[Changes in meal patterns and endogenous chemical determinants related to food intake following intra-ventricle III infusion of mazindol].

To clarify suppressive effects of mazindol on food intake in rats, changes in body weight, meal patterns as well as 24-hr food intake, and endogenous chemical substances were investigated following the intra-ventricle III infusion of 0.03 mumole mazindol. Experiments were carried out under the condition of a 12: 12 light-dark cycle (light: 0800-2000 hr). Mazindol decreased food intake as well as body weight after a 12 hr starvation. Reduced food consumption was observed during 12 hr following the injection. Weight reduction, however, lasted over all 3-tested days. When food was available ad. lib., mazindol decreased meal size during the 4 hr after injection and prolonged postprandial intermeal interval during the 4 hr period starting 2 hr after the injection. The infusion of mazindol also produced relative hyperglycemia and decreased free fatty acids, which was not accompanied with hyperinsulinemia. These findings, together with other reports, indicates that mazindol, unlike amphetamine and fenfluramine, possesses inhibitory actions on the hypothalamic feeding center.

Changes in meal pattern and endogenous feeding related substances following mazindol administration.

In order to ascertain direct central anorectic actions of mazindol (MZD), subtle changes in meal patterns and endogenous feeding related chemical substances were examined in rats following intra-third ventricle injection of 0.03 mumole MZD. In ad lib feeding, MZD decreased meal size and prolonged postprandial intermeal interval during a 4 hr period starting 2 hr after injection. The magnitude of anorectic actions of MZD was depressed by hunger. Although the action of MZD is short in duration, long duration anorexia was achieved by chronic infusion for 8 days. Infusion of MZD starting at 5:50 p.m. decreased plasma insulin, leaving glucose and glucagon unaffected, although no change in plasma glucose or insulin was observed following injection at 11:30 a.m. These findings, together with other reports, can be explained by MZD's direct effects on the hypothalamic hunger and satiation centers.

Synthesis and expression of RNase T1 gene.

In order to obtain knowledges about structure-function relationship of ribonuclease T1, we synthesized a structural gene for RNase T1 and several its modified genes. Using amino acid codons frequently used in Escherichia coli we designed genes consist of 328 X 2 bases. Synthesis of oligodeoxynucleotides with 9-20 base lengths was performed by 1% polystyrene supported triester approach and resulting 42 oligomers were joined together using T4 DNA ligase. The product was analyzed and utilyzed to construct expression vectors, which produced effectively fused proteins.

[Effect of elcatonin on experimental osteoporosis induced by ovariectomy and low calcium diet in beagles].

Effect of elcatonin, a synthesized analogue of eel calcitonin, on experimental osteoporosis in beagles induced by ovariectomy and a low calcium diet was studied. Twelve female beagles, 15-18 months old, were divided equally into 3 groups. The first group of beagles were ovariectomized and then fed a low calcium diet, Ca: 0.08%, (ovariectomy plus low Ca diet group); the second group was subjected to the same procedure and diet plus daily injection of elcatonin, 1.0 u/kg, i.m., (elcatonin group); and the third group was fed a standard diet, Ca: 1.36%, (control group) for 6 months. The undecalcified bone sections from the iliac crest were analyzed using histomorphometric methods following in vivo double labeling with Calcein. In the ovariectomy plus low Ca diet group, there were a decrease in trabecular bone volume and an increase in both resorption and formation surface of the trabeculae compared with the control group. These changes of trabecular bone volume and resorption surface were significantly prevented by daily injection of elcatonin. The increase of formation surface of trabeculae and mineral appositional rate was noted in the elcatonin group in comparison with the ovariectomy plus low Ca diet group. These results suggested that elcatonin could prevent bone loss caused by deficiency of sex hormone and dietary calcium.

Phorbol ester increases the level of interleukin 2 mRNA in mitogen-stimulated human lymphocytes.

TPA alone did not induce the production of IL 2 in human tonsillar lymphocytes but enhanced the PHA-induced IL 2 production by seven-fold. That the effect of TPA was due to an increase in IL 2 mRNA was demonstrated by examining the amount of IL 2 mRNA translatable in Xenopus laevis oocytes, and by Northern blotting analysis using IL 2 cDNA as a probe. In these ways, it was shown that TPA alone did not induce any significant IL 2 mRNA synthesis, but when added together with PHA it increased the level of IL 2 mRNA by at least 10-fold, as compared with that induced by PHA alone.

Cloning of interleukin 2 mRNAs from human tonsils.

Human interleukin 2 (IL-2) mRNAs were cloned from a cDNA library prepared from mitogen-stimulated tonsillar mononuclear cells. One of these clones was sequenced and, using this cDNA as a hybridization probe, Southern blot analysis of the human placental DNA was performed. Our results indicate that there is only one IL-2 gene in the human genome.

Evidence for a malignant proliferation of IgE-class specific helper T cells in a patient with Sézary syndrome exhibiting massive hyperimmunoglobulinemia E.

Peripheral blood mononuclear cells (PBM) from a patient with Sézary syndrome exhibiting massive hyperimmunoglobulinemia E were examined in vitro. The patient's PBM and B cells (Bp) but not normal individuals' PBM and B cells (Bn) produced spontaneously large amounts of IgE. The addition of pokeweed mitogen (PWM) did not affect IgE production by both the patient's and normal individuals' PBM. The IgE production by PWM-stimulated Bp was depressed when cocultured with normal T cells but not depressed with the patient's T cells (Tp). When Tp were cocultured with Bn, significantly larger than expected quantities of IgE were produced. Ig assay of the same supernates showed that Tp had significantly less helper activities for IgG, IgA, and IgM production. Almost all Tp possessed the Leu3a and Leu3b antigens which are expressed on the helper/inducer T cell subset. These results indicate that the neoplastic cells in this patient originated from a subset of T cells programmed not for IgG, IgA, and IgM, but for IgE synthesis.

Diagnostic significance of immunological assay of pancreatic elastases in acute pancreatitis.

Pancreatic elastases have been assumed to be implicated in the pathogenesis of the vascular injury in acute hemorrhagic pancreatitis. We reported the purification and some properties of human pancreatic elastases. In the present study, the value of the determination of serum pancreatic elastases was investigated. 39 patients with acute hemorrhagic and acute edematous pancreatitis were studied. Serum elastase 1(E-1) and elastase 2(E-2) were measured by radioimmunoassay recently developed in our laboratory. The molecular form of exogenous and endogenous elastases in serum was studied by gel filtration on Sephadex G-200. The mean and standard deviation of serum pancreatic E-1 and E-2 were 1.49 +/- 0.49 ng/ml and 292 +/- 82 ng/ml, respectively. In acute pancreatitis, both E-1 and E-2 had markedly raised serum concentrations. However, no significant difference was observed between acute hemorrhagic and acute edematous pancreatitis. Endogenous immunoreactive E-1 and E-2 were present in serum in complex form with alpha 1-antitrypsin. In contrast, exogenous elastases were bound to alpha 1-antitrypsin and alpha 2-macroglobulin.

Innervation of capillaries by local neurons in the cat hypothalamus: a light microscopic study with horseradish peroxidase.

The protein tracer horseradish peroxidase (HRP) has been used in an attempt to define the cell bodies of origin of "nonadrenergic" varicose axons which terminate on the walls of hypothalamic capillaries. Capillaries in this region are also known to receive direct axonal contacts from adrenergic neurons in the pontine locus ceruleus. Solutions of HRP were infused into the lateral ventricles of adult cats of either sex and permitted to circulate in the cerebrospinal fluid spaces for 10 min, 20 min, or 2 h. During these periods HRP entered the perivascular spaces around penetrating arterioles and spread into the surrounding extracellular spaces of the hypothalamus. Certain neurons in the periarteriolar neuropil were consistently labeled by the tracer after all three circulation periods. These cells, including all of their processes, could be visualized in detail. Most neurons, by contrast, did not accumulate HRP. The axons of some tracer-filled neurons terminated on the walls of capillaries in the immediate vicinity of the penetrating arteriole. The arrangement and distribution of these cells suggest that they may provide a substrate for local neural influences on the hypothalamic microcirculation.

Translation of human interleukin 2 mRNA in Xenopus laevis oocytes.

Poly(A)-positive mRNA extracted from tonsillar mononuclear cells stimulated with phytohemagglutinin-M and 12-o-tetradecanoyl phorbol 13-acetate was successfully translated into biologically active interleukin 2 (IL-2) in Xenopus laevis oocytes, and secreted into the incubation medium. In control experiments, the extract of oocytes injected with either poly(A)-negative RNA or buffer did not show any IL-2 activity. By sucrose density gradient centrifugation analysis, IL-2 mRNA was found as a single peak corresponding to a sedimentation coefficient of 10-11S.
