A computer program package for storing and retrieving DNA/RNA and protein sequence data.

A computer program package has been made available which contains five compound programs written in FORTRAN 10 (for DECsystem-10). Program DATBAS is for storing and improving DNA sequence data, especially those obtained by the sequencing method using M13 phages. Programs NUCDAT and PROTEN are for analyzing DNA/RNA and protein sequence data, respectively. They contain various options to help users in analyzing DNA/RNA and protein sequence data. With program NUCDAT, it is also possible to get access to the EMBO Nucleotide Data Library. This can be achieved by running program COPY to create two data files from the library. Program LITRAT enables users to prepare a scientific literature file convenient for writing scientific articles, and program STRAIN for storing information concerning bacterial and/or plasmid strains.

Insertion of IS1 into the rpsE gene for ribosomal protein S5 causes cold-sensitivity in Escherichia coli.

The nucleotide sequence of the rpsE gene for the ribosomal protein S5 of a mutant of E. coli showing cold-sensitive growth revealed the presence of an insertion sequence, IS1, near the 3'-end of the gene. This mutant grows very slowly even at the "permissive temperature" (30 degrees C). At temperatures below 20 degrees C, its growth becomes negligible. It is likely that the presence of IS1 disturbs the continued translation of the polycistronic messenger RNA of the "spc-operon" from the point of IS1 insertion downwards, especially at the unfavorable temperatures.

Signet ring cell carcinoma of the breast.

A rare case of signet ring cell (SRC) carcinoma of the breast occurring in a 51-year-old female is presented. She was first admitted under the diagnosis of gastric carcinoma. Incidentally, a right breast tumor was detected and double cancer was thus suspected clinically. Concurrent right mastectomy and partial gastrectomy were performed. Histologic examination on the removed tumors revealed infiltration of numerous SRCs. The signet ring appearance of tumor cells resulted from formation of a large intracytoplasmic globule. From this unique cytologic feature along with the presence of in situ lesion in the breast tumor, it was concluded that the gastric malignancy was a sequela of metastasis of infiltrating lobular carcinoma. Wide-spread metastasis later occurred, especially on the serosal surfaces. The importance for the knowledge about the unique metastatic pattern of SRC carcinoma of the breast was emphasized.

Structure and biological activity of ascamycin, a new nucleoside antibiotic.

A newly isolated strain of Streptomyces sp. produces a new nucleoside antibiotic, ascamycin and the corresponding dealanyl derivative. The structure of ascamycin was determined to be 2-chloro-9-beta-[5-O-(N-L-alanyl)sulfamoyl-D-ribofuranosyl]-adenine. Remarkable selective toxicity of ascamycin compared to the dealanyl derivative was accounted for on the basis of a dealanylating enzyme present in the envelope of sensitive bacteria. After dealanylation, it becomes permeable to cell membrane.

[Effect of extent of lymph node dissection on prognosis of esophageal cancer].

The lymph node metastasis of the esophageal cancer are located in three main regions such as neck (H), thoracic cavity (T) and abdominal cavity (B). We studied on the relation between the extent of lymph node dissection and prognosis, and that between the lymph node metastasis and prognosis in each region (H.T.B). The long-term survival among the cases with adequate dissection, especially with the adequate dissection in the thoracic region, was better than that among the cases without adequate dissection. Furthermore, adequately dissected cases got better survival compared to the cases without adequate dissection with post-operative prophylactic irradiation. However, in the prognosis of the esophageal carcinoma, hematogenous metastasis have to be taken into consideration as well as lymph node metastasis. Therefore, in order to improve the prognosis of esophageal cancer, it is indispensable to treat the multidisciplinary therapy with immunochemotherapy as well as complete lymph node dissection.

Specific binding of chartreusin, an antitumor antibiotic, to DNA.

Chartreusin, an antitumor and antibacterial antibiotic, was found to inhibit negatively superhelical DNA-relaxation catalyzed by prokaryotic topoisomerase I and conversion of the superhelical DNA into unit length linear form catalyzed by single-strand-specific S1 nuclease. The inhibitory effect of the agent was due to the binding to DNA causing the alteration of tertiary structure. To characterize the binding specificity, we investigated the protection of DNA against cleavages by various restriction endonucleases. It was evidenced that the binding of the agent is not at random and correlates to the sequence 5'CGC 3' 3'GCG 5' on DNA stretch.

Pharmacokinetic approach for reconstruction of predetermined plasma insulin profiles by continuous subcutaneous insulin infusion in depancreatized dogs.

A preprogrammable insulin delivery system which can provide the flexibility of varying plasma insulin concentration in a physiological manner has been developed. This system involves a program of continuous subcutaneous infusion of insulin at variable rates to reproduce arbitrarily selected plasma insulin concentration-time profiles. The desired insulin infusion rate profile was calculated from the desired insulin pattern using pharmacokinetic parameters experimentally determined by measurement of immunoreactive plasma insulin following a subcutaneous bolus dose to depancreatized dogs.

[Basic and clinical studies on metastatic cancer--with special reference of multidisciplinary treatment of gastric and colorectal cancer patients with hepatic metastasis].

We studied 161 gastric cancer patients with P0, H(+) and 51 colorectal cancer patients with P0, H(+) from among cancer patients of the digestive organs and obtained the following conclusions. The effective treatment for synchronous hepatic metastasis was regarded as the group with surgical removal of the primary lesion plus hepatic resection plus chemotherapy, demonstrating most favorable prognosis in both gastric and colorectal cancer patients. Prognosis of the group treated with surgical removal of the primary lesion plus hepatic resection plus chemotherapy, was the most excellent and was followed by the group with surgical removal of the primary lesion plus chemotherapy and group with surgical removal of the primary lesions and group surgical removal of the primary lesion in this order. Concerning chemotherapy after surgical removal of the primary lesion, continuous intraarterial infusion therapy with FML regimen combining Lentinan revealed more favorable prognosis also in both gastric and colorectal cancer patients. Hepatic resection with aggressive reduction surgery was of significance in the treatment for the patients with hepatic metastasis of H1 and H2. Long-term survival is also expected for the patients with metachronous hepatic metastasis of H1 by hepatic resection plus chemotherapy.

[Thoracic esophageal cancer in a young adult].

A 34-year-old Japanese male was admitted to the Second Department of Surgery, Chiba University Hospital complaining of swallowing difficulties of three months duration. Radiological examination with barium swallowing revealed thoracic esophageal cancer, classified as EiEa, was 6cm long and of the serrated type. Preoperative combined therapy included Fast Neutron, Pepleomycin, Bestatin and hyperthermia. The operative procedure was total thoracic esophagectomy and ante-thoracic esophago-gastrostomy using the right thoracotomy approach plus laparotomy. Postoperative combined therapy included Linac irradiation and Cisplatin. His postoperative course was uneventful and he was discharged without complications. The tumor was of ill-defined ulcerative type macroscopically; the pathological diagnosis was moderately differentiated squamous cell carcinoma.

Randomly controlled study of chemotherapy versus chemoimmunotherapy in postoperative gastric cancer patients.

From September 1979 through March 1981, a total of 302 patients with gastric cancer and undergoing gastrectomy at the Department of Surgery at Chiba University Hospital and its 14 affiliated hospitals was studied for clinical effectiveness of immunotherapy with Nocardia rubra cell wall skeleton. The patients were stratified by gross stage of cancer and degree of operative curability. They were then assigned randomly to either chemotherapy group or chemotherapy plus immunotherapy group. Immunotherapy used was intradermal injection of 400 micrograms of N. rubra cell wall skeleton which was given weekly for the first month and monthly thereafter. After the specimen was examined microscopically, the patients were classified by histological stage of cancer and radicality of surgical intervention into curative or noncurative groups. The patients were surveyed for survival period in December 1981. The postoperative survival rate was compared in patients of histologically curative or noncurative resection cases between the two treatment groups. No statistical difference was detected between the groups in age, sex, or operative procedures that might influence the patient's survival. As a result, statistical intergroup difference in survival rates was not seen in patients of the curative group, probably due to a short observation period. However, the intergroup difference in survival rates was statistically significant in patients of the noncurative group (p less than 0.01). These results indicate the adjunctive effect of N. rubra cell wall skeleton as an immunotherapeutic agent in patients undergoing gastrectomy for gastric cancer.

Studies on the causes of deaths from esophageal carcinoma.

Statistics on the causes for deaths of 638 patients operated on in our department for resection of cancer of the intrathoracic esophagus (squamous cell carcinoma) during the period from 1959-1979 showed that the major causes for direct operative deaths were pyothorax, pulmonary complications, failure of the sutures, and postoperative hemorrhage. Among operation survivors, recurrence was the most frequent cause of death, responsible for the deaths of as many as 80% of less-than-five year survivors; and recurrence in the cervical, supraclavicular fossa, and superior mediastinal lymph nodes and that in the other organs were the frequent causes for the deaths of two- to three-year survivors. Pulmonary complications were the causes for the deaths of 50%, and recurrence for the deaths of 30% of five- to ten-year survivors. Recurrence in the digestive organs other than the esophagus and cardiovascular diseases were the frequent causes for the deaths of more-than ten-year survivors, while none of these survivors died of recurrence.
