Influence of testosterone on hydronephrosis in the inbred mouse strain DDD.

Hereditary hydronephrosis was detected in all of the male mice of DDD inbred strain maintained at the National Institute of Animal Health Japan, but in only a few of the females. From the standpoint that male hormones are related to the development of hydronephrosis in this strain, the incidence and severity of the disease were investigated in gonadectomized mice treated with testosterone. In the males, the incidence of hydronephrosis was 50% (7/14) in the control (gonadectomized) group, and 73.3% (11/15), 100% (13/13) and 100% (12/12) in the 0.1 mg, 3.3 mg and 10 mg treatment groups respectively. The same tendency was observed in the female animals, though the incidence in each group was not so high. In both sexes the degree of severity increased in proportion to the dose of testosterone administrated. Blood testosterone levels were higher in intact DDD mice than in C57BL/6 mice, which had normal kidneys in both sexes. These results suggest that male hormones play a significant role in the development of hydronephrosis.

A malignant pheochromocytoma with ileus, polyuria and hypercalcemia: a case of recurrence 17 years after the initial operation.

A case of malignant pheochromocytoma, with a recurrence 17 years after the initial diagnosis of benign pheochromocytoma, was presented. The autopsy revealed multiple metastases of pheochromocytoma to the bone marrow of the thoracic and lumbar vertebrae. Of particular note is the fact that the patient was associated with paralytic ileus, polyuria and hypercalcemia and that he died of hypercalcemia crisis. Cases like this appear to be very rare.

Prevalence of hypertension in diabetes mellitus--its relation to diabetic nephropathy.

The prevalence of hypertension was studied in 374 patients with non-insulin dependent diabetes mellitus (NIDDM) and in 1197 non-diabetic controls. The diagnosis of hypertension was made when the mean systolic pressure of three measurements on different occasions was 151 mmHg or greater, or the mean diastolic pressure was 91 mmHg or greater. The prevalence was 42.8% in the diabetics and 17.8% in the controls. It showed a significant difference over age 31 (p less than 0.05). Proteinuria (p less than 0.001), abnormal ECG (p less than 0.01), hyperlipidemia (p less than 0.05) and hypertensive or sclerotic changes of the retina (p less than 0.001) were more frequently observed in the diabetics than in the controls. Hypertension was found in 71% of those with proteinuria, 48% with diabetic retinopathy, 61% with abnormal ECG and 54% with hyperlipidemia in the diabetics. The incidence of proteinuria was 22.8% in the diabetic hypertensives and was 8.3% in the non-diabetic hypertensives (p less than 0.001). 24 subjects out of 119 diabetics, who were normotensive at their initial visits, became hypertensive within 10 years (N-H), and 95 remained normotensive (N-N). 38% of N-H showed proteinuria already on their initial examinations and 3% of N-N did. 73% of those who showed proteinuria on their initial examination became hypertensive and 13% of those who were free from proteinuria did (p less than 0.001). The results suggest that diabetic nephropathy plays an important role in developing hypertension in diabetics.

[A study of prostatic tissue levels of cefoperazone].

Prostatic tissue levels of Cefoperazone (CPZ) were studied after the administration of 3 g of CPZ, in 16 patients with prostatic hypertrophy who underwent retropubic prostatectomy. The CPZ concentration in the surgical capsule at 60 min. after the administration of CPZ was about 2 times higher than that in the gland. As compared with the histological type of removed gland, the levels in the glandular hyperplasia were slightly higher than that in the fibromuscular and mixed type hyperplasia, but the difference was not statistically significant. CPZ was found to be effective for the treatment of post-operative and bacterial prostatitis, since the CPZ levels of prostatic tissue were considerably high and above the minimal inhibitory concentration level for most bacteria.

Factors influencing the healing rate of gastric ulcer in hospitalised subjects.

A multiple linear regression analysis was carried out on 75 inpatients with gastric ulcer. In order to elucidate the effects of various factors - endoscopic and roentgenological findings, age, sex, medical history, and drugs such as antacids, anticholinergics or both - on the healing rate, these factors were compared between those with ulcer which healed within eight weeks after treatment and those which did not. In patients over 50 years of age, alcohol consumption of over 60 g per day until admission, duration of present ulcer pain for over three months, single ulcer, ulcer located in the lesser curvature and uneven elevation around the ulcer, there was significant delaying effect on ulcer healing. Drug ingestion, sex, smoking habits until admission, size, depth, and shape of ulcer, coexisting gastritis, and past and family history of ulcer disease had no significant effect on healing after eight weeks. The patients with less than two unfavourable factors (n = 46) had the best healing rate (100%) compared with those with three (n = 20) or four or more (n = 9) unfavourable factors. The healing rate of the latter two groups was 60% and 22%, respectively (p less than 0.001). A prognostic score based on these six factors represents the severity of gastric ulcer disease with regard to the healing rate in patients prescribed antacids, and/or anticholinergic drugs.

Response selectivity of neocortical neurons to specific odors in the rabbit.

Odor responses of neocortical neurons in rabbits under light pentobarbital anesthesia were studied in a restricted area in the lateral half of the prefrontal cortex close to the frontal pole. A preevent cumulative sum chart provided a visual display of the change in unit discharge rate following each odor stimulation, and critical levels on each chart provided a visual display of statistical significance. The neocortical neurons studied were activated predominantly by biologically significant odors, such as feces, urine, and dry food. Of those same neurons, 90% failed to respond to pure-chemical odors, and no neurons were found that responded to pure-chemical odors alone. Some neurons responded exclusively to one of the animal-product odors. Some odor-sensitive neurons were affected by electrical stimulation of the mediodorsal thalamic nucleus. Responses to the animal-product odors were observed after ablation of the trigeminal and/or vomeronasal inputs. The responses to these pheromone-like substances are discussed in the context of a central role for odor-sensitive neocortical neurons in cognitive aspects of discrimination of specific odor patterns in the mammals.

Inheritance of hydronephrosis in the inbred mouse strain DDD.

The mode of inheritance of hydronephrosis was investigated by crossing inbred DDD mice having a high incidence of hydronephrosis and C57BL/6 mice having normal kidneys. In the males, incidences of hydronephrosis in F1 animals were intermediate between the two parental strains at a rate of 32.6% in (DDD x C57BL/6)F1 and 23.4% in reciprocal F1. The same tendency was observed in F2 male animals. In BCF1 males, the number of affected mice was higher in (C57BL/6 x DDD) F1 x DDD (72.4%) than in (DDD x C57BL/6)F1 x C57BL/6 (11.1%). A few affected mice were found among the females of hybrids F1, F2 and BCF1. These results suggested that hydronephrosis in the DDD strain of mice was controlled by polygenes, and that male hormones may have some effect on the occurrence of hydronephrosis.

Odor response characteristics of thalamic mediodorsal nucleus neurons in the rabbit.

Extracellular responses were recorded from neurons in the thalamic mediodorsal nucleus (MD) of the lightly anesthetized rabbit. Eighty-seven neurons responded to electrical stimulation of the lateral olfactory tract (LOT shocks). They were located in the medial portion of the MD. In the same portion, negative field potentials with a short latency were evoked by the electrical stimulation of the olfactory projection area in the neocortex (OPA shocks). Fifty-nine MD neurons responded both to LOT and to OPA shocks. Among them, 17 thalamocortical relay neurons (which responded antidromically to OPA shocks) were found to respond transsynaptically to LOT shocks. Of the 87 LOT-responsive MD neurons, 48 responded to the odors applied. Eight odor-sensitive neurons were found to be the thalamocortical relay neurons. Thus, it was proven for the first time that a portion of the olfactory input to the OPA is mediated via relay neurons in the MD. Characteristics of response of MD neurons to odor stimulation were compared with those of OPA neurons. MD neurons did not show a selectivity of response to odors of urine, feces, or dry food pellets, to which OPA neurons responded exclusively. These results were discussed in relation to the functional role of the MD-OPA projection system in the discrimination of specific odors.

Changes in cytochrome oxidase activity of the rabbit olfactory cortex following unilateral olfactory bulbectomy.

The effects of unilateral olfactory bulbectomy on cytochrome oxidase (CO) activity in the olfactory cortex were studied in adult rabbits. In intact rabbits, layer Ia of the olfactory cortex stained symmetrically for CO activity as a continuous brown band. In animals with unilateral lesion of the olfactory bulb (OB), a great decrease in the level of CO activity was found in layer Ia ipsilaterally. The change became prominent about 12 hr after OB lesion. Thereafter no further decrease in CO activity was observed up to 6 months. A decrease in the level of CO activity was observed in only the denervated lamina to which the OB neurons project directly, but not along the synaptic chain in the olfactory cortex.
