An immunohistochemical study on the innervation of SP-IR nerves in the cerebral arteries of the bent-winged bat.

The overall distribution of substance P (SP) immunoreactive (IR) nerves surrounding the cerebral arteries of the bent-winged bat were investigated immunohistochemically. In this microchiropteran species, the walls from the vertebral artery to the caudal part of the basilar artery have considerably well-developed plexuses of SP-IR nerves, whereas no demonstrable SP-IR fibers were found in the rostral part of the basilar artery, and in more rostrally located arteries the nerve supply was very sparse or occasionally lacking. This innervation pattern has not yet been established for the cerebral arterial systems of other mammals that have been studied under normal conditions, but it is very similar to the pattern of SP-IR innervation observed in the guinea pig and cat of which the trigeminal ganglia have been destroyed. From the combination of this and other immunohistochemical findings, it is suggested that SP-IR nerves innervating the vertebral and basilar arteries of the bent-winged bat originate from the upper cervical dorsal root ganglia (DRG) and enter the cranial cavity along the vertebral artery and through the meninges.

Malignant fibrous histiocytoma in the thoracic region--a clinico-pathologic investigation.

Seven cases of malignant fibrous histiocytoma (MFH); 3 originating in the thoracic region, one which was considered to arise from the lung parenchyma, and 4 which were metastatic to the lungs, are presented herein. Six of these patients underwent surgical excision and analysis under light and electron microscopy revealed the lesions of MFH to be composed of two cell types; a fibroblast-like cell and a histiocyte-like cell. The latter showed histologically characteristic growth in a so-called storiform pattern. In all patients adjuvant chemotherapy was performed, although in only one patient did it prove temporarily effective. Despite the fact that the prognosis of MFH in the thoracic region is poor, the suggested therapy for longer survival is resection with postoperative combination chemotherapy including the use of sensitive anticancerous agents.

Effect of combination therapy of radiation and local administration of OK-432 on a murine fibrosarcoma.

The effects of a combination therapy of radiation and local administration of OK-432, which is one of BRMs (biological response modifiers), were studied using a radioresistant murine fibrosarcoma with weak immunogenicity. Fifty percent tumor control doses were 83.5 (79.6-87.4) Gy in animals given radiation alone and 64.3 (57.9-71.0) Gy in animals given OK-432 immediately after radiation, indicating that this biological response modifier can enhance the radiation dose effectiveness by a factor of 83.5/64.3, that is, 1.30. Histological observations of treated tumors showed that the combination therapy induced a marked infiltration with lymphocytes and prominent degeneration and necrosis of the tumor cells. Examination of subsets of the infiltrating lymphocytes using the monoclonal antibodies showed that Lyt-2 positive lymphocytes were more abundant in the group given the combination therapy than in the radiation alone group on day 5 (p greater than 0.05). Two days later, Lyt-1, Lyt-2, and L3T4 positive lymphocytes increased in the group given the combination therapy, whereas these lymphocytes disappeared in the group given radiation alone.

Biological effectiveness of fast neutrons on a murine osteosarcoma.

The effect of fast neutrons and gamma rays on a murine osteosarcoma was studied. The NROS tumor, a radiation-induced osteosarcoma in a C3H mouse, was transplanted into the right hind legs of syngeneic female mice and locally irradiated with single or four daily doses of either fast neutrons or gamma rays. The NROS contained 13-30% hypoxic cells. It took approximately 7 days for the NROS tumor to show apparent reoxygenation following gamma ray irradiations. Two assays were used to determine the neutrons' relative biological effectiveness (RBE) to gamma rays: tumor growth delay time and tumor control dose. The largest RBE of 4.5 was obtained at the smallest dose of neutrons examined, followed by a gradual decrease down to 2.3. The tumor growth delay assay indicated that the RBE values of 2.6-3.1 after single doses of fast neutrons increased to 3.1-4.5 after four daily fractions. The 50% tumor control doses were 78.5 Gy and 33.0 Gy after single doses of gamma rays and fast neutrons, resulting in an RBE of 2.3. Fractionated doses increased the RBE to 2.6. Mitotic cells disappeared shortly after irradiation but reappeared 7 days after irradiation.

Concentrations of catecholamines and indoleamines in the central nervous system of Wriggle mouse Sagami.

1. The concentrations of norepinephrine (NE), 3-methoxy-4-hydroxyphenylglycol (MHPG), 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) in the central nervous system of Wriggle mouse Sagami (WMS), which is a new ataxic mutant mouse, were studied. 2. NE and MHPG levels were increased most remarkably in the cerebellum. 3. 5-HT and 5-HIAA levels were increased most remarkably in the brain stem and spinal cord. 4. The present results suggest enhancement of catecholamine and indoleamine metabolism in the cerebellum and bulbospinal cord, respectively, of the WMS, and these changes seem relevant to the specific motor dysfunction of the WMS.

Malignant lymphoma of the posterior mediastinum with transverse myelopathy.

A case of malignant lymphoma in the thoracic paravertebral region with a severe spinal cord compression syndrome is presented. The patient was a 59-year-old female who had had right hypochondralgia as initial symptoms for more than one year. Chest radiograph and computed tomographic (CT) scanning on admission to another hospital had demonstrated a mass lesion in the thoracic paravertebral area. Due to the subsequent occurrence of progressive cord compression syndrome, she was transferred to our institution for the purpose of surgical treatment. Myelography demonstrated an obstruction of the spinal canal at the level of the 8th thoracic vertebra. CT-myelography showed a paravertebral tumour which extended through the intervertebral foramen into the epidural space and also spread to the posterior thoracic wall in succession. Tumour extirpation and decompressive laminectomy by single stage operation were performed. The pathological diagnosis was extranodal malignant lymphoma, diffuse, medium-sized cell type, with immunohistological demonstration of B-cell markers. Postoperatively, she was given a combination chemotherapy with VEPA (vincristine, cyclophosphamide, prednisone and doxorubisin), and the functional disorder has improved to some extent. To our knowledge, this is the first case of soft tissue malignant lymphoma in the posterior mediastinum.

Intrabronchial neurilemmoma--review of cases in Japan.

We recently encountered a case of intrabronchial neurilemmoma. The patient was a 15-year-old boy who had a wheeze. After observation with a bronchofiberscope, a wedge resection of the left main bronchus was performed. No postoperative complications nor recurrence was seen. To date, five cases of intrabronchial neurilemmoma have been reported in the Japanese Journals. Surgical removal was performed in 4 of 5 and bronchofiberscopic excision in 1. This paper summarizes the clinical features of these cases as well as our own case.

Plasma concentrations of atrial natriuretic peptide in children with persistent hypertension.

Plasma concentrations of human atrial natriuretic peptide (hANP) were determined in children with persistent hypertension and in age-matched normotensive controls. We studied 40 children 6-7 years of age (Group A), and 74 children 13-14 years of age (Group B). There was no significant difference in plasma hANP concentration between hypertensives and normotensives in group A. In group B, the plasma hANP concentration in hypertensives (86.5 +/- 44.9 pg/ml: mean +/- SD) was significantly higher than in normotensives (58.8 +/- 40.4 pg/ml) (p less than 0.01). In group B, 24-hour urinary excretion of sodium and fractional excretion of sodium were not significantly different between hypertensives and normotensives. The plasma hANP correlated significantly with the fractional excretion of sodium (p less than 0.05) and the 24-hour urinary excretion of sodium (p less than 0.01) in normotensives. No such correlation was evident in the case of the hypertensives. The pathogenesis of increased plasma hANP concentration in older children with persistent hypertension remains to be investigated. In regard to the effects of hANP on renal function, hANP may contribute to the regulation of sodium handling in normotensive adolescents, whereas other regulatory mechanisms need to be considered in the case of hypertensives.

Perivascular innervation of the mesenteric artery in spontaneously hypertensive rats.

Perivascular innervation of the mesenteric arteries of 7-week-old and 6-month-old spontaneously hypertensive rats and normotensive Wistar-Kyoto rats was examined. The densities of neuropeptide Y-containing nerve fibers and adrenergic nerve fibers were increased in the distal regions of mesenteric arteries of spontaneously hypertensive rats as compared with findings in Wistar-Kyoto rats. However, the densities of cholinergic nerve fibers, vasoactive intestinal polypeptide-containing, and substance P-containing nerve fibers in the mesenteric arteries of the spontaneously hypertensive rats were unchanged in comparison with findings in the Wistar-Kyoto rats. Thus, not only adrenergic nerve fibers but also neuropeptide Y-containing nerve fibers may play an important role in the development and maintenance of hypertension in spontaneously hypertensive rats.

Extracorporeal circulation influence on plasma atrial natriuretic peptide concentration in cardiac surgery patients.

An attempt was made to clarify the change of plasma human atrial natriuretic peptide (hANP) concentration before, during, and after cardiac surgery in heart failure with extracorporeal circulation. Plasma hANP concentration did not significantly decrease during total aortic cross clamping (ACC) with complete clamping of the superior and inferior vena cavae. This finding may be explained by the suppression of endopeptidase activities and the response of hANP receptors due to the low body temperature. Plasma hANP concentration strongly increased from 56.6 +/- 8.4 to 208.9 +/- 40.7 pg/ml (n = 5) by the release of total ACC. This strong increase of hANP in the plasma may occur due to the rapid increase of atrial pressure from zero to 12.5 mm Hg caused by releasing the total ACC. The molecular form of plasma hANP obtained after the release of total ACC was alpha-hANP alone, which was estimated by gel permeation chromatography and reverse HPLC.

Norepinephrine responsiveness in patients with borderline hypertension under three different sodium balances.

The pressor effects of intravenous norepinephrine (NE) infusion (100 and 200 ng/kg/min for 15 min) were examined in 17 patients with borderline hypertension (BHT) and 15 age-matched normotensive subjects (NT) under three different sodium balances; the regular customary diet, treatment with diuretics, and the high-sodium diet. Treatment with diuretics decreased and high sodium diet increased the pressor response to NE in both groups but there were no significant differences in NE reactivity between the groups. The increments in mean blood pressure after NE infusion (200 ng/kg/min) during the three experimental periods correlated significantly with the preinfusion plasma NE concentration in both BHT and NT: r = -0.58 (p less than 0.01) and r = -0.54 (p less than 0.01), respectively. Neither the slopes nor the intercepts differed between the two groups. Thus, evidence presented indicates that BHT do not have increased pressor responsiveness to NE, and that NE pressor response depends upon basal sympathetic tone.

KS-501 and KS-502, new inhibitors of Ca2+ and calmodulin-dependent cyclic-nucleotide phosphodiesterase from Sporothrix sp.

New inhibitors of Ca2+ and calmodulin-dependent cyclic-nucleotide phosphodiesterase, KS-501 and KS-502 were isolated from a fungus, Sporothrix sp. KAC-1985. Inhibitory concentration causing 50% inhibition (IC50) values of KS-501 and KS-502 for bovine brain enzyme were 1.8 and 4.3 microM, respectively. The compounds exhibited no or weak inhibition for calmodulin-independent cyclic-nucleotide phosphodiesterases and protein kinase C.

Calphostins (UCN-1028), novel and specific inhibitors of protein kinase C. I. Fermentation, isolation, physico-chemical properties and biological activities.

A novel complex of calphostin (UCN-1028), which specifically inhibits protein kinase C (PKC) has been isolated from the culture broth of a fungi Cladosporium cladosporioides. Purification of individual components was carried out by silica gel, non-porus resin Diaion HP-20SS and Sephadex LH-20 chromatography, leading to isolation of five closely related components. A, B, C, D and I. Calphostins showed cytotoxic activities against various tumor cells, and these cytotoxicities were proportional to their inhibitory activities against PKC.

KS-504 compounds, novel inhibitors of Ca2+ and calmodulin-dependent cyclic nucleotide phosphodiesterase from Mollisia ventosa.

Novel inhibitors of Ca2+ and calmodulin-dependent cyclic nucleotide phosphodiesterase were isolated from the culture broth of a fungus, Mollisia ventosa KAC-1148. They were designated as KS-504a, KS-504b and KS-504d. A degradative product of KS-504a, called KS-504e, was also isolated and found to inhibit the enzyme. The 50% inhibitory concentration (IC50) values of KS-504a, KS-504b, KS-504d and KS-504e for bovine brain enzyme were 122, 109, greater than 500 and 139 microM, respectively.

[Prediction of radiosensitivity by DNA analysis].

The aim of the study is to investigate the relationship between DNA histogram and radiosensitivity on tumor specimens taken directly from patients. Flow cytometric DNA content analysis with PI (Propidium iodide) was performed on specimens of cervical carcinoma. Radiosensitivities of respective tumors were determined on biopsy specimens that were taken 3 weeks after the start of radiotherapy. Five parameters (over 4C, %G2M, %S, PI, DI) were calculated from DNA Histogram. Among these parameters over 4C, %G2M and PI correlated well to radiosensitivity.
