Activation of central neurons by ventral root afferents.

It is a fundamental principle of vertebrate neuronal organization that sensory fibers are restricted to dorsal roots and motor fibers to ventral roots. Recent evidence, however, indicates that there are many sensory fibers in ventral roots. The present report shows that stimulation of these fibers activates neurons in the dorsal horn. This provides evidence at the single-cell level for the importance of ventral root afferents and provides an explanation for the clinical phenomenon of recurrent sensibility.

Thyroid CT number and its relationship to iodine concentration.

Sixty-seven patients with thyroid disease and 24 normal controls were examined with computed tomography (CT). The mean CT number (Hounsfield units +/- SD) in the normal controls (118.1 +/- 12.2) was significantly higher (p less than 0.001) than the mean CT number in patients with diseased thyroids, except for 2 cases of simple goiter (CT numbers 113, 132). The mean CT numbers in the patients with Graves disease (69.5 +/- 17.6) and Hashimoto thyroiditis (61.4 +/- 9.1) were significantly higher than those in patients with adenoma (41.7 +/- 10.6, p less than 0.001), cyst (33.1 +/- 14.8, p less than 0.001), or cancer (48.7 +/- 13.0, p less than 0.01). In 14 patients studied, a significant correlation was observed between thyroid CT numbers and the iodine concentration of the tissue (r = 0.889; p less than 0.001).

Inhibition of thyrotropin-induced adenosine 3'5'-monophosphate increase by immunoglobulins from patients with primary myxedema.

Immunoglobulin G (IgG) fractions prepared from the serum of 18 patients with primary myxedema, 9 patients with goitrous Hashimoto's thyroiditis, and 14 normal controls were tested for their ability to alter TSH stimulation of cAMP production in cultured human thyroid cells and the binding of TSH to its receptor. When compared with the cAMP increase induced by 0.1 mU/ml bovine TSH in the presence of normal IgG, cAMP accumulation was significantly inhibited (P less than 0.005) by the addition of IgG from patients with primary myxedema. TSH-induced cAMP accumulation was not affected by IgG from patients with goitrous thyroiditis. IgG from patients with primary myxedema also inhibited the cAMP increase induced by thyroid-stimulating immunoglobulins, but not against the increase induced by prostaglandin E1. None of the IgG tested affected the basal level of cAMP. Two potent inhibitory IgG were strongly positive for TSH-binding inhibitor immunoglobulins. Excluding these, no significant correlation was found between the thyroid stimulation-blocking activity and the TSH-binding inhibitory activity. These data suggest the presence of at least two different types of antibodies in primary myxedema which block adenylate cyclase stimulation by TSH and might be responsible for thyroid dysfunction and atrophy.

Partial purification and properties of the TSH receptors from human thyroid plasma membranes.

Human thyroid plasma membranes were solubilized with 0.5% Triton X-100 and TSH receptors were purified by using DEAE-Sephadex, Con A and TSH affinity chromatography. A TSH binding activity was bound to DEAE-Sephadex equilibrated with 0.05 M sodium acetate, pH 6.3, 0.2% Triton X-100 and was eluted by a linear gradient of 0.1 M to 1.0 M ammonium acetate, pH 6.3. Eighty-five percent of the activity was absorbed to Con A Sepharose and was eluted with 0.5 M alpha-methyl-D-mannoside, 0.05 M sodium acetate, pH 6.0. Seventy-five per cent of the TSH binding capacity could be absorbed to TSH-affinity column and was eluted with 0.1 M glycine-HCl, pH 3.0. By sequential application of the above procedures, more than 100-fold purification of the receptor activity was attainable. [125I]TSH binding of this fraction was inhibited by addition of unlabelled TSH in a dose-dependent manner. Scatchard analysis gave a curvilinear plot with a high affinity association constant of 0.72 X 10(9)M-1. By using Ultrogel AcA 34 gel filtration, the molecular size of the hormonereceptor complex was estimated to be 180 000.

[Effect of medroxyprogesterone acetate (MAP) and vitamin C on DMBA induced rat mammary cancer in relation to estrogen receptor(ER)].

In mature male and female SD rats, the anti-weight gain effect of durabolin plus vitamin C or vitamin C alone was statistically significant compared to durabolin-treated and control rats. The anti-tumor and anti-weight gain effects of MAP plus vitamin C on DMBA-induced rat mammary cancer were compared with those of MAP and vitamin C in relation to ER. An anti-tumor effect was noted in the MAP treated groups, irrespective of the ER status; it was more pronounced in the MAP (100 mg/kg) plus vitamin C treated group with ER+tumors. Among the ER+tumor groups, weight gain was remarkable in the MAP-treated (20 mg/kg) animals; an anti-weight gain effect was seen in the MAP (20 mg/kg) plus vitamin C group.
