Upregulation of CRH gene expression and downregulation of arginine vasopressin gene expression in the hypothalamus of bilateral nephrectomized rats.

The expression of the corticotropin-releasing hormone (CRH) gene and the arginine vasopressin (AVP) gene in the hypothalamus examined in bilateral nephrectomized rats by in situ hybridization histochemistry. The expression of the CRH gene was significantly increased in the parvocellular part of the paraventricular nucleus (PVN) 12 and 20 h after bilateral nephrectomy in comparison with that after sham operation. The plasma concentration of adrenocorticotropic hormone (ACTH) in nephrectomized rats was significantly higher than that in sham operated rats 20 h after surgery. In contrast, the expression of the AVP gene in both the parvocellular and magnocellular parts of the PVN and throughout the supraoptic nucleus (SON) was significantly decreased 20 h after bilateral nephrectomy in comparison with that after sham operation. These results suggest that nephrectomy-induced upregulation of the CRH gene with elevation of plasma ACTH may be due to the activation of the hypothalamo-pituitary adrenal (HPA) axis.

Development of optical coatings for 157-nm lithography. I. Coating materials.

In a basic study to identify low-loss optics for applications in F2 lithography, five potential coating materials (AlF3, Na3AlF6, MgF2, LaF8, and GdF3) and three deposition methods (thermal evaporation by a resistance heater and by electron beam and ion-beam sputtering) were investigated in the vacuum ultraviolet (VUV) region. Samples were supplied as single-layer coatings on CaF2 substrates by four Japanese coating suppliers. Refractive indices (n) and extinction coefficients (k) of these coatings at 157 nm were evaluated; the transmittance and the reflectance were measured by a VUV spectrometer and were compared. As a result, resistance heating thermal evaporation is seen to be the optimal method for achieving low-loss antireflection coatings. The relation among optical constants, microstructures, and stoichiometry is discussed.

Development of optical coatings for 157-nm lithography. II. Reflectance, absorption, and scatter measurement.

The total loss that can be suffered by an antireflection (AR) coating consists of reflectance loss, absorption loss, and scatter loss. To separate these losses we developed a calorimetric absorption measurement apparatus and an ellipsoidal Coblentz hemisphere based scatterometer for 157-nm optics. Reflectance, absorption, and scatter of AR coatings were measured with these apparatuses. The AR coating samples were supplied by Japanese vendors. Each AR coating as supplied was coated with the vendor's coating design by that vendor's coating process. Our measurement apparatuses, methods, and results for these AR coatings are presented here.

Effects of restricted feeding on the activity of hypothalamic Orexin (OX)-A containing neurons and OX2 receptor mRNA level in the paraventricular nucleus of rats.

We have examined the effects of 3 weeks of food restriction on both the activity of neurons containing hypothalamic orexin (OX)-A and the level of OX receptor type 2 (OX2R) mRNA in the paraventricular nucleus (PVN) of rats. Double immunohistochemistry was used to examine the expression of OX-A and Fos in the lateral hypothalamic area (LHA), and in situ hybridization histochemistry was used to measure levels of OX2R mRNA in the PVN. After the period of restricted feeding, 20-30% of OX-A-containing neurons exhibited Fos-like immunoreactivity (LI). The distribution of OX-A-LI/Fos-LI cells in the food-restricted rats was similar to that observed in glucose-deprived rats after intracerebroventricular (icv) administration of 2-deoxy-D-glucose (2-DG). In addition, 3 weeks of food restriction caused a significant decrease in the expression of the OX2R gene in the parvocellular division of the PVN. These results suggest that the activation of OX-A-containing neurons induced by restricted feeding may be involved in neuroendocrine responses to food restriction.

Mini review: surgical indications for adrenal incidentaloma.

The detection of an adrenal mass as an incidental finding has become so frequent that this trend is considered to be an endocrine epidemic. In these adrenal incidentalomas, a difficult problem is to distinguish between benign and malignant primary or secondary tumors. Another major problem in the management of patients with incidentally discovered adrenal mass is whether and when the mass puts the patient at increased risk of an adverse outcome. A Japanese government-supported study conducted by Professor Miyachi's research committee reviewed the records of all university hospitals and hospitals possessing more than 200 beds in Japan and found 2106 Japanese patients with adrenal incidentaloma during the year 1999. They comprised 1095 males (52%) and 1011 females (48%), mostly distributed between 50 and 59 years of age. The tumor size ranged mostly from 1.1 to 2.0 cm. Almost half (50.3%) of all the incidentalomas were diagnosed as non-functional adenomas, and 5.6% were primary or secondary malignant tumors. In conclusion, accurate assessment of tumor size is an important consideration during preoperative evaluation of adrenal tumors in order to rule out malignancy. Careful evaluation of endocrine functions of the hypothalamic-pituitary-adrenal axis is necessary before surgical treatment in patients with adrenal incidentaloma. Patients with adrenal incidentaloma should be carefully followed, especially regarding size and hormonal conditions.

Centrally administered neuromedin U activates neurosecretion and induction of c-fos messenger ribonucleic acid in the paraventricular and supraoptic nuclei of rat.

We examined the effects of intracerebroventricular (icv) administration of neuromedin U (NMU) on plasma arginine vasopressin (AVP), oxytocin (OXT), and ACTH in rats, using RIA. The induction of c-fos protein (Fos) was examined by immunohistochemical study, and in situ hybridization histochemistry was used to detect c-fos gene expression in the paraventricular (PVN) and supraoptic nuclei (SON). Plasma AVP, OXT, and ACTH were increased in a dose-related manner 15 min after icv administration of NMU. The icv administration of NMU caused a marked induction of Fos-like immunoreactivity (LI) in the SON and the magnocellular and parvocellular divisions of the PVN. In the SON and the magnocellular divisions of the PVN, OXT-LI cells predominantly exhibited nuclear Fos-LI in comparison with AVP-LI cells. The marked induction of the expression of c-fos gene in the PVN and SON was observed 15, 30, and 60 min after icv administration of NMU. Neurosecretion and induction of c-fos gene expression after centrally administered NMU were significantly reduced by pretreatment with anti-NMU IgG. These results suggest that centrally administered NMU activates OXTergic cells in the PVN and SON predominantly as well as hypothalamo-pituitary adrenal axis.

Differential regulation of focal adhesion kinase and paxillin phosphorylation by the small GTP-binding protein Rho in human corneal epithelial cells.

PURPOSE: Lysophosphatidic acid (LPA), which activates the small guanosine triphosphate (GTP)-binding protein Rho, was previously shown to promote migration of the rabbit corneal epithelium in culture. The signaling pathway responsible for this effect of LPA was examined in this study with a human corneal epithelial (HCE) cell line. METHODS: The activation of Rho was detected with a pull-down assay. Tyrosine phosphorylation of paxillin and of focal adhesion kinase (FAK) were examined both by immunofluorescence staining and by immunoprecipitation and immunoblot analyses. Expression of integrins alpha5 and beta1 was evaluated by immunoblot analysis. RESULTS: Incubation of cells with LPA (10 micro M) for 2 min resulted in marked activation of Rho, and this effect was blocked by pretreatment with the Rho inhibitor exoenzyme C3 (2 micro g/ml) for 24 h. Tyrosine phosphorylation of both paxillin and FAK was detected in HCE cells under basal conditions by immunofluorescence staining, immunoprecipitation, and immunoblot analyses. LPA induced a concentration- and time-dependent increase in the tyrosine phosphorylation of paxillin, which was maximal at a concentration of 10 micro M and a time of 2 min. Exoenzyme C3 inhibited LPA-induced paxillin phosphorylation. Neither LPA nor exoenzyme C3 affected tyrosine phosphorylation of FAK or expression of integrins alpha5 and beta1. CONCLUSIONS: LPA induces Rho activation and the consequent tyrosine phosphorylation of paxillin in HCE cells, and these effects likely contribute to the promotion of corneal epithelial migration by this agent.

Additional reduction in intraocular pressure achieved with latanoprost in normal-tension glaucoma patients previously treated with unoprostone.

PURPOSE: To determine whether treatment with latanoprost eye drops is able to further reduce intraocular pressure (IOP) in normal-tension glaucoma (NTG) patients whose IOP has been well controlled with unoprostone. PATIENTS AND METHODS: A total of 34 eyes (34 individuals) with NTG that had been treated with 0.12% unoprostone eye drops twice daily for >or=3 months were switched to treatment once daily with eye drops containing 0.005% latanoprost. IOP was measured before and 1, 2, and 3 months after the switch to latanoprost. RESULTS: The mean IOP of all eyes was decreased significantly by 1.8, 2.9, and 2.3 mmHg at 1, 2, and 3 months after the switch from unoprostone to latanoprost treatment. The IOP of patients with an initial IOP of 12 mmHg was reduced by 11.0 or 19.9%, respectively, after 3 months on latanoprost. The IOP of 30 (88.2%) of the 34 eyes was further reduced by the switch from unoprostone to latanoprost. CONCLUSIONS: Latanoprost reduced the IOP of NTG patients who had already been treated with unoprostone, even though both drugs are prostaglandin-related. Switching to latanoprost might thus achieve a maximal decrease in IOP and thereby better prevent damage to the optic nerve and loss of visual field in NTG patients.
