Detection of incompletely sialylated human chorionic gonadotropin by peanut agglutinin in choriocarcinoma.

Human chorionic gonadotropin (hCG) in normal pregnancy and hydatidiform mole did not bind to the peanut agglutinin (PNA)-Sepharose column, whereas significant amount of hCG in choriocarcinoma adsorbed to the column. Therefore, PNA-Sepharose affinity chromatography is an effective tool for the detection of desialylated hCG and may be useful also for the diagnosis of choriocarcinoma.

Induction of unscheduled DNA synthesis in cultured human oral keratinocytes by sodium fluoride.

The effect of treatment of cultured human oral keratinocytes with sodium fluoride (NaF) has been investigated with respect to induction of unscheduled DNA synthesis (UDS). Oral keratinocytes were isolated from excised buccal mucosa of normal individuals by trypsinization at 4 degrees C overnight, followed by separation of the epithelium of mucosa from lamina propria mucosae with forceps. Isolated cells were cultured in vitro and all experiments were performed with secondary cultures. For detection of UDS, the keratinocytes were cultivated with medium containing 1% fetal calf serum (FCS) for 2 days and then treated with 100-300 micrograms/ml NaF for 4 h in medium containing 1% FCS and 10 mM hydroxyurea (1% FCS-HU medium). Following treatment with NaF, UDS was measured by direct scintillation counting of [3H]thymidine incorporated into DNA of the cells in 1% FCS-HU medium. Significant levels of UDS were induced in a dose-related fashion by NaF treatment. The results suggest that NaF causes DNA damage in cultured human oral keratinocytes.

Diurnal changes in glycogen stores in liver and skeletal muscle of rats in relation to the feed timing of sucrose.

Diurnal changes in tissue glycogen stores were determined in rats in relation to the feed timing of sucrose. Rats were daily meal-fed on a 35% sucrose diet at 20.00-21.00 and a basal diet at 08.00-09.00, or meal-fed on the same diets at the reversed time for 7 weeks. Half of the animals were allowed voluntary wheel-running between 21.00-08.00, but the remaining animals were restricted to exercise for 24 h. At the end of the feeding period, both groups of rats were killed at 4-h intervals. Glycogen stores in liver and soleus muscle of sedentary rats showed sharp increases over 8 h after the sucrose meal regardless of its feed timing. The increases continued for only 4 h in exercised rats given the sucrose diet at the evening meal time. In contrast, glycogen stores in these tissues did not show any noticeable increase after the basal diet regardless of its feed timing. Thus, the feeding of sucrose at the evening meal time seems to be more preferable than at the morning meal time in order to have higher glycogen stores in liver and skeletal muscle during the physically active phase of the day in rats. Diurnal changes of glycogen contents in heart and adipose tissue differed from those in liver and skeletal muscle.

[Results of gonadotropin therapy in hypogonadotropic anovulation].

Combined FSH and HCG treatment was applied to patients in whom anovulation had been found to be the cause of sterility. Results were separately evaluated for patients in normal gonadotrophic conditions, on the one hand, and those in hypogonadotrophic conditions, on the other. They were compared with findings obtained from other groups between which no distinction had been possible due to the use of earlier methods for gonadotrophin determination. Lower RIA values for FSH or LH seemed to be more promising in terms of therapeutic success, with gravidity having been recorded from 44 per cent of hypogonadotrophic patients. The conclusion was drawn that prolactin-, FSH- or LH-RIA methods should be practicable for optimum treatment on a routine basis of patients with sterility due to anovulation. The data reported in this paper were derived from 54 patients in 117 therapeutic series.

In situ and ex situ renal artery aneurysmectomy in the solitary kidneys.

We report 2 cases of renal artery aneurysms in congenital solitary kidneys, which were treated respectively by in situ and ex situ aneurysmectomy. Hyperosmolar intracellular electrolyte solution was valid in preserving renal function during renal ischemia in both cases. The occurrence of an aneurysm in a solitary kidney, ex situ operation and renal preservation during ischemia are discussed.
