Studies on receptor for mouse erythrocytes in lymphocytes of patients with malignant lymphoma.

In a group of 30 healthy subjects and 27 patients with the diagnosis of non-Hodgkins, lymphoma, the rosette test was performed with erythrocytes of mice of the C57 BL strain according to the method of Stathopoulos and Elliott. Lymphocytes were isolated from peripheral blood, and in four cases also from lymph nodes, on an Uropolinum-Polfa gradient. In healthy subjects, 12.6 +/- 6.9% of lymphocytes possessed receptors for mouse erythrocytes. The values in the group of patients were significantly higher (31.8 +/- 26.8%). Moreover, in the group of chronic lymphocytic leukemia, the values (68.0 +/- 20%) were significantly higher than in the group with lymphosarcoma (9.7 +/- 7.7%). Results of the rosette test with lymphocytes isolated from lymph nodes were the same as with blood lymphocytes of these patients: a high level of 68% was observed in a patient with chronic lymphocytic leukemia, and low levels (between 2% and 9%) in three patients with lymphosarcoma. It was concluded that the rosette test with erythrocytes from mice of the C57 BL strain detects low differentiated forms of B lymphocytes and could be useful in the diagnosis of chronic lymphocytic leukemia and differentiation from lymphosarcoma.

The monitoring of lymphocyte subpopulations in non Hodgkin lymphoma patients during chemotherapy.

Numerous examinations of the changes in B, T, null subpopulations were systematically carried out in 20 non Hodgkin lymphoma patients 8, 24, 48 hours after the therapy onset and subsequently at intervals of a few days during and after each chemotherapy cycle (COP, Knospe). T cells were evaluated by examining the receptor for neuraminidase pretreated sheep erythrocytes (nE), B lymphocytes by detecting surface immunoglobulins (SmIg) with normal and Fc deprived anti Fab2 globulin, moreover receptors for mouse erythrocytes (Em) and for C3 (EAC) were examined. A characteristic rise of the dominating subpopulation was observed during the first phase of the drug application which, most probably, was due to an expulsion of the pathological cells from involved lymphatic organs to the blood. On the basis of our clinical observations a conclusion has been drown, that this type of monitoring possibly renders a maximal effective treatment, enables the control of the accompanying immunosuppression and offers an additional criterion of the remission.

Lymphocyte subpopulations bearing different surface markers in non-Hodgkin lymphoma.

Lymphoid cells isolated from peripheral blood and lymph nodes were examined by using the rosette test with sheep erythrocytes (E), the immunofluorescent staining of the surface immunoglobulins (SmIg), the rosette test with mouse C 57 BL erythrocytes (Em and a combined test (SmIg + E). These studies were performed in 143 patients with various subtypes of non-Hodgkin lymphoma and in 110 controls. In the control group the following percentage of cell types was obtained: 1) blood (n = 110): T-68 +/- 12, B- 20 +/- 3, O- 11 +/- 7, BT- 1 +/- 1.4, Em+ cells--9 +/- 7, C3+ cells--21 +/- 3, 2) lymph nodes (n = 23): T- 66 +/- 10, B- 17 +/- 10, O- 15.7 +/- 9, BT- 0.8 +/- 1.7. In non-Hodgkin lymphoma lymph nodes a statistically significant increase of null and B cells- was noted in comparison to the controls (p less than 0.001). Similar changes were observed in the blood of non-Hodgkin lymphoma patients in which the leukocyte count exceeded 10(3)/microliter, whereas in the cases with low leukocythosis the B-T values were within the nomal range. It is concluded that the marker studied does allow to differentiate in most cases between the non-Hodgkin lymphoma and other disorders of the lymphoid system. They also give additional information about the more exact nature of the lymphoid malignancies.

[Transformation of type T lymphoma into type B lymphoma].

A 45-year-old woman with T-cell lymphoma is reported. Enlargement of the peripheral lymph nodes was not observed. Splenectomy was performed for hypersplenism and during the operation the mesenteric and retroperitoneal lymph nodes were found to be greatly enlarged. Three years after the diagnosis of lymphoma malabsorption syndrome developed. After 5 years sudden clinical deterioration appeared, with loss of reaction to effective treatment given as yet. Conversion of the predominating lymphocyte subpopulation from T to B cells was observed owing to repeatedly done monitoring of peripheral blood lymphocytes. The pattern of the newly developed lymphoma was that of a highly malignant B-cell proliferation. The possible mechanisms of the conversion discussed by the authors include: simultaneous proliferation of B and T populations of lymphocytes, B-cell lymphoma induction by chemotherapy, and infection by Epstein-Barr virus.
