Effect of seeding materials and mixing strength on struvite precipitation.

Struvite precipitation has increasing interest as a technology for removing and recovering phosphorus from wastewater streams. Many chemical factors have been studied, such as optimum pH values and component-ion molar ratios, yet, understanding of physical aspects is lacking. Two physical parameters were tested: (1) seeding material addition and (2) mixing. Objectives were to evaluate three seeding materials and to optimize mixing conditions for struvite-crystal precipitation, growth, and subsequent sedimentation. Results confirm that mixing strength and proper seeding materials increase crystal size and improve settleability. For unseeded solutions, optimum phosphorus removal was achieved at a mixing strength of G = 76 s(-1). Struvite crystals that were added as the seeding material provided the best performance with respect to phosphorus removal and crystal-size distribution. Overall, this study provided information to improve the practical application of struvite precipitation as a phosphorous-treatment technology for wastewaters, while generating a marketable slow-release fertilizer as a product.

A 70-year-old woman with acute renal failure. Crystal-storing histiocytosis.

Crystal-storing histiocytosis (CSH) is a rare disorder characterized by accumulation of immunoglobulin light chain within histiocytes in one or more organ systems, typically in association with multiple myeloma or, less frequently, with other conditions such as lymphoplasmacytic lymphoma, the use of certain medications, or even polyclonal gammopathies. The reason why only a few patients with multiple myeloma develop CSH, while the majority do not, is unknown, but it may at least partially reflect a defect in intralysosomal degradation because simple light chain overproduction is not sufficient for the development of CSH. Interestingly, CSH is associated with improved survival in multiple myeloma patients, perhaps due to earlier detection and treatment of myeloma because of the symptoms associated with CSH.

Hodgkin lymphoma-like posttransplantation lymphoproliferative disorder.

Posttransplantation lymphoproliferative disorders (PTLD) are a heterogeneous group of monoclonal or polyclonal lymphoproliferative lesions that occur in immunosuppressed recipients following solid organ or bone marrow transplantation, including 4 categories: (1) early lesions (reactive plasmacytic hyperplasia, and infectious-mononucleosis-like PTLD), (2) polymorphic PTLD, (3) monomorphic PTLD (including B-cell neoplasms and T-cell neoplasms), and (4) Hodgkin lymphoma (HL) and HL-like PTLD in the current World Health Organization classification. Although HL-like PTLD has been grouped with classic HL PTLD, controversy remains as to whether it is truly a form of HL or whether it should be more appropriately considered as a form of B-cell PTLD. The current available literature data indicate the presence of important immunophenotypic, molecular genetic, and clinical differences between HL PTLD and HL-like PTLD, suggesting that HL-like PTLD is in fact most often a form of B-cell PTLD. Distinction from true HL may be important for clinical management and prognosis.

[Effects of sensitized donor lymphocyte infusion on the chimerism and graft-versus-host disease after nonmyeloablative allogeneic stem cell transplantation].

To explore whether the complete donor chimerism could be achieved and graft-versus-host disease could be alleviated by donor lymphocyte infusion which was sensitized by the skin of the recipient, female C57BL/6 mice (H-2(b), B6) as recipients received total body irradiation (TBI) of 5.5 Gy ((60)Co gamma-ray) on day 0 followed by allogeneic hematopoietic stem cell transplantation (allo-HSCT). The allo-grafts consisted of 2 x 10(7) peripheral hematopoietic stem cells from mobilized male BALB/c (H-2(d)) donor mice with the granulocyte colony-stimulating factor (G-CSF). Day 2 after allo-HSCT, the recipient mice were given 200 mg/kg cyclophosphamide intraperitoneally. Afterwards these recipient mice were infused 2 x 10(6) sensitized or unsensitized-donor lymphocytes at the 28 days after transplantation. The results showed that the mice receiving sensitized-donor lymphocyte infusion did not suffer from GVHD and the phenotypic character of the recipient mice (black color) converted to that of the donor mice (white color), and to become full-donor chimerism. It was found that the ratio of CD4(+)/CD8(+) T lymphocytes of them decreased at the earlier period and increased after half month, but which were also lower than that of the normal value. While various grades of acute GVHD was observed in that of the control group and the mixed-chimeras were maintained, though it increased a little, and the ratio of CD4(+)/CD8(+) T lymphocytes increased at first, then decreased to the normal level half month later. It is concluded that sensitized DLI converted mixed to complete donor chimerism without GVHD, and the rate of CD4(+)/CD8(+) has close relation to the incidence of GVHD.

Identification of a unique set of genes altered during cell-cell contact in an in vitro model of prostate cancer bone metastasis.

The interaction between prostate cancer cells and bone marrow stromal cells (BMSC) is critical for survival and proliferation of metastatic cancer cells in the bone microenvironment. In order to study molecular mechanisms of prostate cancer bone metastasis, we established a novel heterotypic co-culture system, in which the role of direct cell-cell contact between prostate cancer cells and BMSC in addition to soluble factors can be analyzed. Using both bi-compartmental (insert) system and heterotypic (contact) system, we identified gene expression profiles of interaction between prostate cancer and bone cells. Analysis of differential gene expressions in these two co-culture systems revealed three distinctive sets of genes: 1) genes that were modified only by soluble factors; 2) genes that were regulated by both soluble factors and physical contact; and 3) genes that were altered only by physical contact. The last group consisted of specific set of genes including collagen III, IV, X, XII, integrin alpha1, alpha2, MMP-2, MMP-9, uPA, biglycan, osteopontin and raf-1 in PC3, and collagen VIII, IX, BMP6, TGFbeta1, Smad6 and Twist in BMSC. Among genes that were modified by both soluble factors and physical contact, the gene expression was affected in the same direction (such as MKK4) or in the opposite direction (such as TGFbeta receptor 3). Overall, this suggests that heterotypic cell-cell contact may act as an independent factor affecting the progression of bone metastasis.

[The cytotoxicity of nickel-refining dusts for chinese hamster lung cells and effects on gap junctional intercellular communication].

OBJECTIVE: To investigate the cytotoxicity of the nickel-refining dusts for Chinese hamster lung (CHL) cells and the effects of nickel-refining dusts on the gap junctional intercellular communication (GJIC) of CHL cells. METHODS: The cytotoxicity of the nickel-refining dusts for the CHL cells was determined in two nickel-refining dusts samples with the CHL cells as the target cells by MTT method while the effects of nickel-refining dusts on the CJIC of the CHL cells were investigated using the scrape-loading and dye transfer (SLDT) technique. RESULTS: There were no significant difference in the CHL proliferation between all dosage groups in the two samples and the control group at 6 and 12 hours (P > 0.05). The survival rate of cells in all dosage groups were all decreased at 36 hours (P < 0.05), presenting the dosage-reaction relationship and the time-reaction relationship. IC(50) was 21.36 and 23.07 micro/ml for the two samples respectively at 36 hours. Compared with the control group, the transport of Lucifer Yellow (LY) from the injury line to the adjacent cells was decreased when the CHL cells were treated with nickel-refining dusts of 25.00, 50.00 and 100.00 microg/ml (P < 0.01). CONCLUSION: The nickel-refining dusts have cytotoxicity for the CHL cells cultivated in vitro, can inhibit the growth of the cells and at a certain concentration can inhibit the GJIC function of CHL cells.

[Surveillance on contamination of total aflatoxins in corn, peanut, rice, walnut and pine nut in several areas in China].

OBJECTIVE: To realize the contamination of total aflatoxins in corn, peanut, rice, walnut and pine nut in China, and provide the base data for establishing a China tolerance limit standard and an international control practice for total aflatoxins. METHODS: The samples of corn, peanut, rice, walnut and pine nut from Chongqing, Fujian, Guangdong, Guangxi, Hubei, Jiangsu, Shanghai and Zhejiang provinces and municipalities were collected randomly from markets, with the totally 284 samples. The samples were grounded and added to acetonitrile/water mixture. After filtering, the extract was transferred into a purifying column and pressed slowly. Then the purified liquid was derivatized with trifluoroacetic acid (TFA) and detected by using a high performance liquid chromatography (HPLC). RESULTS: There was 70.27% corn having been detected out an average level of aflatoxins of 36.51 microg/kg and the highest level was 1098.36 microg/kg. At the same time, there was 14.86% corn exceeding the China national tolerance limit. In peanut, the aflatoxins detected rate was 24.24%. The average level was 80.27 microg/kg and the highest level was 437.09 microg/kg. While there was 3.03% peanut exceeding the China national and Codex tolerance limits. All of the rice, walnut and pine nut samples met the China tolerance limit for aflatoxins. CONCLUSION: Corn and peanut might be the severely contaminated foods with aflatoxins in China. The aflatoxin B(1) in foods might be can not delegate the contamination of aflatoxins completely. Surveillance of total aflatoxins in foods suggested an actual need of establishing the China national and international standards for total aflatoxins.

[Analysis of hematopoietic chimerism after non-myeloablative allogeneic peripheral blood stem cell transplantation].

The aim of this study was to analyze the hematopoietic chimerism after non-myeloablative allogeneic peripheral blood stem cell transplantation (NAPBSCT). 28 patients received NAPBSCT were evaluated. The conditioning regimen included FBC (fludarabine, busulphan, cyclophosphamide) +/- Ara-C. Peripheral blood was collected before and after transplantation in different periods. Semi-quantitative assessment of hematopoietic chimerism was performed by short tandem repeat-polymerase chain reaction (STR-PCR), polyacrylamide gel electrophoresis (PAGE) and silver staining, and analyzed by Image Analysis System. The results showed that on day 30 after transplantation, one patient failed to engraft, but 22 cases formed complete chimerism (CC) and 5 cases were of mixed chimerism. On day 7 after transplantation, the average percentage of donor cells was 74.71%. The time of dominance of the donor-specific allelic pattern preceded the recovery time of neutrophils and platelets. The incidence of aGVHD in group CC was significantly higher than that in group MC (P < 0.05). There was no significant difference in the incidence of cGVHD and disease relapse between group CC and group MC (P > 0.05). One patient relapsed in CC status without a transitional stage of MC. One patient with MC rejected grafts in early stage. 3 patients with MC transferred to CC and got complete remission after early implementation of therapy. It is concluded that sequential and quantitative detection of chimerism may be of great value to evaluate engraftment and to predict graft rejection, disease relapse and GVHD. Furthermore, it may provide a basis for early intervention treatment in the related complications.

[Early clinical evaluation of visual function in patients with age-related cataract].

OBJECTIVE: To investigate the characteristics of visual acuity, contrast sensitivity and glare contrast sensitivity in age-related cataract patients. METHODS: In this study, 115 eyes of 66 age-related cataract patients with a best corrected visual acuity of 0.5 or better, who had been classified into cortical cataract group (50 eyes of 31 subjects), nuclear sclerosis group (32 eyes of 17 subjects), posterior subcapsular cataract group (33 eyes of 18 subjects); and control group (34 eyes of 34 subjects) were examined with a contrast glare tester to assess contrast sensitivity and glare contrast sensitivity. RESULTS: There were statistically significant differences among contrast sensitivity and glare contrast sensitivity in the cataract patients with different visual acuity at low, medium spatial frequency (P < 0.01); there were no statistically significant differences among those at high spatial frequency (P > 0.05). Cortical cataract was associated with the contrast sensitivity at low, medium spatial frequency, where the glare contrast sensitivity fell into at low, medium and high spatial frequency; in the patients with cataract of nuclear sclerosis was associated with the contrast sensitivity at low spatial frequency, where the glare contrast sensitivity was at low, medium and high spatial frequency; in contrast to posterior subcapsular cataract both the contrast sensitivity and glare contrast sensitivity (at low, medium, high spatial frequency) were lower than normal eyes (P < 0.05). Each spatial frequency of contrast sensitivity and glare contrast sensitivity in 57 cataract eyes with a best corrected visual acuity of 0.8 or better were lower than normal eyes. In those eyes the contrast sensitivity at low spatial frequency and glare contrast sensitivity at low and high spatial frequency had statistically significant difference compared with control (P < 0.05); other spatial frequency had no statistically significant difference among groups (P > 0.05). The glare contrast sensitivity in cataract eyes was lower than contrast sensitivity at visual angle of 6.3 degrees, 4.0 degrees, 2.5 degrees, 1.6 degrees, 1.0 degrees, the difference was statistically significant (P < 0.01). CONCLUSION: Contrast sensitivity and glare contrast sensitivity are reliable in evaluating visual function in the patients with early age-related cataract.

[Correlation analysis and identification of G421C in regulatory region of CYP4F2 gene with essential hypertension].

OBJECTIVE: To investigate the correlation between G421C polymorphism in the regulatory region of CYP4F2 gene and essential hypertension and its molecular mechanism. METHODS: Totally 196 hypertensive patients (hypertension group) and 219 normotensive subjects (control group) were genotyped by polymerase chain reaction-restriction fragment length polymorphism. The promoter activity with different alleles was evaluated by reporter assay. A Myb responsive element was identified using gel retardation assay. RESULTS: Significant differences were found in distribution of genotype and allele frequency of G421C between hypertension group and control group (P < 0.05), and homozygous GG genotype was independently associated with hypertension after adjustment for age, gender, body mass index, and other risk factors (odds ratios 1.87, 95% CI 1.11-3.13, P < 0.05). 421G reporter construct showed decreased promoter activity compared with 421C reporter construct. 421G existed in Myb responsive element, whereas 421C damaged this motif. CONCLUSION: G421C polymorphism in the regulatory region of CYP4F2 gene is correlated with essential hypertension. 421G allele inhibits transcription by binding affinity of Myb responsive element.

[Managements of masked mastoiditis].

OBJECTIVE: To appreciate clinically of masked mastoiditis to explored how to reduce the incidence and associated morbidity of otogenic complications. METHODS: Between January 1998 and February 2005, 11 cases of masked mastoiditis were collected retrospectively. Their clinical presentation, results of investigations, and response to treatment were reviewed. RESULTS: Of 11 cases, there were 8 cases with hearing impairment, 5 cases with otalgia, 4 cases with facial nerve palsy, one patient with postauricular subperiosteal abscess, one case with meningitis, and one with thrombosis of the lateral sinus. Computed tomography (CT) scan revealed blurring (haziness) of the mastoid air-cells. After admission, intravenous antibiotics were prescribed and antro-mastoidectomy or mastoidectomy was performed for eradication of infection source. The predominant finding at mastoidectomy was granulation tissue filling the mastoid cavity and antrum. A varying amount of pus and osteitis was found in the 5 cases. The granulations into the antrum were severe, obstructing the drainage into the attic and the middle ear. The mastoid tip cells were filled with granulation tissue which spared the antrum. The patients recovered excellently postoperative, without facial palsy, vertigo or other complications. CONCLUSIONS: To be a contemporary otologist, such severe complications of otologic diseases should not be overlooked. Appropriate intravenous antibiotics and adequate surgeries, as soon as possible, were recommended. Advanced CT scans of the temporal bone were necessary. Failure to identify associated concomitant pathology might result in treatment failure or persistent neurological deficit.

[Effects of mechanical tensile stress on the expression of ICAM-1 mRNA in osteoblasts differentiated from rBMSCs].

OBJECTIVE: To investigate the correlation between mechanical tensile stress and the expression of ICAM-1 mRNA in osteoblasts differentiatd from rBMSCs and elucidate the mechanism for osteoclastogenesis regulated by osteoblasts in bone modeling and remodeling during the process of orthodontic tooth movement. METHODS: rBMSCs-derived osteoblasts were isolated and cultured in vitro, and were subjected to static mechanical tensile stress of 1,3,5 kPa or dynamic tensile stress of 3, 5 kPa at 0.017 Hz with the use of cellular tension-stress system for 24 h. Controls were without any stress. Cells were collected at 0 h, 3 h, 6 h, 9 h, 12 h, 24 h and 48 h after stress loading. The expression patterns of ICAM-1 mRNA were examined by semiquantitative RT-PCR assay. RESULTS: ICAM-1 mRNA level significantly decreased after mechanical tensile stress loading, either dysamic or static, compared with controls; the effects of inhibition positively correlated with the magnitude of stress (5 kPa>3 kPa>1 kPa). The inhibition effects of dynamic tensile stress groups exceeded the corresponding static stress (3,5 kPa dynamic stress>3,5 kPa static stress respectively). The expression of ICAM-1 mRNA significantly decreased at 3 h, reached the minimun transcription, as low as 23% of that of control, at 12 h, and then slightly rebounded and stayed at a considerably lower and stable level. CONCLUSION: Mechanical tensile stress can regulate osteoclastogenesis by inhibiting the expression of ICAM-1 in osteoblasts derived from rBMSCs, it can lead to a better understanding of the molecular basis for osteoblast-osteoclast communication in bone resorption induced by application of mechanical tensile stress during orthodontic tooth movement.

[Effects of osteoporosis on experimental tooth movement in aged rats].

OBJECTIVE: To examine the experimental tooth movement and recruitment of new osteoclasts in aged variotomized (OVX) rats. METHODS: Eighty-two 3-month female virgin health rats were divided into two groups. Each group was divided into seven sub-groups of tooth movement: 0,1,3,5,7,10 and 14 days. The tooth movement and the population of osteoclasts were measured and comparatively analyzed. RESULTS: The osteoporosis rats showed larger and faster tooth movement (P<0.05). In the osteoporosis group, the instantaneous tooth movement lasted longer time and traversed larger distance, the delay period was shorter, the population of osteoclasts was larger and displayed a faster recruitment. CONCLUSION: These findings indicate that osteoporosis accelerates tooth movement in aged OVX rats.

[Platelet-derived microparticles stimulate proliferation of granulocyte-macrophage progenitor cells from umbilical cord blood].

This study was aimed to investigate the effect of platelet-derived microparticles (PMP) on stimulating the proliferation of granulocyte-macrophage progenitors (CFU-GM) from umbilical cord blood. Different concentrations of thrombin were adopted to activate the platelets for releasing PMP. Flow cytometry was adopted to evaluate the efficiencies of different concentrations of thrombin to produce PMP. Umbilical cord blood mononuclear cells (MNC) were obtained from healthy donors and the MNC were isolated by Ficoll density gradient centrifugation. MNC were cultured in 2.7% methylcellulose containing different concentration of PMP and colonies were counted under an inverted microscope after 7 days. The result showed that the release rate of PMP activated by 2.0, 1.5, 1.0 and 0.5 U/ml thrombin were 28.7%, 47.7%, 50.1% and 43.9% respectively. The PMP enhanced colony formation in dose-dependent manner. The number of colonies per 2 x 10(5) MNCs in groups of PMP at different concentrations (10, 50 and 100 microg/ml) were 119.8 +/- 32.2,142.8 +/- 45.2 and 180.8 +/- 85.1 respectively. The number of colonies in the groups of PMP at 100 microg/ml and 50 microg/ml were statistically significant when compared with control group (103.0 +/- 24.8) (P < 0.05). The number of colonies per 2 x 10(5) MNC in the group of PMP (10 microg/ml) was 119.8 +/- 32.2 which was higher than that in control group, but there was no statistical significance between two groups. It is concluded that platelet activated with 1.0 U/ml thrombin can get the best release efficiency of PMP and PMP can enhance the proliferation of granulocyte-macrophage progenitor cells of umbilical cord blood.

[Primary experimental studies on differentiation of marrow mesenchymal stem cells into skin appendage cells in vivo].

OBJECTIVE: To investigate the feasibility of differentiation of the marrow mesenchymal stem cells (MSCs) into the cells of the skin appendages and the mechanism of their involvement in the wound healing. METHODS: The bone marrow was collected from Wistar rats by the flushing of the femurs, MSCs were isolated and purified by the density gradient centrifugation. Then, the MSCs were amplified and labelled with 5-bromo-2'-deoxyuridine (BrdU). The full-thickness skin wounds with an area of 1 cm x 1 cm were made on the mid back of the homogeneous male Wistar rats. At the same time, 1 x 10(6)/ml BrdU-labelled MSCs were infused from the penile vein. The specimens were harvested from the wound tissues on the 3rd day and the 7th day after operation and were immunohistochemically stained by either BrdU or BrdU and pan-keratin. RESULTS: The BrdU positive cells appeared in the hypodermis, the sebaceous glands, and the hair follicles of the wounds, as well as the medullary canal of the femurs. The double-staining showed the BrdU positive cells in the sebaceous glands and the hair follicles of the wounds expressed pan-keratin simultaneously. CONCLUSION: During the course of the wound healing, MSCs are involved in the wound repair and can differentiate into the cells of the skin appendages under the microenvironment of the wound.
