sinusoidal volume flow was calculated from these two parameters. 3. Results show that phenylephrine causes a sinusoidal constriction and an increased sinusoidal blood flow, whereas isoprenaline causes the opposite effects on the sinusoids. 4. All the sinusoidal responses to phenylephrine and isoprenaline were dose-dependent and were possibly related to the direct effect of these drugs on the sinusoids.

Circulatory changes in cold-acclimation and cold stress.

1. The cardiovascular changes of conscious cold-acclimated (CA) and warm-acclimated (WA) rats during exposure to 5 degrees C or 28 degrees C were studied. 2. The cardiac output, heart rate and stroke volume of Ca rats exposed to 5 degrees C and of WA rats during cold stress were significantly greater, and their calculated total peripheral resistance significantly less than those of WA rats ats exposed to 28 degrees C. These results show that circulatory changes participate in cold acclimation and cold stress. The circulatory changes in the two conditions were compared and the mechanism of the observed differences were discussed. 3. CA rats exposed to 28 degrees C showed a striking decrease of oxygen consumption and arterio-venous O2 difference, but significant circulatory changes were decreased heart rate and cardiac index only, indicating that the response was mainly metabolic.
