Expression of cytoskeletal-associated protein tyrosine phosphatase PTPH1 mRNA in human hepatocellular carcinoma.

We investigated the mRNA expression of cytosolic protein tyrosine phosphatase (PTPH1), which has a homologous domain to cytoskeletal-associated proteins, in human hepatocellular carcinomas (HCCs) by using reverse transcriptase-polymerase chain reaction (RT-PCR). PTPH1 mRNA was detected in all HCC cell lines (n = 6), and HCC and adjacent noncancerous tissues (n = 8) examined, indicating that PTPH1 was expressed in HCCs and hepatocytes. There was no remarkable difference in the level expression of PTPH1 mRNA between HCC and adjacent noncancerous tissues. We also performed RT-PCR single-strand conformation polymorphism (SSCP) analysis in HCC cell lines and tissues in the C-terminal region of the catalytic domain of PTPH1. In the cHc4 cell line and a HCC tissue specimen, a shifted band was detected, although it was not found in the non-cancerous tissue of the HCC specimen. Nucleotide sequence analysis showed a common mutation from T to C at the third letter of codon 919 which did not lead to amino acid substitution. These results suggest that another mutation leading to the development of HCC could occur in some region of PTPH1 other than that investigated in this study.

Shape of the haemoglobin-oxygen equilibrium curve and oxygen transport efficiency.

The sigmoid shape of the oxygen equilibrium curve (OEC) of haemoglobin permits large changes in oxygen saturation (S) upon narrow variations of the partial pressure of oxygen (P). The slope of the Hill plot (n) calculated from the Adair constants for human adult haemoglobin is usually maximised at S values greater than 0.5, implying that co-operative interactions of haemoglobin subunits are maximised above half-saturation. Our analysis of OECs based on the Adair equation shows that the slope of the OEC, S', which is proportional to the capacitance coefficient beta (Piiper et al., Respir. Physiol. 13: 292-304, 1971) and is a direct measure for oxygen-transport efficiency of haemoglobin, is maximised at S values smaller than 0.5, usually at S = 0.38. The analysis also gives the relations: Pdmax < P50 < Pnmax and Sdmax < 0.5 < Snmax where Pdmax, P50 and Pnmax are P at which S' is maximised, P at half-saturation and P at which n is maximised, respectively, and Sdmax and Snmax are S at Pdmax and S at Pnmax, respectively. Thus the most efficient point of OEC does not coincide with the point of maximal cooperativity nor with the oxygen saturation level of mixed venous blood in resting conditions (S approximately = 0.75). The steep portion of the OEC around S = 0.38 is exploited for large oxygen demands under conditions of exercise. The most efficient oxygen unloading region around S = 0.38 could be revealed by the Adair analysis but not by analysis using the conventional Hill equation.

Recipient-mediated effect of a traditional Chinese herbal medicine, ren-shen-yang-rong-tang (Japanese name: ninjin-youei-to), on hematopoietic recovery following lethal irradiation and syngeneic bone marrow transplantation.

Ren-shen-yang-rong-tang (Japanese name: Ninjin-youei-to, NYT), a traditional Chinese herbal medicine, was evaluated for recipient-mediated effect on hematopoietic recovery in a murine model of syngeneic bone marrow transplantation (BMT). BALB/c recipient mice were preconditioned with a lethal total body irradiation (TBI) at a dose of 6.5 Gy and transplanted with syngeneic bone marrow (BM) cells. NYT treatments, given intraperitoneally (i.p.) once per day for 3 consecutive days in a dose of 0.625 mg, were performed either before or after TBI and BMT to assess any recipient-mediated effect of this compound. NYT pretreatment was as effective as NYT posttreatment in enhancing the total number of colony-forming unit erythroid (CFU-E) and colony-forming unit granulocyte-macrophage (CFU-GM) per marrow and spleen after TBI and BMT. NYT pretreatment caused a significant increase in marrow and splenic CFU-E and CFU-GM numbers over a prolonged period following TBI and BMT, and affected late-stage erythropoiesis (CFU-E) more profoundly than early-stage erythropoiesis (burst-forming unit erythroid, BFU-E). NYT pretreatment significantly accelerate recovery of not only erythrocyte and leukocyte counts but also platelet counts after transplantation with a limited number (1 x 10(5)) of BM cells. The same treatment, however, was significantly less effective in hematopoietic recovery after transplantation with a minimal number (1 x 10(4)) of BM cells, indicating that NYT accelerates recovery of donor-derived rather than recipient-derived cells.(ABSTRACT TRUNCATED AT 250 WORDS)

The core and complementary sequence responsible for biological activity of the diapause hormone of the silkworm, Bombyx mori.

To evaluate the structure-function relationship of the diapause hormone of the silkworm, Bombyx mori, the entire molecule and selected fragment and deleted analogues were chemically synthesized to compare their biological activity. The C-terminal pentapeptide amide was the shortest fragment that elicited 11% diapause eggs at maximum, indicating that this sequence is the core-active structure required for a biological response. The full biological response of about 70% diapause eggs was expressed by the C-terminal hexapeptide amide. However, an ED50 value of this peptide amide was 1000-fold higher than that of the parent molecule. The serial elongation of peptide chain lengths toward the N-terminus brought about the sudden decrease in ED50 values at two positions between Arg9-Gly10 and Thr1-Asp2. The deletion of duplicated sequence(s) located in the middle part of the molecule or the truncation of N-terminal region of the parent molecule increased ED50 values but had no effects on response. Thus, N-terminal region and duplicated sequences act as the complementary structures for full potency of diapause hormone.

Silkworm diapause hormone, structure-activity relationships indispensable role of C-terminal amide.

To determine the structure-activity relationships of the silkworm diapause hormone, a series of peptide analogs having different chain lengths starting from the parent C-terminus and analogs having identical sequences with free acid C-termini were chemically synthesized by solid-phase Fmoc methodology and were further purified by HPLC. Bioassay showed that the analogs with free acid C-termini were non active. The retained activities of those shorter chains were shown only with the amidated C-terminal analogs among which the potency depended on the length of the chain. The active peptides required two minimal elements; namely the sequence near and the amidation of the C-terminus. There was no difference in enzymatic digestion of the C-terminally amidated or free acid analogs in pupal haemolymph. Hence the absence of DH activity of the free acid analogs was not because of being selectively hydrolyzed faster than the C-terminally amidated peptides. This suggested that existence of a certain higher order structure could be involved in expressing hormonal activity, or that the negative charge of the free acid terminus may be deleterious to a proper ligand receptor interaction. Since most of the hydrophobic amino acids were located near the C-terminal portion, both the hydrophobicity of the portion near and the amidation of the C-terminus were indispensable structures for diapause hormone activity.

Expression and localization of aminopeptidase N, neutral endopeptidase, and dipeptidyl peptidase IV in the human placenta and fetal membranes.

OBJECTIVE: Our purpose was to determine the distribution of membrane-bound cell surface peptidases, namely aminopeptidase N, neutral endopeptidase, and dipeptidyl peptidase IV in the human placenta and fetal membranes. STUDY DESIGN: Frozen tissue sections of the first-trimester chorionic villi, term placentas, and term fetal membranes were stained by indirect immunofluorescence with specific monoclonal antibodies. RESULTS: In the first trimester chorionic villi cytotrophoblasts expressed both neutral endopeptidase and dipeptidyl peptidase IV, but syncytiotrophoblasts expressed only neutral endopeptidase. Stromal cells in the chorionic villi expressed the three peptidases at various intensities. In the term placentas villous syncytiotrophoblasts expressed neutral endopeptidase weakly, and the villous stromal cells expressed large amounts of both aminopeptidase N and dipeptidyl peptidase IV but neutral endopeptidase weakly or faintly. In the term fetal membranes amniotic epithelial cells and chorion laeve expressed both neutral endopeptidase and dipeptidyl peptidase IV. Decidual cells in the decidua parietalis moderately or highly expressed aminopeptidase N. CONCLUSION: Three peptidases, aminopeptidase N, neutral endopeptidase, and dipeptidyl peptidase IV, are expressed by different cell populations in the human placenta and fetal membranes, suggesting their respective and important roles at the maternofetal interface.

In vivo effects of transurethral balloon laser prostatectomy on the canine prostate.

Twenty-three dogs received prostatectomy by transurethral balloon laserthermia, and the in vivo effects and morbidity were evaluated. The prostate was heated transurethrally, and tissue temperature at a depth of 5 mm. from the urethral mucosa was maintained at 60C for 20 minutes. Immediately after the treatment, coagulative necrosis was observed around the urethra to an average depth of 5 mm., and the tissue started to slough off within 1 week. Cavity formation and reepithelialization were complete after 4 weeks. Histologically, no tissue damage was found in the bladder neck or the urethral sphincter. The cavity formation threshold was 46.1C for 20 minutes. There was no incontinence or macroscopic hematuria in any case. No abnormality was found in hematological or biological examinations after treatment. From these data, the clinical use of transurethral balloon laserthermia for prostatectomy is considered both safe and effective.

Effect of sugars-addition on the survival of vitrified bovine blastocysts produced in vitro.

We investigated the effect of addition of sugars to a vitrification solution on the survival rate of bovine blastocysts produced in vitro. In vitro-matured (IVM) and in vitro-fertilized (IVF) bovine Day-6 to Day-8 bovine blastocysts were classified into 3 developmental stages: early blastocysts, blastocysts and expanded blastocysts. The blastocysts were cryopreserved in 1 of 3 vitrification solutions: 1) 25% glycerol25% ethylene glycol (GE); 2) 20% glycerol20% ethylene glycol3/4 M sucrose (GES); and 3) 20% glycerol20% ethylene glycol3/8 M sucrose3/8 M dextrose (GESD). The basic solution was Dulbecco's PBS supplemented with 20% of fetal calf serum. Embryos were exposed to each vitrification solution in 3 steps, and after loading into 0.25-ml straws, were plunged into liquid nitrogen. After warming in water bath at 20 degrees C, cryoprotectants were diluted in 1/2 M and 1/4 M sucrose each for 5 min. Equilibration and dilution procedure except warming were conducted at room temperature (23 to 27 degrees C). After dilution, the embryos were cultured in Ham's F10 medium0.1 mM beta-mercaptoethanol20% fetal calf serum. Survival rates of embryos at 48 h of incubation of each of the 3 developmental stages (early blastocysts, blastocysts and expanded blastocysts) exposed to the 3 types of the vitrification solutions (GE, GES and GESD) were 23.5, 33.3, 65.8% (early blastocysts, blastocysts and expanded blastocysts respectively) in GE, 55.6, 71.9, 90.5% in GES and 84.6, 83.3, 95.8% in GESD respectively. These results indicate that a mixture of 25% glycerol25% ethylene glycol is not suitable for vitrification of early bovine blastocysts; however, addition of sugars to the solution significantly (P<0.01) improved the survival rate of the vitrified blastocysts, independently of their stage of development.

Effects of interleukin-6 on hematopoiesis in allogeneic and syngeneic bone marrow chimeras.

Effects of interleukin-6 (IL-6) on hematopoietic progenitor cells were analyzed in murine bone marrow chimeras. When IL-6 was injected into syngeneic [C3H/He-->C3H/He] bone marrow chimeras from day 1 to day 12, the numbers of highly proliferative potential colony-forming units (CFU-HPP) or colony-forming units mix (CFU-Mix) in spleen cells and bone marrow cells increased on day 14 although there was a marked increase in spleen cells but not in bone marrow cells on day 21. The numbers of CFU-HPP increased in spleen cells from allogeneic [BALB/c-->C3H/He] bone marrow chimeras injected with IL-6 on days 14 and 21. In syngeneic bone marrow chimeras, the numbers of colony-forming units granulocyte/macrophage (CFU-GM) and burst colony-forming units (BFU-E) increased similarly to those of CFU-HPP and CFU-Mix on day 14. On day 21, these were mainly increased in spleen cells. In allogeneic bone marrow chimeras, IL-6 decreased the numbers of CFU-GM and BFU-Mix dose-dependently on day 14. Only 10 micrograms of IL-6 increased the numbers of CFU-GM and BFU-E on day 21. In our previous work, we showed that platelet counts increased on day 14 in syngeneic bone marrow chimeras injected with IL-6, whereas platelet and leukocyte counts increased on days 14 and 24 in allogeneic bone marrow chimeras injected with IL-6, correlating inversely with the numbers of hematopoietic progenitor cells. Overall, primitive hematopoietic progenitors (i.e., CFU-HPP and CFU-Mix) existed primarily in spleen cells of allogeneic bone marrow chimeras on day 14, whereas those in spleen cells of syngeneic bone marrow chimeras were found on day 21. These findings indicate that the effect of IL-6 on hematopoiesis in allogeneic bone marrow chimeras is completely different from that in syngeneic bone marrow chimeras, probably via graft-versus-host reaction (GVHR) but not GVH disease (GVHD).

Effects of 15-deoxyspergualin on proliferative responses and cytokine gene expression in vitro.

15-deoxyspergualin (DSG)-treated BALB/c spleen cells showed increased spontaneous proliferation and increased alloreactive mixed lymphocyte reactions (MLRs) when a 3-h treatment was carried out. However, when spleen cells were treated with DSG for 5 days without washing out DSG, decreased spontaneous proliferation was observed, although alloreactive MLRs against C3H/He and C57BL/6 alloantigens were increased. In contrast, cyclosporin A (CsA) induced markedly decreased alloreactive MLRs. Decreased concanavalin A (Con A)- and pokeweed mitogen (PWM)-induced responses were observed in spleen cells treated with DSG for 3 h; whereas increased phytohemagglutinin (PHA)-induced responses were observed. On the other hand, increased Con A- and PHA-induced responses were observed in spleen cells treated with DSG for 2 days, whereas PWM-induced responses were decreased. CsA-treatment induced markedly decreased mitogen-induced responses. These results suggest that the immunosuppressive mechanism of DSG differs from that of CsA. Reverse transcriptase-polymerase chain reaction (RT-PCR) method showed that interleukin 1 beta (IL-1 beta), IL-2, IL-3, IL-4, IL-5, and leukemia inhibitory factor (LIF) mRNA expression in DSG-treated spleen cells were increased by Con A stimulation, thus indicating that DSG modulates cytokine gene expression and inducing immunosuppressive mechanisms different from CsA.

In vivo administration of interleukin-6 in murine allogeneic bone marrow chimeras: early and delayed enhancement of hematopoiesis accompanied with split tolerance but not with graft-versus-host disease.

Interleukin-6 (IL-6) induced increased, leukocyte and platelet counts on around day 20 when it was administered into [BALB/c-->C3H/He] bone marrow chimeras from day 1 to day 12. Increased leukocyte counts and hemoglobin (Hb) levels were also observed at around day 60 and from day 41 to 80, respectively. On the other hand, hematopoietic recovery in [C3H/He-->C3H/He] bone marrow chimeras injected with IL-6 was different from that in [BALB/c-->C3H/He] bone marrow chimeras, showing no delayed and long-lasting increase in Hb levels but showing an early and transient increase in Hb levels and platelet counts. Sera from [BALB/c-->C3H/He] bone marrow chimeras injected with IL-6 showed predominant productions of IL-3 and/or IL-4. Reverse transcriptase polymerase chain reaction (RT-PCR) showed that stem cell factor (SCF) mRNA expression was increased in bone marrow or spleen cells from [BALB/c-->C3H/He] bone marrow chimeras injected with IL-6 on day 36. Furthermore, we analyzed influence of IL-6 on graft-versus-host disease (GVHD) in [BALB/c-->C3H/He] bone marrow chimeras injected with IL-6. Decreased survival days and body weights were not observed when compared with the control. Histopathological changes of the liver due to GVHD were also not obvious. However, alloreactive mixed lymphocyte reactions (MLRs) were readily detected although cytotoxic T cells were not generated. Since H-2 typing showed that donor-type chimerism was predominantly observed, it was suggested that split tolerance might be induced by IL-6 administration. Increased IL-2 levels were not detected in sera from [BALB/c-->C3H/He] bone marrow chimeras injected with IL-6 whereas IL-4 was detected in the same sera, indicating that type 2 helper T (TH2) cells appeared to be predominantly generated. These results suggest that IL-3/IL-4 and SCF appeared to synergistically support delayed effects on hematopoiesis in [BALB/c-->C3H/He] bone marrow chimeras injected with IL-6 although early effects appeared to be mediated mainly by IL-6 directly or indirectly. Furthermore, IL-6 could induce split tolerance in [BALB/c-->C3H/He] bone marrow chimeras via a preferable activation of TH2 type cells without inducing severe GVHD.

Morbidity rates of complications in persons with spinal cord injury according to the site of injury and with special reference to hypertension.

It has been reported that since 1982 the incidence in persons with spinal cord injury (SCI) of hypertension is commoner than it is in the general population of the same age groups in Japan. In the current study, we examined outpatient morbidity rates and standardised outpatient morbidity ratios (SOMR) according to the site of injury, as well as blood pressure levels and history of disease, and compared the incidences with those for the general population. The subjects consisted of 195 men with SCI. All were engaged in light work at special centres while living with other persons at the centres. The mean age was 49.5 years old, and the average post-injury period was 17.9 years. With respect to the site of injury, 19 patients had had injuries at the level of C-T5, 24 at T6-T10, 139 at T11-L1, and 13 at L2 or lower. The SOMR (general population = 100) for hypertension was closely related to the site of injury, i.e. 0 at C-T5, 250 at T6-T10, 221 at T11-L1 and 308 at L2 or below. Among the patients treated with antihypertensive agents (41 persons), 17.1% were under treatment for renal diseases, 4.9% for diabetes, and 4.9% for hepatic disorders. In 68% of the SCI persons examined, however no disease (such as renal disease, diabetes mellitus, hepatic disease or endocrine abnormality) could be regarded as a cause of secondary hypertension. In addition, the survey revealed that the body weight of SCI persons was lower than that of the general populations.

Mandibular reconstruction using computer-generated three-dimensional solid models.

In mandibular reconstruction, it is necessary to know the exact, three-dimensional extent of the mandible and its defect; the bone graft must be the exact size and dimension of the defect, to assure a precise three-dimensional configuration of the mandible. Previously, the bone graft had to be reshaped during the operation by trial and error, often a time-consuming procedure. The operative procedure has been simulated in advance using three-dimensional, solid models, which has shortened the operating time required.

Tumour necrosis factor alpha inhibits in-vitro decidualization of human endometrial stromal cells.

Interleukin-1 (IL-1) has been reported previously to inhibit the in-vitro decidualization of human endometrial stromal cells as assessed by progesterone-induced prolactin production and morphological transformation. In this study we examined whether other cytokines, such as tumour necrosis factor-alpha (TNF alpha), interferon-beta (IFN beta), IFN gamma or granulocyte-macrophage colony-stimulating factor (GM-CSF), could affect the decidualization of human endometrial stromal cells in vitro. Of these cytokines, TNF alpha significantly suppressed prolactin production in a dose-dependent manner, with no apparent effect on cell number. The morphological transformation of endometrial stromal cells was also inhibited by TNF alpha. TNF alpha and IL-1 significantly suppressed cAMP-stimulated prolactin production by endometrial stromal cells. Neither the progesterone concentration in the supernatant of the endometrial stromal cell culture system nor intracellular calcium concentration of the endometrial stromal cells were affected by the addition of TNF alpha or IL-1. These results indicated that TNF alpha and IL-1 suppress both progesterone-induced and cAMP-mediated prolactin production in endometrial stromal cells, and that this inhibition was not attributable to direct effects on progesterone metabolism or related to Ca(2+)-mediated signal transduction. These experiments suggested that a local increase of TNF alpha and IL-1 under certain pathological conditions in vivo may disturb blastocyst implantation and/or the maintenance of pregnancy by inhibiting the decidualization of endometrial stromal cells.

Establishment and characterization of mouse-human chimeric monoclonal antibody to erbB-2 product.

A mouse-human chimeric antibody for erbB-2 product was established by a new procedure using heavy chain loss mouse mutant hybridoma and human immunoglobulin expression vector. The E401 hybridoma secreted anti-erbB-2 product monoclonal antibody (MoAb) (IgG1, kappa). The gene of the mouse variable regions of heavy chain was amplified and cloned by the polymerase chain reaction technique directly from the E401 hybridoma RNA. The variable region of heavy chain was joined with the expression vector, which contains human gamma 1 constant gene. The expression vector was transfected into heavy chain loss mutant cells E401-12, which produced only murine immunoglobulin light chains. A chimeric monoclonal antibody CH401 retained full binding reactivity to erbB-2 product, compared with murine E401 parental antibody. Furthermore, the chimeric MoAb CH401 was much more efficient in supporting antibody dependent cell-mediated cytotoxicity activity against erbB-2-bearing human adenocarcinoma cells than murine MoAb E401. These suggest that a chimeric monoclonal antibody CH401 may be a potent reagent for therapy of human adenocarcinomas.

Expression of sulfated carbohydrate chains detected by monoclonal antibody 91.9H in human gastric cancer tissues.

Expression of sulfated carbohydrate chains in intestinal metaplasia (IM) and gastric cancer tissues was immunohistochemically evaluated by using a monoclonal antibody (MAb) 91.9H. While normal gastric tissues were negative for MAb 91.9H, IM and cancer tissues were positively stained with high frequency. The incidence was 67% for IM with chronic gastritis (n = 12), 95% for IM with gastric cancer (n = 21), 77% for well and moderately differentiated adenocarcinoma (n = 13), 48% for poorly differentiated adenocarcinoma (n = 23), 89% for signet-ring cell carcinoma (n = 9) and 100% for mucinous adenocarcinoma (n = 7). When poorly differentiated adenocarcinoma cases were divided into two groups, solid (n = 13) and non-solid types (n = 10), the incidences were 8% and 100%, respectively. These data suggest that MAb 91.9H could be of practical use as a new marker for IM and gastric cancer, and may be valuable for subgrouping poorly differentiated adenocarcinomas. Analyses of the core proteins for 91.9H epitope were then carried out. Comparison of immunostaining of ten poorly differentiated adenocarcinoma cases by MAb MUSE11 against MUC1 gene product with that by MAb 91.9H suggested that 91.9H epitope is not expressed on MUC1. Northern blot analysis of 10 pairs of gastric cancer and adjacent normal tissues with a MUC2 cDNA probe showed that the expression level of MUC2 mRNA was below the limit of detection. Thus, 91.9H epitope may be expressed on other proteins than MUC1 or MUC2 core proteins in gastric cancer.
