Biopsy specimens of lymph nodes or bone marrow taken from two cases, which seemed to have received intravenous infusions of polyvinylpyrrolidone (PVP) more than 15 years ago, were examined histopathologically, histochemically or electron microscopically. In the lymph nodes, proliferation of foam cells, often with multinuclear giant cells, and basophilic vacuolar massive deposition were found markedly in the lymphatic sinuses and sporadically in the parenchyma. In the bone marrow, basophilic foam cells predominated, together with infrequent appearance of multinuclear giant cells and basophilic vacuolar deposits. The basophilic materials stored in the foam cells and multinuclear giant cells, as well as basophilic vacuolar massive deposits, were histochemically proved to stain with the special stains for demonstration of PVP. Electron microscopically, storage inclusions observed in the cytoplasm of PVP-storing macrophages were comprised of electrondense, amorphous substances or of vacuolar, fine reticulogranular, flocculent materials and were bounded by a single delimitating membrane. Most of the inclusions, particularly osmiophilic ones, showed intense acid phosphatase activity, while larger, vacuolar inclusions had a reduced or no enzyme activity. Extracellularly, vacuolar storage inclusions with no enzyme activity were found. The possible mechanism of long-term PVP storage in the tissues was briefly discussed.

To investigate the mechanism by which various biological action of licorice root are brought about, the effects of echinatin as a small constituent of Glycyrrhiza echinata and several related compounds on mitochondrial energy transfer reactions were examined. The results obtained were as follows: 1) Echinatin, 4'-hydroxychalcone, chalcone and 3,4'-dihydroxychalcone at a low concentration cause deterioration of respiratory control and oxidative phosphorylation of isolated rat liver mitochondria. 2) Chalcone and 4'-hydroxychalcone stimulate both latent and DNP-ATPase activity of mitochondria. Echinatin inhibits DNP-ATPase activity while stimulating range latent ATPase activity in the low concentration. 3) Chalcone and 4'-hydroxychalcone induce a rapid potassium release from mitochondrial vesicles, while echinatin and 3,4'-dihydroxychalcone have lesser effect than the former two substances. From these results, it can be concluded that echinatin and several related compounds disturb the mitochondrial energy transfer reactions and membrane permeability.

Whole-embryo culture techniques have advanced to the point where the study of normal and abnormal primary palate development in vitro is possible. The purpose of this study was to ascertain whether localized administration of tunicamycin (TM) an inhibitor of protein glycosylation, into the region of the developing primary palate would induce cleft lip in culture. Rat embryos were explanted on late day 11 of gestation and cultured with open yolk sacs for 40 h. TM was administered by implanting a sterile eyelash soaked in TM into the nasal placode region. The contralateral placode was used as the control by implanting an eyelash lacking TM. Under these conditions, TM-treated placodes were found to develop cleft lip in 14 out of 15 embryos compared to 0 for the controls. These experiments demonstrate that localized administration of TM results in cleft lip formation in whole embryo culture. The technique of localized administration of drugs and teratogens in whole embryo culture should prove useful for similar studies on embryonic development.

Dental examinations and tooth measurements were conducted on 16 mothers, 10 fathers, and 23 affected males in 15 families with X-linked hypohidrotic ectodermal dysplasia. Small teeth and congenital missing teeth were sufficiently consistent findings in obligate heterozygotes to suggest that carriers can usually be recognized by clinical criteria.
