[Incidence of postoperative adhesions following use of various methods of coagulation. An animal experiment study].

In 5 groups of 20 rats each, equal areas of the uterus horns and of the corresponding peritoneum of the anterior abdominal wall were coagulated resp. vaporized. To achieve this endocoagulation, mono- and bipolar high frequency current, and CO2- and Neodym-YAG-lasers were used. After 2 resp. 4 weeks, no intraabdominal adhesions were found in two animals after endocoagulation and in one animal each after coagulation with bipolar high frequency current and with CO2-laser. The lowest adhesion rates were seen after endocoagulation and coagulation with bipolar high frequency current. After application of the YAG-laser, to the uterus horns adhesions were present in 100%, applying the YAG-laser to the anterior abdominal wall for a short time there were produced few adhesions. After coagulation resp. vaporization of equal areas of the anterior abdominal wall, significantly more adhesions were found after CO2-laser-application, compared to all other techniques (p less than 0.01). Adhesions to the abdominal wall were significantly less frequent than adhesions to the tubes (p less than 0.01) with all techniques except with the CO2-laser.

[The production of hybrid cells producing monoclonal antibodies against Toxoplasma gondii].

Hybridomas that secrete monoclonal antibody to Toxoplasma gondii have been developed. Two groups of 10 female BALB/c mice each were immunized either over a shorter (71 d) or longer (117 d) period at first with Toxoplasma lysate antigen and afterwards with intact tachyzoites of the RH strain. Higher titres of antibody were obtained with the long-period immunization. The fusion experiments have shown that both schemes of immunization approximately result in the same number (16 and 14% respectively) of hybridoma cell lines producing antigen-specific monoclonal antibodies. Hybridoma cultures secreting antitoxoplasma monoclonal antibodies were screened parallel by indirect immunofluorescence antibody test (IFAT) and enzyme immunoassay (EIA). 16 of the hybridoma cell cultures produced positive results only in the IFAT, 112 reacted only in the EIA and 21 were positive in both tests. The monoclonal antibodies 5B10, 5G6 and 1B2, which are positive in the IFAT form a chemical compound with the major antigens on the surface of RH tachyzoites. The patterns of fluorescence produced by these monoclonal antibodies are in conformity with those produced by using polyclonal sera of Toxoplasma gondii infected hosts (mouse, rabbit, man).

Influence of the specific antagonist of PAF-acether, BN 52021, and of Ginkgo extract on the rejection of murine tail skin allografts and the PAF-acether mortality in mice in particular consideration of the role of TXB2.

An oral application of 2 x 10 mg/kg daily of the PAF antagonist, BN 52021, significantly prolonged the distal allograft survival in the murine tail skin model of cell-mediated allograft rejection. BN 52021 diminished the TXB2 content in the skin at and near to the transplantation site by two thirds whereas PAF-acether significantly enhanced the TXB2 content in murine tail skin. Similar to the tail skin BN 52021 also reduced by more than 70% the TXB2 release from human granulocytes stimulated by calcium ionophore A 23187. The PAF-acether mortality (LD75) in NMRI mice was decreased to 20% by BN 52021 with a lower dose than by BN 52020. Comparing Ginkgo total extract in doses which significantly diminished the 75% mortality by PAF-acether in mice with BN 52021 and BN 52020 in doses that produced the same inhibitory effect, a potentiated effect of Ginkgo extract resulted with respect to the share of BN 52021 and BN 52020 in Ginkgo extract.

Influence of inhibitors of the eicosanoid metabolism, of antagonists of the eicosanoids and of PAF on mortality assayed in three biochemically characterized shock models.

Experiments were carried out to lower the mortality (LD70-90) of rats in ovalbumin-induced anaphylactic (DA) shock and in endotoxin-induced (ET) shock, and of mice after injection of Platelet-activating Factor (PAF shock) comparing the effects of the cyclooxygenase (COX)-inhibitors aspirin (ASA), indomethacin, of the COX-/lipoxygenase (LOX)-inhibitors nordihydroguajaretic acid (NDGA), phenidone and X 86 (analogue of BW 755c), of the inhibitor of thromboxane (TX) synthesis HOE 944, of the TX-antagonist BM 13177, of the PAF-antagonist BN 52021 and of ketotifen. Ketotifen was strongly effective in DA shock, COX- and LOX-inhibitors only slightly. Combined COX- and LOX-inhibitors and BN 52021 showed good effects in the ET shock. Ketotifen was inefficacious. All the used substances influenced the PAF shock. The shock syndromes were biochemically characterized by determination of isocitratedehydrogenase (ICDH) activity, lactate, glucose, haematocrit, numbers of thrombocytes and leucocytes, TXB2 and 6-keto-Prostaglandin(PG)F1 alpha.

Influence of prostaglandins and PG-analogues on the mortality of PAF-acether in mice and the TXB2 release from human granulocytes.

The PAF-acether mortality in female mice varied with strains: AB mice were very resistant, NMRI mice showed dose-dependent effects. We used an i.v. injection of 75 micrograms/kg PAF-acether resulting in a 75% mortality in NMRI mice. Nileprost and PGE1 in one tenth of the doses of iloprost or PGI2 were able to reduce the PAF-acether mortality. Nalador, PGE2 and flunoprost were ineffective. 10 min after PAF application the TXB2 content in tail skin was increased significantly. The inhibitory effect on the TXB2 release induced by A 23187 from human granulocytes was stronger by nileprost than by iloprost. Nalador was ineffective, flunoprost enhanced the release. The lowered TXB2 formation is a possible explanation for the prevention of the PAF-acether induced death by PGs.

[Everyday psychiatry--a contribution to the hermeneutics of diagnostic and therapeutic clinical practice].

The relations between diagnostic and therapeutic practice and the events of everyday life are investigated in a two-fold regard. The first view is directed towards the signification of "psychiatry" and of the psychiatric view-points in the everyday life of society; the second view is the experience of everyday life inside psychiatric institutions. Under both aspects an interest in the development and the both aspects an interest in the development and the process of mental illness is pursued, which is also an interest in correct diagnostic insight and in promoting therapeutic consequence that positively influences the process.

[Cardia cancer: attempt at a therapeutically relevant classification].

On the basis of a prospective analysis of 107 adenocarcinomas of the gastro-esophageal junction we propose a differentiation into adenocarcinomas of Barrett's esophagus (endobrachyesophagus), "real" carcinoma of the cardia, and subcardial carcinoma of the fundus. This classification allows the determination of an adequate surgical resection. Therefore the operative risk can be kept low. Total esophagogastrectomy is reserved only for advanced "real" carcinomas of the cardia in this concept.

Rescue of a tk-plasmid from transgenic mice reveals its episomal transmission.

This communication demonstrates the usefulness of the plasmid rescue procedure for recovery of plasmids from transgenic mice. We have microinjected the plasmid pSK1 harbouring the Herpes simplex virus thymidine kinase gene into fertilized mouse oocytes and succeeded in recovering plasmids from newborns by transformation of E. coli either with HindIII cut cellular DNA or with uncut DNA. The majority of the rescued plasmids were indistinguishable from pSK1 by restriction analysis. The rescued plasmids proved to be functionally active in a transient expression assay in mouse Ltk- cells. The pSK1 DNA sequences were inherited by up to 90% of the second generation progeny mice, which is not in agreement with a Mendelian transmission of heterozygous markers integrated into a single site of the chromosome. These data support the assumption that germ line transmission of non-integrated episomal plasmids can occur.

Evaluation of the Coulter S-Plus V three-part differential in a community hospital, including criteria for its use.

The Coulter S-Plus V three-part differential was evaluated for its precision, correlation with 100-cell manual differentials, ability to process routine specimens without flagging, and ability to detect abnormalities as defined by manual differential. The instrument's precision is markedly greater than that of manual methods, especially in the mononuclear category. Correlation with manual differential lymphocyte, mononuclear, and granulocyte percentages was 0.91, 0.54, and 0.94, respectively. Eighty-four percent of routine samples could be processed by the instrument without further review. With the use of a combination of S-Plus V-generated flags and preset laboratory criteria, only 9% of abnormal manual differentials would have been undetected. Three-fourths of these undetected cases involved low-frequency abnormalities of questionable clinical significance. The three-part differential should be able to decrease the number of manual differentials that are performed, which would result in a cost savings to the laboratory.
