Heterogeneity in the expression of HLA and tumor-associated antigens by surgically removed and cultured breast carcinoma cells.

Surgically removed normal and malignant mammary tissues and human breast carcinoma cell lines were tested in binding assays with monoclonal antibodies to HLA-A,B,C antigens, beta 2-microglobulin, HLA-DR antigens, and tumor-associated antigens; the latter included a Mr 280,000, a Mr 94,000, and a Mr 85,000 membrane-bound glycoprotein and a cytoplasmic antigen. HLA-A,B antigens, beta 2-microglobulin, HLA-DR antigens, and the cytoplasmic antigen are expressed by normal mammary cells. Their malignant transformation may be associated with quantitative changes in the expression of these antigens and with the appearance of Mr 94,000 and Mr 85,000 glycoproteins. The Mr 280,000 glycoprotein was detected on only one of the breast carcinoma cell lines tested. Analysis of primary tumors and autologous axillary lymph node metastasis from 13 patients has shown differences in the expression of all the antigens tested between primary and metastatic lesions.

[Experimental and clinical studies on intravesical instillation therapy for bladder cancer].

Intravesical instillation therapy was performed in 155 cases of urinary bladder cancer. In 60% of the patients treated by therapeutic intravesical instillation, antitumor effects were observed. The patients who were given only the instillation therapy frequently had recurrence of the tumor in the same site within one year. Therefore, in such cases prophylactic instillation seems to be necessary. The preventive effect of intermittent instillation therapy given over a long period was superior to that of concentrated instillation therapy given during a short period. The rate of recurring bladder tumors decreased after instillation therapy. Eight cases that were cystoscopically nonvisual tumors but indicated positive cytology were treated by intravesical instillation therapy. In 5 of these cases cytology was negative after treatment.

[10 cases of congenital cystic dilatation of the common bile duct].

Ten cases of congenital cystic dilatation of the common bile duct (including insufficiency form) were examined for mode of pancreatico-biliary anastomosis and cholangiographic findings. The results were as follows. Male to female ratio was 1:9 in this series. Regarding the mode of pancreatico-biliary anastomosis, type I (pancreatic duct flowing into common bile duct) tended to occur frequently in patients 30 years old or more, while type II (common bile duct flowing into pancreatic duct) in patients under 30 years old. With respect to clinical symptoms, no cases had triad (jaundice, abdominal pain and palpation of mass), but abdominal pain occurred in all the cases. Jaundice and palpation of mass were seen in 30% and 10% of the cases, respectively. Regarding the age at the onset and the duration of the disease, type I occurred in elderly cases with a longer duration of disease, while type II occurred in young cases with a shorter duration of disease. The degree of distention increased in proportion to the severity of the peripheral stenosis of the biliary duct. Complication with biliary tract carcinoma was seen in 2 type I cases. Among the type I cases, those with mild distention were considered to be insufficient form.

[Monoclonal antibodies to digestive-tract cancer].

There have been few reports concerning monoclonal antibodies to human digestive-tract cancers and CEA. Recently, we established several monoclonal antibodies to either gastric cancer, colorectal cancer or CEA using a CEA-secreting gastric cancer cell line or a purified CEA preparation as immunogen. In this review, we mainly described serologic and molecular characterization of both CEA-related and CEA-unrelated antigens detected by the monoclonal antibodies originated in our laboratory. We also introduced some approaches to the clinical application of the monoclonal antibodies described in the literature.
