Reduced spontaneous activity of mice defective in the epsilon 4 subunit of the NMDA receptor channel.

In an attempt to examine the functional significance of the molecular diversity of the N-methyl-D-aspartate (NMDA) receptor channel, we generated mutant mice defective in the epsilon 4 subunit by gene targeting technique. The epsilon 4 subunit mutant mice grew and mated normally. No epsilon 4 subunit protein was detected in the homozygous mutant mice, and the amount of the epsilon 4 subunit protein of 155 kDa was reduced in the heterozygous mice. The expressions of the other NMDA receptor channel subunit mRNAs were not appreciably affected by the mutation. The mutant mice exhibited no obvious histological abnormalities in the various brain regions and in the formation of whisker-related neuronal patterns (barrels, barreloids and barrelettes). In an open field test, however, the epsilon 4 subunit mutant mice showed a reduced spontaneous activity. No significant difference was found between the heterozygous and mutant mice in motor activity and anxiety tests. These results suggest that the epsilon 4 subunit of the NMDA receptor channel plays a role in vivo in controlling the spontaneous behavioral activity.

Hemolysis and heat generation in six different types of centrifugal blood pumps.

What the most causative factor affecting hemolysis is still controversial. To resolve this problem, we investigated the relationship between hemolysis and heat generation in six types of centrifugal blood pumps (Bio-Pump, Delphin, Capiox, Nikkiso, Isoflow, and Toyobo). The analyzed parameters were index of hemolysis in fresh goat blood, pumping performance, and heat generation in a thermally isolated mock circuit. These parameters were analyzed at a flow rate of 5 L/min by changing the pressure head (100 mm Hg and 500 mm Hg). At 500 mm Hg of pressure head, the Bio-Pump needed the highest rotation number and showed the highest hemolytic rate and heat generation. The index of hemolysis is well correlated to heat generation (r2 = 0.721). Heat may originate from the motor by conduction, hydraulic energy loss, and mechanical friction between the shaft and seal. We strongly suspect that hemolysis was caused by a factor such as mechanical friction which generates heat locally.

Expression of a tumor necrosis factor-alpha transgene in murine lung causes lymphocytic and fibrosing alveolitis. A mouse model of progressive pulmonary fibrosis.

The murine TNF-alpha gene was expressed under the control of the human surfactant protein SP-C promoter in transgenic mice. A number of the SP-C TNF-alpha mice died at birth or after a few weeks with very severe lung lesions. Surviving mice transmitted a pulmonary disease to their offspring, the severity and evolution of which was related to the level of TNF-alpha mRNA in the lung; TNF-alpha RNA was detected in alveolar epithelium, presumably in type II epithelial cells. In a longitudinal study of two independent mouse lines, pulmonary pathology, at 1-2 mo of age, consisted of a leukocytic alveolitis with a predominance of T lymphocytes. Leukocyte infiltration was associated with endothelial changes and increased levels of mRNA for the endothelial adhesion molecule VCAM-1. In the following months, alveolar spaces enlarged in association with thickening of the alveolar walls due to an accumulation of desmin-containing fibroblasts, collagen fibers, and lymphocytes. Alveolar surfaces were lined by regenerating type II epithelial cells, and alveolar spaces contained desquamating epithelial cells in places. Platelet trapping in the damaged alveolar capillaries was observed. Pulmonary pathology in the SP-C TNF-alpha mice bears a striking resemblance to human idiopathic pulmonary fibrosis, in which increased expression of TNF-alpha in type II epithelial cells has also been noted. These mice provide a valuable animal model for understanding the pathogenesis of pulmonary fibrosis and exploring possible therapeutic approaches.

The factor structure of the general health questionnaire (GHQ-30) in Japanese middle-aged and elderly residents.

We conducted Goldberg's 30-item General Health Questionnaire (GHQ) on 1,216 Japanese general population aged 40-92. Among them, 9.8% of males and 13.7% of females scored over the cut-off point which is used to indicate minor psychiatric disorders. Factor analysis was carried out using the Likert method and eight factors labelled as follows were selected: depression, anxiety and tension, anergia, interpersonal dysfunction, difficulty in coping, insomnia, anhedonia and social avoidance. The mean value of the standardized scores for each age-sex group indicated that changes in sex-age social roles with age affect the mental health of the general population.

Three-dimensional organization of lymphatics in the dog small intestine: a scanning electron microscopic study on corrosion casts.

Casts of the lymphatics of the canine small intestine were made by direct injection of a low viscosity methacrylate resin into a large lymphatic in the submucosal layer, and observed under a scanning electron microscope. The lymphatics started with rod-like central lacteals in the villi of the jejunum and leaf-like ones in those of the ileum. The bases of the lacteals were connected by slender lymphatics forming a three-dimensional network, tentatively called the "superficial lamina propria lymphatic plexus". From this plexus, a few straight branches descended through the lamina propria to drain into a well developed "deep lamina propria lymphatic plexus", which was extended two-dimensionally closely above the lamina muscularis mucosae. From this plexus, a few short lymphatics extended and penetrated the muscularis mucosae and drained into the "superficial submucosal lymphatic plexus", a coarse mesh work of thick lymphatics. From this plexus, a few slender lymphatics descended to drain into a second lymphatic plexus, called the "deep submucosal lymphatic plexus", which extended two-dimensionally on the circular muscle layer. This deep submucosal plexus was a coarse network of thick knotty lymphatics. A large collecting lymphatic was occasionally seen running through the mesh. The tunica muscularis contained tubular lymphatics extending horizontally parallel to the muscle fiber, both circular and longitudinal.

Familial hypobetalipoproteinemia associated with hypothyroidism.

A 55-year-old Japanese woman with familial hypobetalipoproteinemia associated with hypothyroidism was admitted to the hospital because of orthopnea and congestive heart failure. Thyroid function testing revealed hypothyroidism, but she exhibited low levels of serum cholesterol (111 mg/dl) and apolipoprotein (apo) B (48.5 mg/dl). No abnormal bands were detected with gradient sodium dodecyl sulfate-polyacrylamide gel electrophoresis of very low density lipoprotein, low density lipoprotein and high density lipoprotein followed by immunoblotting with anti apo B polyclonal antibody. In this case, apo B structural abnormality could not be identified.

Ultra-high-resolution scanning electron microscopic studies on the membrane system of the rat parietal cells after tetragastrin stimulation.

The membrane system of the rat parietal cells in the resting state and after stimulation with tetragastrin (gastrin) was examined by ultra-high-resolution scanning electron microscopy after removal of the cytoplasmic matrix by the aldehyde-osmium-DMSO-osmium procedure. The intracellular canaliculus was lined with numerous microvilli. Viewed from the cytoplasmic side, the intracellular canaliculi appeared as an arborized system of cactus-like structures with numerous holes about 100 nm in diameter corresponding to the basal openings of the microvilli. The intracellular canaliculi were more developed after gastrin stimulation than in the resting state. In resting cells, most of the tubulovesicles were isolated, 100-200 nm in diameter, spherical or tubular in shape. After gastrin stimulation, these structures were interconnected by slender tubules of about 30 nm in diameter forming together tubulovesicular network. Stereo SEM views clearly demonstrated that the tubulovesicular network was connected with the intracellular canaliculus by the slender connecting tubulus. The increase in the canalicular membrane area and the depletion of tubulovesicles is explained by the transfer of the tubulovesicular membrane to the intracellular canaliculus. In the resting parietal cell, the microvilli are slender and their interior is packed with microfilaments. After gastrin stimulation, the microvilli are swollen and their interior is edematous. These morphological changes seem to indicate the accumulation of fluid in the microvilli after gastrin stimulation.

[Mental health activities for evacuees of the volcanic eruption of Mt. Unzen-Fugen--crisis intervention of a psychiatrist].

In a regional mental health activity for residents who have been evacuated for over three years on account of the volcanic eruption of Mt. Unzen-Fugen, a crisis intervention of the author took place in 76 people (21 males and 55 females) between June 1992 and November 1994. Most of them were over 41 in both sexes and half of female subjects was over 61. Using ICD-10, 57.9% of total were tentatively diagnosed as having depressive disorders (F32, F34.1, F41.2 and F43.2). Of the 76 cases, 49 people improved with counseling, 22 people received a prescription of psychotropics from general practitioners and 5 people required outpatient care or hospitalization at psychiatric or geriatric institutions. The outcome as of November 1994 was 60 remitted cases including 6 relapse cases, 6 worsened cases and 10 unchanged cases. The case history and the reality of crisis intervention was introduced on some cases and the evacuated people's peculiar psychology, that mental scars and grief after experiencing the disaster are unspoken and kept locked in their hearts, was discussed. On the basis of the above results and discussion, I considered the meaning of visiting the evacuees' shelters and emphasized the importance of the following basic strategies of crisis intervention to evacuated people: 1. to give them counseling so that they can accept their devastating experiences as part of their life, 2. to give support to the whole family, 3. to work in close cooperation with general doctors in cases requiring drug therapy. Furthermore, it was insisted that it is necessary to establish a disaster mental health system to give support to evacuees who need psychiatric help on a long time scale not just immediately after the disaster. Finally, I pointed out the undeveloped level of disaster mental health activities in Japan and referred to future problems.

[Experimental and clinical study on intraoperative local infusion chemotherapy (ILIC) for advanced gastric carcinoma].

An intensive chemotherapy combined with surgery, termed "intraoperative local infusion chemotherapy (ILIC)", was devised. The ILIC procedure is to infuse 50 mg of cisplatin through the feeding artery to the tumor, which has been isolated from blood flow by clamping the stomach and blood vessels, before radical surgery. An experiment in dogs showed considerable free Pt transfer into thoracic lymph after ILIC. Sum total Pt in the thoracic lymph after ILIC (294.5 micrograms/3 h) was 4.3 times that after IV administration (68.3 micrograms/3 h). In ILIC, AUC of free Pt in the thoracic lymph was about 5.8 times that in peripheral blood. ILIC was applied in 24 advanced gastric cancer patients, consisting of 6, 2, 4 and 12 in pTNM stage II, IIIA, IIIB and IV, respectively. Pt concentration of the tissues was high in the tumor (23.22 micrograms/g) and the regional lymph nodes (2.95 micrograms/g) compared to that in the serum (0.45 microgram/ml). The survival rate (Kaplan-Meier) in the ILIC patients was significantly higher than that of control patients (matched pair method) from among patients treated by surgery alone. No fatal complications were encountered in any patient treated by ILIC.

Three-dimensional configuration of crypts of different types of colorectal adenomas.

The three-dimensional configuration of isolated crypts of normal human colonic mucosa and colorectal adenomas was examined by scanning electron microscopy. For isolation of the crypts, the digestion method with HC1 was used for formalin fixed tissues, and the separation method with ethylenediaminetetraacetic acid (EDTA) following ultrasonication was applied to fresh tissues. In a comparative study, the NaOH cell-maceration method, which visualized the sub-basal laminal collagen sheath, was applied. The isolated crypts from the normal colon were visualized as a single straight tubule resembling a test tube. Most isolated crypts of the tubular adenomas were visualized as elongated fan-like structures with several protuberances and a few short branchings. Their average length was more than twice that of the normal colonic mucosa crypts. Most crypts of the villous adenomas were visualized as slender tubules without protuberances and short branchings, and their average length was three times that of the tubular adenoma crypts. Most crypts of the tubulovillous adenomas were long and triangular with several longitudinal folds and protuberances, and the average length was about three times that of the tubular adenoma crypts.

Scanning tunneling microscopic study of osmium-impregnated collagen.

The vapor-phase osmium impregnation method was applied to increase the conductivity of the collagen specimens used in scanning tunneling microscopy (STM), since the application of STM to biological materials has been limited by the poor conductivity, irregular sample geometry, and instability of these materials. The impregnation method was applied to collagen fibrils attached to highly oriented pyrolytic graphite, and the STM images obtained were compared with those of routinely processed uncoated specimens. The osmium-impregnated collagen fibrils showed a periodicity of about 67 nm, which was also observed on the negative-stained specimens by transmission electron microscopy. In addition, the periodicity of 3.1 nm, which corresponded to the helical cycle of the 3 alpha-chains within collagen, was clearly demonstrated by STM. Moreover, the comparative observations of collagen specimens without osmium impregnation disclosed the left-handed helical structure with a periodicity of 3.1 nm in a single collagen filament.

Use of strip biopsy for early duodenal cancer--a case report.

A 67-year-old man was referred for investigation of an abdominal mass. Upper gastrointestinal endoscopy revealed a polypoid lesion with a central depression in the duodenum. CT scan demonstrated a cystic lesion anterior to the pancreas. Examination of a biopsy specimen of the duodenal lesion suggested it was a carcinoma. Strip biopsy was therefore performed, and histological examination revealed a tubular adenocarcinoma with invasion limited to the mucosa, indicating that the lesion was completely resected. Subsequent 24-month follow-up did not indicate recurrence of the cancer. Strip biopsy thus appears to be a safe and efficient procedure for the management of early duodenal cancer.

Phosphorylation of Rabphilin-3A by calmodulin-dependent protein kinase II.

Rabphilin-3A is a putative target protein for Rab3A small GTP-binding protein which is implicated in neurotransmitter release. We have examined here whether Rabphilin-3A is phosphorylated by calmodulin-dependent protein kinase II (CaMKII). Recombinant Rabphilin-3A was phosphorylated by CaMKII purified from rat brain. Two moles of phosphate were maximally incorporated into one mole of Rabphilin-3A. The phosphorylation sites were Ser34, Thr205, Thr209, and Thr537. These results suggest that the CaMKII-catalyzed phosphorylation of Rabphilin-3A may modulate neurotransmitter release.

Involvement of the glutamate receptor delta 2 subunit in the long-term depression of glutamate responsiveness in cultured rat Purkinje cells.

An antisense oligonucleotide against the glutamate receptor delta 2 subunit mRNA, which is selectively expressed only in Purkinje neurons, suppressed the induction of long-term depression (LTD) of glutamate responsiveness in the rat cerebellar culture. LTD of glutamate response is induced by pairing glutamate application and depolarization of a Purkinje cell. Treatment of the culture with the antisense oligonucleotide exerted no appreciable effect on basic physiological and morphological properties of Purkinje cells, except for LTD induction and reduction of delta immunoreactivity which was intense in distal dendrites. Sense and missense oligonucleotides, which were used as controls, did not block LTD induction. These results suggest that the glutamate receptor delta 2 subunit is involved in the cerebellar LTD.

GDP/GTP exchange reaction-stimulating activity of Rabphilin-3A for Rab3A small GTP-binding protein.

Rabphilin-3A is a putative target protein for Rab3A, a small GTP-binding protein particularly implicated in neurotransmitter release. Rabphilin-3A interacts more preferentially with GTP-Rab3A than with GDP-Rab3A. Moreover, Rabphilin-3A shows a weak activity to stimulate the GTPase activity of Rab3A and a strong activity to inhibit the Rab3A GTPase-activating protein (GAP)-stimulated GTPase activity of Rab3A. Here, we show that Rabphilin-3A has another activity to stimulate the GDP/GTP exchange reaction of Rab3A. Rabphilin-3A may keep Rab3A continuously in the GTP-bound form by converting again GDP-Rab3A, which may be converted from GTP-Rab3A by Rab3A GAP, to GTP-Rab3A, until the function of Rab3A is accomplished.

Characterization of two new human apolipoprotein A-I variants: apolipoprotein A-I Tsushima (Trp-108-->Arg) and A-I Hita (Ala-95-->Asp).

Two unrelated subjects with new variants of apolipoprotein (apo) A-I were found during screening with isoelectric focusing (IEF) gel analysis. In the first case, apo A-I Tsushima, sequencing following amplification by the polymerase chain reaction (PCR) revealed a residue 108 missense mutation (TGG-->CGG, Trp-->Arg) in exon 4. The proband of apo A-I Tsushima was heterozygous for this mutation. The second case, apo A-I Hita, revealed a residue 95 missense mutation (GCC-->GAC, Ala-->Asp) in exon 4. The proband of apo A-I Hita was compound heterozygous with apo A-I (Ala-37-->Thr). These two subjects exhibited normal plasma concentrations of apo A-I and HDL cholesterol. In screening normal high school students (n = 198), we used a PCR-mediated site-directed mutagenesis to rapidly detect the substitution of G to A at codon 37 because the apo A-I (GC-->ACC, Ala-37-->Thr) mutation is unrelated to the charge difference on IEF. The frequency of the A allele was 0.04: the substitution G to A at codon 37 did not affect the plasma concentrations of lipids and lipoproteins.
