[Laboratory and clinical studies on cefroxadine (author's transl)].

Cefroxadine (CXD) was applied to infectious diseases in children and the following results were obtained. 1. Serum concentration and urinary excretion: CXD was given orally in dose of 10 mg/kg in dry syrup from 30 minutes after meals. The peak serum concentration was at 1 hour after administration in 3 cases and at 2 hours in 1 case, and the average peak serum concentration was 16.41 microgram/ml in these 4 cases. The average urinary excretion rate of the antibiotic during 6 hours after administration was 93.9% in another 4 cases. 2. Antibacterial activity: The MIC of CXD against E. coli was slightly superior to that of CEX, but against S. aureus, S. epidermidis, Str. pyogenes, Str faecalis, Str. pneumoniae, Kleb. pneumoniae, Kleb. oxytoca, Pro. mirabilis, Pro, vulgaris, H. influenzae, H. parainfluenzae, H. parahaemolyticus the MICs of CXD were almost equal to those of CEX. 3. Clinical study: Thirty seven patients with bacterial infections (7 cases of urinary tract infection, 2 of cystitis, 10 of tonsillitis, 9 of scarlet fever, 7 of bronchitis and 2 of bronchopneumonia) were orally treated with CXD dry syrup, 15 approximately 48 mg/kg/day divided into 3 doses. The overall efficacy rate was 91.7%, and side effects inclusive of abnormal laboratory findings were not observed.

Platelet-dependent serum factor that stimulates the proliferation of epidermal cells in vitro.

Agglutinated platelets are known to release a factor that stimulates the growth of arterial smooth muscle cells or dermal fibroblasts. The present study was designed to see the effect of this platelet factor on epidermal cells. The growth of cultured epidermal cells and dermal fibroblasts obtained from guinea pig ear skin was observed using 10% platelet-rich serum or platelet-poor serum in the culture medium. These sera were prepared from fresh human blood according to the method described by Ross. The cell growth was checked by counting the number of cells in culture or by the uptake of labeled thymidine by the cells. The platelet-rich serum significantly stimulated the growth of epidermal cells as well as fibroblasts as compared with the platelet-poor serum. This implies that platelet factor released after injury to cutaneous vessels in trauma or inflammation plays an important role in initiating proliferation of epidermal cells and dermal fibroblasts.

Culture of normal human epidermal cells with 3T3 feeders on millipore filters.

Human epidermal cells were cultured with lethally irradiated 3T3 cells and grew as large colonies both on plastic and millipore filters. Subcultures were possible. The grown colonies were studied morphologically and histologically. The whole intact crossview of epidermal colonies in paraffin sections was obtained from the cell culture on millipore filters. The epidermal cells derived from younger persons had higher efficiency of colony formation than those from older ones. The membrane filter was shown to be good substratum for epidermal cell growth as well as plastic.

Effectiveness of a new drug, bredinin, on canine kidney allotransplant survival.

BR has remarkable effectiveness on canine kidney allograft survival. Its effectiveness in suppression of the immune responses is dependent on the dosage. Atrophy of the spleen and the mesenteric lymph nodes is prominent in treated dogs. Septic complications are slight, with no reduction in the number of peripheral white blood cells. Erosive changes of the mucosa in the entire bowel are the major side effects of the drug treatment. Liver functional abnormality has not been observed.

Hydroxyapatite-reactive salivary protein revealed by iso-electrofocusing electrophoresis.

Salivary protein involvement in the formation of acquired enamel pellicle, so far, has been discussed in terms of hydroxyapatite (HA)-reactive salivary proteins only from the parotid gland. This study was undertaken to seek this type of protein in the human whole (mixed) saliva and to investigate its normal and pathological variations. Several kinds of hydroxyapatite, either biogenous or synthesized by solid phase reaction, were used as a powder (250 mesh). HA was incubated with concentrated whole saliva at 25 degrees for 30 min. After centrifugation and filtration salivary proteins were analysed on a Multiphor isoelectrofocusing gel electrophoresis. The control salivary proteins were separated into three major groups; acidic (A1-A8), neutral neutral (N1-N4), and basic (B1-B3) isoelectric point (pI). In the HA incubated sample, one of the major neutral bands (NI) preferentially disappeared at about pI 7.5. This NI band was missing or scarce in the parotid saliva and had an amino acid composition rich in glycine, lysine, serine, glutamic acid, aspartic acid, and histidine. This protein was considered to be one of the major HA-reactive proteins in human whole saliva.

Cyclic alteration of electrophoretic property associated with growth circle in HeLa S3 cells.

Changes of electrical cell-surface charge associated with cell cycle were examined electrophoretically in the synchronous culture of HeLa S3 cells. The electrophoretic mobility of the cells increased abruptly in the mitotic phase as high as 45% compared with that of cells in the S phase. This difference of mobility was obtained by comparing pure population of mitotic cells and S-phase cells. Mitotic cells were also found to be more susceptible to neuraminidase than S-phase cells in term of electrophoretic mobility, a greater amount of sialic acid being correspondingly released from the former. The mechanism of the electrokinetic change associated with the processes of cell cycle is discussed, with special reference to the distribution of electric charge in the cell surface membrane.

The role of prostaglandin E, cyclic AMP, and cyclic GMP in the proliferation of guinea-pig ear skin stimulated by topical application of vitamin A acid.

Daily treatment of guinea-pig ear skin with topical 0.5% retinoic acid in acetone produced erythematous scaly dermatitis. Histologic sections revealed bandlike thickening of the epidermis on days 2 to 4, psoriasiform acanthosis, papillomatosis and increased mitotic activity on days 5 to 6. Also seen were dilatation of the upper dermal blood vessels and a fibroblastic, histiocytic reaction in the dermis. Prostaglandin E, cyclic AMP, and cyclic GMP levels were increased in the treated skin and thymidine incorporation was enhanced. Cyclic AMP and GMP levels peaked on day 5 simultaneous with maximal epidermal hyperplasia, increased mitotic activity and dermal reaction. Tritiated thymidine uptake peaked on days 4 and 5, and prostaglandin E levels continued to increase up to day 6. Cyclic AMP phosphodiesterase activity of treated skin on day 5 did not appear to be significantly different from control.

Epidermal synthesis of prostaglandins and their effect on levels of cyclic adenosine 3', 5'monophosphate.

Extracts of guinea-pig and human skin epidermis were analyzed for prostaglandins PGE1, E2, and F2alpha by radioimmunoassy, and found to contain a total of 62.0 (guinea pig) and 144.7 (human) ng/gm wet weight. the three prostaglandins occurred in approximately equal amounts. Guinea=pig epidermal homogenates converted labeled arachidonic acid to PGE2 and PGF2alpha, the rate of formaiton being 10 and 2.5 pmoles per mg protein in O.K hr, respectively. Conversion in the dermis occurred to a much smaller extent. Homogenates of univolved and involved epidermis from 10 subjects with psoriasis produced PGE2 from arachidonic acid at rates of 6.48 and 2...

Reduced epidermal cyclic AMP accumulation following prostaglandin stimulation: Its possible role in the pathophysiology of psoriasis.

Since prostaglandins are known to be one of the modulators of cyclic AMP, a study of the role of prostaglandins and their relation to the regulation of the nucleotide in psoriasis was initiated. Guinea-pig epidermal preparations were incubated with prostaglandins (PGs) in the presence of theophylline. PGE1 increased cyclic AMP levels 3.5-fold whereas a 2-fold increase was seen with PGE2 and PGA1. The increase in cyclic AMP accumulation was linear with increasing concentration of PGE1, from 10(-11) to 10(-5) M. PGE1 significantly increased cyclic AMP in psoriatic epidermis in vitro; however, the stimulation of cyclic AMP accumulation was significantly less in involved epidermis as compared with uninvolved tissue. The specificity of this stimulation, its occurrence at physiologic levels, and the decreased responsiveness of the cyclic AMP system in psoriatic epidermis to PGE1 stimulation suggest that the altered response of epidermis to PGs may be one of the factors in the pathophysiology of psoriasis.
