Presynaptic inhibition of tooth pulp afferents in the trigeminal nucleus during REM sleep.

In order to find whether presynaptic inhibition of tooth pulp afferents is enhanced in any phase of sleep or wakefulness, the orthodromic mass response of the trigeminal nucleus to electrical stimulation of the ipsilateral tooth pulp and the antidromic mass response of the tooth pulp to stimulation of the trigeminal nucleus were recorded in the cat. Both responses were stable in amplitude during different phases of sleep and wakefulness. However, attenuation of the orthodromic response and augmentation of the antidromic one were sometimes observed during the burst of rapid eye movements during paradoxical sleep, suggesting that presynaptic inhibition is massively enhanced only on those occasions.

Subcutaneous fat necrosis associated with pancreatitis and gastric carcinoma.

Subcutaneous fat necrosis associated with acute pancreatitis in a 54-year-old man was reported. The acute pancreatitis was proved histologically after gastrectomy for coincident early gastric carcinoma. The patient was successfully treated with prednisolone and his skin lesions subsided within a month.

Convergence of sleep-wakefulness subsystems onto single neurons in the region of cat's solitary tract nucleus.

Neurons in the region of cat's solitary tract nucleus were classified into three groups on the basis of their spontaneous discharge rate during slow wave sleep (SS) as compared with wakefulness (W): group I: SS greater than W; group II: SS less than W; and group III: SS divided by W. These three groups were further examined with regard to the modulation of discharge rate i) during motor activity in W, ii) at the transitional phase from SS to paradoxical sleep (PS), iii) during rapid eye movements (REMs) in PS, and iv) following electrical stimulation of the midbrain reticular formation (MRF). During motor activity in W, increase in unit discharge was frequently observed in group II, while group I and III were often associated with suppression. At the transitional phase from SS to PS many neurons showed a reduction in discharge rate. During REMs in PS, about a half of the neurons of each group responded with burst discharge. The responsiveness to electrical stimulation of the MFR was highest for the group I neurons and the lowest for the group III. The inhibitory responses tended to be smaller in magnitude during SS than during W. During PS, both excitatory and inhibitory responses were, in general, markedly diminished. The analysis of the results has suggested that groups I and II neurons may be involved in more multiple functions than group III neurons which have a tendency to maintain steady discharge rate.

Fetal activity determination and neosynephrine test for evaluation of fetal well-being in high risk pregnancies.

At the special outpatient clinic for high risk pregnancies, a total of 197 patients were evaluated with fetal activity acceleration determination (FAD) as a non stress test. FAD was done on 197 patients, of these there were 8 fetal distress (4.7%) and 3 low Apgar scores (1 minute Apgar score less than 6) (1.8%) of 169 positive FAD, on the other hand, 14 fetal distress (50%) and 10 low Apgar scores (35.7%) of 28 negative FAD. Neosynephrine Test was performed in 62 cases as a stress test and the correlation between these two tests was evaluated. In 51 instances FAD was positive and the simultaneous Neosynephrine Test was normal. In 2 cases of negative FAD with positive Neosynephrine Test, there were 2 fetal distress (100%) and 2 low Apgar scores (100%). All the two tests are reliable for fetal evaluation in high risk pregnancies, because FAD and Neosynephrine Test are less time consuming and have fewer contraindications. It is concluded that the combination of these two tests are the most useful screening methods for fetal evaluation in high risk pregnancies.

Mechanism of spermine stimulation of rat-liver isoleucyl-tRNA formation.

The mechanism of spermine stimulation of rat-liver isoleucyl-tRNA formation has been studied by Cleland's method [Biochim. Biophys. Acta, 67, 104--137 & 173--197 (1963)]. It was demonstrated that isoleucyl-tRNA formation proceeded through a ping-pong mechanism in the presence of spermine and magnesium. From a study of the effect of spermine on both the formation of isoleucyl-AMP . enzyme complex and on the subsequent formation of isoleucyl-tRNA from the isoleucyl-AMP . enzyme complex, evidence is presented that the stimulatory effect of spermine functions mainly through the spermine preventing the tRNA inhibition of isoleucyl-AMP . enzyme complex formation.

Effect of polyamines on isoleucyl-tRNA formation by rat-liver isoleucyl-tRNA synthetase.

The effect of polyamines on rat-liver isoelucyl-tRNA formation was studied using isoleucyl-tRNA synthetase purified by column chromatography successively on Sephadex G-200, DEAE-Sephadex A-25, and tRNA-Sepharose 4B. In the presence of 50 mMK+, isoleucyl-tRNA formation was inhibited markedly by 1.5 mM or higher concentrations of Mg2+. However, the addition of spermine to the reaction mixture prevented the inhibitory effect of Mg2+. In the presence of 200 mMK+, the addition of spermine to the reaction mixture stimulated isoleucyl-tRNA formation in the presence of Mg2+ concentrations from 0 to 5 mM. Although the effective concentration was different, spermidine exhibited a similar stimulative effect. The effective concentration of spermine required for stimulation was higher when larger amounts of tRNA were used. The stimulatory effect of isoleucyl-tRNA formation by polyamines was shown to reflect on polypeptide synthesis. When formaldehyde-treated poly(A,U) was used as messenger RNA, polypeptide synthesis from amino acids was stimulated by polyamines, but that from aminoacyl-tRNAs was not stimulated by polyamines.

Whole body bone scintigram and stage classification of the patients with adenocarcinoma of lung.

One hundred and six patients having adenocarcinoma of the lung were studied by whole body bone scanning with 99mTc-phosphates in connection with their clinical stages. Abnormal accumulation was found in 64 patients (60%). There were 64 patients who received surgical treatment. Among these 64 patients, 38 underwent bone scanning within 3 months after the establishment of their clinical stages. Twenty-two of them were classified as Stage I clinically. Pathological stage classification after the surgery revealed these 22 to be classified into 3 groups; 6 for Pathological Stage I, 4 for Pathological Stage II and 12 for Pathological Stage III. None of these 6 in Stage I showed abnormal accumulations. However, 1 of 4 in Stage II and 6 of 12 in Stage III were found to have abnormal accumulations. Therefore, if bone scintigrams had been taken into consideration for clinical stage classification, 7 patients would have never been placed into Clinical Stage I and could be expected more precise treatment. The authors recommend application of the results of whole body bone scanning to stage classification of adenocarcinoma of the lung. Needle biopsy is also recommended to confirm the metastasis at the site of radioisotope accumulation in the rib.
