[2 cases of bullous pemphigoid and Bowen's disease associated with esophageal cancer].

There are many published reports on the combination of malignant tumors of the digestive tract and various skin diseases. To date, 8 cases with combined bullous pemphigoid and malignant tumor, or Bowen's disease and malignant tumor, have been reported in Japan. In only 2 cases was the esophageal cancer combined with skin disease. The pathophysiological relationship between skin diseases and malignant tumors of the digestive tract remains unknown.

[Hypothalamic-pituitary-ovarian function in puerperal women].

Hypothalamic-pituitary-ovarian function in women during the first month postpartum has been studied. The plasma LH-RH level in puerperal women was very low on days 5 and 10 postpartum and thereafter increased, but was still significantly lower on day 30 postpartum than that in eumenorrheic women on day 8 or 9 of the normal cycle. Serum FSH reached a very low level on days 5 and 11 postpartum, then gradually increased to a slightly higher level on day 30 postpartum than that in eumenorrheic women. The serum estradiol level fell immediately after delivery and remained low until day 30 postpartum. No response of serum FSH or LH to LH-RH was found on day 11 postpartum. However, daily injections of LH-RH from days 5 through 10 postpartum caused an increased secretion in FSH on day 11 postpartum, both basally and in response to LH-RH. On day 20 postpartum, serum FSH responded excessively to LH-RH in all women, but the response in serum LH was restored in a half of them. The mean basal level of serum FSH was significantly higher in cases demonstrating the response in serum FSH and LH than that in cases demonstrating the response in only serum FSH. Serum FSH and LH showed excess and normal responses respectively to LH-RH on day 30 postpartum. It is concluded that hypothalamic secretion of LH-RH decreases during the first month postpartum, especially for the first 10 days, and the low basal level of serum FSH and unresponsiveness of the pituitary to LH-RH observed in the early puerperium may result from hypothalamic hypofunction.

Spontaneous age-associated amyloidosis in senescence-accelerated mouse (SAM).

Morphological studies on spontaneous systemic amyloidosis were conducted on 222 senescence-accelerated mice (SAM) (P) and on 150 mice in the senescence-resistant series (R). Among the pathologic findings, amyloidosis showed the highest incidence in both SAM (79.7%) and R (32.7%). Although an extensive deposition of amyloid was evident in some aged mice in the R series, a more severe amyloidosis occurred with a higher incidence in the P series. There was a statistical significance between the incidence of amyloidosis and age, in both the P and R series. There were no differences in organ distribution and mode of amyloid deposition between the P and R series or between the sexes. In about 60% of the amyloid-positive cases in the 28 killed SAM and 7 mice in the R series, there were no signs of inflammation or neoplasm. The morphological features in SAM more closely resembled those seen in cases of murine spontaneous senile amyloidosis than the features seen in cases of experimentally induced amyloidosis. This model is expected to be a valuable tool with which to assess the relationship between amyloid deposition and the aging process or senescence, perhaps even cases of human senile amyloidosis.

Oxytocin-induced Ca-free contraction of rat uterine smooth muscle: effects of preincubation with EGTA and drugs.

1. The effect of the time of preincubation with 3 mM EGTA on oxytocin-induced Ca-free contraction of rat uterus was investigated. It was found 10(-2) unit/ml oxytocin induced sustained Ca-free contraction only after preincubation for 50 min or more. 2. The tension developed showed a biphasic relation to the time of preincubation with a minimum at 25 min. 3. Sustained Ca-free contraction could be repeated with only slight reduction in magnitude. 4. Oxytocin-induced Ca-free contraction was relaxed by isoproterenol. The effect of isoproterenol was inhibited by 3 X 10(-4) M 1,1-diphenyl-3-piperazino-1-butanol hydrochloride.

A cyclohexanecarboxylic acid utilizing yeast: isolation, identification, and nutritional characteristics.

A yeast capable of utilizing cyclohexanecarboxylic acid as sole carbon and energy source, strain KUY-6A, was isolated from soil by enrichment cultures. Taxonomical studies indicated that strain KUY-6A was Trichosporon cutaneum. Strain KUY-6A grew on a number of carboxylic acids. Among the cyclic compounds tested, cyclohexanecarboxylic acid was the best substrate. Cyclopentanecarboxylic acid, cycloheptanecarboxylic acid, cyclopentanone, cyclohexanone, and cyclopentanol also supported growth. In addition, the organism used the monocarboxylic acids, butyric, valeric, and caproic; the dicarboxylic acids succinic, glutaric, adipic, pimelic, and suberic; and the aromatic acids, benzoic and o-, m-, and p-hydroxybenzoic. The yeast did not require any vitamins for growth, although thiamine gave slight stimulation. The cell dry weight yield was 0.75 g from 1 g cyclohexanecarboxylic acid used.

A novel amyloid fibril protein isolated from senescence-accelerated mice.

A protein was isolated from amyloid fibrils extracted from the liver of an inbred strain of mice (senescence-accelerated mouse) characterized by a high frequency of age-associated systemic amyloidosis. This 5200-dalton protein has a different electrophoretic mobility from the murine protein AA. Its amino acid composition also differs from that of murine protein AA and from the amyloid protein in SJL/J, the only strain for which the spontaneous occurring amyloid has been characterized biochemically and immunochemically. Sequence analysis of the protein also revealed a blocked N-terminus. Immunochemically, the protein did not react with antisera against mouse immunoglobulin components. The antiserum against the protein did not react with murine protein AA, mouse immunoglobulin components, or with mouse normal liver protein. Thus, we have characterized an amyloid protein that is distinguishable from previously reported murine amyloid proteins and is not related to immunoglobulins. The possible relationship between this protein, designated ASSAM, and ASc in human senile cardiac amyloidosis is considered.

[Clinical trial of netilmicin in infections of respiratory organs and studies on its penetration into pleural fluid II. (author's transl)].

To 10 cases with respiratory infections, 200 mg, twice daily, of netilmicin was administered without other antibiotics and the following results were obtained. 1) Netilmicin was administered to 2 cases of pneumonia and 8 cases of bronchitis for 7 to 30 days, and 4 remarkably effective and 6 effective cases were observed, that is, netilmicin was effective in all cases. 2) Abnormal laboratory test values were found in 2 cases; 1 case showed slightly elevated creatinine value, and 1 case showed slightly increase GTP value, and these values were normalized rapidly without any treatment after discontinuation of netilmicin administration. Netilmicin 100 mg was intramuscularly injected to 15 patients with pleural effusion to see the time-course distribution of the drug to serum and to pleural fluid by determining the concentration of netilmicin. 1) Netilmicin concentrations in serum reached the peak at 30 minutes after the intramuscular injection and it gradually decreased, while in the pleural fluid, it reached the peak at 3 hours after the injection, and the peak value in the pleural fluid in average was 2.63 +/- 1.98 micrograms/ml, and it was still detectable at 24 hours after the injection. 2) The ratio of netilmicin concentrations in the serum and pleural fluid at the peak was 31.7 +/- 23.4%, and distribution of netilmicin into the pleural fluid was considered to be high enough.

A new murine model of accelerated senescence.

Five senescence-prone series of mice (P-1, P-2, P-3, P-4 and P-5) and three senescence-resistant series (R-1, R-2 and R-3) were obtained by continuous sister-brother breeding from five original litters of AKR mice with severe deterioration, and the three original litters of AKR mice with normal aging, respectively. A grading score system was adopted to evaluate the degree of senescence of these mice and a steady and irreversible increase in this grading score was seen with advancing age in both the R and P series. The high grading score in the P series was due to an earlier onset of loss of passivity and reactivity, loss of skin glossiness and increased coarseness, hair loss, periophthalmic lesions, increased lordokyphosis of the spine and a more marked increase in their severity with advancing age as compared to the R series. Among the P series, P-2 showed a 100% incidence of systemic amyloidosis after 6 months of age and P-3 a 70% incidence of cataract over 16 months of age. The life span in the P series was shortened by about 26% of that of the R series. In view of the evidence obtained from the survivors, the growth rate and Gompertz function, the aging pattern in the P series was considered to be an acceleration of senescence. The P series has been named "SAM" ("Senescence Accelerated Mouse").
