Superoxide release by zymosan-stimulated rat Kupffer cells in vitro.

Kupffer cells were isolated from pronase-perfused rat livers and were maintained as a monolayer culture in a state of high purity and viability. Immediately after contact with zymosan particles, O2 uptake of the Kupffer cells increased fivefold; about 50% of the net oxygen consumed was accounted for as superoxide released into the medium. Concomitantly, a transient burst of luminol-dependent chemiluminescence, an increased activity of NAD(P)H oxidase and a stimulation of the flow of glucose through the hexose monophosphate shunt were observed. Chemiluminescence and O2- production were almost completely inhibited by superoxide dismutase and iodoacetate. Zymosan-induced chemiluminescence was not inhibited in the presence of the non-penetrating thiol reagents, 5,5'-dithio-bis-2-nitrobenzoate and iodoacetyl-sepharose. Iodoacetate acted on the cytosolic glucose-6-phosphate dehydrogenase rather than on NAD(P)H oxidase of the cell membrane.

Ganglioside biosynthesis in rat liver. Characterization of UDP-N-acetylgalactosamine -- GM3 acetylgalactosaminyltransferase.

UDP-N-acetylgalactosamine--GM3 acetylgalactosaminyltransferase (GM2-synthase) was studied in a Golgi-rich fraction from rat liver. Activity in a cell-free system required the presence of detergents; octyl glucoside was found to be the most effective in stimulating the enzyme. Optimal activity of GM2-synthase was obtained at pH 7.2, in the presence of 0.8% octyl glucoside, 10 mM Mn2+ and 5 mM CDP-choline. The latter was used to counteract the rapid sugar nucleotide hydrolysis caused by a nucleotide pyrophosphatase activity in the Golgi fraction. The apparent Km values for UDP-N-acetylgalactosamine and added GM3 were 0.035 mM and 0.1 mM, respectively. Different results were obtained if endogenous GM3 only was used as the glycolipid acceptor. In this case, the apparent Km value for UDP-N-acetylgalactosamine was 0.18 mM and Co2+ and Fe2+ exceeded Mn2+ in activating GM2-synthase. Under optimal assay conditions and in the presence of added GM3 and 5 mM CDP-choline, the specific activity of the enriched Golgi fraction was measured to be 25-30 nmol X mg protein-1 X h-1; with endogenous GM3 as the sole glycolipid acceptor, V was calculated to be 9 nmol X mg protein-1 X h-1.

Properties and biosynthetic connection of the nucleotide pyrophosphatases of rat liver plasma membrane and endoplasmic reticulum.

The detergent-solubilized nucleotide pyrophosphatases of the rat liver plasma membrane and endoplasmic reticulum fractions were purified by lectin affinity chromatography. They have the same molecular mass of 148 000 dalton; their catalytic properties are also very similar and correspond to those of the trypsin-solubilized activities from the same membrane preparations. Pulse-chase experiments on isolated perfused livers using [3H]leucine indicated different labelling kinetics of the proteins isolated from plasma membrane and endoplasmic reticulum. The plasma membrane enzyme became only slightly labelled in the presence of 100 microM vinblastine. The data support a precursor-product relationship of the nucleotide pyrophosphatases from endoplasmic reticulum and plasma membrane.

[Angiography of the orbit (author's transl)].

Advances in angiography of the orbit are presented. Angiography is done by selective catheterisation of the internal carotid artery or of the branches of the external carotid artery supplying the periorbital area. Since the procedure is done without heavy sedation or even general anesthesia, the patient can follow instructions and move his eyeballs or open or close his eyelids if wanted for the procedure. Magnification angiography and subsequent subtraction is necessary to obtain good images of the globe permitting visualisation of even the smallest vessels in the choroid layer.

[Selective angiography of the external carotid artery and branches (author's transl)].

Selective angiography of the external carotid artery and superselective angiography of single branches can be used for demonstration of blood supply of dural tumors, tumors of the skull, skull base and of facial parts of the skull. Especially malformations of vessels and their blood supply can be shown. Improvements in x-ray technique and apparatus enable movement angiography, which can be very helpful for the diagnosis of special changes. In special kinds of headache influences from outside to the external carotid artery, to the maxillary artery, to the ascending pharyngeal artery, to the posterior auricular artery, to the occipital artery and the meningeal branches can be of higher importance than was assumed before. Selective angiographies of the external carotid artery and radiographic investigations of the anatomical situation are giving the basis for planning of embolisations. For diagnosis of intra- and extra-cranial changes the neuroradiologist is the connecting person between all specialists for skull and brain.

The therapy of anaerobic infections in obstetrics and gynecology.

The sites of infection and causative organisms found at the Department of Obstetrics and Gynecology of the University of Tübingen are reviewed. About 10% were wound infections, and 10% of all organisms were anaerobes. The principles of treatment of obstetrical and gynecological infections by surgery and chemotherapy are discussed. In cases of mixed infections, good antibiotic coverage of both aerobes and anaerobes should be provided.

Cell-free synthesis of a flavoprotein containing the 8 alpha-(N3-histidyl)-riboflavin linkage.

6-Hydroxy-D-nicotine oxidase is an inducible flavorprotein of Arthrobacter oxidans in which one mole of FAD is bound covalently to the polypeptide chain. During cell-free translation of polysomes from nicotine-induced A. oxidans cells in the presence of an Escherichia coli (MRE 600) supernatant fraction, labelled FAD, leucine and histidine were incorporated into 6-hydroxy-D-nicotine oxidase in the same ratio found in the enzyme isolated from whole cells. This indicates that one mole FAD is covalently attached per mol of 6-hydroxy-D-nicotine oxidase synthesized in vitro. In the native enzyme the coenzyme molecule is bound via its 8 alpha-methyl group to the N-3 atom of a histidyl residue. From the translation products an aminoacyl-riboflavin was isolated and its identity with synthetic 8 alpha-(N3-histidyl)-riboflavin was shown.

[Nosocomial infections (author's transl)].

Within 3 years 2546 nosocomial infections were registered among 18 897 hospitalised patients of a gynaecological-obstetric university hospital, Results were obtained in a prospective study on the use of nurses specially trained in hygiene. The incidence of nosocomial infections was 13.5% including predominantly asymptomatic bacteriurias and 3.9% without bacteriurias. It was higher after major surgical intervention and lowest in pregnant women, postnatal patients, vaginal deliveries and minor operations. Bacteriurias were the most common infection in all patient groups. They made up 71% of the total, followed by postoperative pelvic infections (7.3%), phlebitis at the site of infusion (6.5%) and abdominal wound infection (4.2%). During the control programme, introduction and reinforcement of various hygienic measures and continuous instruction and control of the wards and treatment rooms by the specially trained nurses the incidence of all nosocomial infections decreased by 48.1% and of urinary tract infections by 42.7%.
