The frequency in Japanese of genetic variants of 22 proteins. I. Albumin, ceruloplasmin, haptoglobin, and transferrin.

This paper presents the results of an electrophoretic survey of approximately 4000 individuals from the cities of Hiroshima and Nagasaki, Japan, for four serum proteins: albumin, ceruloplasmin, haptoglobin and transferrin. The haptoglobin gene frequencies obtained for the HP1-HP2 polymorphism are in agreement with earlier reports. Rare electrophoretic variants of albumin, ceruloplasmin and haptoglobin occur with frequencies of 2-48, 0-50 and 0-58 per 1000 determinations, respectively. The noteworthy finding of 8 distinct transferrin variants in these populations, with a combined frequency of 20-90 per 1000 determinations, is also presented. Four of these variants (Dchi, B1, B3, and DHIR2 which corresponds electrophoretically to D4) have been reported in other populations in Japan, but the other five have not previously been differentiated.

Determination of e antigen and antibody to e by means of passive hemagglutination method.

A simple and sensitive method was developed for the detection of e antigen and antibody to e (anti-e) by means of the passive hemagglutination (PHA) method. Sheep erythrocytes were fixed by glutraraldehyde and coated with either e antigen or anti-e which had been specifically purified from plasma of asymptomatic carriers of hepatitis B antigen by affinity column chromatography, and then by gel filtration on Sephadex G-200. A gain in specific activity of more than 2000-fold for e antigen and 100-fold for anti-e was achieved as compared with the original plasma pool which was used as the source for purification. The PHA method was at least 300 times more sensitive than the conventional immunodiffusion method both for e antigen and anti-e assays. When the PHA method was applied for neat serum samples of 80 asymptomatic carriers, e antigen was found in 21.3% and anti-e in 62.5%, at rates much higher than those determined by immunodiffusion for the identical serum samples at 3-fold concentration (16.3 and 30.0%, respectively).

An epidemic of infantile papular acrodermatitis (Gianotti's disease) in Japan associated with hepatitis-B surface antigen subtype ayw.

An epidemic of infantile papular acrodermatitis (I.P.A.) (Gianotti's disease) occurred in Matsuyama City, in south-east Japan in 1974-75. Patients ages ranged from less than one year to eight years. Hepatitis-B surface antigen (HBsAg) was detected by an immune adherence haemagglutination method in the serum samples of 48 of the 54 patients tested. HBsAg subtypes were determined by a haemagglutination-inhibition method. ayw antigens were identified in 42 patients and adr antigens in 3; it was not possible to determine subtypes in the remaining 3 patients because antigen titres were too low. Since subtype ayw and I.P.A. are extremely rare in Japan, the association of the disease with HBsAg subtype ayw is regarded as being most significant.

Echocardiographic diagnosis of endocardial cushion defects.

Sixteen patients with endocardial cushion defects were studied by echocardiography and ultrasonocardiotomography. Three characteristic findings, common to both complete and partial forms, were observed. They were the ostium primum defect of the interatrial septum, the left ventricular outflow narrowing and the atrioventricular valve abnormalities which are the anatomic bases of endocardial cushion defects. Several additional findings were observed in patients with complete form. Most strikingly, the anterior leaflet echo locating in the left ventricle had the direct connection to that in the right ventricle without the interposition of the interventricular septum by echocardiogram. Ultrasonocardiotomogram, on the other hand, demonstrated beautifully the common anterior leaflet and the high inteventricular septal defect. There were, in addition, the absence of the paradoxical motion of the interventricular septum and the diffuculty in detecting the interatrial septum. The intact interatrial septum continuous with the anterior mitral leaflet was demonstrated in patients with the interventricularseptal defect of atrioventricular canal type, in which there were the typical abnormalities in the atrioventricular valves and the left ventricular outflow narrowing as well. This study indicates that ultrasonic examination may provide a direct, yet noninvasive method in the diagnosis of various types of endocardial cushion defects.

Comparative studies on subacute sclerosing panencephalitis virus and a usual measles virus with special reference to their capacities to produce viral hemagglutinin in Vero cells.

Various comparative studies were made on the capacities of a cell-associated subacute sclerosing panencephalitis agent (SSPE agent, Niigata/1 strain) and a usual measles virus (Toyoshima strain) to produce measles virus hemagglutinin (MV-HAnim) in Vero cells. The results obtained by various methods, such as the hemagglutination test (HA), hemadsorption test (HAd), antibody blocking test, antibody inducing test and fluorescent antibody technique (FAT) using monospecific antibody to MV-HAnin, consistently suggested that the SSPE agent was an incomplete measles-like virus having little if any MV-HAnin, but sharing immunologically closely related viral components other than MV-HAnin with those of the usual measles virus. The significance of these findings is discussed with respects to the latency of the SSPE agent.
