Traction forces of cytokinesis measured with optically modified elastic substrata.

Animal cells dividing in culture undergo a dramatic sequence of morphological changes, characterized by cytoskeletal disassembly as cells round up, redistribution of actin, myosins and other cytoplasmic and surface molecules into the cleavage furrow, and respreading, before daughter cells finally separate at the mid-body. Knowledge of forces governing these movements is critical to understanding their mechanisms, including whether formation of the cleavage furrow results from increased force generation at the equator or relaxation at the poles, and whether traction force subsequently mediates cytofission of the intercellular bridge. We have quantitatively mapped traction forces in dividing cells, by extending the silicone-rubber substratum method to detect forces of nanonewtons to micronewtons. We used a new silicone polymer to fabricate substrata whose compliance can be adjusted precisely by ultraviolet irradiation. We show that traction force appears locally at the furrow in the absence of relaxation at the poles during cleavage. Force also rises as connected daughter cells respread and attempt to separate, suggesting that tension contributes to the severing of the intercellular bridge when cytokinesis is completed.

Donor-specific transfusions have long-term beneficial effects for human renal allografts.

From April, 1980 to November, 1988, 163 one or two haplotype-mismatch living related (LR) or living unrelated (LUR) potential renal transplant recipients received three 200 ml aliquots of donor specific transfusion (DST) at biweekly intervals with concomitant azathioprine (2 mg/kg/day). Following transplantation, only prednisone and azathioprine were given for immunosuppression. The results for the DST group are compared with those for HLA identical living recipients (57 patients) transplanted during this same interval (1980-1988). Comparison is also made with a group of one or two haplotype-mismatched living donor recipients (54 patients) not treated with DST but with triple drug therapy (prednisone, azathioprine, cyclosporine) and antithymocyte globulin (ATG) or OKT-3 induction. Permanent T cell crossmatch sensitization occurred in 11 of 163 patients (7%). Successful DST donor transplants were performed between 121 one HLA haplotype-mismatched, 14 two HLA haplotype-mismatched LR, and 7 two haplotype-mismatched LUR pairs. Actual one- and five-year graft survivals were 94%, 100%, 100%, and 72%, 85%, and 71%, respectively, for these three subgroups of DST treated patients. The graft survival for all DST pretreated recipients at one, five, and ten years was comparable to the HLA-identical group (94%, 79%, 64% vs. 91%, 80% and 77%). At a mean follow-up of 10 1/2 years, 54% (80 patients) of the entire group of 147 patients transplanted after DST have functioning transplants with a mean serum creatinine of 1.7 mg/dl. Fifteen percent of DST patients (21 patients) died with a functioning graft 2 to 132 months after transplantation, 26% (37 patients) rejected the DST graft after 1 to 128 months, and 6% (9 patients) were lost for nonimmunological reasons. No lymphoproliferative disease developed in the DST group and the incidence of cytomegalovirus sepsis was only 2% (3 patients). The long-term beneficial effects of DST on renal allograft survival and function and the lower incidence of the complications of nonspecific immunosuppression should encourage increased utilization of DST in renal transplantation.

Family history of severe cardiovascular disease in Marfan syndrome is associated with increased aortic diameter and decreased survival.

OBJECTIVES: We attempted to determine whether a family history of severe cardiovascular disease in patients with the Marfan syndrome is associated with increased aortic dilation or decreased survival, or both. BACKGROUND: The prognostic importance of a family history of severe cardiovascular disease in patients with the Marfan syndrome has been incompletely examined. We hypothesized that such a family history would correlate with increased aortic dilation and would be associated with decreased survival. METHODS: One hundred eight affected patients and 48 unaffected family members from 33 multigenerational families with the Marfan syndrome underwent echocardiographic measurement of the aortic root, arch and mid-abdominal aorta. Date of birth and age at death ascertained from family pedigrees were used to perform life table analysis and estimate survival. RESULTS: Aortic root and arch diameters were significantly greater in patients with a family history of severe cardiovascular disease than in patients without such a family history. Of subjects in the highest quartile for aortic size, > 80% had such a family history in contrast to < 10% of those in the lowest quartile (chi-square 57.37, p < 0.00001). Mean age at death and cumulative probability of survival were significantly lower in patients with such a family history. CONCLUSIONS: Among patients with the Marfan syndrome, aortic dilation is greater and life expectancy shorter in those with a family history of severe cardiovascular manifestations. These data suggest that such a family history is an important risk factor for cardiovascular events in patients with the Marfan syndrome.

Identification of source of oscillations in apparent sarcomere length measured by laser diffraction.

The most widely used technique for dynamic estimates of sarcomere length in muscle is laser light diffraction. We have identified conditions under which artifactual oscillations can arise in apparent sarcomere length measured by this technique and report methods to reduce the effect. Altringham et al. (1984) first reported that the diffraction angle can exhibit one cycle of oscillation for each sarcomere length displacement of the illuminated portion of the fiber. We find that the amplitude of similar oscillations is strongly dependent on the intensity of light scattered from objects near the fiber and on the spacing between fiber and scatterer. The oscillations can be eliminated by minimizing scattered light and positioning the fiber a few millimeters from sources of scattering. A theoretical description shows that oscillations of this kind are expected from interference of scattered and diffracted light. Interference fringes were observed along the meridian of the pattern, and these moved during translation of either a fiber or a grating. The movement of fringes across the diffraction order shifts the centroid back and forth and, when associated with steady shortening, can give rise to "steps" and "pauses" in apparent striation spacing.

Beneficial effects of donor-specific transfusions on long-term renal allograft function.

The induction of immunologic unresponsiveness to improve renal allograft survival was attempted in 163 patients by the pretransplant administration of donor-specific whole blood or buffy coat in conjunction with continuous Aza immunosuppression. All donor-recipient combinations were at least one-haplotype disparate, and 21 were two-haplotype disparate. Transient sensitization occurred in 2% and permanent sensitization in 7%. Over a 10-year period, the DST+Aza allograft survival rate is similar to the HLA-identical sibling transplants. The CMV sepsis rate was only 2%, and there were no lymphoproliferative neoplasms. The low rate of sensitization (7%) has permitted almost all patients to undergo eventual renal transplantation from the specific blood donor. This and the low rate of early rejection (3%) argues for a modification of the immunologic response.

Adenine transport in Escherichia coli.

The purP gene for energized high-affinity adenine uptake in Escherichia coli is localized to a restriction fragment between tnaB and bglR. Comparison with DNA sequence data (Burland et al. 1993) indicates the involvement of the open-reading frame f445 (86003 < 84666). A non-energized transport route of lower affinity also occurs. The purP system is known to require metabolic removal of transported adenine which would diminish futile cycling by the two transport systems and toxicity by external adenine. Analysis of a conventional kinetic model for permeases suggests that internal adenine might inhibit transport sufficiently to account for the lack of accumulation.

Malignant sex and aggression: an overview of serial sexual homicide.

Serial murderers have attracted considerable attention in the popular press and criminal justice field, but scientific literature about these individuals is limited. This article provides an overview, from a psychiatric perspective, of serial sexual homicide, one type of serial killing. Characteristics of this type of murder and of these offenders are discussed. Defining qualities and diagnoses applicable to serial sexual killers are reviewed. Various etiologic theories are discussed, with emphasis on the role of fantasy and psychodynamic explanations. Governmental agencies involved in combating this type of crime, along with the role of mental health professionals in criminal profiling, are presented. Finally, the authors explore the reaction of society to this phenomenon.

An improved method of stature measurement for quantitative determination of spinal loading. Application to sitting postures and whole body vibration.

A refined procedure for measuring stature is described; this provides a reproducibility error of 0.4 mm. The procedure accommodates the natural diurnal change in stature and permits estimation of the net stature change caused by a change in spinal loading. A series of measurements done with a cohort of 20 young and middle-aged persons showed that stature decrease was related linearly to the quasi-static load on the spine. The coefficient of proportionality between load and height loss was inversely proportional to the cross-sectional area of the lumbar discs. This method was used to investigate sitting postures and whole-body vibration to demonstrate the applicability of the procedure to quantify spinal strain (and, therefore, estimate comparative loading) in applied ergonomics. Sitting invariably led to an increase in stature, regardless of the type of chair used or the posture maintained. Whole-body vibration did not induce any loss of stature. Thus, this novel approach was able to enhance understanding of spinal behavior under different loading conditions.

Child sexual abuse and forensic psychiatry: evolving and controversial issues.

Child sexual abuse has received growing attention in recent years, and the topic continues to spark controversy among mental health and legal professionals as well as in the popular media. This paper will review the concept of child sexual abuse, cover relevant definitions, address the clinician's role, and then will address the principal evolving and controversial areas. These areas include psychic damages, false allegations, improper investigatory techniques, use of anatomical dolls, admissibility of expert testimony, hearsay testimony, and the competency of minors to testify.

Measuring flexibility.

Intuitively one would think that good flexibility (both extent and dynamic) would reduce the risk of injury as well as enhance performance, and the limited evidence available would tend to support this view. However, much of this evidence is based on studies of young fit sportspersons; we cannot conclude that the same principles apply to the general population until well-designed studies, using suitable instruments, have been performed. Reliable tools for measurement of flexibility are described, together with statistical methods considered appropriate for quantifying reliability. Recently described methods present the opportunity to investigate further, and it seems likely that dynamic flexibility will be a useful parameter to study. In all probability, prediction of fitness-for-task should be based not just on flexibility measures but on multivariable models; flexibility measurement is but one of a number of parameters to be viewed interactively in relation to the specific task or sport under consideration.

Crossbridge activity monitored from the state of polarization of light diffracted by activated frog muscle fibres.

The polarization properties of the first diffraction order have been measured when single frog fibres are illuminated by laser light. The relative difference in the amplitudes of the orthogonal electric field polarization components (differential field ratio) as well as their phase shift normalized by the pathlength (birefringence) have been obtained from fibres at rest and during fixed-end twitches and tetani. The differential field ratio decreased during contraction and the change during a single twitch averaged 69% of that during a companion tetanus. The birefringence of the first order averaged 2.80 +/- 0.59 x 10(-3) (mean +/- SD) at rest and the average decrease during a tetanus was 8.4% +/- 6.4%. The decrease in the differential field ratio upon activation was a decreasing function of sarcomere length, maximum at rest length and falling to zero at about 3.7 microns. Differences between the two first diffraction orders were observed for both the differentiated field ratio and the birefringence. At the time when force had risen to half the value reached at the end of the fast rise of tension, the change in the differential field ratio lead the tension by about 10-15 ms. The differential field ratio returned to its resting value after the fall of tension. The above results suggest that the differential field ratio is a sensitive indicator of intact fibre structure. The temporal lead in the differential field ratio with respect to tension rise supports models in which crossbridges initially attach in a non-force-producing state.

Sarcomere length behaviour along single frog muscle fibres at different lengths during isometric tetani.

A detailed investigation of sarcomere lengthening and shortening during fixed-end tetani has been made along frog muscle fibres stretched over a large range of sarcomere lengths. A variety of sources of error common in such measurements are quantitated and give an uncertainty in sarcomere length of about 53-62 nm. The difference in sarcomere length calculated from the left and right first orders at rest was 21 nm +/- 16 nm and this is suggested to be a measure of 'Bragg artefact'. The laser diffraction measurements showed that the shortening end regions decrease in size during contraction and that the magnitude of shortening is increased at greater fibre extensions. The average length change and sarcomere length of the central and end regions was 0.10 microns (2.85 microns) and 0.37 microns (2.66 microns), respectively. The sarcomere length of the end regions at the end of creep was regularly observed to be less than 2.1 microns. An unexpected finding was the occasional observation of striations in the transition zone between lengthening and shortening regions which remained nearly isometric during a period of tension rise during creep. Measurements of diffraction order linewidth do not suggest increased sarcomere length dispersion in these areas. A smooth transition from shortening to lengthening was always observed. Although our data are in general agreement with the models proposed by Morgan, Mochon and Julian (Biophys. J. 39 (1982) 189-96) and Edman and Reggiani (J. Physiol. (Lond.) 351 (1984) 169-98), specific differences which do exist are discussed.

Polarization states of diffracted light. Changes accompanying fiber activation.

Measurement of the state of optical polarization of light diffracted from single, skinned and intact fibers of anterior tibialis muscle from Rana pipiens revealed a dependence upon rigor, activation, and sarcomere length (SL) change. Changes in total birefringence, delta nT, and differential field ratio value, rT, were determined. In a relaxed, skinned fiber the total birefringence value, delta nT, decreases as sarcomere length is increased from 2.1 microns to approximately 2.8-3.0 microns. From there it increases significantly to a value of approximately 1.8 x 10(-3) at a sarcomere length of 3.6 microns. The differential field ratio, rT, also shows a biphasic response to increasing sarcomere length, first exhibiting a rapid decrease over shorter SL and leveling out after the SL is beyond 3.0 microns. In comparison, relaxed intact fibers change substantially less upon sarcomere length change, showing little change in birefringence and a small bi-phasic change in rT. Skinned fibers were activated using a solution that has the same ionic strength as the relaxing solution and allows repeatable, and sustained activation. A decrease in both delta nT and rT was observed upon fiber activation. The decrease in delta nT and rT was slightly larger at shorter sarcomere lengths than at longer lengths. Relaxed fibers placed in rigor showed changes in delta nT and rT similar to those observed in activated fibers. These results are consistent with the hypothesis that, after activation, a significant portion of the thick filament cross-bridges rotate towards the actin filament resulting in redistribution of the interfilament mass content. They are also consistent with an average orientation of crossbridges in the overlap region different from that in the nonoverlap region.

Diagnostic criteria for intraarticular T.M. disorders.

Epidemiological investigations about the prevalence of TMJ ID and osteoarthritis have largely reported the symptoms and signs associated with these disorders without specific criteria to establish the diagnosis. In this study, specific diagnostic criteria have been developed, using historical, clinical and tomographic items that are predictive of the presence and stage of TMJ internal derangement. The diagnostic criteria were compared to arthrotomography to assess their diagnostic accuracy. Overall, the percent agreement was 75%. The predictability of specific stages of TMJ ID varied. Overall, sensitivity and specificity of the criteria were acceptable. The diagnosis of osteoarthritis was predicted only with tomography. Utilization of these diagnostic criteria in epidemiological studies is valid due to their high overall predictiveness when applied to a large population.
