Studies on aculeacin. II. Isolation and characterization of aculeacins B, C, D, E, F and G.

Six new antibiotics were isolated as the minor components related to aculeacin A from the culture broth of Aspergillus aculeatus M-4214 and named as aculeacins B, C, D, E, F and G. Their physico-chemical properties were analogous to those of aculeacin A and they showed significant activity against fungi. All of the minor components liberated palmitic acid on alkaline hydrolysis. Amino acid analysis showed that threonine and hydroxyproline are common constituents of aculeacins.

Serum globulin assay of estrogen-binding ability during endocrine treatment of postmenopausal advanced breast cancer.

Estrogen-binding ability of sex hormone binding globulin (SHBG) was assayed in sera of 18 postmenopausal patients with advanced breast cancer during the treatment of diethylstilbestrol by agar gel electrophoresis at low temperature. In eight cases showing high level of SHBG, seven cases (88 per cent) responded to the estrogen therapy. On the contrary, only one case (13 per cent) responded out of eight patients with low levels of SHBG. This result may indicate that the level of estrogen binding ability of SHBG (SEBA) is a useful tool in predicting response to estrogen therapy for breast cancer.

Biosynthesis of adrenodoxin in mouse adrenal tumor cells.

ACTH and cAMP stimulate steroidogenesis and the mitochondrial electron transport system for steroid hydroxylation in cultured mouse adrenal cortex tumor cells. During this stimulation, the biosynthesis of adrenodoxin, a non-heme iron protein which is one of the electron transport enzymes, was examined. 14C-labeled adrenodoxin was isolated by employing a purified rabbit antibody to bovine adrenodoxin. The antibody-adrenodoxin precipitates were further purified by acrylamide gel electrophoresis. It was observed that the biosynthesis of adrenodoxin was stimulated in response to ACTH induction and that this stimulation was completely inhibited with cycloheximide and partially inhibited with chloramphenicol. As a result, it was concluded that adrenodoxin requires both mitochondrial and cytosol ribosomal activities for its synthesis and integration into adrenal mitochondria.
