Experimental allergic encephalomyelitis-prophylactic and therapeutic treatment with the cyclooxygenase inhibitor piroxicam (Feldene).

Prophylactic administration of Piroxicam (Feldene), a reversible inhibitor of prostaglandin biosynthesis, significantly reduced the occurrence of paralytic signs and the amount of antibodies against myelin basic protein in the model of mild acute experimental allergic encephalomyelitis in the Lewis rat. Mononuclear infiltration of the central nervous system remained unaffected. A therapeutic intervention with piroxicam, however, increased paresis and CNS pathology. Immunohistochemical studies revealed an increased proportion of ED1-positive macrophages and monocytes in the infiltrates of the spinal cord in animals treated with piroxicam. Possible reasons for the different effects of the prophylactic and therapeutic treatment are discussed in the study.

Quantification of thioguanine-resistant lymphocytes from mice irradiated in vivo.

Adult mice were Co-60 gamma irradiated, and 7 months later splenocytes were isolated, cultured in microwells, and the frequency of hprt-deficient mutants was determined by measuring the cloning efficiency in media with 6-thioguanine. The mutant frequency at 2, 4, and 6 Gy was 1.6 x 10(-5), 4.4 x 10(-5), and 12.7 x 10(-5), respectively. The frequency of spontaneous mutants was 2.5 x 10(-6). The effect of metabolic cooperation on the cloning efficiency of thioguanine-resistant T-cells in selective medium was evaluated in co-cultures with wild-type T-cells. We found that the growth of hprt-deficient T-cells is supported in the presence of thioguanine-inactivated wild-type splenocytes up to a cell density of 5 x 10(5) cells per well. When cell density was higher, cell growth was inhibited. Possibilities and limitations of cloned lymphocytes for the analysis of somatic mutations that occur in vivo are discussed.

[Efforts toward harmony within the scope of the European Community. What has been accomplished, what remains to be done?].

Report about harmonization of surgical training in the E.C. countries. Responsibility of the U.E.M.S. (European Union of Medical Specialists), in particular of the section of surgery. Each country is represented by two members. Annual meetings. A harmonization committee has been founded under direction of Prof. Gruwez, Leuwen/Belgium. A first report on the harmonization of surgical training in the E.C. countries is available. Standards of quality shall be reached. Consideration to found a European Board of Surgery.

Effect of atrial natriuretic peptide on the adenylate cyclase activity in the anterior and neurointermediate pituitary lobes of rats.

The effect of two atrial natriuretic peptides, rat ANP (3-28) and synthetic ANP (Arg101-Tyr126), on pituitary adenylate cyclase activity was measured in rats. Neither one was effective on basal, forskolin- or guanylylimidodiphosphate (GMP-PNP)-stimulated adenylate cyclase levels measured in homogenates of anterior and neurointermediate pituitary lobes. High concentrations (10(-7) and 10(-6) M) of ANPs further stimulate NaF-induced high adenylate cyclase activity - ANP (3-28) in both lobes, ANP (Arg101-Tyr126) only in the anterior lobe-but they were without effect in 10(-8) M or lower concentrations. These data indicate that ANP may not influence adenylate cyclase/cAMP system in the rat pituitary gland.
