Antigenic structure of chimeras of type 1 and type 3 polioviruses involving antigenic sites 2, 3 and 4.

Chimeric polioviruses have been made in which regions of the type 1 Sabin strain corresponding to antigenic sites 2, 3 and 4 have been replaced by the corresponding regions of the type 3 Sabin strain. Manipulation of one site or a component of it generally did not affect the reactions of the others, suggesting that they form independent structural features. The extent to which the inserted site expressed the antigenic properties of type 3 could be assessed by reaction with polyclonal or monoclonal antibodies, or by immunogenicity. Site 2 could be expressed on infectious virus and site 3 on heated non-infectious virus (C antigen), but not on the native virion. The results are consistent with the view that sites consisting of a continuous sequence of amino acids may be presented on chimeras, whereas more complex sites, such as site 4 or site 3 of the native virion, are transferred less readily from type 3 to type 1.

Seizure promotion and protection by D-1 and D-2 dopaminergic drugs in the mouse.

Mice injected with pilocarpine (100-400 mg/kg plus 1 mg/kg methylscopolamine), picrotoxin (0.75-6 mg/kg) or strychnine (0.75-6 mg/kg) exhibited clonic or clonic/tonic convulsions. Pretreatment with the D-1 agonist CY 208-243 (0.375-1.5 mg/kg) dose-dependently potentiated the convulsions elicited by 100 mg/kg pilocarpine, but had neither a convulsant nor anticonvulsant effect in mice receiving picrotoxin (3 or 6 mg/kg) or strychnine (0.75 or 1.5 mg/kg). This facilitatory effect of CY 208-243 was abolished by the D-1 antagonist SCH 23390 (0.2 mg/kg). SCH 23390 by itself (0.05-0.8 mg/kg) dose-dependently protected mice against pilocarpine (400 mg/kg) seizures. Stimulating D-2 receptors with LY 171555 (0.167-4.5 mg/kg) dose-dependently protected mice against seizure activity induced by pilocarpine, but neither protected nor sensitised mice given picrotoxin or strychnine. The neuroleptics haloperidol (1-4 mg/kg), sulpiride (10-50 mg/kg), metoclopramide (1.25-6.25 mg/kg), thioridazine (0.5-2 mg/kg) and clozapine (0.5-2 mg/kg) had no effect on the seizure threshold to 100 mg/kg pilocarpine by themselves, although 10 mg/kg thioridazine and clozapine caused 100% convulsions, possibly through a toxic action. When administered in conjunction with a minimally effective quantity of CY 208-243 (0.375 mg/kg), however, all five neuroleptics interacted synergistically with the D-1 agonist to promote convulsions to pilocarpine (100 mg/kg). No such interaction occurred between submaximally protective doses of the D-1 blocker SCH 23390 (0.05 and 0.2 mg/kg) and a wide range of doses of the D-2 stimulant LY 171555 (0.167-4.5 mg/kg).(ABSTRACT TRUNCATED AT 250 WORDS)

Antigenic structure of chimeras of type 1 and type 3 poliovirus involving antigenic site 1.

Chimeric polioviruses have been prepared in which part of the antigenic site 1-encoding sequence of the Sabin strain of type 1 poliovirus has been replaced by sequences based on those found in the homologous region of the Sabin type 3 strain. The chimeras were analysed for their reaction with polyclonal and monoclonal antibodies raised against type 1 and type 3 viruses, and with polyclonal antipeptide sera, as well as for their immunogenicity in animals. The effectiveness with which the type 3 site was presented antigenically varied in ways which were partially predictable, based on the behaviour of type 3 mutants with monoclonal antibodies. However, other factors were implicated which may include conformational effects and other components of the site in addition to those altered in the chimeras. The ability of the chimeras to induce antibodies reacting with type 3 polioviruses paralleled their antigenic reactivity, and evidence is presented for the induction of strain-specific antibodies.

An evaluation of external jugular vein patch angioplasty after carotid endarterectomy.

Seventy-six patients had 91 internal carotid endarterectomies closed with patch angioplasty. There were 36 redo and 55 primary procedures. External jugular vein was used as the patch material in 38, facial vein in nine, Dacron in seven, polytetra flour ethylene (PTFE) in nine, and saphenous vein in 28 operations. There were no perioperative deaths. Five patients required reoperation, but only one required replacement of a defective facial vein. There were five postoperative strokes, but two were related to a contralateral occlusion. Two of the three ipsilateral strokes had a temporary indwelling shunt. Only one patient returned with recurrent neurologic symptoms. Five other restenoses of greater than 75 per cent were noted in three patients. One false aneurysm occurred three months after an external jugular vein patch angioplasty. External jugular vein, although requiring careful handling, is conveniently located in the operative field, does not appear to be associated with increased complications, has excellent long-term results, and allows preservation of the saphenous vein for other bypass surgery.

Tight clustering of human hepatitis B virus integration sites in hepatomas near a triple-stranded region.

The open circular genome of human hepatitis B virus (HBV) is known to contain a partially double-stranded DNA with a single-stranded gap region of variable length. This circular structure of the genome is maintained by base-pairing of the 5' ends of the two DNA stands, the long or L(-) strand and the short or S(+) strand. By cloning, mapping, and sequencing studies, we have localized three recombinational junctions of the integrated HBV in two hepatoma samples, HT14 and FOCUS. Breakpoints of recombination derived from these results and those of others appear to be clustered and coincidental with the identified 5' or the deduced 3' end of the long-strand DNA, respectively. Statistical analysis of these results supports the hypothesis that integration preferentially occurs in an extremely narrow region on the HBV genome. This site-specific recombinational mechanism appears to be conserved among different HBV subtypes. No extensive sequence homology was found between each pair of the recombining parental molecules; however, at the site of crossover, 2- to 3-base-pair junctional homology was consistently observed. Examination of the patterns of the integrated HBV DNAs allowed us to categorize these various patterns into four different groups according to their end specificity and strand polarity. The molecular form of relaxed circle is proposed to be one major substrate for HBV integration. The effect of free strand in the integration of HBV is emphasized in this model. Unlike any other known DNA animal viruses, the site specificity of HBV integration appears to be similar to that of the retroviruses.

Mutations resulting in promoter-like sequences which enhance the expression of araC in Salmonella typhimurium.

The araC gene in Salmonella typhimurium is autogeneously regulated. Nine non-self-regulated mutants were isolated by selecting for increased expression of an araC-lacZ fusion in the presence of a repressing AraC protein. S1 mapping experiments demonstrated that the effect of the mutations was to increase the amount of araC mRNA in the cell. The 5'-end of the major araC transcript was the same in the wild-type (wt) and mutant strains. DNA sequence analysis showed that all nine mutations occurred in the araC promoter. Two mutations were a G-to-T transversion at position -25, six were a G-to-T transversion at position -47 and one was a single bp deletion at position -83. The data suggest that the mutations have created a new RNA polymerase binding site which enhances transcription from the wt start point.

A holistic program for chronic schizophrenic patients.

A 10-week, inhospital holistic health program for male schizophrenic patients was compared with an equally intense social skills training program. The holistic program included training in the stress reduction techniques of exercise and meditation as well as education in stress management. Patients were also encouraged to explore the growth potential of their psychotic experiences and to develop positive beliefs about the outcome of their illness. Both groups showed similar significant decreases in psychopathology from admission to discharge, but the use of medication and a token economy milieu by all patients confounds the interpretability of this finding. After the holistic patients were discharged into the community, there was no maintenance of any of the holistic techniques. The 2-year relapse rate did not differ significantly between the two treatments. Findings from various studies associating schizophrenic relapse with stressful life events and familial tension make further experimentation with stress reduction techniques for the treatment of schizophrenia worthwhile.

Work function analysis of vegetarian entrée production.

Data on labor time for food production can be used as an effective management tool. It is essential for foodservice managers to know how labor time is being used (1). A continuous time study was conducted to determine total labor time for the production of eight vegetarian entrées in a hospital foodservice system. Two work areas were observed: the ingredient assembly area and the cooks' production area. Times were recorded by work function to identify how labor time was distributed. Results showed (a) observed frequency for each work function, (b) time expended in seconds per portion for each work function, (c) percentage distribution of labor time by work function, (d) total time for each employee involved in entrée production, and (e) percentage of total time in which each employee was involved in the production of each entrée. Total labor time varied by type of entrée, ranging from 39.97 to 19.33 seconds per portion. Entrées with the highest labor time required the largest amount of hand labor. A one-way analysis of variance indicated significant differences in mean labor time among the eight vegetarian entrées for direct labor time (p = .0009), and total labor time (p = .0018). No significant differences were found among entrées for indirect labor or delay time.

Receptors for complement of Langerhans cells.

A population of guinea pig epidermal cells, identified as Langerhans cells, formed rosettes with sheep erythrocytes coated with IgM immunoglobulin and C3b (EAC1-3b) but not with intermediates carrying only C1, C14b, or C14b2a. These findings were unusual since most other cells expressing C3b receptors also express C4b receptors. To investigate whether these results were the consequence of a limited number of C4b molecules on the cells, EAC14b cells were prepared with increased amounts of C4b (15 to 250 micrograms on 10(9) cells). EAC14b rosette formation was found with a maximal number when there were 3,000 molecules per cell, a 6-fold excess over that found in previous experiments. One could inhibit this rosette formation by fluid phase C3b as well as by fluid phase C4b, suggesting proximity or possible identity of these 2 receptors.

Performance of task functions by A.D.A. dietetic technicians.

To meet the professional demands of their positions, dietitians must delegate less demanding responsibilities and routine tasks to educationally qualified dietetic technicians. This research was conducted to determine the degree to which A.D.A. qualified dietetic technicians were performing identified tasks.

Comparison of the bronchodilating effects of SCH 1000 with isoproterenol in patients with bronchial asthma.

The effect of SCH 1000 and isoproterenol was evaluated in 10 patients with extrinsic asthma. The onset of action for the SCH 1000 dose compared to the baseline response occurred at or before 15 minutes (FEV1 and MEFR). The onset of action for the isoproterenol dose occurred at or before one minute for all three pulmonary function variables. At the 1-, 3-, 6- and 15-minute intervals the response to the isoproterenol dose was significantly greater than the response to the SCH 1000 dose for FEV1, FVC and MEFR. At 30 and 60 minutes, however, the response to SCH 1000 was progressively increasing, whereas the response to isoproterenol was decreasing. The maximum improvement in FEV1 for isoproterenol was 49% at three and six minutes, whereas for SCH 1000 was 28% at 60 minutes. In patients with chronic bronchitis or bronchial asthma SCH 1000 may be considered as a therapeutic alternative to beta-adrenergic bronchodilators and in patients who also have cardiac arrythmias or angina pectoris and are adversely affected by sympathomimetic drugs.
