Cardiovascular effects of morphine and opioid peptides following intracisternal administration in chloralose-anesthetized rats.

Beta-Endorphin (0.9--2.0 nmol), morphine (11--250 nmol) and D-ala2-met-enkephalinamide (17--33 nmol) administered intracisternally produced preferential vasodepressor responses and bradycardia. Leu- (1.8--180 nmol), met-enkephalin (17--520 nmol) and alpha-endorphin (5.7--57 nmol) administered in the same way produced preferential vasopressor effects and the latter two peptides also produced bradycardia. Results obtained with naloxone (300 nmol) given intracisternally indicate that the pressor and depressor actions as well as the bradycardia are mediated through opiate receptors. The results indicate that opioid peptides may be involved in central cardiovascular control.

Ergot drugs and central monoaminergic mechanisms: a histochemical, biochemical and behavioral analysis.

The effects of ergot drugs on central monoaminergic mechanisms, particularly dopaminergic mechanisms, have been investigated in histochemical, biochemical and behavioral studies. It is concluded that the behavioral effects and the antiparkinsonian properties of ergot drugs can to a large extent be explained by the hypothesis that ergot drugs act at various dopamine receptor sites in the brain as partial agonists, the ratio of agonist-antagonist activity varying from one dopamine receptor population to another.

Prolactin-like immunoreactivity: localization in nerve terminals of rat hypothalamus.

Antibodies to rat prolactin were used in immunohistochemical studies of the hypothalamus and preoptic area of the rat. Evidence was obtained that a protein immunochemically related to prolactin was stored in networks of nerve terminals of many hypothalamic areas such as the arcuate nucleus, the dorsomedial hypothalamic nucleus, and periventricular regions of the hypothalamus and preoptic area. The neuronal storage of a prolactin-like protein in the hypothalamus was unaffected by hypophysectomy.

Localization and characterization of phenylethanolamine N-methyl transferase in the brain of various mammalian species.

The distribution of PNMT activity in various regions and nuclei of the rat, monkey and human brain was investigated. The distribution of PNMT activity in the rat brain correlates with the distribution of PNMT immunofluorescence. The PNMT activity in the primate brain is more widely distributed than in the rat brain. High and intermediate enzyme activity values were dound in the same regions of the primate brain as in the rat brain. Intermediate or low levels were also found in various other regions of the primate brain, e.g., basal ganglia, amygdala, septum, habenula. The brain PNMT has the same substrate specificity and similar kinetic properties as the adrenal enzyme. Immunotitration studies revealed cross-reactivity between the homologous adrenal and brain PNMT.

Enhancement of sexual behavior in the female rat by nicotine.

The effects of nicotine alone or in combination with mecamylamine, and the effects of both in combination with DA agonists and antagonists and d-LSD (5-HT agonist) have been studied on the sexual behaviour of castrate, estrogen-treated female rats. The results show that nicotine (as low as 50 mug/kg) significantly increases sexual receptivity. The pharmacological analysis suggests this effect to be mediated by a central, nicotine-like cholinergic receptor whose relationship to DA and 5-HT pathways known to exert inhibitory influences on receptivity in the female rat is also discussed.
