[A study on the cell differentiation induced by tanshinone IIA and its molecular mechanism in retinoic acid: resistant acute promyelocytic leukemia].

OBJECTIVE: To investigate retinoic acid-resistant acute promyelocytic leukemia (APL) cell differentiation induced by tanshinone IIA (Tan IIA) and its molecular mechanism. METHODS: NB4 cells treated with 0.5 mg/L Tan IIA was regarded as positive control. After in vitro incubation of MR-2 cells with Tan IIA at the concentration of 1.0 mg/L for 4 days, the cell differentiation was observed by growth status, cytomorphology, and nitroblue tetrazolium test. Cell cycle, membrane cluster differentiation (CD) antigens (CD(33), CD(11b)) and expression of some oncogene (c-myc, c-fos, p53 and bcl-2) were analysed by flow cytometry. RESULTS: The growth of MR-2 and NB4 cells was inhibited after Tan IIA treatment, the inhibition rate were 73.5% and 67.7% respectively (P < 0.01, P < 0.01) without significant difference. After Tan IIA treatment, MR-2 and NB4 cells were induced to undergo morphological differentiation, which exhibited small cell bulk decreased nucleus/cytoplasm proportion, rough chromatin, disappearance of nucleolus and formation of azurophil granules and anomalous nucleus. MR-2 cells could be induced to metamyelocyte while NB4 could be induced to band form. NBT reduction of MR-2 and NB4 cells treated with Tan IIA showed that positive cells accounted for (95.30 +/- 0.76)% and (93.20 +/- 1.04)% respectively; but the positive rate of either group of the treated positive cells was significantly higher than that of untreated, being (3.50 +/- 1.32)% and (2.80 +/- 0.29)% respectively (P < 0.01). Flow cytometry showed that the expression of CD(33) was reduced, while that of CD(11b) was increased. The quantity of treated cells in G(0)/G(1) phase increased but that in S phase decreased. The proliferous index was also decreased. After treated with Tan IIA, the expressions of anti-oncogene p53 and c-fos were up-regulated while those of oncogene bcl-2 and c-myc were down-regulated (P < 0.01). CONCLUSIONS: 1.0 mg/L Tan IIA could inhibit proliferation of MR-2 cells and induce differentiation of MR-2 cells into mature granulocyte, the effectivity was the same as 0.5 mg/L Tan IIA treated NB4 cells. Its possible molecular mechanism might be related to modulation of oncogene expressions associated proliferation and differentiation as well as inhibition of DNA synthesis. Tan IIA probably can be applied to treat the patients with APL, particularly to the relapsed and drug resistant patients with broad prospect.

[Experimental study on gelatin-chondroitin sulfate-sodium hyaluronate tri-copolymer as novel scaffolds for cartilage tissue engineering].

OBJECTIVE: To fabricate a novel gelatin-chondroitin sulfate-sodium hyaluronate tri-copolymer scaffold and to confirm the feasibility of serving as a scaffold for cartilage tissue engineering. METHODS: Different scaffolds was prepared with gelatin-chondroitin sulfate-sodium hyaluronate tri-copolymer by varying the freezing temperatures (-20 degrees C, - 80 degrees C and liquid nitrogen). Pore size, porosity, inter pores and density were observed with light microscopy and scanning electron microscopy (SEM). The load-stiffness curves were compared between different scaffolds and normal cartilage. The number of MSCs attaching to different scaffolds and the function of cells were also detected with MTT colorimetric microassay. RESULTS: The pore size was 300 +/- 45, 230 +/- 30 and 45 +/- 10 microm; the porosity was 81%, 79% and 56%; the density was 9.41+/-0.25, 11.50 +/- 0. 36 and 29.50 +/- 0. 61 microg/mm3 respectively in different scaffolds fabricated at - 20 degrees C, - 80 degrees C and liquid nitrogen; the latter two scaffolds had nearly the same mechanical property with normal cartilage; the cell adhesion rates were 85.0%, 87.5% and 56.3% respectively in different scaffolds and the scaffolds can mildly promote the proliferation of MSCs. CONCLUSION: Gelatin-chondroitin sulfate-sodium hyaluronate tri-copolymer scaffold fabricated at -80 degrees C had proper pore size, porosity and mechanical property. It is a novel potential scaffold for cartilage tissue engineering.

[Expression of osteoclast differentiation factor and intercellular adhesion molecule-1 of bone marrow mesenchymal stem cells enhanced with osteogenic differentiation].

OBJECTIVE: To investgate whether the expression of osteoclast differentiation factor (ODF), intercellular adhesion molecule-1 (ICAM-1) depended on the stage of osteoblastic differentiation from rat bone marrow mesenchymal stem cells (BMSC). METHODS: BMSC (4 passage) were selected for osteogenic differentiation by treated with osteogenic supplements (OS). Cells were harvested by day 0, 3, 6, 9, 12, 18 respectively after osteogenic inducement. In each experiment, control and OS-treated cells were processed in parallel. ODF and ICAM-1 mRNA were analyzed by semiquantitave RT-PCR assay. RESULTS: Expression of ODF was enhanced with osteogenic differentiation guadully. whereas, expression of ICAM-1 was activated at OS-treated day 6, then keeping at a stable level. CONCLUSION: This study indicated that BMSC undergoing osteogenic inducement was an ideal model for studying the differentiation and maturation of osteoblasts. During the early stage of differentiation along osteoblasts from stem cells to osteocytes, BMSC or osteoprogenitor react somewhat differently from osteoblasts, suggesting the ability of osteoblasts to regulating differentiation and maturation of osteoclasts have been improved with osteogenic culture.

[Study on the association of ABCA1 gene common variants with the risk of coronary atherosclerotic heart disease].

OBJECTIVE: To investigate the association of R219K and M883I polymorphisms of ATP binding cassette transporter 1 gene with lipid metabolism and the susceptibility to coronary atherosclerotic heart disease in Chinese population. METHODS: Genotypes were determined by PCR-restriction fragment length polymorphism and Primer introduced restriction analysis-PCR techniques, respectively, in 248 unrelated CHD-free controls and 224 CHD cases. RESULTS: Smoking, high blood pressure and high serum glucose were independent risk factors for CHD. Multivariate logistic regression analysis revealed that individuals carrying at least one 219K variant allele (RK + KK genotypes) had a significantly decreased risk for CHD (adjusted OR = 0.41; 95% CI = 0.27-0.61) compared with the wild-type genotype (219RR) and only 883II homozygotes displayed a decreased risk for CHD (adjusted OR = 0.54; 95% CI = 0.26-1.11) compared with 883MM and 883MI genotypes. Furthermore, compared with individuals with both wild genotypes (219 RR and 883 MM or 883 MI) other individuals with all other assembly genotypes had a significantly decreased risk (adjusted OR = 0.39, 95% CI = 0.26-0.60). Plasma HDL-C in 219K allele carriers were markedly higher than those in 219 K non-carriers in controls (P = 0.037). CONCLUSION: The ABCA1 R219K polymorphism may be involved in the variability of serum HDL-C and the susceptibility to coronary artery disease.

Sonocatalytic degradation of methyl orange in the presence of (nanometer and ordinary) anatase TiO2 powders.

The nanometer and ordinary anatase titanium dioxide (TiO2) powders were adopted as the sonocatalysts for the degradation of methyl orange used as a model compound for the first time. It was found that the sonocatalytic degradation effect of methyl orange in the presence of TiO2 powder were much better than that without TiO2, but the sonocatalytic activity of the nanometer anatase TiO2 particle was obviously higher than that of ordinary anatase TiO2 particle. Although there are many factors influencing sonocatalytic degradation of methyl orange, the experimental results showed that the best degradation ratio of methyl orange could be obtained when the experimental conditions were: initial concentration 15 mg/L, nanometer anatase TiO2 adding amount 750 mg/L, ultrasonic frequency 40 kHz, output power 50 W, pH = 3.0 and temperature 40 degrees C within 150 min. In addition, the catalytic activity of reused nanometer anatase TiO2 catalyst was also studied and found to decline gradually comparing with initial nanometer anatase TiO2 catalyst. All experiments indicated that the method of the sonocatalytic degradation of organic pollutants in the presence of TiO2 powder was an advisable choice for non- or low-transparent organic wastewaters.

[Efficacy of NO regimen and NP regimen on advanced non-small cell lung cancer: a prospective randomized trial].

BACKGROUND & OBJECTIVE: Oxaliplatin (LOHP) is an effective drug in treatment of non-small cell lung cancer (NSCLC) with mild toxicities to gastrointestinal tract, kidney, and bone marrow. Cisplatin (DDP) plus vinorelbine (NVB) constitute the first-line regimen (NP regimen) for NSCLC. This study was to compare the short-term response, long-term outcome, and adverse events between advanced NSCLC patients received NO regimen (LOHP plus NVB) and NP regimen. METHODS: A total of 90 patients with advanced NSCLC were randomized into NO group (58 patients, 25 mg/m(2) of NVB, day 1 and day 8; 130 mg/m(2) of LOHP, day 1) and NP group (32 patients, 25 mg/m(2) of NVB, day 1 and day 8; 50 mg/m(2) of DDP, day 2 and day 3). The short-term response, long-term outcome, adverse events, and survival status of the 2 groups were observed. RESULTS: The response rates were 33.33% in NO group, and 35.48% in NP group, but no significant difference was detected between the 2 groups (P > 0.05). The clinical benefit response rate was significantly higher in NO group than in NP group (80.70% vs. 64.52%, P < 0.05). The median time to progression (TTP) was 17 weeks in NO group, and 15 weeks in NP group; the median time of remission was 21 weeks in NO group, and 19 weeks in NP group; the median survival time was 39 weeks in NO group, and 37 weeks in NP group; the 1-year survival rate was 37.93% in NO group, and 31.25% in NP group. No significant differences were detected between the 2 groups. The incidence rates of phlebitis and grade I-II peripheral neuritis were significantly higher in NO group than in NP group (77.59% vs. 50.00%, P<0.01; 43.10% vs. 15.63%, P<0.01). The incidence rate of grade III-IV nausea/vomiting was significantly higher in NP group than in NO group (31.25% vs. 3.45%, P<0.05). CONCLUSIONS: The efficacy of NO regimen on advanced NSCLC is similar to that of NP regimen, but the clinical benefit response rate is higher in NO group than in NP group. In short, NO regimen may be recommended as the first-line chemotherapy regimen for advanced NSCLC.

[Influence of heart and brain disease on multifractal singularity spectrum of synchronous 12-lead ECG signals].

We analyzed the multifractal singularity spectrum of synchronous 12-lead ECG (electrocardiogram) signals from heart and brain disease patients, and found that multifractal curves of different leads do not overlap each other. After calculating the scope of the singularity strength, we noticed that the averages of scope are not the same in different subjects, and the dispersing degree is also different in someone's different leads. Both the deltaalpha, the average of the deltaalpha, and the dispersing degree deltaalpha (be defined as standard deviation) of the deltaalpha of every one's 12-lead ECG were computed. Then, a comparison was made between the spectrum of the healthy subjects and the heart disease patients. The results showed that their deltaalpha are close, but their deltaalpha are markedly different. Also the healthy subjects and brain disease patients were compared, we found that their deltaalpha are close, but their deltaalpha are markedly different. These indicate that the character of multifractal spectrum is controlled by both the neurosystem of body and the self-syntonic property of cardiac structures. Furthermore, the deltaalpha is related with the neuroautonomic control of people's body on the ECG, and the deltaalpha is related with the anisotropy of the heterogeneous tissue and the electric signal propagation in heart.

[Ultrastructural changes of rat ischemic myocardium after bone marrow mesenchymal stem cell transplantation].

OBJECTIVE: To observe the ultrastructural changes of ischemic myocardium of rats after bone marrow mesenchymal stem cells (MSCs) transplantation. METHODS: Rat models of myocardial ischemia were established by ligating the descending anterior branch of the left coronary artery. The isolated and in vitro cultured MSCs labeled with 4' 6-diamidine-2-phenylindole (DAPI) were injected into the rats via the tail vein and the hearts of the rats were taken 1 week and 8 weeks after transplantation, respectively, for observation under fluorescence microscopy. The ultrastructural changes of the ischemic myocardium of the recipient rats were observed by transmission electron microscope. RESULTS: The third passage of cultured MSCs growing in colonies possessed good homogeneity. One week and 8 weeks after transplantation, DAPI-labeled cells were observed in the heart of the recipient rats, but not in the hearts of control rats. One week after transplantation, ultrastructural observation identified a small number of cells in the peripherals of the infarct area of the recipient rats with similar morphology to that of MSCs cultured in vitro. At 8 weeks,a large number of capillaries were seen in the ischemic myocardium on the peripheral of the infarct area. Ultrastructural observation also revealed some immature myocytes surviving in the ischemic region. CONCLUSION: MSCs is capable in vivo of homing to the ischemic myocardium via the blood circulation, and promote regeneration of the myocardium and blood vessels.

[A prospective cohort study on body mass index and mortality among middle-aged and elderly men in urban Shanghai].

OBJECTIVE: To examine the association between body mass index (BMI) and mortality as well as the effect of age on it among middle-aged and elderly men in urban area of Shanghai. METHODS: A total of 18,244 male subjects aged 45-64 years resided in urban area of Shanghai were enrolled in the study during January 1, 1986 through September 30, 1989, and were actively followed under annual visits. 'Cox proportional hazards model' was used to estimate the relative risks (RR). RESULTS: By the end of the follow-up process in 2002, a total number of 235,762 person-years was accumulated in the cohort, with an average of 12.9 years per subject. A total number of 3365 deaths including 1381 from cancer and 1165 from cardio- and cerebro-vascular diseases (CVD), was identified during the follow-up period. Compared with those under normal BMI (BMI 18.5-23.9), the RRs of death for all causes of death among groups at low BMI (BMI < 18.5), overweight (BMI 24-27.9) and obesity (BMI > or = 28) were 1.20, 1.12 and 1.61, respectively, among non-smokers after adjustment for age, alcohol consumption and level of education. After excluding the numbers observed during the first 5 years of follow-up, the corresponding RRs became 1.01, 1.12, and 1.75, respectively. The risk of deaths from colon cancer or CVD increased along with the increase of BMI, while the risk of non-cancer and non-CVD deaths, mostly deaths from infectious diseases, increased significantly in the group of low BMI. Among those aged > or = 55 years at baseline survey, the risk for all causes of death increased more significantly with those having obesity. However, among those who were younger than 55 years of age, no significant correlation between BMI and overall mortality was noticed. CONCLUSION: A positive relationship between obesity and total mortality was observed in the middle-aged and elderly men in urban Shanghai. The association was more obvious among the elderly while the risk of deaths from colon cancer or CVD rose along with the increase of BMI. The risk of death from infectious disease increased significantly in the group with low BMI.

[Accommodation in pseudophakic eyes with the 1CU accommodative intraocular lens].

OBJECTIVE: To investigate the clinical effect of accommodating intraocular lens (IOL) on the accommodation of patients with IOL implantation. METHODS: 75 cases (94 eyes) had phacoemulsification and implantation of Human-Optics accommodating 1CU intraocular lens. The main outcome measures were subjective refraction, uncorrected distance vision acuity, spectacle-free near visual acuity, best corrected distance vision acuity, distance corrected near vision acuity at 30 cm, best corrected near vision acuity at 30 cm, and amplitude of accommodation with subjective techniques of pushing-up test (near point) and defocusing with minus glasses. Patients were being follow-up for 1 to 12 months, the data was collected at 7, 30 and 90 days after surgery. RESULTS: Postoperatively, better uncorrected distance vision acuity, best corrected distance and near vision acuity were obtained in all of patients studied. At different time points after surgery near visual acuity with Jaeger 5 or better without spectacle was 81.9%, 85.1%, and 84.0% at 7 days, 30 days and 90 days respectively. The near visual acuity with Jaeger 5 or better after distance vision acuity correction in all of cases was 78.7% at 7 days, 79.8% at 30 days and 74.5% at 90 days. The amplitude of accommodation was (1.96 +/- 0.63) D (range 0.75 to 4.50 D) by push-up test, (1.74 +/- 0.59) D (range 0.75 to 4.50 D) by defocusing at 7 days; (1.68 +/- 0.47) D (range 0.75 to 3.25 D) by push-up test, (1.89 +/- 0.54) D (range 0.75 to 3.25 D) by defocusing at 30 days; (1.77 +/- 0.53) D (range 0.75 to 2.75 D) by push-up test, (1.66 +/- 0.50) D (range 0.75 to 2.75 D) by defocusing at 90 days. The differences in amplitude of accommodation among 7, 30 and 90 days were not statistically significant (P > 0.05). Distance corrected near acuity and amplitude of accommodation showed a positive correlation. CONCLUSION: The implantation of 1CU accommodative IOL provided both better distance and near vision acuity.

[Heavy metal stabilization in municipal solid waste incineration fly ash using soluble phosphate].

The physico-chemical characteristics of municipal solid waste incineration fly ashes was analyzed. It indicated that the main elements of fly ashes are Ca, Cl, K, S and Si, and many heavy metals such as Pb, Zn, Cu, Mn and Cr can be found in fly ashes, and the heavy metal leaching toxicity such as Pb, Cu, Zn and Cd is much higher than the standards. A new kind of stabilization agent-soluble phosphate is chosen to treat with fly ash, different influence factors including agent dosage, curing time and pH are taken into consideration to test its stabilization for the stabilized fly ash. The experimental results indicate that fly ash treated with soluble phosphate can have excellent stabilization effect, and the leaching toxicity for Pb, Cd and Zn can be reduced by 97.5%, 91.6% and 95.5% at a phosphate dosage of 3%; curing time can not influence its stabilization; and, stabilized fly ash using soluble phosphate can keep long-term stabilization at a broad pH value.

[The influence of several anions on sonoluminescence].

1.1 MHz ultrasound was used to induce sonoluminescence in air-saturated solutions dissolved five kinds of anions in different ranges: NO3-: 0.2-20 mg x L(-1), F- : 4-20 mg x L(-1), Cl- : 5-25 mg x L(-1), HCO3- : 8-30 mgx L(-1), SO4(2-) : 0.4-40 mg x L(-1). It was found that four kinds of the anions, Cl- , NO-, HCO3- and SO4(2-) at the above mentioned concentrations have no influence on sonoluminescence. However, only F- in the range of 4-20 mg x L(-1) can quench the sonoluminescence. The quenching mechanism was analyzed.
