Mutations in ftsZ that confer resistance to SulA affect the interaction of FtsZ with GTP.

Mutations in the essential cell division gene ftsZ confer resistance to SulA, a cell division inhibitor that is induced as part of the SOS response. In this study we have purified and characterized the gene products of six of these mutant ftsZ alleles, ftsZ1, ftsZ2, ftsZ3, ftsZ9, ftsZ100, and ftsZ114, and compared their properties to those of the wild-type gene product. The binding of GTP was differentially affected by these mutations. FtsZ3 exhibited no detectable GTP binding, and FtsZ9 and FtsZ100 exhibited markedly reduced GTP binding. In contrast, FtsZ1 and FtsZ2 bound GTP almost as well as the wild type, and FtsZ114 displayed increased GTP binding. Furthermore, we observed that all mutant FtsZ proteins exhibited markedly reduced intrinsic GTPase activity. It is likely that mutations in ftsZ that confer sulA resistance alter the conformation of the protein such that it assumes the active form.

Synaptonemal complex analysis of domestic sheep (Ovis aries) with Robertsonian translocations. I. Pachytene karyotype substaging of normal sheep.

Meiotic chromosome pairing behaviour in three normal sheep was surveyed by synaptonemal complex (SC) analysis at the electron microscope level. The SC karyotype of normal rams is composed of a sex pair, three metacentric or submetacentric pairs, and 23 acrocentric autosomal bivalents, with the total autosomal SC length ranging from 309 to 315 mircrons. Five nucleoli are terminally located on the three metacentric, and one large and one small acrocentric autosomal bivalents. XY morphology was used to divide pachytene into five substages. Although pairing abnormalities (mostly SC interactions) were recorded in an average of 16% of the spermatocytes, bivalent-XY association occurred in only 4.7% of the cells.

Synaptonemal complex analysis of domestic sheep (Ovis aries) with Robertsonian translocations. II. Trivalent and pairing abnormalities in Massey I and Massey II heterozygotes.

Zygotene and pachytene spermatocytes from Massey I (t1 5;26) and Massey II (t2 8; 11) translocation heterozygotes each contained one trivalent, often delayed in pairing, while cells from double Massey translocation heterozygotes had two such trivalents. As meiosis progressed, trivalents became fully paired, with acrocentric axes in a cis configuration. Abnormal pairing configurations often resulted from interactions between unpaired chromosome axes or segments. However, when two Massey trivalents were present in the same nucleus, there was no pairing interaction between them. In different Massey translocation heterozygotes, trivalent-involved pairing abnormalities occurred in 14-28% of cells, with XY-trivalent and XY-bivalent-trivalent associations being as high as 7.1-23.1%. In spermatocytes from single and double Massey translocation heterozygotes with normal-sized testes, the total SC abnormality frequency was 34.4% for the t1 heterozygotes, 27.1% for the t2 heterozygotes, and 21.4% for the double heterozygote. One Massey II heterozygote with one normal and one small testis had significantly higher SC abnormality frequency (54%) than normal rams. A trisomic cell was recorded in one ram and two hyperdiploid cells in another ram, but these were unrelated to the translocations. It is suggested that resolution of pairing abnormalities by synaptic adjustment is important in reducing the effects on fertility of the translocations.

Synaptonemal complex analysis of domestic sheep (Ovis aries) with Robertsonian translocations. III. Deficient pairing and NOR role in Massey III heterozygotes.

Spermatocytes from rams heterozygous for Massey translocation III (t(3)7;25) contained one trivalent and those from the rams heterozygous for both Massey translocations I and III (t1t3) contained two trivalents. Where two trivalents and the XY pair are present in the same nucleus they may form a synaptonemal complex (SC), but no SC segment is found between the two trivalents. The frequency of pairing abnormality in four t3 rams and two t1t3 rams did not significantly differ from that in the normal sheep, but t3 trivalents with diverse abnormal configurations were recorded. A hypothesis was proposed that nucleolar organizer regions (NORs) or nucleoli could account for deficient pairing behaviour and hence chromosome rearrangement, because the NOR region on an NOR-bearing chromosome was usually delayed in pairing, and unpaired regions tended to associate with other unpaired axes or SCs.

Escherichia coli cell division protein FtsZ is a guanine nucleotide binding protein.

FtsZ is an essential cell division protein in Escherichia coli that forms a ring structure at the division site under cell cycle control. The dynamic nature of the FtsZ ring suggests possible similarities to eukaryotic filament forming proteins such as tubulin. In this study we have determined that FtsZ is a GTP/GDP binding protein with GTPase activity. A short segment of FtsZ is homologous to a segment in tubulin believed to be involved in the interaction between tubulin and guanine nucleotides. A lethal ftsZ mutation, ftsZ3 (Rsa), that leads to an amino acid alteration in this homologous segment decreased GTP binding and hydrolysis, suggesting that interaction with GTP is essential for ftsZ function.

Cloning and characterization of ftsN, an essential cell division gene in Escherichia coli isolated as a multicopy suppressor of ftsA12(Ts).

A new cell division gene, ftsN, was identified in Escherichia coli as a multicopy suppressor of the ftsA12(Ts) mutation. Remarkably, multicopy ftsN suppressed ftsI23(Ts) and to a lesser extent ftsQ1(Ts); however, no suppression of the ftsZ84(Ts) mutation was observed. The suppression of ftsA12(Ts), ftsI23(Ts), and ftsQ1(Ts) suggests that FtsN may interact with these gene products during cell division. The ftsN gene was located at 88.5 min on the E. coli genetic map just downstream of the cytR gene. ftsN was essential for cell division, since expression of a conditional null allele led to filamentation and cell death. DNA sequence analysis of the ftsN gene revealed an open reading frame of 319 codons which would encode a protein of 35,725 Da. The predicted gene product had a hydrophobic sequence near its amino terminus similar to the noncleavable signal sequences found in several other Fts proteins. The presumed extracellular domain was unusual in that it was rich in glutamine residues. A 36-kDa protein that was localized to the membrane fraction was detected in minicells containing plasmids with the ftsN gene, confirming that FtsN was a membrane protein.

The proper ratio of FtsZ to FtsA is required for cell division to occur in Escherichia coli.

Interactions among cell division genes in Escherichia coli were investigated by examining the effect on cell division of increasing the expression of the ftsZ, ftsA, or ftsQ genes. We determined that cell division was quite sensitive to the levels of FtsZ and FtsA but much less so to FtsQ. Inhibition of cell division due to an increase in FtsZ could be suppressed by an increase in FtsA. Inhibition of cell division due to increased FtsA could be suppressed by an increase in FtsZ. In addition, although wild-type strains were relatively insensitive to overexpression of ftsQ, we observed that cell division was sensitized to ftsQ overexpression in ftsI, ftsA, and ftsZ mutants. Among these, the ftsI mutant was the most sensitive. These results suggest that these gene products may interact and that the proper ratio of FtsZ to FtsA is critical for cell division to occur.

A complex pattern of antigen receptor gene rearrangements in a case of Philadelphia positive acute lymphoblastic leukaemia.

The paper describes a case of Philadelphia (Ph) positive acute lymphoblastic leukaemia (ALL) presenting with high white cell count and central nervous system involvement. Immunophenotypically the case was characterized as common ALL. The t(9;22) abnormality corresponded to a rearrangement within the breakpoint cluster region gene, while antigen receptor gene studies showed multiple rearrangements of the immunoglobulin heavy chain gene (IGH) concomitant with a single rearrangement of the T cell receptor beta chain gene (TCR beta). We speculate that this case represents the neoplastic transformation of a stem cell, the Ph abnormality being involved in the early steps of transformation. It is conceivable that the IGH but not the TCR beta gene was accessible to recombination within the malignant clone, thus generating the multiple rearrangements observed. If this is the case, these findings would appear to be compatible with the hypothesis that antigen receptor gene rearrangements may be partly dependent on the accessibility of the corresponding genetic loci.

ftsZ is an essential cell division gene in Escherichia coli.

The ftsZ gene is thought to be an essential cell division gene in Escherichia coli. We constructed a null allele of ftsZ in a strain carrying additional copies of ftsZ on a plasmid with a temperature-sensitive replication defect. This strain was temperature sensitive for cell division and viability, confirming that ftsZ is an essential cell division gene. Further analysis revealed that after a shift to the nonpermissive temperature, cell division ceased when the level of FtsZ started to decrease, indicating that septation is very sensitive to the level of FtsZ. Subsequent studies showed that nucleoid segregation was normal while FtsZ was decreasing and that ftsZ expression was not autoregulated. The null allele could not be complemented by lambda 16-2, even though this bacteriophage can complement the thermosensitive ftsZ84 mutation and carries 6 kb of DNA upstream of the ftsZ gene.

The life experience and status of Chinese rural women from observation of three age groups.

Interview data gathered during 2 surveys in Anhui and Shejiang Provinces in 1986 and 1987 are used to depict changes in the social status and life situation of rural women in China in 3 age groups, 18-36, 37-55, and 56 and over. For the younger women, marriage increasingly is a result of discussion with parents, not arrangement, but 3rd-party introductions are increasing. They are active in household and township enterprises and aspire to more education and economic independence. The middle-aged group experienced war and revolution and now work nonstop under the responsibility system of household production, aspiring to university education for sons and enterprise work for daughters. The older women, while supported by their sons, live a frugal existence. In general, preference for sons is still prevalent and deep-seated. At the same time, the bride price and costs of marriage are increasing and of widespread concern. Rural socioeconomic growth is required before Confucian traditions are overcome.

[Quantitative evaluation of human balance function and its significance].

A system of computerized balance function evaluation is introduced. 86 normal chinese adults divided into two age groups tested by the system under 6 standing postures, and a series of normal values of 7 balance parameters and 3 curve diagrams were then obtained. No statistical difference could be found in the values of parameters between male and female. A comparison between the old-age group and the middle-aged and young group revealed no significant difference in each parameter at the time of open eyes with the two feet standing side by side. In terms of closed eyes with the two feet standing side by side, one foot standing in front and the other behind, and single foot standing, the parameters between the two age-groups showed significant difference (p less than 0.05-0.01). The advantages of the system and the value of its application in medical practice, sport medicine, aesthetic medicine, judicature and especially in geriatrics are discussed.

Enzyme-linked immunosorbent assay for the detection of antibodies to bovine viral diarrhea virus in bovine sera.

A specific and sensitive enzyme-linked immunosorbent assay (ELISA) was established for the detection of antibodies to bovine viral diarrhea virus (BVDV) in bovine sera. Polyethylene-glycol concentrated, equilibrium density gradient purified BVDV was used as test antigen at an optimal amount of 1 microgram/well, whereas the optimal concentration of conjugate was at 1/2000 dilution. The standardized test encountered no non-specific reaction with test sera at a starting dilution of 1/10. A total of 50 bovine serum samples was assayed for the presence of antibodies against BVDV by ELISA and serum neutralization test (SNT). A positive correlation between the 2 tests was found. However, ELISA could be as much as 500-fold more sensitive than SNT in detecting low levels of BVDV antibodies.

On the detection of messages carried in arterial pulse waves.

The hypothesis is made that a disturbance in blood flow at one place can be detected in the arterial pulse waves at a distant site. This hypothesis was motivated by the traditional Chinese medicine which uses arterial pulse waves as a principal means of diagnosis. We formulated a test by asking whether a disturbance to the blood flow in a leg can be detected by changes in the pulse waves in the radial arteries. In particular, we ask whether the radial artery can differentiate a disturbance in the right leg from that in the left leg. We put force transducers on the radial arteries, depressed them by a specific amount, and recorded the force waves in response to a 2-min occlusion of the blood flow in the right or left tibial artery. The results show that the radial artery force waves do change in response to the flow disturbance. For a given individual, the force varies with the location of the force transducer on the radial artery, the specific amount of initial depression, and the right or left leg occlusion. Generally, an occlusion in the right leg reduces the force level in both radial arteries, the more so in the right radial artery than in the left. Although the discrimination is not very strong, the phenomenon is novel, and warrants further investigation.

[The intelligence structure of the mental retardates: an examination by means of the WAIS (author's transl)].

A comparison was conducted, with the purpose of investigating the intelligence structure of the mental retardates, on the results of an administration of Wechsler Adult Intelligence Scale (WAIS) to the groups of both normal and mentally retarded persons. Intercorrelations among the WAIS subtest scores for each group were separately analyzed by the method of principal component analysis. Three common factors were found for the two groups: (1) the factor of general intelligence, (2) the factor separating verbal subtests from all other subtests, and (3) the factor separating numerical subtests from all other subtests. There were no clear difference in intelligence structure between the mental retardates and the normals.

FtsZ and cell division.

The ftsZ gene in Escherichia coli is thought to be an essential gene and to play a pivotal role in cell division. Gene disruption experiments confirmed that ftsZ is an essential gene. Examination of cellular responses to FtsZ depletion indicated that FtsZ was required for division but not for nucleoid segregation. Analysis of mutations within the ftsZ, gene, selected for resistance to the cell division inhibitor SulA, revealed that they also conferred resistance to MinCD. This raises the possibility that ftsZ is the target of these two cell division inhibitors. Analysis of the ftsZ gene from Bacillus subtilis revealed that the gene was required for both septation during vegetative growth and asymmetric septation during sporulation.
