Cryopreservation of organs by vitrification: perspectives and recent advances.

The cryopreservation of organs became an active area of research in the 1950s as a result of the rediscovery of the cryoprotective properties of glycerol by Polge, Smith, and Parkes in 1949. Over the ensuing four decades of research in this area, the advantages of vitrification, or ice-free cryopreservation, have become apparent. To date, experimental attempts to apply vitrification methods to vascularized whole organs have been confined almost entirely to the rabbit kidney. Using techniques available as of 1997, it was possible to vitrify blood vessels and smaller systems with reasonable success, but not whole organs. Beginning in 1998, a series of novel advances involving the control of cryoprotectant toxicity, nucleation, crystal growth, and chilling injury began to provide the tools needed to achieve success. Based on these new findings, we were first able to show that an 8.4M solution (VMP) designed to prevent chilling injury at -22 degrees C was entirely non-toxic to rabbit kidneys when perfused at -3 degrees C and permitted perfusion-cooling to -22 degrees C with only mild additional damage. We next investigated the ability of the kidney to tolerate a 9.3M solution known as M22, which does not devitrify when warmed from below -150 degrees C at 1 degrees C/min. When M22 was added and removed at -22 degrees C, it was sometimes [corrected] fatal, but when it was perfused for 25min at -22 degrees C and washed out simultaneously with warming, postoperative renal function recovered fully. When kidneys loaded with M22 at -22 degrees C were further cooled to an average intrarenal temperature of about -45 degrees C (about halfway through the putative temperature zone of increasing vulnerability to chilling injury), all kidneys supported life after transplantation and returned creatinine values to baseline, though after a higher transient creatinine peak. However, medullary, papillary, and pelvic biopsies taken from kidneys perfused with M22 for 25min at -22 degrees C were found to devitrify when vitrified and rewarmed at 20 degrees C/min in a differential scanning calorimeter. It remains to be determined whether this devitrification is seriously damaging and whether it can be suppressed by improving cryoprotectant distribution to more weakly perfused regions of the kidney or by rewarming at higher rates. In conclusion, although the goal of organ vitrification remains elusive, the prospects for success have never been more promising.

Judging egocentric distance on the ground: occlusion and surface integration.

On the basis of the finding that a common and homogeneous ground surface is vital for accurate egocentric distance judgments (Sinai et al, 1998 Nature 395 497-500), we propose a sequential-surface-integration-process (SSIP) hypothesis to elucidate how the visual system constructs a representation of the ground-surface in the intermediate distance range. According to the SSIP hypothesis, a near ground-surface representation is formed from near depth cues, and is utilized as an anchor to integrate the more distant surfaces by using texture-gradient information as the depth cue. The SSIP hypothesis provides an explanation for the finding that egocentric distance judgment is underestimated when a texture boundary exists on the ground surface that commonly supports the observer and target. We tested the prediction that the fidelity of the visually represented ground-surface reference frame depends on how the visual system selects the surface information for integration. Specifically, if information is selected along a direct route between the observer and target where the ground surface is disrupted by an occluding object, the ground surface will be inaccurately represented. In experiments 1-3 we used a perceptual task and two different visually directed tasks to show that this leads to egocentric distance underestimation. Judgment is accurate however, when the observer selects the continuous ground information bypassing the occluding object (indirect route), as found in experiments 4 and 5 with a visually directed task. Altogether, our findings provide support for the SSIP hypothesis and reveal, surprisingly, that the phenomenal visual space is not unique but depends on how optic information is selected.

Structure-based preliminary analysis of immunity and virulence of SARS coronavirus.

The research on SARS-associated coronavirus (SARS-CoV) has not stopped since its discovery, but the pathogenesis of SARS is still unclear. To explore the possible molecular mechanisms of the invasion and virulence of SARS-CoV, we investigated the structural basis of the viral proteins using computational biology. Forty-five motifs relating to superantigens, toxins and other bioactive molecules were detected in the proteins of SARS-CoV. The results showed that the distribution of the motifs varied in different proteins. Enzyme-like motifs were located in the R protein, while ICAM- 1-like and toxin-like molecules were located in the spike, envelop, nucleocapsid, PUP1, PUP 2 and PUP 4 proteins. Comparison of SARS-CoV with other viruses (OC43, PEDV, HRSV, HHerpV and HAdenoV) showed that each group of motifs was different for each type of virus. Data suggest that the proteins of SARS-CoV with toxic motifs might play crucial roles in targeting host cells and interfering with the immune system. This study provides new information for drug and vaccine design, as well as therapeutic strategies against SARS.

Gene expression of early hypertrophic scar tissue screened by means of cDNA microarrays.

BACKGROUND: Hypertrophic scar is an excessive healing response that often follows thermal injury. The most outstanding morphologic change is the overdeposition of collagen, which is caused by imbalance between synthesis and metabolism of collagen. Previous studies also found that transforming growth factor-beta was the key factor controlling scar formation. However, neither anti-transforming growth factor-beta nor other methods could completely control scar formation and contraction. This fact suggests a multifactorial cause. Fortunately, cDNA microarray throws light on the general alteration at the gene level, and thus could allow us to find some new clues for understanding scar formation and contraction. METHODS: In this article, we report the results obtained from the scanning of gene expression of hypertrophic scar by means of cDNA microarray. Five cases of early human postburn hypertrophic scars were selected. Total tissue RNA was extracted from each hypertrophic scar sample and the corresponding uninjured region skin tissue; mRNA was further purified by Oligotex and then was reversely transcribed to cDNAs with the incorporation of fluorescent dUTP to prepare the hybridization probes. The mixed probes were hybridized to the cDNA microarray containing 4,096 genes on a type of chemical material-coated glass slide. After high-stringent washing, the hybridized slides were scanned for fluorescent signal detection. Then, the expression and distribution of cytoskeletal genes such as alpha-smooth muscle actin (alpha-SMA) gene; fibroblast tropomyosin TM30(pl) gene; vimentin gene; profilin gene; and BM40 gene of hypertrophic scar at 3, 6, 9, and 12 months age were further quantitatively studied by in situ hybridization or immunohistochemistry. RESULTS: Our data indicated that there were 94 genes overexpressed and 3 genes down-regulated in early postburn hypertrophic scar. These altered genes were related to proto-oncogenes, apoptosis, immune regulatory genes, cytoskeletal elements, metabolism, and so forth. We also found that the detected cytoskeletal gene expression was much more intense at all time points than the control group. Consistent with clinical observation, cytoskeletal genes reached a peak at an early stage and gradually decreased. CONCLUSION: Our study implied that multiple genes are involved in scar formation and contraction. Interferon is an autosecreted cytokine that might be responsible for self-control of overgrowth of cells in wounds. The early period of hypertrophic scar formation might be a good time for preventing overgrowth and contraction of hypertrophic scar by gene therapy.

Molecular expression of a cysteine proteinase of Clonorchis sinensis and its application to an enzyme-linked immunosorbent assay for immunodiagnosis of clonorchiasis.

We produced a recombinant cysteine proteinase of Clonorchis sinensis and tested its value as an antigen for serologic diagnosis of C. sinensis infections. The predicted amino acid sequence of the cysteine proteinase of C. sinensis was 58, 48, and 40% identical to those of cathepsin L cysteine proteinases from Paragonimus westermani, Schistosoma japonicum, and Fasciola hepatica, respectively. Western blotting analysis showed that sera from patients infected with C. sinensis strongly reacted with the recombinant protein and that sera from patients infected with S. japonicum weakly reacted with the recombinant protein. Antibody against the recombinant protein stained proteins migrating at about 37 and 28 kDa in C. sinensis adult worm crude extracts. Immunostaining revealed that the cysteine proteinase of C. sinensis was located in the intestinal epithelial cells of the adult parasite and in intrauterine eggs. The specificity and sensitivity of the recombinant antigen or C. sinensis adult worm crude extracts were assessed by an enzyme-linked immunosorbent assay (ELISA) using serum samples from humans infected with different parasites, including 50 patients with clonorchiasis, and negative controls. The sensitivities of the ELISA with the recombinant antigen and C. sinensis adult worm crude extracts were 96 and 88%, respectively. The specificities of the ELISA with the recombinant antigen and C. sinensis adult worm crude extracts were 96.2 and 100%, respectively. The results suggested that the recombinant cysteine proteinase-based ELISA could provide a highly sensitive and specific assay for diagnosis of clonorchiasis.

The influence of St John's Wort on CYP2C19 activity with respect to genotype.

Induction of cytochrome p450 isozymes is the major cause for clinical drug interactions of St. John's wort. The relationships of St. John's wort to cytochrome p450 isoforms have been fully investigated, but its effect on CYP2C19 is lacking. Thus, the aim of the present study was to observe the effect of St. John's wort on CYP2C19 activity using CYP1A2 as a control. Twelve healthy adult men-6 extensive metabolizers of CYP2C19 (2C19(*)1/2C19(*)1) and 6 poor metabolizers (4 2C19(*)2/2C19(*)2 and 2 2C19(*)2/2C19(*)3)-were enrolled in a two-phase, randomized, crossover manner. All subjects took a 300-mg St. John's wort tablet or placebo three times daily for 14 days, and then the activities of CYP2C19 and CYP1A2 were measured using mephenytoin and caffeine. It was found that St. John's wort treatment significantly increased CYP2C19 activity in CYP2C19 wild-genotype subjects, with urinary 4'-hydroxymephenytoin excretion raised by 151.5% +/- 91.9% (p = 0.0156), whereas no significant alteration was observed for CYP2C19 poor metabolizers. Repeated St. John's wort administration did not affect the CYP1A2 phenotypic ratio for both CYP2C19 genotype subjects. In conclusion, St. John's wort is an inducer to the human CYP2C19, and clinicians should pay great attention when St. John's wort is added to or withdrawn from an existing drug regimen containing substrates for such enzymes.

[Inhibition of 8-chloroadenosine on proteasome activity of HL-60 cells].

BACKGROUND & OBJECTIVE: The proteasome is a multicatalytic proteinase complex in eukaryotic cells, and it can be used as a new target for screening anti-tumor drugs according to certain previous researches. The inhibition activity of 8-CA (8-chloroadenosine, a new anti-tumor drug synthesized in our lab) in sarcoma 180, hepatocarcinoma 22, and human gastric carcinoma xenograft tumor has been confirmed. However, the relationship between the mechanism of the inhibition and proteasome is still unclear. The present study was designed to explore the effect of 8-CA on 3 kinds of enzyme activities (chymotrypsin-like activity, trypsin-like activity, and peptidyl-glutamyl- peptide hydrolyzing activity) of 20S proteasome in human progranulocyte leukemia HL-60 cells. METHODS: HL-60 cells were treated with 8-CA at different concentrations for 24, 48, and 72 hours. Then total protein of HL-60 cells was extracted, and the three enzyme activities of 20S proteasomes in the total protein was examined. And the chymotrypsin-like activity of the purified proteasome from the total protein was also examined by immunoprecipitation. The enzyme activity was indicated by fluorescence absorbance (A) produced by degradation of specific substrates by proteasomes. RESULTS: In HL-60 cells, three enzyme activities of the proteasomes were significantly inhibited after exposure to 0.1 micromol/L 8-CA for 48 hours; and the inhibition effect was concentration dependent. When the concentration of 8-CA reached 5 micromol/L, the inhibition rates of three proteasome activities (chymotrypsin-like activity,trypsin-like activity, and peptidyl- glutamyl-peptide hydrolyzing activity) were 44.96%, 54.52%, 48.36% for 48 hours and 67.53%, 70.48%, 64.08% for 72 hours, respectively. The inhibition rate of 1 micromol/L and 5 micromol/L 8-CA on chymotrypsin-like proteasome activity of HL-60 cells for 48 hours were 68.14% (P< 0.01) and 92.75% (P< 0.001), respectively. CONCLUSION: 8-CA inhibits proteasome activity of HL-60 cells with a time- and concentration-dependent manner.

Effect of Maotai liquor on the liver: an experimental study.

BACKGROUND: Epidemiology investigation showed that no worker drunk Maotai liquor for nearly 30 years died of hepatic diseases, and no obvious hepatic fibrosis and cirrhosis were found in 99 workers who had drunk Maotai liquor for a long period by epidemiology investigation and needle biopsy. The same finding was detected in rats that were drunk by Maotai liquor continued for 56 days. This study was to investigate the effects of Maotai liquor on the liver and its mechanism of preventing hepatic fibrosis. METHODS: After ingestion of Maotai for 56 consecutive days, male SD rats were killed for detecting the levels of metallothionein and malondialdehyde (MDA) in liver tissues. Rat hepatic stellate cells (HSCs) and human HSCs were cultured in vitro to observe the effect of Maotai on HSCs proliferation and collagen synthesis. After ingestion of Maotai for 14 consecutive weeks, the livers of male SD rats were harvested for pathohistological examination. RESULTS: The level of metallothionein in the liver of Maotai-induced rats increased by 22 folds, whereas the levels of hepatic lipid peroxide and MDA was decreased significantly (P<0.05) in Maotai-induced animals suffering from CCl4. Maotai demonstrated obvious inhibitory effect on proliferation of HSCs and the inhibition was concentration-dependent. Gene expression and protein secretion of collagens could also be inhibited by Maotai. In alcoholic group, typical liver cirrhosis was observed. In Maotai group, however, though fatty degeneration of hepatocytes and mild fibrosis of the interstitium were observed, no obvious hepatic fibrosis and cirrhosis were found. CONCLUSION: It might be an important mechanism of interfering the progress of hepatic fibrosis that Maotai increases the level of metallothionein in the liver and inhibits the activation of HSCs and the synthesis of collagen proteins.

[Effect of Astragalus polysaccharides in promoting neutrophil-vascular endothelial cell adhesion and expression of related adhesive molecules].

OBJECTIVE: To explore the detoxication and tissue generation effect of Astragalus (As) in wound healing and its relation with inflammatory reaction, through observing the effect of Astragalus polysaccharides (AP) on neutrophil-endothelial cell adhesion and expression of related adhesive molecules. METHODS: Human polymorphonuclear leucocyte (PMN) or human umbilical vein endothelial cell (HUVEC) was treated separately with AP, AP plus interleukin 1 (IL-1) and tumor necrosis factor (TNF) to study the effect of AP on PMN adhesion with HUVEC by rose bengal staining, and that on expression of superficial adhesive factor by means of Cell-ELISA and APAAP method. RESULTS: When AP acted on HUVEC, it could significantly promote the adhesion of HUVEC with PMN, while when AP acted on PMN, the adhesion would not increase. When HUVEC was treated by AP plus IL-1, the IL-1 induced PMN adhesion with HUVEC could be strengthened, and the expression of HUVEC superficial adhesive factor ICAM-1 induced by IL-1 and TNF was strengthened also, but when PMN treated with AP, it showed no effect on the expression of adhesive factor CD18. CONCLUSION: AP promotes the adhesion between neutrophil and endothelial cell by way of promoting the expression of superficial I-CAM-1 on surface of endothelial cells, so as to improve the inflammatory reaction in the wound healing course, it possibly is one of the biological bases of the detoxication and tissue generation effects of AP.

[The induction of differentiation into dendritic cells from HL-60 cells by calcium ionophore].

AIM: To explore whether the promyelocytic leukemia cell line HL-60 may differentiate into activated dendritic cells (DCs) by A23187, a calcium ionophore. METHODS: The HL-60 cells were cultured in common medium alone or with various concentrations of A23187 (25-1 600 microg/L) and rhGM-CSF (100 microg/L). After culture for 24-96 hours, the cellular morphological change was observed under light microscope and electron microscope. Surface makers on treated HL-60 cells were analyzed by flow cytometry. The proliferation of allogeneic human T cells was detected by MTT colorimetry. RESULTS: Under the condition of a suitable dose (200 microg/L) of A23187 treatment of HL-60 cells for 24 hours, the expression of CD83 molecule, a characteristic marker on DCs, was highest. The typical dendritic outgrowth appeared at a time when HL-60 cells were treated with A23187 for 72 hours. However, when HL-60 cells were treated with A23187 for 96 hours, the expressions of CD80 (B7.1), CD86 (B7.2), MHC-class II molecule and CD54 on HL-60 cells reached peak, and marked activation of allogeneic T cells occurred. CONCLUSION: Calcium ionophore A23187 can induce the HL-60 cells to differentiate into activated DCs-like cells.

[Prognosis of three-dimension change at middle facial soft tissue with distraction osteogenesis].

PURPOSE: To evaluate the changes in middle facial soft tissue caused by distraction osteogenesis. METHODS: A three-dimensional finite element model was established based on the simulation of a 13-year-old female with skeletal class III malocclusion. Three approaches of osteotomy, namely Le Fort I, Le Fort II and Le Fort III, were simulated and the maxillary complex was advanced by 5mm in the direction of FH plane, functional occlusal plane and functional occlusal plane with deviation of counter-clockwise 10 degrees, respectively, some premeters indicating the contour of the facial soft tissue were evaluated. RESULTS: The three kinds of osteotomy in which different levels of osteotomied maxillary complex area were advanced. Changing in detail is that the nose tip moved forward 1.25 mm, upper lip moved forward 4.9 mm, and the angle of Cm-Sn-U1 tended to increase after Le Fort I osteotomy; and nose tip moved forward 4.9 mm after Le Fort II osteotomy without increase of the angle of Cm-Sn-U1; besides changes that happened after Le Fort II osteotomy, lower eyelid moved forward 2mm after Le Fort III osteotomy. CONCLUSION: By computer surgery simulation using three-dimensional finite element model, we can obtain not only the amount of the three-dimensional change of the facial soft tissue, but also the three-dimensional effected graphics after operations.

Transplantation tolerance mediated by regulatory T cells in mice.

BACKGROUND: With potent suppressive effect on responder T cells, CD(4)(+)CD(25)(+) regulatory T (Treg) cells have become the focus of attention only recently and they may play an important role in transplantation tolerance. However, the mechanism of action is not clear. This study was designed to assess the possibility of using CD(4)(+)CD(25)(+) Treg cells to induce transplantation tolerance and to investigate their mechanism of action. METHODS: CD(4)(+)CD(25)(+) Treg cells were isolated using magnetic cell separation techniques. Mixed lymphocyte reactions were used to assess the ability of Treg cells to suppress effector T cells. Before skin transplantation, various numbers of CD(4)(+)CD(25)(+) Treg cells, which have been induced using complex skin antigens from the donor, were injected into the host mice either intraperitoneally [0.5 x 10(5), 1 x 10(5), 2 x 10(5), 3 x 10(5), 4 x 10(5), or 5 x 10(5)] or by injection through the tail vein [5 x 10(3), 1 x 10(4), 2 x 10(4), 5 x 10(4), 1 x 10(5), 2 x 10(5)]. Skin grafts from two different donor types were used to assess whether the induced Treg cells were antigen-specific. The survival time of the allografts were observed. Single photon emission computed tomography was also used to determine the distribution of Treg cells before and after transplantation. RESULTS: Treg cells have suppressive effect on mixed lymphocyte reactions. Grafts survived longer in mice receiving CD(4)(+)CD(25)(+) Treg cell injections than in control mice. There was a significant difference between groups receiving intraperitoneal injection of either 2 x 10(5) or 3 x 10(5) CD(4)(+)CD(25)(+) Treg cells and the control group (P < 0.05, respectively). Better results were achieved when Treg cells were injected via the tail vein than when injected intraperitoneally. The transplantation tolerance induced by CD(4)(+)CD(25)(+) Treg cells was donor-specific. Analysis of the localization of Treg cells revealed that Treg cells mainly migrated from the liver to the allografts and the spleen. CONCLUSIONS: CD(4)(+)CD(25)(+)Treg cells can induce donor-specific transplantation tolerance. Cell-to-cell contact may be the primary mechanism by which Treg cells act on effector T cells.

[Signal transduction in the differentiation of human peripheral blood mononucleocytes towards dendritic cells induced by calcium ionophore].

AIM: To explore the intracellular signal transduction pathway in the differentiation of human peripheral blood mononucleocytes (PBMCs) towards dendritic cells (DCs) induced by calcium ionophore (CI). METHODS: PBMCs isolated from a healthy donor were cultured with A23187 plus rhGM-CSF, 100 microg/L each. In some experiments, PBMCs were cultured for 30 minutes with W-7 (10 micromol/L), CsA(0.5 mg/L) or KT5926(1 micromol/L) before addition of rhGM-CSF and A23187. After culture for 40 hours, morphological change of the cells were observed under phase contrast microscope; surface markers on treated PBMCs were analyzed by flow cytometry; the proliferation of allogeneic human T cells stimulated by the treated PBMCs was detected by MTT colorimetry. RESULTS: PBMCs of the healthy donor cocultured with rhGM-CSF plus CI for 40 hours had the typical morphology of DCs, with decreased CD14 expression, and increased CD83, CD80 and CD86 expressions. The proliferation of allogeneic T cells stimulated by PBMCs treated with A23187 plus rhGM-CSF was strengthened. But the morphological changes, surface marker expressions and the ability to enhancing proliferation of allogeneic T cells were inhibited to different degrees by W-7, CsA or KT5926. CONCLUSION: The differentiation of PBMCs towards DCs by CI may be modulated by Ca (2+)/calmodulin and multiple signal transduction pathways downstream.
