[Anti-tumor activity of peripheral lymphocytes of tumor-bearing rats by in vitro culture].

This basic study was performed to determine whether the anti-tumor effect of lymphocytes obtained from the peripheral blood of a tumor bearing host can be increased when cultured in vitro with Mitomycin C (MMC)-treated AH109A tumor cells, using interleukin-2 (IL-2). Donryu rats were used as tumor bearing hosts. The following results were obtained. Weak anti-tumor activity of lymphocytes was noted 7 days after lymphocytes were cultured in the medium to which only IL-2 was added. Anti-tumor activity was augmented by adding antigen (MMC treated tumor cells) to the above (1). Anti-tumor activity was further augmented by adding to (2) above antigen presenting cells such as intraperitoneal exudate macrophages or peripheral hole leukocytes. Anti-tumor activity of lymphocytes was detected when IL-2 and MMC treated antigen were added to the peripheral hole leukocytes containing the lymphocytes.

[Experimental studies of plasma exchange therapy for malignant tumor].

We conducted the experiment to evaluate the effect of plasma exchange in treatment of cancer. Donryu rats of 8-10 week old female were used: 1 X 10(6) AH109A were inoculated subcutaneously in the back of rats. We performed comparative study on four groups: 1) untreated group; 2) plasma exchanged group; 3) MMC treated group; and 4) plasma exchange + MMC treated group. Plasma exchange was conducted from 8th day of the tumor inoculation for three consecutive days. The plasma contained in 6ml blood was taken out from the tumor-bearing rats and same volume of fresh frozen normal serum was replaced. MMC (0.5 mg/kg/day) was administered intraperitoneally from 13th day of the tumor inoculation for five consecutive days. Efficacy of plasma exchange therapy was evaluated by tumor inhibition effect as well as by 50% survival time. Plasma exchanged group showed no tumor inhibition effect, however a considerable extention of survival of 36 days was observed compared with the untreated group (29.3 days). Five out of 14 cases of MMC treated group showed tumor inhibition effect and survived for 36.5 days. Six out of 10 cases of plasma exchange + MMC treated group showed tumor inhibition effect and the survival time was 49 days. The above findings indicate: plasma exchange therapy has effects on prolongation in survival time, but has no effects on tumor inhibition and; a combination of plasma exchange and chemotherapy (MMC) produces greater tumor inhibition effect and longer survival compared to plasma exchange therapy alone and chemotherapy alone.

Magnesium ion induced proton release as a probe for the polyelectrolytic structure of ribosomal RNAs and subunits.

E coli ribosomes and rRNA's released 20 to 50 protons upon jump of magnesium ion concentration from 1 mM to 20 mM. The Mg2+-induced proton release was measured separately for 16S rRNA, 23S rRNA, 30S subunit, and 50S subunit by a new spectrophotometric method that had a much better sensitivity than the pH-stat method. The proton release from the subunits and rRNA's were similar in the number of protons, the pH dependence that had a minimum at neutral pH, and the upward concaveness of the Scatchard plot. From these results, the main source of protons in ribosomal subunits was assigned to nucleotide bases of rRNA's that showed a downward pKa shift upon Mg2+-ion binding. The subunits and rRNA's, however, differed in the proton release. 16S rRNA released protons somewhat more effectively than 23S rRNA, while 30S subunit released protons 2 to 5 times more effectively than 50S subunit. The marked difference between the two subunits suggest that ionizable bases in 16S and 23S rRNA's are covered and their pKa values are shifted by ribosomal proteins to different extents. The association of 30S and 50S subunits induced little proton release, showing that few ionizable groups with pKa near neutral pH are involved in the association. E. coli tRNA and poly U also showed Mg2+-induced proton release. The amounts of protons released from rRNA's, tRNA, and poly U were roughly proportional to the amount of bases not hydrogen bonded. The Mg2+-induced proton release from the natural and synthetic RNA's can be explained by the electrostatic field effect of polyphosphate backbones on bases not hydrogen bonded, as proposed in a previous paper. It also reflects the conformational structure of each RNA molecule.

[Morphological and endocrinological study of theca cell tumor during pregnancy].

With a theca cell tumor of the ovary associated with pregnancy, an extremely rare occurrence in a 30 years old primigravida at 15 weeks of gestation, morphological and endocrinological studies were performed. The tumor was composed of cell-nests scattered among a fibromatous structure. The round or polygonal cells are rich in eosinophilic cytoplasm, resembling theca internal cells. The histochemical activity of 3 beta-HSD and G-6-PDH was demonstrated in these cells. They had the ultrastructures characteristic for steroid-producing cells, in some of which immature and mature Reinke crystalloid-like structures were identified. Mature ones were infrequently observed in contrast to the numerous immature ones. It is considered that a few cells might have differentiated into Leydig cells in the course of differentiation into thecomatous cells from ovarian stroma or mesenchyme. Judged from the rapid enlargement in early pregnancy, this tumor may depend on hCG. Serum estradiol was decreased immediately after removal of the tumor even at 15 weeks of gestation. Histochemical, ultrastructural and endocrinological findings suggest that the tumor was secreting mainly estradiol at the time of the operation.

[The influence of bromocriptine-induced pregnancy and delivery on infertile cases with pituitary microadenoma].

Two consecutive pregnancies were induced in three hyperprolactinemic primary infertile patients bearing intrasellar microadenoma without visual disturbance. Pretreatment prolactin levels, 224 -496ng/ml, were normalized by 10-22.5mg/day of bromocriptine and the initial conceptions occurred. No abnormal sign due to pituitary enlargement was detected throughout the period of gestation and they delivered single full-term babies. Since no significant change was observed in puerperium, it was decided to give the second bromocriptine therapy. The prolactin levels, 80-190ng/ml, measured before the second therapy were apparently lower than those seen prior to the first therapy and the minimum effective dose used to induce the second pregnancy (5-7.5mg/day) was identical to 1/3-1/2 of the first one. The course of the second pregnancy of each case was also uneventful and the babies were all normal and mature. The magnitude of the rise in the prolactin level during the second pregnancy was smaller than that observed during the first pregnancy. The present longitudinal study throughout two consecutive pregnancies in three cases indicates that resolution of prolactinoma was resulted in some extent after withdrawal of stimulative factors associated with pregnancy. Hence, hyperprolactinemic infertile cases with intrasellar microadenoma can be subjected to bromocriptine therapies inducing consecutive pregnancies under intensive follow-up.

[Visual and neurological disturbances accompanying with pregnancies of hyperprolactinemic patients].

Twenty-three hyperprolactinemic infertile cases become pregnant after surgical treatment and/or bromocriptine administration, and 18 of them accomplished normal full-term deliveries. Serial ophthalmological examination which had been performed monthly throughout pregnancy revealed that four cases bearing pituitary adenomas developed visual disturbances which manifested themselves in the latter part of gestational period. The objective signs detected in these cases were bitemporal hemianopsia, temporal and nasal visual defects and the disappearance of central isopter. Another patient with microadenoma complained of headache at 16 weeks of gestation and it became more severe as gestation progressed. Bromocriptine (1.25 mg/day) was effective in diminishing this symptom. Although all these cases recovered from visual disturbances and headache after delivery, two of them were operated on transsphenoidally when radiological evidence of pituitary enlargement was found in the postpartum period. It is generally accepted that nearly 20% of hyperprolactinemic patients have pituitary adenomas. In view of the limited accuracy of techniques used in diagnosing pituitary tumor, it is legitimate to postulate that most hyperprolactinemic infertile patients have microadenoma to some extent. For this reason, a detailed follow up of visual field and neurological signs throughout pregnancy of hyperprolactinemic patients is important in early detection of changes in pituitary size.

Synthesis and structure--activity relationship of nonyl 3-acyldithiocarbazates and related compounds for uncoupling activities.

Various nonyl 3-substituted-dithiocarbazates, methyl and dimethyl derivatives of nonyl 3-benzoyldithiocarbazate, nonyl 2-substituted-dithiocarbamates, and benzaldehyde nonyldithiocarbohydrazone were synthesized and their uncoupling activities of oxidative phosphorylation in mitochondria were examined. The results indicate that the presence of the thiocarbamoyl structure with the potential SH group is a requisite for uncoupling activity. The presence of a C=O group and a hydrophobic aromatic ring significantly increases the uncoupling activity.

Neuritogenic determinant of bovine P2 protein in peripheral nerve myelin.

Experimental allergic neuritis (EAN) is an experimentally produced demyelinating disease of peripheral nervous system. Several peptides of bovine P2 protein were tested for neuritogenic activity in Lewis rats. The hexacosapeptide CiT4 (residues 53--78 of bovine P2 protein) showed the highest neuritogenic activity among the peptides tested. The nonapeptide (residues 70--78) and the tridecapeptide (residues 66--78) were synthesized using the liquid phase peptide synthesis technique. The tridecapeptide showed mild, but definite activity in inducing EAN in the rats, while the nonapeptide was inactive. The localization of the neuritogenic determinant of bovine P2 protein in Lewis rats is discussed.
