A new human pleomorphic hepatocellular carcinoma cell line, KYN-2.

A new human hepatocellular carcinoma (HCC) cell line, KYN-2, has been established from a surgical specimen obtained from a 52-year-old Japanese male HCC patient. The originally resected HCC was classified as pleomorphic HCC corresponding to Edmondson-Steiner's grade III with a thick trabecular to solid arrangement. The cell line has been maintained for 17 months through 35 passages. Morphologically, the KYN-2 cells have retained the characteristics of the original HCC, being pleomorphic and composed of various types such as cells with relatively small, polygonal, eosinophilic cytoplasm and oval-shaped nuclei with a marked tendency to pile up, flat cells with abundant clear cytoplasm and oval-shaped nuclei, and many multinucleated giant cells, proliferating in a pavement-like cell arrangement. Some junctional complexes and a number of microvilli are evident between the cells by electron microscopy. Functionally, these cells were found to secrete albumin, alpha 1-acid glycoprotein, alpha 1-antitrypsin, ceruloplasmin, transferrin, complement C, fibrinogen, fibronectin, prothrombin, retinol-binding protein (serum type), alpha-fetoprotein (AFP), carcinoembryonic antigen (CEA), ferritin and beta 2-microglobulin in chemically defined medium (CDM). The secretion of AFP and CEA is apparently dependent upon culture medium and passage. The doubling time of cells growing in serum-containing medium at the 14th passage was 84 h, and those of cells in serum-containing medium, HB101 (serum-free medium) and CDM at late passage were 28, 68, and 42 h, respectively. Chromosome analysis revealed that the chromosome number ranged from 56 to 69 without a mode, and the presence of marker chromosomes. HB virus DNA sequence was not detected by hybridization analysis. The tumorigenicity of KYN-2 cells was identified by development of tumors in nude mice after subcutaneous injection of the cells; the tumors showed an appearance basically similar to that of the original HCC. Thus, these findings suggest that the KYN-2 cell line is available as a new human HCC cell line and should be useful for various studies on HCC.

Noninvasive evaluation of right coronary artery-right atrial fistula using two-dimensional echocardiography, pulsed Doppler echocardiography and color flow mapping.

The case of a patient with right coronary artery-right atrial fistula was reported, with special reference to the noninvasive evaluation. On two-dimensional echocardiography in the short axis view, it was observed that the fistula had an entrance from the right coronary cusp, with the body of fistula sigmoid-like appearance running along the tricuspid ring and atrioventricular groove. In the subxyphoidal approach the exit of fistula into the right atrium was demonstrated. Intrafistular blood flow and drainage flow into the right atrium were identified by color flow mapping. The potential usefulness of the combination of two-dimensional, pulsed Doppler echocardiography and color flow mapping were discussed.

Sequence of preparatory set for response movement.

Electromyographic reaction times (EMG-RTs) of the right biceps brachii muscle were examined for two movement patterns, elbow flexion and forearm supination, in 8 healthy male subjects under simple and complex RT conditions with varied preparatory intervals (PIs): 0, 200, 400, 600, and 800 msec. In the simple RT condition, the subject was informed of the movement patterns to be performed prior to beginning the trials. In the complex RT condition the subject had to choose one of the two movement patterns at the time of the presentation of a warning signal. The results indicated that: (1) compared with the simple RT condition a delay of about 100 msec. in over-all mean EMG-RT was observed at PI = 0 msec. in the complex RT condition; (2) the difference of over-all mean EMG-RT between the two RT conditions disappeared when PI = 400 msec.; and (3) the difference in EMG-RTs between flexion and supination in the complex RT condition became the same as that in the simple RT condition when PI = 700 msec. It is assumed that the preparatory set for response movements is organized in an order, resulting in the differentiation of RT.

Induction of peroxisome proliferation in rat liver by dietary treatment with 2,2,4,4,6,8,8-heptamethylnonane.

1. Exposure of rats to 1% (w/w) of 2,2,4,4,6,8,8-heptamethylnonane in the diet for 2 weeks resulted in marked induction of liver peroxisome proliferation as judged from electron micrography, elevated activities of hepatic catalase (36%), cyanide-insensitive palmitoyl-CoA oxidase (10-fold), carnitine acetyl transferase (9.6-fold), lauric acid hydroxylase (12.4-fold), and the induction of the 80 K protein in SDS-polyacrylamide gel electrophoresis (4.1-fold). 2. 2,2,4,4,6,8,8-Heptamethylnonane dicarboxylic acid, a non-beta-oxidizable fatty acid, was detected as the major metabolite in the liver, an example of an unmetabolizable lipophilic anion as a peroxisome proliferator.

Biomechanical analysis of stability and fixation strength of total shoulder prostheses.

The Neer I (polyethylene glenoid), Neer II (plastic glenoid surface with metal backing), Cofield, and Gristina monospherical total shoulder prostheses were tested for joint subluxation resistance and glenoid component fixation strength. Synthetic glenoid models with consistent structural properties and standard under-cutting geometry were used for glenoid component fixation with bone cement. It was found that joint subluxation resistance varied linearly with axial compressive force applied. Higher subluxation strength was associated with the amount of curvature of the glenoid articulating surface. Consequently, joint stability was consistently higher for the Gristina design, and all had lower resistance in the anteroposterior direction. After complete joint dislocation under high axial compressive force, the polyethylene material at the site of subluxation experienced gross plastic deformation. The fixation strength of the glenoid component was lowest for the Neer I design and highest for the Cofield design. Fatigue loading at the joint did not seem to affect the glenoid component fixation strength. In general, all glenoid components appeared to have sufficient fixation strength against normal shoulder joint forces except the Neer I design. Loosening and fracture of the plastic glenoid component may occur under excessive loads.

[Determination of various tumor markers, with special reference to sialyl SSEA-1 antigen in lung cancer].

Tumor markers including sialyl SSEA-1 antigen (SLX) were measured in 77 patients with lung cancer and 116 patients with benign respiratory diseases. Increased levels of serum SLX were observed in 57.6% (19/33) of adenocarcinoma of the lung, and an overall positive rate was 42.9% (33/77) in lung cancer cases. The false positive incidence of SLX in the sera from the patients with non-malignant diseases was as low as 8.6%. Among other tumor markers determined, serum CA 19-9, CEA had a relatively high positive rate, but the false positive incidence were not low. In cases of poor prognostic cases of with adenocarcinoma of the lung, SLX levels became levels higher than the previous ones, but not in cases of other cell types were not. Not only in the patients with lung cancer but also in those with benign respiratory diseases such as bronchiectasis and lung fibrosis, high levels of SLX seemed to suggest poor prognosis. These results indicate that SLX may contribute for monitoring and prognosis of the cases of lung cancer and other respiratory diseases.
