Gel chromatography for control purposes of human albumin preparations.

Information on molecular composition of human albumin preparations is important for characterization of these protein mixtures. Permeation chromatography in gel columns is best suited for separative analysis with respect to time consumption and experimental and instrumental expenditure. Standardization, precision and reproducibility of this quality control method have been especially considered in our investigations and the best conditions for practical performance and evaluation have been established. Results will be discussed in comparison to those of sedimentation analysis in the ultracentrifuge.

Toxin-specific ultrastructural alterations of the mouse liver after burn injuries and the possibility of a specific antitoxic therapy.

Electron-microscopic studies in mice revealed similar and comparable mitochondrial alterations of the liver cells 5-7 days after either a sublethal controlled burn injury or an i.p. injection of an equivalent dose of a burn toxin. Electron microscopy 5 days after i.p. application of different amounts of the burn toxin in rats showed that the extent of the liver alterations correlates directly to the applied dose (occurrence of: cristolysis--intramitochondrial vacuolization--total vacuolar changes of mitochondria(. Controls with the non-toxic/"native" compound isolated from normal skin or excision of a skin piece identical in size to the sublethal burn showed no ultrastructural changes in the liver of mice or rats. In a 2nd series of experiments the therapeutic effect of an antitoxic IgG raised in sheep was tested. The first 3 days after a standard burn or an i.p. injection of 15 mg burn toxin mice obtained 10 mg of the antitoxic IgG (2x/day). Controls injected with the "native" compound or excised as described were treated in the same way. The results showed a specific complete immunological protection from mitochondrial alterations by either the toxin or the burn injury. These results suggest the possibility of an antitoxic IgG-therapy in severe burns.

Immunoelectron microscopic localization of T proteins in the cell wall of Streptococcus pyogenes.

T proteins of Streptococcus pyogenes (group A streptococci) were localized by means of immunochromatographical isolated anti-T-antibodies. For the electron microscopical detection both the direct and the indirect immunoferritin techniques were used. The arrangement of the ferritin particles showed, that the T proteins are evenly distributed on the whole cell surface. They are immediately bound to the outer layer of the cell wall or to only short filaments. On isolated cell walls the T protein was detected only on the outer surface.

Gel chromatography applied to quantitation of components of IgG preparations.

Investigations were undertaken to standardize gel chromatography for quality control of IgG preparations. Great importance was attached to convenience of procedure. With application of the optimal type ofgel, Ultrogel AcA 34, the influence on quality of separation of various experimental parameters such as pH and ionic strength of the eluant, volume and concentration of the sample, flow rate and direction, and temperature were tested. Various methods of evaluation were applied. According to the demands, quantitation of components may be performed by collection of fractions and determination of peak volumes and protein concentrations, by determination of peak areas on absorbance recordings, or automatically by use of an integrator or a computer. The results obtained by gel chromatography are discussed and compared with the results of analytical ultracentrifugation.

Improved methodology for the quantitative analysis of mixtures by band centrifugation: reversible equilibration in immunoglobulin preparations.

Double-sector aluminium centerpieces are modified for use in analytical high-speed band centrifugation up to 60,000 rpm. A method has been developed for testing IG preparations according to administrative regulations with special regard to precision and reproducibility. The proposed band-forming centrifugation method with scanner registration in UV absorption optics and special evaluation of the curves gives values in comparison to well-known procedures by Svedberg and Pedersen. Subjective and systematic errors are especially estimated with this method and the sensitivity is best suited to automatic computerization and results are therefore readily available at the end of the centrifuge run within a few minutes. Investigations have been undertaken to analyze reversible equilibration in immunoglobulin preparations; the results are discussed.

[Pathophysiology of burns].

Ultrastructural and metabolic alterations of the liver were studied in mice and rats after standardized "in vivo" burn injuries or i.p. injections of a specific cutaneous burn toxin. The main findings of the ultrastructural investigations were delayed and specific intramitochondrial vacuolizations appearing not before the 5th-7th day after burning or toxin-injections. -By applying the Schimassek model of the isolated and perfused liver for investigating liver metabolism we found a drastically reduced ATP/ADP-quotient on the 5th day after burns or toxin-injection as compared to the normal controls. The total content of adenonucleotides, however, remained unchanged. The urea and glucose release into the perfusate was also significantly decreased in both groups of animals and the glucose synthesis was not adequately stimulated by an additional lactate load. The results show a direct effect on the burn toxin on the hepatic metabolism as well as the hepatic cell structure suggesting a marked alteration of the oxydative phosphorylation.

[Radioimmunological determination of aldosterone in the serum. New method for the localization of aldosteronism].

It is described a radioimmunologic method for the determination of aldosterone in the plasma and the urine. The method possesses a high specifity, so that a chromatographic prepurification can be given up. The achieved sensitivity allows an analysis in 0.2 ml plasma. This radioimmunoassay can be used for the determination of the aldosterone concentration in the blood after side-separated taking from the adrenal veins by means of the catheter technique. With this a localisation diagnostics of the aldosteronoma is possible.
