Tumor induction by a single subcutaneous injection of N-methyl-N'-nitro-N-nitrosoguanidine and its derivatives in newborn mice.

N-Methyl-N'-nitro-N-nitrosoguanidine (MNNG), or its derivatives N-hexyl-N'-nitro-N-nitrosoguanidine (HNNG) and 1,6-bis(N'-nitro-N-nitrosoguanidinyl)-n-hexane (HxBNNG) were given to newborn ICR/JCL mice by a single subcutaneous injection in 1% gelatin suspension. In an acute toxicity study, the maximum tolerated dose of MNNG, HNNG or HxBNNG was 62 microgram/g, 555 microgram/g, or 500 microgram/g of body weight, respectively. In a chronic study, a single subcutaneous injection of MNNG to newborn mice at a dose of 62, 31, or 3 microgram/g of body weight induced tumors of the lung and hemangioendotheliomas in both sexes, and tumors of the liver in males. Other pathologic findings, such as deformities of the spine and alopecia of the skin, were frequently observed. The incidences of tumors in each group were clearly dose related. HNNG and HxBNNG were not tumorigenic within the observation period.

A radioimmunoassay of plasma unconjugated and conjugated estetrol.

A radioimmunoassay for the measurement of both unconjugated and conjugaged estetrol in plasma has been developed. The antiserum obtained after 6 months of immunization with 6-oxoestetrol-6-(O-carboxy-methyl)oxim-BSA was used at a final dilution of 1:90,000 and showed almost no cross reaction with other steroids except for estriol at 1.24%. Esterol-glucosiduronate was synthesized by incubating with adrenalectomized rat liver homogenate and uridine diphosphoglucuronic acid. Then, plasma estetrol-glucosiduronate was measured in the same manner for unconjugated estetrol after hydrolysis with beta-glucuronidase. Sephadex LH-20 column chromatography (7X110 mm, benzene:methanol, 85:15) was employed for accurate assessment. The sensitivity was 10 pg and the smallest amount measurable was 40 pg/sample. The method bland was consistently negligible. The intra and inter assay precision was 11.8% and 14.2% for unconjugated estetrol and that for estetrol-glucosiduronate was 13.5% and 17.1%.

Induction of tumours by administration of N-dibutylnitrosamine and derivatives to infant mice.

Pulse doses of N-dibutylnitrosamine(DBN), N-butyl-N-(4-hydroxybutyl)nitrosamine(BBN) and N-butyl-N-(3carboxypropyl)nitrosamine(BCPN) suspended in 1% gelatin, were administered s.c. to infant CDF1 mice, and the experiment terminated at one year of age. Tumours were induced in lungs and liver. The incidences of lung adenomas were 73-95% in all treated mice, with no sex differences. Hepatocellular adenomas and a carcinoma were found with an incidence of 81% (21/26) in DBN, 59% (13/22) in BBN, and 32% (9/28) in BCPN-treated males and the incidence was 23% (5/22) in DBN-treated females. Only one papilloma of the fore-stomach was induced in mice treated with DBN. These results indicated that the s.c. administration of DBN, BBN, and BCPN induced tumours of the lung and liver, but no tumours of the urinary bladder, under these experimental conditions. The carcinogenic effect on mice at the treated dose level was DBN greater than BBN greater than BCPN.

Volatile metabolites of halothane in the rabbit.

To date, carbon dioxide is the only volatile metabolite that has been identified to result from the biotransformation of halothane. This study was undertaken to determine whether other volatile metabolites might be formed. Expiratory gas from four rabbits given halothane by inhalation and from three rabbits into which the halothane was injected intraperitoneally was analyzed by gas chromatography. Qualitative analysis of the metabolites was made by injecting 50-70 microliter of the expired halothane condensed in an ultralow-temperature device (-80 C) attached to the mass spectrometer. Gas chromatography revealed two volatile metabolites between the air peak and the halothane peak. They were identified by mass spectra to be CF2:CHCl and CF2CH2Cl. These volatile metabolites appeared immediately after the beginning of anesthesia. The present investigation suggests the possible existence of a previously unknown metabolic pathway of defluorination and debromination occurring in the early stage of halothane biotransformation. These volatile metabolites may be toxic, highly reactive intermediates that undergo further biotransformation.

[The influence of stimulation of the glossopharyngeal nerve on the gastric inhibitory and excitatory motor reflexes (author's transl)].

The influence of electrical stimulation of central cut end of the glossopharyngeal nerve on the vagal inhibitory and excitatory reflexes of gastric movements were investigated. Frogs and bullfrogs were employed in all experiments under urethan anaesthesia and also immobilized with 1% solution of d-tubocurarine chloride. Bilateral splanchnic nerves were previously severd, and so the stomach was innervated through the vagus nerves only. 1. The vago-vagal inhibitory reflex of gastric movements was depressed or abolished during the maintenance of stimulation of the glossopharyngeal nerve. But, the vago-vagal excitatory reflex of gastric movements was augmented in the one example and in the other example it was depressed during the glossopharyngeal stimulation. 2. The labyrintho-vagal excitatory reflex of gastric movements was excited during the maintenance of stimulation of the glossopharyngeal nerve.

[Clinical experience with sustained release cefalexin (S-6437) for pediatric use in pediatrics (author's transl)].

S-6437 is a preparation of sustained release cefalexin consisting of non-enteric coated granules and enteric coates ones and it is reported that the drug shows good effectiveness with b.i.d. regimen. Various infections in pediatrics were treated with S-6437 and the following results were obtained. Dose used was 40 approximately 100 mg/kg/day at 2 divided doses after breakfast and dinner. 1) Twenty patients (87.0%) of the 23 with respiratory tract infections and 3 (60.0%) of the 5 with the other infections responded to the drug. Overall effectiveness was 82.1% (23/28 cases). 2) As for side effects, tentative diarrhea was observed in 5 patients (17.2%) of the 29 but no other severe side effects were found. 3) S-6437 is considered to be a drug which overcomes inconvenience of giving regular cefalexin four times a day.

Osteogenesis imperfecta: biochemical studies of bone collagen.

The biochemical properties of tibial bone collagen, obtained from patients with osteogenesis imperfecta, were studied by investigating amino acid composition, subunit composition and crosslink formation. Direct comparison of this bone was made with normal bone, age and sex matched, which had been removed from the tibiae of individuals within 48 hours after accidental death. The amino acid compositions of OI and normal bone collagen were almost identical and the pepsin solubilized collagen fraction as well as the CNBr peptides of insoluble bone collagen were very similar, indicating that no differences in collagen genetic type occurred in OI compared to normal. The crosslink contents and the specific radioactivities of the insoluble collagens were determined following NaB3H4 reduction. The specific radioactivity values of OI bone collagen were found to average 50 per cent higher than normal collagen. Analyses of OI collagen showed abundant formation of the major reducible aldehydes and crosslinks. Compared to the controls there were much higher proportions of the reduced aldehyde, dihydroxynorleucine and the reduced crosslink, dihydroxylysinonorleucine. These results may indicate delayed maturation of crosslinking in OI bone collagen and may reflect diminished stability of such collagen during bone development.
