Molecular conformation dictates signaling module formation: example of PKCepsilon and Src tyrosine kinase.

Our laboratory has conducted multiple functional proteomic analyses to characterize the components of protein kinase C (PKC)epsilon cardioprotective signaling complexes and found that activation of PKCepsilon induces dynamic modulation of these complexes. In addition, it is known that signal transduction within a complex involves the formation of modules, one of which has been shown to include PKCepsilon and Src tyrosine kinase in the rabbit heart. However, the cellular mechanisms that define the assembly of PKCepsilon modules remain largely unknown. To address this issue, the interactions between PKCepsilon and Src were studied. We used recombinant proteins of wild-type PKCepsilon (PKCepsilon-WT) and open conformation mutants of the kinase (PKCepsilon-AE5 and PKCepsilon-AN59), the regulatory and catalytic domains of PKCepsilon, along with glutathione-S-transferase (GST) fusion proteins of Src (GST-Src) and two domains of Src (GST-SH2 and GST-SH3). GST pulldown assays demonstrated that Src and PKCepsilon are binding partners and that the interaction between PKCepsilon and Src appears to involve multiple sites. This finding was supported for endogenous PKCepsilon and Src in the murine heart using immunofluorescence-based confocal microscopy and coimmunoprecipitation. Furthermore, PKCepsilon-WT and GST-Src interactions were significantly enhanced in the presence of phosphatidyl-L-serine, an activator of PKC, indicating that Src favors interaction with activated PKCepsilon. This finding was confirmed when the PKCepsilon-WT was replaced with PKCepsilon-AE5 or PKCepsilon-AN59, demonstrating that the conformation of PKCepsilon is a critical determinant of its interactions with Src. Together, these results illustrate that formation of a signaling module between PKCepsilon and Src involves specific domains within the two molecules and is governed by the molecular conformation of PKCepsilon.

Formation of protein kinase C(epsilon)-Lck signaling modules confers cardioprotection.

The epsilon isoform of protein kinase C (PKCepsilon) is a member of the PKC family of serine/threonine kinases and plays a critical role in protection against ischemic injury in multiple organs. Functional proteomic analyses of PKCepsilon signaling show that this isozyme forms multiprotein complexes in the heart; however, the precise signaling mechanisms whereby PKCepsilon orchestrates cardioprotection are poorly understood. Here we report that Lck, a member of the Src family of tyrosine kinases, forms a functional signaling module with PKCepsilon. In cardiac cells, PKCepsilon interacts with, phosphorylates, and activates Lck. In vivo studies showed that cardioprotection elicited either by cardiac-specific transgenic activation of PKCepsilon or by ischemic preconditioning enhances the formation of PKCepsilon-Lck modules. Disruption of these modules, via ablation of the Lck gene, abrogated the infarct-sparing effects of these two forms of cardioprotection, indicating that the formation of PKCepsilon-Lck signaling modules is required for the manifestation of a cardioprotective phenotype. These findings demonstrate, for the first time to our knowledge, that the assembly of a module (PKCepsilon-Lck) is an obligatory step in the signal transduction that results in a specific phenotype. Thus, PKCepsilon-Lck modules may serve as novel therapeutic targets for the prevention of ischemic injury.

U.S. national trends in labor induction, 1989-1998.

OBJECTIVE: To examine the epidemiology of labor induction in the United States. STUDY DESIGN: We used U.S. natality data from 1989 to 1998 and examined the rate of labor induction by year, geographic region, maternal characteristics and pregnancy complications. RESULTS: Between 1990 and 1998, the rate of labor induction increased from 9.5% to 19.4% of all births nationwide. However, the induction rate varied widely by state. White race, higher education and early initiation of prenatal care were associated with a higher rate of induction. For all gestational ages, a significantly increased induction rate occurred during the study period. The increase for clinically indicated induction was significantly slower than the overall increase, suggesting that elective induction has risen much more rapidly. CONCLUSION: The rate of induction of labor more than doubled in the U.S. nationwide in the decade from 1989 to 1998. The increased use of labor induction may be attributable to both clinically indicated and elective induction.

Retrospective analysis of percutaneous transluminal coronary angioplasty and coronary stenting.

OBJECTIVE: To examine long-term efficacy of percutaneous transluminal coronary angioplasty (PTCA), coronary stenting and to assess the factors affecting its efficacy. METHODS: A total of 790 patients who underwent successful PTCA and PTCA + stent in this hospital were followed by direct interview or letter. The rate of follow-up was 84.2% and the period of follow-up was 0.9 - 12.7 (3.5 +/- 2.4) years. RESULTS: During follow-up, 4 (0.5%) patients died, 22 (2.8%) had nonfatal acute myocardial infarction, 10 (1.3%) had coronary artery bypass surgery, and 98 (12.4%) had repeat PTCA. The rate of recurrent angina pectoris was 31.1%. The cardiac event-free survival rate calculated by the Kaplan-Meier method was 88.2% at 1 year and 80.6% at 12.7 years. Cox regression analysis showed that there was a positive correlation between AMI history, stent implantation and the risk of cardiac events, and there was a negative correlation between the number of diseased arteries and the risk of cardiac events. Compared to the PTCA group, patients with PTCA + stent had significantly lower rates of total cardiac events. CONCLUSION: The long-term efficacy of PTCA, especially PTCA + stent in Chinese patients was very satisfactory, suggesting that PTCA + stent therapy should be the major treatment for revascularization in patients with coronary heart disease.

[Diagnosis, treatment and prognosis of thymoma: analysis of 116 cases].

OBJECTIVES: To study the diagnosis and treatment of thymoma and to assess prognosis factors. METHODS: The clinical data on 116 patients with thymoma were collected. A retrospective analysis was performed by comparison of their survival rates computed by the actuarial method and rate of recurrence and metastasis. RESULTS: Chest radiograph was used chiefly for the preoperative diagnosis of thymoma; myasthenia gravis (MG) (25.0%, 29/116) was the most common paraneoplastic disease. An extensive and radical resection was carried out to reduce the recurrence rate of thymoma with stage I and stage II (chi(2) = 4.941 P = 0.0219). The survival time was prolonged by postoperative radiotherapy and chemotherapy. A strong correlation was noted between the clinical stage and histologic subtype of M-H classification, by which the invasive behavior of thymoma was predicted (r = 0.385, P = 0.007). The 3-, 5-, and 10-year survival rates were 81.2%, 67.9% and 40.5%, respectively. Statistical analysis showed a significant negative correlation between stage and survival rate (r = -0.897, P = 0.0000). CONCLUSION: The prognosis of thymoma depends mainly on the histologic subtype, clinical stage and multimodality treatment rather than paraneoplastic diseases.

[Ecological effect of pendimethalin on soil microbe].

The effect of pendimethalin, a herbicide, on microbial activity in soil was studied here. The results showed that the amount of emission CO2 from three treatment of 1, 5 and 10 micrograms.g-1 of pendimethalin had no significant differences from the CK. The study of the influences on the degradation of the cellulose demonstrated that pendimethalin can stimulate the cellulose degradation in the soil. In contrast to the CK, the half-life (T0.5) of 5, 10 micrograms.g-1 treatments were increased to 1.32 times and 2.14 times, respectively. The activities of dehydrogenase were activated in some degrees, and the higher the pendimethalin concentration the greater and longer the influence. The results also showed that the soil urease activity was restrained, and then, was stimulated by pendimethalin. The level and the sustained time of inhibition and stimulation was proportional to the concentration of the treatment.

[Tagging and mapping of QTLs controlling lint yield and yield components in upland cotton (Gossypium hirsutum L.) using SSR and RAPD markers].

Using interval mapping and marker simple regression methods, the QTLs of yield and its components in (Simian 3 x TM-1) F2 and F2:3, were tagged and Mapped with 39 SSR and 10 RAPD markers having polymorphism between parents screened from 301 pair SSR primers and 1040 RAPD primers. Simian 3 is being grown extensively in Yangtze River cotton-growing valley characterized as high productivity with more bolls and higher lint percent, whereas TM-1, Genetic standard in Upland cotton with more heavy boll weight. In the present report, two QTLs controlling boll size with 18.2% and 21.0% phenotype variance explained in F2:3 generation, one QTL controlling lint percent with 24.9% phenotype variance explained in F2 generation and 5.9% in F2:3 generation and one QTL controlling 100-seed weight with 15.6% phenotype variance explained in F2:3 generation were mapped in Chromosome 9. Additionally, another QTL responsible for 100-seed weight was identified and mapped at the same position in Chromosome 9 in F2:3 generation. It is worth for further to be studied whether it is one QTL for pleiotrophism or two closely linked QTLs. The molecular markers mapped and tagged closely with main QTLs of yield traits in this paper can be used for MAS in cotton high-yield breeding program.

[The study of aggregate of the ORF2 peptide of hepatitis E virus expressed in Escherichia coli].

A fragment of hepatitis E virus open reading frame-2(ORF2), located from amino acid residues 394 to 604, was expressed in E. coli. The recombinant protein NE2 was found to form homodimer mostly in SDS-PAGE, which can be dissociated to monomers when treated with urea, and it was recognized more strongly in its dimeric form than the monomer by HEV reactive human serum in Western blotting. Besides, many aggregated form of NE2 from dimer to at least hexamer can be seen in MALDI-TOF-MS. And when the hydrated dynamic semidiameter of NE2 moleculars in PBS was measured as about 4 nm by Dynamic Light Scattering (DLS), being equal to tetramer, but with high polydispersity, which suggested that the NE2 moleculars were existed in PBS in many different sizes. These results suggested that the recombinant NE2 can aggregate into several oligomer forms, the association in the dimer is most strong, and dimers can assemble further to form some super-structure.

[Preliminary study of apoptosis of human hepatocarcinoma cell line HepG2 induced by topotecan].

BACKGROUND & OBJECTIVE: Topotecan, a semisynthetic water-soluble derivative of camptothecin, is a potent inhibitor of DNA topoisomerase I and cell cycle specific antitumor agent; The incidence rate of hepatocarcinoma ranks the third in all types of malignant tumor in China. The clinical curative effect of the present treatments and drugs for hepatocarcinoma are not so satisfactory. The inducing effect of topotecan on apoptosis of hepatocarcinoma cell line HepG2 and its cytotoxicity on HepG2 were studied in this paper. METHODS: Microculture tetrazolium assay (MTT), HE staining, transmission electron microscope (TEM), flow cytometer (FCM) and immunohistochemistry were employed to determine and observe the apoptosis of HepG2 induced by topotecan and its lethal action on HepG2. RESULTS: Topotecan killed HepG2 cell by inducing apoptosis. Topotecan had strong anti-HepG2 activity in vitro, suggesting distinct dose- and time-dependent relationship. The drug IC50 was about 95 micrograms/L. Topotecan block HepG2 in S-phase, then further initiate apoptosis program. The morphologic characteristic of apoptosis showed that it is a typical interphase apoptosis process: the nuclei showed chromatin pycnosis, and clustered on the inner border of karyotheca, cytoplasm condensed, many vacuoles occurred in it. The expression of bcl-2 were not affected in the course of HepG2 apoptosis induced by topotecon (P > 0.05). CONCLUSION: Topotecan had significant cytotoxicity on HepG2 by inducing apoptosis.

[Effects of chronic alcohol intake on serum glucose and insulin of rats].

In order to study the effects of chronic alcohol intake on serum glucose, insulin and glucose tolerance test (GTT), a oral administration of alcohol solution (6 ml/kg.d) in different concentration (10%, 30% and 50%) is given to rats for 60 days. The results showed that the fast serum glucose of rats in the groups orally administrated with 30% and 50% alcohol solution increases obviously in comparison with that in control group. There exist abnormalities in 1 hour, 2 hour and, 3 hour GTT (P < 0.05) in these groups and the difference in gender in both groups is statistically significant. It is also shown that the level of serum insulin of rats decreased along with the increase of alcohol dosage and is significantly lower in the group of rats administered 50% alcohol solution than that in the normal control group (P < 0.05). These results demonstrated that chronic alcohol intake causes the increases of fast serum glucose and abnormality of glucose tolerance test (GTT), and likely, the final abnormality of glucose metabolism in which the decrease of insulin secretion might be one of the contributing factors to the pathogenesis.

[Acute aging model induced by gamma-ray irradiation].

In order to develop a quick and convenient model of acute induced-aging, gamma-ray (3.0 Gy/time for 4 min 53 s for 5 days) is used to irradiate a group of rats, and galactose (40 mg.kg-1.d-1 for 40 days) is injected into a group of mice. At the end of the experiment, the activities of superoxide dismutase (SOD) and content of malondiadehycle (MDA) in the serum of experimental animals are assayed, showing that the gamma-ray irradiation leads to the reduction of SOD activity and an increase in MDA content in the serum of the rats, while galactose injection leas to the reduction of SOD activity and an increase in MDA content in the serum of the mice. The results from this investigation indicate that an acute aging model can be induced by gamma-ray irradiating on rats.

Giant electric field modulation of double exchange ferromagnetism at room temperature in the perovskite manganite/titanate p-n junction.

We report on the electrical modulation of double exchange ferromagnetism at room temperature in hole-doped manganites of a metal oxide p-n junction. In this (La0.9Ba0.1)MnO(3)/Nb doped SrTiO3 p-n junction, the temperature dependence of the junction resistance shows a metal-insulator transition whose temperature, corresponding to that of ferromagnetic transition, is hugely modulated from 290 to 340 K by a bias voltage increasing from +1.0 to +1.8 V. The magnetoresistance can also be modulated electrically.

Analysis of in vivo patterns of caspase 3 gene expression in primary hepatocellular carcinoma and its relationship to p21(WAF1) expression and hepatic apoptosis.

AIM:To detect the expression of caspase 3 gene in primary human hepatocellular carcinoma (HCC) and investigate its relationship top21(WAF1) gene expression and HCC apoptosis.METHODS:In situ hybridization was employed to determine caspase 3 and p21(WAF1) expression in HCC.In situ end-labeling was used to detect hepatocytic apoptosis in HCC.RESULTS:Twenty-one of 39 (53.8%) cases of HCC were found to express caspase 3 transcripts, while 46.2% of HCC failed to express caspase 3.Non-cancerous adjacent liver tissues showed more positive caspase 3(87.5%, 7/8) as compared with HCC (p < 0.05). The expression of caspase 3 is correlated with HCC differentiation, 72.2% (13/18) of moderately to highly differentiated HCC showed caspase 3 transcripts positive, while only 38.1% of poorly differentiated HCC harbored caspase 3 transcripts (italic>p < 0.05). No relationship was found between caspase 3 expression and tumor size or grade or metastasis, although 62.5% (5/8) of HCC with metastasis were caspase 3 positive and a little higher than that with no metastasis (51.6%, p> 0.05). Expression of caspase 3 alone did not affect the apoptosis index (AI) of HCC. The AI was 7.12 in caspase 3 positive tumors (n = 21), while in caspase 3-negative cases (n = 18) 6.59 (italic>p > 0.05). Expression of caspase 3 clearly segregated with p21(WAF1) positive tumors as compared with p21(WAF1) negative cases (16 of 23, 69.6% versus 5 of 16, 31.3%) with statistical significance (p = 0.017).In the cases with positive caspase 3 and negative p21(WAF1), the AI was found slightly higher, but with no statistical significance, than that with expres-sion of p21(WAF1) and caspase 3 (7.21 vs 6.98 , p>0.05).CONCLUSION:Loss of caspase 3 expression may contribute to HCC carcinogenesis, although the expression of caspase 3 does not correlate well with cell apoptosis in HCC.p21(WAF1) may be merely one of the inhibitors which can reduce caspase 3 mediated cell apoptosis in HCCs.

[Curative Effect of Lishengsu on Leukopenia and Infection after Chemotherapy in Tumor Patients]

To examine the effect of Lishengsu (rhG-CSF) on the recovery of leukopenia and control of infections after chemotherapy, twenty tumor patients were observed. When blood WBC decreased to less than 2.5 x 10(9)/L and ANC less than 1.5 x 10(9)/L after chemotherapy, Lishengsu was administered in a dosage of 150 micro g/day for 5 days or until WBC and ANC recovered to normal level. The changes of hemogram, body temprature, the signs of oral mucous membrane and lung were examined. The recovery of WBC and ANC in the Lishengsu group was much faster than that in the control group. The difference between the two groups was statistically significant (P < 0.01). The duration of fever was shorter and the healing of oral ulcers was faster in Lishengsu group than that in the control group (P < 0.05). The results indicated that Lishengsu in a low dosage and a short administration period could enhance the recovery of WBC and ANC efficiently, shorten the duration of fever, quicken the recovery process of oral ulcers and reduce the needs for supporting treatment in tumor patients after chemotherapy.

A critical examination of the sensitivity of unidimensional subscales derived from the Hamilton Depression Rating Scale to antidepressant drug effects.

Unidimensional subscales for assessment of major depression may be more sensitive to antidepressant drug effects than the Hamilton Depression Rating Scale (HAM-D). To further examine this possibility, we analyzed pooled data from eight comparable, well-controlled clinical trials of venlafaxine and compared such subscales and the 17-item HAM-D (HAM-D(17)) based on effect size and number of patients required for 80% power. Symptoms of depression were assessed using the HAM-D among intent-to-treat patients (2045) randomly assigned to receive venlafaxine (immediate release, n = 474; extended release, n = 377), one of several selective serotonin reuptake inhibitors (SSRIs) (n = 748), or placebo (n = 446) for up to 8 weeks. With SSRIs or venlafaxine vs. placebo, subscales yielded effect sizes (0.328-0.528) 16 to 76% larger than the HAM-D(17) did (0.237 and 0.396, respectively), and required 31 to 64% fewer patients for 80% power. With venlafaxine vs. SSRIs, the subscales showed no advantage over the HAM-D(17); all devices yielded comparable, positive effect sizes (0.183-0.195). Final subscale scores significantly predicted (all P < 0.05) whether patients met criteria for remission (eg, HAM-D(17) score of < or = 7). These findings suggest that unidimensional subscales are more sensitive to antidepressant drug effects than the HAM-D(17) is, but only in active agent/placebo comparisons. Our data further suggest the subscales can predict the presence of remission. Given these findings, prudent use of these subscales may be appropriate, cost-effective, and informative.

Synaptic organization of GABAergic amacrine cells in the salamander retina.

The synaptic organization of GABA-immunoreactive (GABA-IR) amacrine cells in the inner plexiform layer (IPL) of salamander retina was studied with the use of postembedding immuno-electron microscopy. A total of 457 GABA-IR amacrine synapses, with identified postsynaptic elements, were analyzed on photomontages of electron micrographs covering 3,618 microm2 of the IPL. GABA-IR amacrine synapses were distributed throughout the IPL, with a small peak at the proximal margin of sublamina a. The majority of the output targets (81%) were GABA(-) neurons. Most of the contacts were simple synapses with one postsynaptic element identified as a process of an amacrine cell (55%), bipolar cell (19%) or ganglion cell (26%), and serial synapses were very rare. Of the 89 postsynaptic bipolar terminals, 63% participated in a reciprocal feedback synapse with the same presynaptic GABA-IR amacrine profile. There appeared to be no preference between GABA-IR amacrine contacts with rod- or cone-dominated bipolar cells (9.1% vs. 8.9%) or in the total number of amacrine synapses in sublaminas a and b (52% vs. 47%). The preponderance of amacrine cell input to bipolar cells in the OFF layer was derived from GABA-IR cells. These findings provide ultrastructural support to the existing physiological studies regarding the functional roles of the GABAergic amacrine cells in this species. Our results have added to the data base demonstrating that, in contrast to mammals, GABA-IR amacrine cells in amphibians and other nonmammals contact other amacrine cells more frequently, suggesting greater involvement of GABAergic amacrine cells in modulating lateral inhibitory pathways.
