Suppression of human melanoma growth in nude mice injected with anti high-molecular-weight melanoma-associated antigen monoclonal antibody 225.28S conjugated to purothionin.

The cytotoxic agent purothionin purified from barley was covalently conjugated to the anti-human high-molecular-weight melanoma-associated antigen (HMW-MAA) monoclonal antibody (MoAb) 225.28S by utilizing water-soluble carbodiimide. Injection of the conjugate (0.1 mg/injection) significantly increased the life span of nude mice injected with ascitic-form human melanoma cells (Colo 38), and caused 40% inhibition on day 25 of the growth of solid-form human melanoma cells in nude mice. The effect is specific and is markedly influenced by the site of growth of tumors and by the schedule of administration of the conjugate.

[Evaluation of bacterial growth rate by the ATP-bioluminescence method (luciferase assay)--comparison with conventional methods and application to clinical studies].

Bacterial growth rate was evaluated by estimating the increase in ATP content of cultures of bacterial suspensions using luciferase assay, and the method was applied to the estimation of bactericidal activity of serum and anti-bacterial activity of antibiotics. The results obtained were as follows: The bacterial numbers or bacterial growth rate determined by the ATP-Bioluminescence method was in accordance with those by CFU assay and spectrometrical assay by optical density readings. Furthermore this method was confirmed to be non-time-consuming and more simple than other methods. Using this method, anti-bacterial activity of fresh serum was evaluated. The serum suppressed the growth rate of E. coli, Str. fecalis, CBS, Kleb. pneumoniae and Staph. aureus at least for 2 hours. Heat-inactivated serum and absorbed serum with bacteria tended to lose such suppressive effect. Thus the main effective factors seem to be complements and specific antibody. The anti-microbial activity of antibiotics was enhanced by addition of fresh serum to bacterial cultures, and the results were not in accordance with those by the conventional Disc method. Thus the sensitivity test under the influence of fresh serum is recommended in order to know the clinical effect of antibiotics. These results suggest the usefulness of ATP-Bioluminescence method in the clinical laboratory examinations or investigations.

[An analysis of 103 patients with renal injuries].

Between 1971 and 1983, 103 patients with renal injury were admitted to the Department of Urology, Tatebayashi Kosei Hospital. There were 87 males and 16 females. The most frequent age group seen (27.2% of patients) was the 10 to 19 year group. The various causes included traffic accidents (55 patients), falls (16 patients), athletic injuries (8 patients), labor accidents (7 patients), blows (6 patients) and others (11 patients). Associated injuries were present in 52.4% of the patients, including thoracic injuries such as rib fracture (20 patients), lumbar vertebra and transverse process fractures (18 patients), and head injuries (12 patients). Evaluation of the renal injuries was made by excretory urogram (IVP) and conservative management was for most, contusion and rupture, and operation for laceration as a principle: 95 patients (97 renal injuries) were treated by conservative management and 7 patients were treated surgically, 5 patients by nephrectomy and 2 patients by perirenal drainage. Nine patients who had associated injuries were treated surgically. In the follow-up study of 41 patients who had received conservative management and operation (perirenal drainage), followed for more than 6 months and had IVP, urinalysis and blood pressure measurement, representative changes were calyceal deformity (3 patients), renal calculus formation (2 patients) and non-functioning kidney (2 patients) in urograms.

Haemoglobin Warwickshire (beta 5 [A2] Pro----Arg). A possible 'fine tuning' of 2,3-DPG affinity by beta 5 Pro.

The blood of a 67-year-old Scotsman who was admitted to hospital with an abdominal carcinoma showed an abnormal red cell lysis in the oxidase channel of the Technicon H6000 analyser. This is seen regularly with Hb C and sometimes with Hb S and on further investigation a new haemoglobin variant--Hb Warwickshire--was found. When the family was examined, other examples were revealed. The haematological parameters were normal. Hb Warwickshire is mildly unstable and whilst in the absence of phosphates its oxygen dissociation does not differ from that of Hb A, a small fall of oxygen affinity is noted when phosphates are present.

[Studies of progestin specific binding protein in the human prostate [I]: Studies on basic conditions for the accurate quantitative assay method].

Specific binding of the synthetic progestin 17 alpha-methyl-[3H]-promegestone (R5020) in the cytosol of human benign prostatic hypertrophy was studied to determine the accurate quantitative assay method. No significant effect was observed between Tris and Phosphate buffer during a buffer composition investigation. The addition of either glycerol or mercaptoethanol was effective in the enhancement of R5020 specific binding. When sodium molibdate was added into the incubation buffer, the obvious increase of 7-8S components in the SDG analysis was observed. R5020 specific binding was suppressed by the addition of CaCl2. No enhancement effect was observed by the addition of EDTA (0.1-6mM). Under the high concentration of EDTA (10-50mM), it was suppressed. SDG assay by the vertical rotor was superior to that by the swing rotor. It seemed that the long incubation time was an important factor to obtain 7-8S, which was sufficiently bounded. This estimation, however, is not assertive. Further information on the incubation time frame will be presented in the next report. It was evident that the 7-8S saturable peak alone was less than the value calculated by the charcoal assay. It was concluded that not only 7-8S but also 4S binding was included in N value by the charcoal assay. The SDG assay is recommended for the clinical receptor study, since there is not enough information concerning the character of 4S and 7-8S saturable binding in the human prostate.

Sensitive detection of amino acids in human serum and dried blood disc of 3 mm diameter for diagnosis of inborn errors of metabolism.

Sensitive high-performance liquid chromatographic determination of amino and imino acids in human serum (5 microliters) and dried blood (2.6-2.8 microliters) on a paper disc (3 mm diameter) of normal and abnormal newborns with inborn errors of metabolism (phenylketonuria, maple syrup urine disease and tyrosinosis) is described. Amino and imino acids in the biological specimens were extracted with ethanol and derivatized with 4-fluoro-7-nitrobenzo-2-oxa-1,3-diazole at 60 degrees C and at pH 8.0 for 1 min. The fluorescent derivatives were separated on mu Bondapak C18 and detected fluorometrically (530 nm/470 nm). The method was about one order of magnitude more sensitive than the similar method using o-phthalaldehyde. The amino acid contents obtained by the proposed method were comparable to those obtained by the amino acid analyser with use of o-phthalaldehyde.

A randomized trial of chemoimmunotherapy of acute nonlymphocytic leukemia in adults using a protein-bound polysaccharide preparation.

The effect of immunotherapy with a protein-bound polysaccharide preparation termed PSK on remission duration and survival of adults with acute nonlymphocytic leukemia (ANLL) was studied in a prospective randomized cooperative trial. After having achieved complete remission and receiving a consolidation therapy, 73 patients were randomized either to maintenance chemotherapy or to maintenance chemotherapy plus immunotherapy with PSK. Ultimately 36 patients in the chemotherapy group and 31 in the chemoimmunotherapy group were evaluable. Six months after the last entry, immunotherapy with PSK showed a borderline beneficial effect on remission duration (P = 0.089) and on duration of survival (P = 0.062). When the data were analyzed 12, 18, and 24 months after the last entry there were no significant differences in duration of remission and survival between the two groups. However, analysis of the data of patients who had maintained complete remission for more than 270 days revealed that immunotherapy had a suggestive beneficial effect (P = 0.105), prolonging the 50% remission period by 418 days (885 vs 467 days). Thus, immunotherapy with PSK seems to be active in the treatment of adult ANLL when used for maintenance therapy in combination with chemotherapy, especially in patients with a good prognosis.

Eimeria tenella, E. necatrix, E. acervulina, and E. maxima: anticoccidial activity of 1,6-dihydro-6-oxo-2-pyrazinecarboxylic acid 4-oxide.

The anticoccidial activity of an orotic acid analog, 1,6-dihydro-6-oxo-2-pyrazinecarboxylic acid 4-oxide (carboxyemimycin), was tested in battery experiments, utilizing 9-day-old Single-Comb White Leghorn cockerels. Carboxyemimycin, at 125 ppm and more in feed, exhibited marked anticoccidial activities against Eimeria tenella, E. necatrix, E. acervulina, and E. maxima. High doses of carboxyemimycin--up to 1000 ppm--did not cause any reduction in weight gains. The battery and in vitro studies with delayed and restricted medications revealed that carboxyemimycin affected the development of E. tenella in first and second generation schizogony and in gametogony.

Effect of catecholamines on the activity of rat brain dipeptidyl aminopeptidase.

The inhibitory effect of catecholamines, their metabolic intermediates and derivatives on the activity of rat brain dipeptidyl aminopeptidase was studied. The enzyme was extracted from a crude mitochondrial fraction containing lysosomes (and also synaptosomes) and purified by three-step column chromatography. The activity of the purified enzyme was inhibited by a low concentration (IC50 2 approximately 5 mM) of dopamine, norepinephrine and epinephrine. Dihydroxyphenylalanine or dihydroxyphenylacetic acid, each having a carboxyl group, showed only a weak inhibitory effect (IC50 greater than 10 mM). These results suggest the possibility that brain dipeptidyl aminopeptidase activity may be regulated by the changing of the concentration of catecholamines in the central nervous system.

Determination of total captopril in dog plasma by HPLC after prelabelling with ammonium 7-fluorobenzo-2-oxa-1,3-diazole-4-sulphonate (SBD-F).

A HPLC method has been developed for the determination of captopril, an angiotensin-converting enzyme (ACE) inhibitor, in dog plasma. The plasma was deproteinized with trichloroacetic acid (TCA) containing N-(3-mercapto-1-oxopropyl)-piperidine-2-carboxylic acid SQ 25,233 as an internal standard, adjusted to pH 9.5 with borate buffer containing tri-n-butyl phosphine (TBP) to reduce the oxidized captopril, and treated with a thiol specific reagent, SBD-F. The fluorescent derivatives of captopril and the internal standard were separated from other interfering peaks on a reversed-phase column and determined by HPLC. The proposed method was simple, selective and sensitive. The total plasma concentration of captopril reached a maximum of 3.04 +/- 0.54 microg/ml (mean +/- S.E.) 60 min after administration of the drug.

Zinc concentration and progestin receptor in human benign hyperplastic prostate.

The effect of cationic zinc on the specific binding of the synthetic progestin, promegestone (17 alpha-21-dimethyl-19-nor-pregna-4,9-diene-3,20-dione, R5020), has been investigated in cytosols from human benign hyperplastic prostate. Zinc was analyzed using flameless atomic absorption spectrometry. Progestin specific binding was assayed using sucrose density gradient centrifugation in a vertical tube rotor. The concentration of endogenous zinc in benign hyperplastic prostate cytosols was 0.53 mM +/- 0.26 (range 0.15 mM to 0.88 mM). No significant relationship was observed between zinc concentration in cytosols and the amount of progestin specific binding in the 7-8S peak in sucrose density gradients. Addition of excess zinc to cytosols (1.6 mM) in the presence or absence of monothioglycerol had no effect on the total amount of 7-8S progestin specific binding observed. Removal of free zinc by gel filtration of cytosols on Sephadex G-25 did not change the amount of 7-8S cytosolic receptor binding observed. The addition of the chelating agents, 2,2',2''-tripyridine, 9,10-phenanthroline or EGTA, after the gel filtration step had no apparent effect on the 7-8S progestin specific binding. The anionic chelator, EDTA, inhibited the 7-8S specific binding in a dose dependent manner. However, this inhibition was not reversed by the addition of excess zinc (6 mM) and was observed at very high concentrations of EDTA. This suggests that the effect of EDTA was due to its anionic effect rather than to its effect as a chelator. We conclude that cationic zinc in concentrations up to 2 mM does not change the amount of 7-8S progestin specific receptor binding in cytosols from BPH tissue.

Role of Ferrous Ions in Synthetic Cobaltous Sulfide Leaching of Thiobacillus ferrooxidans.

Microbiological leaching of synthetic cobaltous sulfide (CoS) was investigated with a pure strain of Thiobacillus ferroxidans. The strain could not grow on CoS-salts medium in the absence of ferrous ions (Fe). However, in CoS-salts medium supplemented with 18 mM Fe, the strain utilized both Fe and the sulfur moiety in CoS for growth, resulting in an enhanced solubilization of Co. Cell growth on sulfur-salts medium was strongly inhibited by Co, and this inhibition was completely protected by Fe. Cobalt-resistant cells, obtained by subculturing the strain in medium supplemented with both Fe and Co, brought a marked decrease in the amount of Fe absolutely required for cell growth on CoS-salts medium. As one mechanism of protection by Fe, it is proposed that the strain utilizes one part of Fe externally added to CoS-salts medium to synthesize the cobalt-resistant system. Since a similar protective effect by Fe was also observed for cell inhibition by stannous, nickel, zinc, silver, and mercuric ions, a new role of Fe in bacterial leaching in T. ferrooxidans is proposed.
