Peptidomimetic inhibitors of Ras farnesylation and function in whole cells.

The ras protooncogene is involved in regulation of cell growth. Mutations that activate the protein result in uncontrolled cell growth. Ras undergoes a series of posttranslational processing events, the first of which, farnesylation, is crucial for the function of the protein. Inhibitors of the farnesyltransferase enzyme are therefore potential candidates for the development of anticancer drugs. Tetrapeptides have been reported to be good inhibitors of this enzyme in vitro. We have synthesized analogs of the tetrapeptide Cys-Val-Phe-Met by replacement of the amino-terminal amide bonds. One inhibitor, B581, is permeable to the cell membrane. In the cell, it inhibits processing of two farnesylated proteins, H-ras and lamin A, but it does not inhibit processing of a geranylgeranylated protein, Rap 1A. Microinjection of B581 into frog oocytes inhibits maturation induced by activated, farnesylated H-ras but not maturation induced by activated, geranylgeranylated H-ras or by progesterone. These results demonstrate that this peptide mimic inhibits farnesylation selectively in the cell. The inhibition of farnesylation results in inhibition of H-ras function.

[A concept of social support for acute schizophrenic patients. Time series analysis of daily fluctuations of psychotic markers].

Findings of 12 single case studies which documented and analyzed the course of features typical in an acute psychotic episode during a milieu-therapeutic treatment are reported (criteria: DSM-III-schizophrenia). Three different stages in treatment are distinguished (relaxation-activation-integration). Each stage can be described as a decrease in stimulus protection with a concurrent continual increase in psychosocial demands. The purpose of this study is to verify whether stable differences in psychotic characteristics exist between the disparate stages, and whether stage assignment is sensitive to changes in psychotic features. The results of the statistical data analysis according to the ARIMA-intervention-model demonstrate a significant reduction in psychotic features in relation to treatment stages (10 and 8 out of 12 time series respectively). Hypothetical changes were specified as starting abruptly and as being permanent.

Light-induced GTPase activity and GTP[gamma S] binding in squid retinal photoreceptors.

Illumination greatly increases the GTPase activity in homogenates of squid (Loligo) whole retinas or rhabdomeric membranes. Adenylylimidodiphosphate inhibits the light-insensitive (but not the light-sensitive) GTPase activity in these homogenates. Illumination also greatly increases the binding of GTP[gamma S] to the rhabdomeric membranes. This binding at saturating illuminations indicates that there are approximately 10-100 times more rhodopsin molecules than G-protein molecules in squid photoreceptors. Each light-activated rhodopsin molecule activates about 10 G-protein molecules which might provide amplification for the first stage of the phototransduction cascade.

Synthesis of sedoheptulose from non-dialyzable, endogenous substrates in mammalian tissue extracts.

Sedoheptulose was observed to be formed at the rate of 12-120 nmol/g tissue/h in dialyzed rat and bovine tissue homogenates. The compound was identified by its gas-chromatographic retention time and by its mass spectrum. A standard of [14C]-sedoheptulose was prepared from D-[14C]fructose 6-phosphate and D-erythrose 4-phosphate for use in radioactive gas-chromatographic analysis in studies of possible precursors of the sedoheptulose. The reaction was stimulated by NAD+. Evidence is presented to show that non-dialyzable sedoheptulose 7-phosphate may be the endogenous precursor. These results show that care must be exercised when using crude enzyme preparations and extremely sensitive analytical methods so that chromatographically unresolved products formed from non-dialyzable, endogenous substrates of low molecular weight do not introduce significant error in the analysis of the expected product.

[Possibilities for the use of lasers in dental surgery].

The use of the Nd:YAG laser in everyday oral surgery in patients with a hemorrhagic diathesis is reported. It proved successful in various hemorrhagic disorders mild forms of hemophilia A. The most essential including step forward seems to be that this may simplify the so far complicated treatment of this group of patients in a hospital, as it may enable outpatient therapy to be performed and considerably reduce the cost of treatment. In addition, the risk of hepatitis is smaller for all those patients in whom missing clotting factors have had to be substituted up to the present.
