An alpha-galactosyl residue in the large carbohydrates of teratocarcinoma cells: the antigenic determinant recognized by sera from patients with ovarian germ cell tumors.

Certain patients with ovarian germ cell tumors develop a specific antibody reacting with glycoprotein-bound large carbohydrates of murine teratocarcinoma cells. The antigenic determinant was found to involve an alpha-galactosyl residue, since alpha-galactosidase from coffee bean, but not other glycosidases abolished the antigenic activity of the large glycan isolated from F9 and OTT6050 cells. Several evidences excluded the possibility that the antigen is blood group B or P1 antigen. These results indicate tumor-associated expression of an unusual alpha-galactosyl residue in human ovarian germ cell tumors.

Establishment and properties of a human choriocarcinoma cell line of ovarian origin.

A human nongestational choriocarcinoma cell line of ovarian origin (IMa) was established in vitro. This cell line had been subcultured serially more than 22 times over 18 months. Small polygonal cells with a prominent nucleus were dominant and a sparsity of cytoplasmic organelles was an ultrastructural characteristic of the IMa cells. The production and secretion of human chorionic gonadotropin and its subunits were identified by radioimmunoassay. The IMa cells were transplantable in the hamster cheek pouch and the histological diagnosis was choriocarcinoma. A newly established ovarian choriocarcinoma cell line can be considered useful for clarifying the biological differences between nongestational and gestational choriocarcinoma cells.

A newly established human gestational choriocarcinoma cell line and its characterization.

The cell biological characteristics of a newly established human gestational choriocarcinoma cell line, HM, were examined and compared with four other gestational and two nongestational choriocarcinoma cell lines. Striking heterogeneity in the production and secretion of hCG and its subunits was observed among the cell lines. The HM cell line consisted mainly of cytotrophoblast-like cells, and the hCG and its subunit values in both the cells and culture fluid were extremely low. The syncytiotrophoblast-like multinucleated giant cells which were occasionally seen in other cell lines were scarce in the HM cell line in vitro. On the other hand, deviation of the LDH isozyme pattern to the heart-type (LDH1) was characteristic of the HM cells in contrast to the other cell lines in which LDH3 was dominant. The HM cell line represents useful material for investigating the cell biological heterogeneity of human choriocarcinomas.

[Autopsy case of malignant carcinoid tumor of the ascending colon].

A 72-year-old female operated for ascending colon cancer was histologically diagnosed as having co-existing goblet cell-like carcinoid and rosette-forming carcinoid. The tumor cells were positive for the argyrophil reaction, but negative for the argentaffin reaction. A few tumor cells had both argyrophil granules and sulfated acid mucin in their cytoplasm. Despite combination chemotherapy she expired 10 months after the operation. Autopsy revealed local recurrence and metastases to the liver, lung, pancreas, left adrenal and many lymph nodes.

Antibodies to large glycopeptides in sera from patients with ovarian germ cell tumours.

Antibodies reactive with a murine teratocarcinoma cell line (F9) were detected by immunofluorescence staining in sera from patients bearing ovarian germ cell tumour. Immunochemical studies revealed that antibodies binding to the cell surface of F9 cells react with large glycopeptides which are known to be components of F9 antigens defined by murine anti-F9 antibodies. In addition, treatment of F9 cells with retinoic acid, which induces differentiation of embryonal carcinoma cells, distinctly reduced both the ability of these antibodies to stain F9 cells and the biosynthesis of large glycopeptides by the cells. These findings indicate that the large glycopeptides precipitable by the patients' antibodies are differentiation associated antigens on characteristic embryonic cells.

[A new serological diagnostic method for malignant ovarian tumors of germ cell origin. Detection of antibodies to large glycopeptides isolated from F9 cells].

Whether the presence of antibodies in patients sera to large glycopeptides (LGP) isolated from a murine teratocarcinoma cell line (F9) can be useful in diagnosing malignant ovarian tumors of germ cell origin was examined by Farr's assay. We measured the binding ability to LGP by Farr's assay of human sera from 188 patients with various ovarian tumors and 110 normal volunteers. All of the 9 patients with dysgerminoma showed negative results (less than 200 cpm). Except the cases with dysgerminoma, 9 (64%) of the 14 patients with malignant ovarian tumors of germ cell origin were found positive (more than 200 cpm). Of these 9 positive cases, 4 cases with endodermal sinus tumor (EST), 2 cases with immature teratoma, one each with immature teratoma plus EST, choriocarcinoma, and mature solid teratoma were included. In contrast, all of the sera from benign ovarian tumors (111 cases), other malignant ovarian tumors (54 cases) and normal nonpregnant women (110 cases) were found negative. These result indicated that Farr's assay of anti-LGP antibodies in sera is useful in diagnosing ovarian germ cell tumors.

[Case of primary infiltrating carcinoma in the right breast with metastatic ovarian dysgerminoma].

A 33-year-old woman whose left ovarian dysgerminoma had been resected presented with left axillar swelling and a right breast tumor about three years after surgery. Histological examination of the mastectomized tumor revealed primary infiltrating carcinoma into the muscle layer and a metastatic ovarian dysgerminoma showing well-defined proliferation. Despite chemotherapy, she died 7 months later of breast cancer. To our knowledge, this is the only reported case of its kind in the world literature.

[Epidemiologic studies on multiple cancers in Japan].

During a 21-year period, 3040 patients with malignant tumors were treated at the Division of Surgery, Fukuyama National Hospital. Of these 86 (2.8%) were diagnosed as having double cancers. The simultaneous double cancer rate was 27.3%. Statistical study showed that the etiology had a relation to family history and previous therapy, but not to immunology. The prognosis was good and diagnosis by RI was non-specific.

A study of fetal growth retardation in teratological tests: relationship between body weight and ossification of the skeleton in rat fetuses.

The normal development of preterm rat fetuses (day 19.0 to day 21.0) was investigated with respect to body weight and ossification of the sacrococcygeal vertebrae, supraoccipital bone, sternebrae, and proximal phalanges in the forepaw. Normal standard growth curves were established for these indexes in rat fetuses. A method was devised for the analysis of the relationship between low body weight and retarded ossification induced by teratogenic agents. In normal fetuses, mean body weight and number of ossified sacrococcygeal vertebrae and the ossification stage of the supraoccipital bone increased approximately linearly with advancing fetal age. The number of ossified sternebrae and proximal phalanges in the forepaw increased curvilinearly with advancing fetal age. By use of the standard curves, fetal growth retardation observed in teratological experiments may be expressed as retardation in relation to the standard in hours. The characteristic pattern of growth retardation induced by an agent may be evaluated by comparison of the degree of retardation expressed in a common "hour" scale among various growth indexes. This method was applied to experimental data of fetal growth retardation induced by maternal fasting. The degree of retardation was found to differ among growth indexes; body weight and ossification of the sacrococcygeal vertebrae were the most severely retarded; ossification of the supraoccipital bone and the sternebrae was moderately retarded; ossification of the proximal phalanges in the forepaw was least retarded.

A study of fetal growth retardation in teratological tests: an examination of the relationship between body weight and ossification of coccygeal vertebrae in mouse and rat fetuses.

We examined the relationship between body weight and ossification of coccygeal vertebrae in normal full-term fetuses of mice and rats to reliably evaluate fetal growth retardation in teratological tests. Correlation coefficients between body weight and number of ossified coccygeal vertebrae were positive in mice and rats. However, the coefficients were variable between groups and were not statistically significant in some groups. Averages of body weight and ossified coccygeal vertebrae were variable in the normal fetuses. However, the coefficient of variation in each group was nearly constant, and the component of variance between groups (sigma 1(2)) was small as compared with that within groups (sigma 0(2)). Therefore, the relationship between low body weight and retardation of ossification observed in teratological experiments may be evaluated by measuring relative differences of body weight and ossification between the treated group and the control group with the variance within groups (sigma 0) as a scale.

[Analytical studies of HSG on cases of tuboplasty and their histopathological findings (author's transl)].

Tubal shadows in HSG after the tuboplasty were analysed in relation to their histology, for the betterment of diagnosis of tubal factors and for the establishment of appropriate therapeutic indication in infertile clinics. 91 cases of tuboplasty were followed up more than one year postoperatively. One set of HSG consist of four films; immediately after injection of contrast medium 3-5 ml (1), additional 6-10 ml (2), their side snap (3) and 5 minutes after the end of injection (4). Tubal partial resection was performed in some of them for microscopic and SEM studies correlating with HSG figures. Passage recovery was observed more than one year, in 73 tubes (52.1%) out of 140, in 91 cases. Pregnancy was successfully brought about in 16 (17.6%). HSG with abnormal direction and shape in tubal shadows suggested adhesion, endometriosis or tumor in their corresponding portions. Abnormal morphology of tubal shadow was associated with histopathological changes in high occurrence rate. Posttuboplatic pregnant cases have indicated physiological range both in their postoperative HSG and in their histological changes.
