Multivariable conditional analysis for complex trait and its components.

Methods of multivariable conditional analysis were proposed for analyzing contribution of component traits to complex target trait based on mixed linear model approaches. The contribution ratio and the contributed genetic effect were defined. The contribution ratio could measure the proportion of genetic variation of the given component trait on the target trait. The contributed genetic effect could quantify genetic effect of the given component traits contributed to the target trait. A worked example from the cotton data is given to illustrate the application of the new methods for analyzing the contribution of three yield components to lint yield.

[Effects of delayed opening of infarct-related artery on late left ventricular remodeling in patients with acute anterior myocardial infarction].

OBJECTIVE: To assess the effect of delayed opening of the infarct-related artery (IRA) by percutaneous coronary intervention (PCI) on the late left ventricular remodeling after acute anterior myocardial infarction (AAMI). METHODS: Sixty four patients with initial Q-wave AAMI and with the total occluded IRA conformed by angiogram at 9.1 +/- 2.3 (2 - 14) days after the onset were divided into successful PCI group and control group (not receiving PCI or the IRA not re-opened). Two-D echocardiogram was performed at acute phase (about 3 weeks), 2 and 6 months after onset of AAMI respectively to detect the left ventricular function and left ventricular wall motion abnormality (VWMA). The total congestive heart failure events were recorded during 6 months follow-up. RESULTS: VWMA scores, left ventricular ejection fraction (LVEF), left ventricular end-diastolic and end-systolic volume indexes (LVEDVI and LVESVI) were similar in 2 groups at acute phase and 2 months after the onset of AAMI. There were no differences between the parameters above at acute phase and 2 months in each group too. VWMA scores and LVEF did not changed significantly at 6 months in each group compared with those at acute phase and 2 months (P > 0.05). But LVEDVI and LVESVI were significantly smaller in the successful PCI group than those in the control group (P < 0.01, P < 0.05). The rate of congestive heart failure events was 19% in control group and 2.0% in successful PCI group (P > 0.05) respectively. CONCLUSIONS: Delayed opening of IRA in AAMI could prevent the late phase but not the early phase of left ventricular remodeling after AMI.

[Clinical characteristics and prognosis of diffuse large B-cell lymphoma].

OBJECTIVE: To investigate the clinical characteristics of diffuse large B-cell lymphoma (DLBCL) and the factors affecting its prognosis. METHODS: From 1994 to 2002, 138 patients with DLBCL were confirmed by morphological and immunohistochemical examination. Sex, age, clinical stage, performance status (PS), serum lactate dehydrogenase (LDH), number of extranodal lesions, treatment response, cycles of chemotherapy, B symptom, erythrocyte sedimentation rate (ESR), 5-year survival rate and median survival time (mST) were included as the analysis indeces. RESULTS: Lymph nodes were involved in 87.7% of the patients, and extranodal lesions were found in 60.1%. Five-year survival rate was 41.3% for the entire group. Age, stage, PS, serum LDH, number of extranodal lesions, international prognostic index (IPI) and remission rates were significantly correlated with overall survival (OS) and mST (P < 0.05), However, sex, chemotherapy cycles, B symptom, ESR were not related to OS and mST (P > 0.05). Age, stage, remission rates were identified as independent factors affecting the prognosis. Combination of surgery and chemotherapy was quite impressive in the prolongation of survival of patients with extranodal lesions and gastrointestinal lymphoma compared to those by chemotherapy alone. CONCLUSION: Age, stage, PS, serum LDH, number of extranodal lesions, IPI, chemotherapy cycles and remission rates are significant factors affecting the prognosis in DLBCL patients. Age less than 40 years or >/= 65 years, Stage III-IV, partial remission or progressive disease are demonstrated as poor prognostic factors. Combined treatment is the strategy suggested for DLBCL patients with extranodal lesions.

Mutation analysis of HOXD13 gene in a Chinese pedigree with synpolydactyly.

OBJECTIVE: To study the clinical features and to identify homeobox D13 (HOXD13) gene mutation of the affected individuals in a Chinese synpolydactyly (SPD) kindred. METHODS: Clinical data and peripheral blood samples of SPD family members were obtained through field investigation. For every member of this pedigreeìthe fragment containing mutational hot spots of HOXD13 was amplified by PCR for mutation screening. To examine whether there is any other mutation within coding sequence of HOXD13, exon 1 and exon 2 of HOXD13 were also amplified by PCR. All the amplified fragments were electrophoresed on 2% agarose gels and then the mutant fragments were electrophoresed on 5% polyacrylamide gels to be separated. Purified PCR products of normal and selected mutant alleles were directly sequenced. RESULTS: Comparing the HOXD13 coding sequence of the affected individuals with HOXD13 sequence in the GenBank and with that of the unaffected, an inserted segment coding 8 alanine residues within HOXD13 was found segregating with the disorder. This mutation is also termed polyalanine expansion. The 8-alanine expansion can be interpreted as a reduplication of normal alanines 5-12. CONCLUSION: The results suggest that synpolydactyly in this kindred may be caused by polyalanine expansion in HOXD13.

Constructing linkage maps with achiasmatic gametogenesis.

Maximum likelihood (ML) approach is used for estimating recombination frequency based on the achiasmatic model and the corresponding software package is developed for constructing linkage maps for achiasmatic organisms (F2 populations). The detection of sex-linked markers is done through a chi-square test. Monte Carlo simulations were conducted for comparing estimation of recombination frequency and mapping powers between these two genetic models (chiasmatic and achiasmatic models) when the achiasmata occurs. Simulation results showed that the achiasmatic model could provide unbiased estimations, while the chiasmatic model (without correction) gave under-estimates. The powers of grouping and ordering by the achiasmatic model were greater than those by the chiasmatic model (without correction) for all cases. ML approach based on the achiasmatic model can be used without correcting the data to obtain desirable linkage map powers in achiasmatic organisms.

Population genetics for Y-STRs haplotypes of Chinese Ewenki ethnic minority group.

Chinese Ewenki ethnic minority group, with a population of 26,300 in a statistical survey conducted in 2000, has a lifestyle of nomad, and is distributed across seven counties in the Inner Mongolia Autonomous Region and in Nahe County of Heilongjiang Province of China. The Ewenki people have no written script but a spoken language with three dialects belonging to the Manchu-Tungusic group of the Altai language family. Raising reindeers is the key livelihood of Ewenkis.

[The relationship between expression of fas mRNA and apoptosis after traumatic brain injury in rats and the role of GM-1 in protecting the brain].

OBJECTIVE: To explore the mechanism of apoptosis after traumatic brain injury (TBI) in rats and elucidate the role of GM-1 by detecting the expression of Fas mRNA and apoptosis in hippocampi. METHODS: After creating the model of Marmarou cranio-cerebral trauma and offering GM-1 therapy, we observed the expression of Fas mRNA and apoptotic cell death using in situ hybridization and terminal deoxynucleotidyl transferase-mediated dUTP-biotin in situ nick end labeling (TUNEL) technique. RESULTS: Increased expression of Fas mRNA and increased apoptotic cells in hippocampi after TBI were observed. GM-1 could decrease the expression of Fas mRNA and apoptotic cell death. CONCLUSION: The increased expression of Fas mRNA may be a noteworthy cause of apoptotic cell death after traumatic brain injury. GM-1 may play a protective role by way of decreasing the expression of Fas mRNA and apoptotic cell death.

[Bioinformatic analysis of the 14-3-3 gene family in rice].

Using two-step HMM (hidden markov model) scan strategy,eight 14-3-3-like proteins were identified by searching the Oryza sativa L. ssp. japonica protein database. From them four genes were newly detected in this study. We confined the genes expressing in Nipponbare by EST search. Expression analysis also showed each gene expressed diversely within any individual,this tends suggested specific function of particular gene. Alignment of amino acid sequences suggested that there could be isoform function of the specific residues. The analyses of gene structure and chromosome location indicated that rice genome contains both epsilon and no-epsilon forms of 14-3-3 proteins. In addition,we analyzed the evolution of the rice 14-3-3 protein family.

[Dynamic observation of pathological changes and expression of nuclear factor-kappaB in the intestinal mucosa of rats after diffuse brain injury].

OBJECTIVE: To investigate the dynamic pathological changes in the intestinal mucosa of rats and the role of activation of nuclear factor-kappaB (NF-kappaB) in the alteration in intestinal mucosal barrier dysfunction after diffuse brain injury. METHODS: The animal model established by Marmarou was used to produce diffuse brain injury. One hundred and fifty male Wistar rats were randomly divided into nine groups in terms of postinjury time: 1, 2, 4, 8, 12, 24, 48, 72, 168 hours after injury, and control group. Mucosal thickness, villous height and width were estimated under light microscope, and expression of NF-kappaB was assessed with immunohistochemical staining in control group to compare with those in injury groups. RESULTS: Mucosal thickness, villous height and width were diminished and the optical density of expression of NF-kappaB was much higher in the injury groups than those of control group. CONCLUSION: The structure of intestinal mucosa is damaged in the earlier stages after diffuse brain injury. NF-kappaB is activated by cellular stress, which may be involved in the pathogenesis of secondary intestinal lesion.

[Molecular cloning and expression of anti-tumor adhesion peptide (beta3)].

To block tumor cell adhesion, inhibit tumor metastasis and recurrence, the anti-adhesion peptide-trimeric beta peptide (DLYYLMDLSYSMKGGDLYYLMDLSYSMKGGDLYYLMDLSYSMK, beta3) was designed. The DNA fragment of beta3 was cloned into expression vector pET-His and the fusion protein His-beta3 was expressed in E. coli. BL21(DE3)plysS. After 1.5 hours' induction with IPTG, His-beta3 peptide was expressed significantly amounting to 10% of the insoluble proteins and 4% of the total proteins. 20mg of beta3 peptide was obtained from one litter culture medium after purification by using metal-chelating sepharose 6B FF. The purity of beta3 is 92.2% according to Gel-Pro analysis. The anti-adhesion effects of beta3 peptide, beta1 peptide (DLYYLMDLSYSMK) and GRGDS on the hepatocellular carcinoma cell line SMMC-7721 and the high metastasis hepatocellular carcinoma cell line HCCLM6 were studied. The result showed the beta3 blocked the adhesion of HCCLM6 cells and SMMC-7721 cells to fibronectin (FN) specifically. The inhibition effect was dose-dependent and time-dependent and the inhibition rate of beta3 was higher than three times concentration of beta1 and GRGDS. This suggested that pET-His-beta3/BL21(DE3)plysS was a suitable expression system for beta3, and the expressed beta3 specially inhibited the adhesion of cancer cells.

[An epidemiological investigation on gastroschisis in China during 1996 to 2000].

OBJECTIVE: To study the epidemiological characteristics of gastroschisis in China from 1996 to 2000. METHODS: The birth defects monitoring program was undertaken in 450-466 hospitals from 31 provinces, cities and autonomous regions of China. Data about births including intrauterine death and stillbirth of 28 weeks of gestation was collected to report through the medical records over the time period 1996 to 2000. The maximal diagnosis time was up to 1 week after the delivery. The international Classification of Diseases of Ninth Revision with some modifications was used to code the birth defects. RESULTS: A total of 569 cases with gastroschisis was found in 2,218,616 births monitored. The overall birth incidence of gastroschisis was 2.56 per 10,000 births during the monitored period. The increasing trend of incidence of gastroschisis hadn't been found (P > 0.05). The birth incidence of gastroschisis in rural areas was significant higher than that in urban areas (3.93 vs.1.98 per 10,000, P < 0.01). There was also significantly difference on birth incidence between female and male (2.64 vs. 2.15 per 10,000 births, P < 0.05). The birth incidence of gastroschisis in the mothers less than 20 years old was the highest among all mothers age, which were at 5.4 times increased risk relative to that of mother age of 25-29 years old. The overall perinatal mortality rate of gastroschisis was 74.69 percent, and 41.46 percent of gastroschisis were diagnosed before birth. 27.44 percent of cases were accompanied with other major defects. CONCLUSIONS: There was no increasing trend on the incidence of gastroschisis to be found in China during 1996 to 2000. The mother age less than 20 years old was at an increasing risk of fetal gastroschisis. The cases with gastroschisis had a high perinatal mortality rate.

[Analysis of under 5 years old children mortality and the leading death cause in China from 1996 to 2000].

OBJECTIVE: To study the trend of under 5 years old children mortality and the leading cause of the deaths in China from 1996 to 2000. METHODS: The data presented in this report were obtained from the national child mortality surveillance network, including 116 counties (cities) throughout China. The target population was all children under 5 years old in the monitored areas whose mothers or fathers had resided in the area for at least one year. The data were collected and reported by health workers at the three-level network. RESULTS: The neonatal mortality rate (NMR), infant mortality rate (IMR) and under 5 years old mortality rate (U(5)MR) in China dropped to 22.8, 32.2, 39.7 per 1,000 live births in 2000, respectively (they were 24.0, 36.0, 45.0 respectively in 1996), which declined 5.0%, 10.6%, 11.8% from 1996 to 2000, respectively. In urban areas, NMR, IMR and U(5)MR dropped to 9.5, 11.8, 13.8 per 1,000 live births in 2000, respectively (they were 12.2, 14.8, 16.9 respectively in 1996), which declined 22.1%, 20.3%, 18.3% from 1996 to 2000, respectively. In rural areas, NMR, IMR and U(5)MR dropped to 25.8, 37.0, 45.7 per 1,000 live births in 2000, respectively (they were 26.7, 40.9, 51.4 respectively in 1996), which declined 3.4%, 9.5%, 11.1% from 1996 to 2000, respectively. There was a steady decline in the U(5)MR due to diarrhea, pneumonia, neural tube defects and drowning in China. CONCLUSION: In urban/rural areas, the overall decline in NMR, IMR and U(5)MR from 1996 to 2000 was spectacular. Especially the U(5)MR due to avoidable deaths such as pneumonia and diarrhea was dropped markedly in rural areas.

[Passive remote sensing of VOC in atmosphere by FTIR spectrometry].

The paper expatiates the passive remote sensing experiment of VOC in atmosphere by FTIR spectrometry, a mode of simulated spectra for pollution gas in a complicated environment is proposed, and the detecting limit of passive FTIR remote sensing is discussed. Using this measurement technique and the data analysis method, the authors can obtain column density and effective radiation temperature of target-gas that does not require a previously measured background spectrum.

[Genetic contribution of agronomic traits to yield in flue-cured tobacco (Nicotiana tabacum L.)].

In order to understand the genetic contribution of six agronomic traits to yield, 14 flue-cured tobacco varieties (or breeding lines) and their 41 F1 crosses were used for multivariable conditional analysis. The contribution of additive variance of plant height to yield was larger than other agronomic traits. The largest contribution of dominant variance to yield was due to the length of middle leaves. All agronomic traits investigated had small contribution to yield due to additive x environment interaction effects and dominant x environment interaction effects. No identical trait of different parents showed the largest contribution to additive effect of yield. This could be resulted from the fact that each parent had its own genetic and developmental characterization. The dominant effects of yield were mainly influenced by length of middle leaves in most crosses. Length of middle leaves could be served as ameasurement to indirectly select the cross parent having high dominant effect of yield.

[Comparison between R-CHOP regimen and CHOP regimen in treating naive diffuse large B-cell lymphoma in China--a multi-center randomized trail].

BACKGROUND & OBJECTIVE: CHOP regimen is a standard treatment for patients with diffuse large B-cell non-Hodgkin's lymphoma (NHL), and its 5-year overall survival (OS) rate is 30%-40%. Rituximab is a chimeric monoclonal antibody (MoAb) directly against CD20-positive B cells, and has good effect on diffuse large B-cell NHL. Rituximab combined with standard chemotherapy has been approved for treating aggressive B-cell NHL in Europe and the US. This study was to determine efficacy and safety of the combination of Rituximab and CHOP regimen in treating Chinese patients with CD20-positive diffuse large B-cell NHL. METHODS: From Sep. 2003 to Nov. 2004, a total of 63 patients in 9 centers were enrolled. All the patients were randomized into 2 groups: 32 received CHOP regimen alone (CHOP group), and 31 received Rituximab and CHOP regimen (R-CHOP group). All patients signed informed consent. The complete response rates, overall response rates, and side events of the 2 groups were compared. RESULTS: The complete response rates were similar in R-CHOP and CHOP groups (41.9% vs. 37.5%, P=0.719); the overall response rates were slightly higher in R-CHOP group than in CHOP group (83.8% vs. 65.6%, P=0.096). Disease progression during treatment was reported for 7 (21.9%) patients in CHOP group and 1 (3.2%) patient in R-CHOP group (P=0.026). The occurrence rates of adverse events were similar in R-CHOP and CHOP groups (65.6% vs. 67.7%, P=0.859). The most common adverse event was leukopenia; fever and chills were rather common in R-CHOP group. Clinically relevant toxicity was similar in both groups. CONCLUSION: When compared with standard CHOP alone, the addition of Rituximab to standard CHOP regimen reduces the risk of treatment failure in patients with diffuse large B-cell NHL, and doesn't increase the occurrence of chemotherapy-related adverse events.

[Study on ultrasound-assisted decolorization of methyl orange with Fenton reagent].

Employed with ultrasound, Fenton reagent, ultrasound-Fenton methods, performance and mechanism of decolorization and COD removal were investigated for methyl orange. Results indicated that little decolorization phenomenon was observed with ultrasound solely. For 400mg/L methyl orange solution, 67.51% decolorization ratio and 63.15% COD removal ratio were obtained with Fenton reagent for 30 min. Combined the same operation conditions with ultrasonic field, decolorization ratio and COD removal ratio are 86.91% and 80.32%, respectively. Ultrasound and Fenton reagent have synergistic action on degradation of methyl orange solution. It is oxidative reaction for methyl orange solution with Fenton reagent in strong acidic condition, and it is reduction process for the degradation when pH>5.
