[Expression of intestine-specific transcription factor CDX2 in different subtypes of intestinal metaplasia and gastric carcinoma].

BACKGROUND & OBJECTIVE: Intestinal metaplasia (IM) is thought as the precancerous lesion of gastric carcinoma, and CDX2 gene plays important roles in development and differentiation of intestinal epithelium, and maintenance of intestinal phenotype. Recent studies found that CDX2 were expressed aberrantly in IM of chronic atrophic gastritis (CAG) and some gastric carcinomas, which implied that CDX2 may play an important role in IM formation and gastric carcinogenesis. This study was to investigate the roles of CDX2 in the development and progression of IM and gastric carcinogenesis, and determine the correlation of IM to gastric carcinogenesis. METHODS: A tissue microarray containing 46 cases of CAG with IM, 40 cases of gastric carcinoma, and 32 cases of IM foci in paracancerous tissues was constructed. High iron diamine/alcian blue (HID/AB) and HE staining were used to classify IM and gastric carcinoma, and the expression of CDX2 protein and mRNA in different gastric lesions was assessed with immunohistochemistry and in situ hybridization, respectively. RESULTS: The proportion of type III IM was significantly higher in IM foci in paracancerous tissues than in CAG with IM (56.25% vs. 21.74%, P<0.01). The positive rates of CDX2 protein were 69.56% in IM foci in CAG, 53.13% in IM foci in paracancerous tissues, and 42.50% in gastric carcinomas, and the positive rates of CDX2 mRNA were 63.04%, 46.87%, and 35.00%, respectively. The positive rates were significantly lower in gastric cancer than in IM in CAG (P<0.01), but there was no significant difference between gastric cancer and IM foci in paracancerous tissues (P>0.05). The expression of CDX2 protein and mRNA was significantly higher in intestinal-type gastric cancer than in diffuse-type gastric cancer (54.55% vs. 27.78%, 45.45% vs. 22.22%, P<0.05). The expression of CDX2 protein was significantly lower in type III IM than in type I IM (46.42% vs. 79.31%, P<0.05). CONCLUSIONS: CDX2 may play important roles in the development and progression of IM and gastric carcinogenesis.

[Sited-directed mutagenesis of hCu, Zn-SOD gene and its expression in Synechococcus sp. PCC7942].

The Cys111 genetic code of human copper/zinc superoxide dismutase (hCu, Zn-SOD) gene in the pESOD plamid was mutated into the Ala111 code with site-directed mutagenesis, and then the plamid pESODT111 which contained groESL promoter, mutated hCu, Zn-SOD gene, rbcS-polyA terminator and reporter gene (Kanr) was constructed and transduced into Synechococcus sp. PCC7942 with homologous recombination platform. The results of PCR and DNA sequence analysis showed that the target nucleotide had been genetically integrated into genome DNA of the host cell. SDS-PAGE, Western blot and Pyrogallol autoxidation assay confirmed that the transformant strains expressed the mutated hCu, Zn-SOD protein. And the level of the mutated hCu, Zn-SOD protein reached a value of 3.61% of the total soluble protein. Furthermore, the transformants still retained 95% activities of SOD after 30 minutes at 80 degrees C environment, it indicated that the mutated hCu, Zn-SOD protein could endure higher temperature than the natural one.

[Experimental study on repairing damage of corneal surface by mesenchymal stem cells transplantation].

OBJECTIVE: To investigate the survival, migration and differentiation of the cultured human mesenchymal stem cells derived from bone marrow (hMSCs) after transplanted onto the alkaline burn rabbit cornea. METHODS: Alkaline burn on rabbit corneas was induced with NaOH solution. One month later, hMSCs cultured with a feeding of amniotic membrane were transplanted onto the surface of alkaline burn rabbit corneas. Amniotic membrane alone transplantation was used at the same time as control group. One month after transplantation, the changes of corneal morphology were analyzed by clinical observations and HE staining. Immunohistochemistry was carried out with the antibodies against human nuclei and cytokeratin 12 to investigate the distribution and differentiation of hMSCs. RESULTS: The rabbit cornea became totally opaque one month after alkaline burn. Blood vessels could be seen within the superficial layer and stroma. The surface of cornea was rough and dry. There were many goblet cells found in the corneal epithelial layer. One month after transplantation of hMSCs, the surface of alkaline burn cornea became smoother. The amount of the new blood vessels of the cornea reduced and the goblet cells disappeared. Anti-human nuclei antibody staining showed positive cells on the surface layer and superficial stroma while cytokeratin 12 positive cells were only present in the epithelium layer. In the amniotic membrane transplantation control group, no clinical improvement of the cornea was found. Goblet cells were still seen on the corneal surface. Both anti-human nuclei antibody and cytokeratin 12 staining were negative. CONCLUSIONS: hMSCs survive and migrate to stroma after transplanted onto the surface of alkaline burn rabbit cornea. No rejection is detected. hMSCs on the corneal surface differentiate to corneal epithelium, where those migrated to the stroma differentiate to cells other than epithelium. hMSCs transplantation decrease corneal conjunctivization after alkaline burn.

[Preparation and characterization of monoclonal antibody against Vibrio cholerae O139].

AIM: To prepare monoclonal antibody (mAb) against Vibrio cholerae O139, which would be used as the gold colloidal reagent strip for rapid detection of O139, and to determine its biological characterization. METHODS: BALB/c mice were immunized by inactivated Vibrio cholerae O139. Anti-O139 mAbs were prepared by using hybridoma technique. The specificity of mAb was determined by indirect ELISA and Western blot. The indirect ELISA was used to identify Ig subgroup, detect its titer in ascites and relative affinity, and analyze antigen-binding epitope of mAbs. RESULTS: Two hybridoma cells (O4D7 and O4D10), secreting anti-O139 mAbs were obtained. The Ig subgroups of O4D7 and O4D10 were IgG2b and IgG3, respectively. The mAb's titer in ascites was 1:10(7). The relative affinity of mAb O4D7 was more than 10(5) and that of O4D10 was more than 10(4). The result of additive ELISA showed that two mAbs could recognize different antigen epitopes. CONCLUSION: The successful preparation of two anti-O139 mAbs provides a powerful tool for the development of a rapid method of detecting Vibrio cholerae O139 infection.

[Quantification study of bone adaptive model based on experiment of rapid-growing rats in different stress environment].

Presented is a method to set up a quantification model of bone growing and remodeling adaptation, which integrates animal experiments, parameter identification of mathematical functions and technique of computer simulation. By designing a new animal experiment, we investigate the effects of growing and remodeling of the rat femurs in different stress environments, gather the bone mineral density (BMD) of proximal femur in the same interval for the unknown parameter (B and K) inversion of bone growing and remodeling equation and create the femur three-dimensional geometrical model based on CT images. The model in this paper can not only numerically measure the relation between outer stimulus and the femur BMD variation of rapid growing rats, but also predict the growth trend of rat femur under different stress environments in its whole lifecycle. The thought and method of creating the model in this paper can be used for reference to modeling human bone growth and remodeling.

[Spectra analysis of [MnHg(SCN)4 (H2O)2 x 2C4H9NO (MMTWD) crystals].

The factor group symmetry analysis method was used to predict lattice vibrational modes of MMTWD single crystal, and it was found that there are no more than 297 and 148 theoretically observable Raman peaks and infrared reflection bands, respectively. The Raman and IR spectra were measured at room temperature. All the Raman and IR spectra peaks were assigned on the basis of the group theory and lattice dynamics theory. According to the Raman and IR spectra, the molecular structure of MMTWD crystal was discussed. From the results of the present theoretical and experimental works, the three-dimensional network structure of MMTWD crystal was identified. This structure provides the crystal with a large interaction force between molecules, which in turn induces high stability and macroscopic nonlinear optical properties.

[Error permissibility of neural network used for renal corpuscle area enhancement].

In the automatic analysis system of the kidney-tissue image, boundary enhancement for glomerulus area is a vital step. Complex characteristics of kidney-tissue image leads to the difficulty in boundary features description. This paper suggests a kind of feature template under the special boundary definition. A nonlinear threshold surface is constructed by neural network, then the proper surface can be selected to enhance boundary with the influence of error permissibility being taken into account. Experimental results indicate that this learning method with error permissibility can enhance the boundary of glomerulus and suppress noises at the same time, so it can obtain good processed effects and have a fine performance highly adaptive to various sample images.

[The relationship between histone acetylation modification and IFN-gamma responsive gene regulation].

OBJECTIVES: To study the role of histone modification in the regulation of IFN-gamma-activated gene using chromatin immunoprecipitation technique. METHODS: Real-time PCR was used to measure the mRNA level of interferon-gamma-inducible protein 10 (IP-10) in Hela cells. Chromatin immunoprecipitation combined with Real-time PCR was used to check the histone H4 acetylation level at IFN-stimulated response element (ISRE) locus of IP-10 gene. RESULTS: IP-10 was strongly activated by IFN-gamma. The histone H4 deacetylation happened at the ISRE locus when IP-10 was induced by IFN-gamma. The activation of IP-10 and the deacetylation of histone H4 at the ISRE site induced by IFN-gamma were inhibited or blocked by histone deacetylase (HDAC) inhibitor trichostatin A (TSA). CONCLUSION: The histone H4 deacetylation at the ISRE site is related with the activation of IP-10 by IFN-gamma.

[Expressions of plasma SDF-1 and its receptor CXCR4 in peripheral blood mononuclear cells of children with Kawasaki disease].

OBJECTIVE: This study examined the expressions of plasma stromal cell derived factor-1 (SDF-1) and CXCR4 mRNA in peripheral blood mononuclear cells (PBMC) of children with Kawasaki disease (KD) and aimed to explore the significance of SDF-1 and CXCR4 mRNA in KD. METHODS: Fifty-six children with KD (12 cases complicated by coronary artery lesions) and 60 age and gender-matched healthy children (normal controls) were enrolled in this study. Plasma SDF-1 levels and CXCR4 mRNA expression in PBMC were measured using ELISA and real-time quantitative PCR at the acute and convalescence stages of KD. RESULTS: Plasma SDF-1 levels (1833 +/- 395 ng/L vs 1126 +/- 408 ng/L; P < 0.05) and the CXCR4 mRNA expression in PBMC (6.57 +/- 2.81 vs 2.58 +/- 1.01; P < 0.01) in KD patients were significantly higher than those in normal controls at the acute stage. Both plasma SDF-1 levels and CXCR4 mRNA expression in KD patients decreased significantly at the convalescence stage, but nevertheless remained higher than those in the normal controls. The patients with concomitant coronary artery lesions showed higher CXCR4 mRNA levels than without at the acute stage (8.19 +/- 2.39 vs 6.13 +/- 2.77; P < 0.05). CONCLUSIONS: Plasma SDF-1 concentration and CXCR4 mRNA expression in PBMC increased in KD patients. CXCR4 mRNA might be involved in the development of coronary artery lesions in KD.

[Immune response induced by combinant immunization of different HBsAg vaccines in mice].

AIM: To investigate the specific humoral and cellular immune response induced by prime-boost immunization of HBsAg protein vaccine (P), recombinant vaccinia virus vaccine (V) and DNA vaccine (D) in mice. METHODS: Groups of BALB/c mice were primed by one of the three vaccines P, V or D and boosted by another vaccine at 2, 5, 8 and 11 week later, thus 9 immune combinations were made: PP, PV, PD, VP, VV, VD, DP, DV and DD. Serum samples were collected at week 2, 5, 8 and 11 and levels of anti-HBsAg IgG antibodies and their sub-isotypes were determined. Seven days after every boost, spleen cells of vaccinated mice were separated and the specific CTL lysis ratio of P815S cells were determined. RESULTS: Among the three HBsAg vaccines P, V and D, V could induce the quickest humoral immune response. The memory humoral immune response induced by P was the strongest. D induced the weakest antibody titer. The ratio of specific IgG1/IgG2a indicated that antibody induced by PP was more polarized to Th2. The other groups induced balanced Th1/Th2 immune response. Among all the groups, VD and DV induced the strongest CTL response, and the specific lysis ratio of P815S cells was 71% and 64%, respectively. CONCLUSION: The experimental results suggested that among all the immune combinations, PV, PD, VP and VD can induce better humoral immune response while DV and VD can induce stronger CTL response in BALB/c mice.

[Preliminary study on the conditions of solid-phase screening phage antibody library].

AIM: To explore the conditions of solid-phase screening phage antibody library and to provide the experimental basis for the design of screening project. METHODS: Diverse antibodies including HEV NE2-specific and non-specific humanized phage antibodies were used to study the screening conditions, such as the binding time of phage antibodies to antigen, the concentration of coating antigen, the washing times and elution method. RESULTS: The best binding time of positive phage antibody to antigen was 1 min. The highest positive rate of screening was obtained under the conditions of washing for 20 to 30 times and pH value of the washing solution being 5. The concentration of the coating antigen had no obvious influence on the positive rate of screening. Higher positive rate was obtained by using 10 mg/L antigen to competitively elute for 1 h. CONCLUSION: Solid-phase screening of the phage antibody library is a very complex process, in which there are close relationship between the conditions, therefore, appropriate readjustment should be made for screening conditions according to concrete conditions.

[Application of continuous renal replacement therapy in the rescue of MODS patients].

OBJECTIVE: To investigate the effect of continuous renal replacement therapy (CRRT) on patients with emergency and serious diseases. METHODS: The patients were divided into 2 groups: Group A [71 patients with multiple organ dysfunction syndrome (MODS)] were treated by CRRT for 175 times. Group B (30 patients) were treated with common hematodialysis (HD). Blood urea nitrogen (BUN), serum creatinine (Scr), natrem (Na(+)), kalium (K(+)), chlorine (Clj), power of hydrogen (pH), bicarbonate (HCO3-), carbon dioxide partial pressure (PCO(2)), and oxygen pressure (PO(2)) were measured before and after the treatment. RESULTS: After the treatment, all patients' general state of health, water-electrolyte and acid base disorder were improved. The levels of BUN, Scr, K(+) (All P(s)<0.01), and Na(+) (P<0.05 in Group A, P<0.01 in Group B) were all lower and the levels of CO(2)CP (P<0.01 in Group A, P<0.05 in Group B) were higher than that before the pretherapy in both Group A and Group B. There was significant difference on the levels of BUN, Scr, K(+)(All P(s)<0.01), Cl(-) and CO(2)CP (All P(s)<0.05) between Group A and Group B, which indicated the therapeutic effect in Group A was better than that in Group B. Furthermore, the levels of pH, PO(2) and HCO(3)(-) were higher (P<0.01) and PCO(2) was lower (P<0.05) obviously than that before the CRRT in 30 patients with acute respiratory distress syndrome (ARDS) in Group A. Among the 9 patients with poisoning, 7 were alleviated completely and 2 died after the CRRT plus hemoperfusion (HP). The survival time of patients was lengthened in Group A. No blood or bleed aggravation occurred in patients with bleeding inclination or 48 h before the operation after using the method of heparin soaking the blood filter. CONCLUSION: CRRT has better curative effect on removing the toxin effectively, rapidly correcting the turbulence of water electrolyte and sour-alkali balance for patients compare with HD. At the same time, it can increase the level of PO(2) and reduce CO(2) retention, improve the lung function and brain edema, and increase the patient's survival rate. Using CRRT as early as possible is very important and it can improve the prognosis of the patients who had MODS or other dangers because the death rate of MODS patients is very high.
