Isolation and short term cultivation of swine hepatocytes for bioartificial liver support system.

BACKGROUND: The demand for the clinical use of hepatocytes is increasing. The aim of this study was to develop a method for procurement of high qualitative pig hepatocytes and to evaluate the state of freshly isolated and cultured hepatocytes. METHODS: The domestic extracorporeal circulating perfusion apparatus was used to isolate and harvest swine hepatocytes by the two-step perfusion method with EDTA and collagenase. The viability, function and morphology of the freshly isolated and cultured cells were evaluated and observed by the trypan blue exclusion test, biochemical measurements, phase contrast microscopy and transmission electron micrography (TEM). RESULTS: The total yield of isolated hepatocytes reached to 1.5(+/-0.4)X10(10) per liver with a viability of 92(+/-5)%, and the purity of hepatocytes reached to 98%. Immediately after isolation, phase-contrast microscope and TEM showed that undamaged hepatocytes appeared bright, translucent and spherical in shape, with a characteristic well-contrasted border. After 24 hours, the concentrations of alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), albumin (ALB), creatinine (Cr) and blood urea nitrogen (BUN) in the fluid of culture were declined significantly. CONCLUSION: This method of procuring swine hepatocytes could get high quality cells with active metabolic function.

[Cloning of alpha-tubulin gene from Fusarium graminearum and analyzing its relationship with carbendazim-resistance].

The full-length nucleotide sequence of alpha-tubulin gene from each of 6 Fusarium graminearum strains from China which had different carbendazim (MBC) sensitivity phenotypes were separated using PCR with 4 primer sets designed in accordance with nucleotide sequence of the gene from the reference isolate, NRRL 31084 (PH-1). The DNA sequence comparison showed that there was no difference in the nucleotide sequence of alpha-tubulin gene amongst 3 sensitive and 3 resistant strains from China. This result demonstrates that there is no relationship between MBC-resistance and alpha-tubulin gene. The full-length of the gene spans 1718 bp, including 6 introns, encoding 449 amino acids. With 99% homology, there is 5 nucleotide differences in alpha-tubulin gene between PH-1 isolate and the 6 strains from China. The homology of the deduced amino acid sequence of the gene is 99.78% between the 6 strains and PH-1 isolate, and 37%-86% between the 6 strains and other 6 species of fungi.

[The expression of heat shock protein 27 in retinal ganglion cells in the rat glaucoma model].

OBJECTIVE: To observe the expression of heat shock protein 27 (HSP27) in retinal ganglion cells in experimental rat glaucoma model and to study the relationship between HSP27 expression and hypertension using serum autoantibodies. METHODS: Forty nine Wistar rats were randomly divided into ocular hypertension group, sham control group and normal control group. Intraocular pressure (IOP) was elevated by electrocoagulation of at least 3 episcleral and limbal veins in the right eye of each rat. Contralateral eyes were used as the controls. Serum immunoreactivity against HSP27 was examined in these three groups by means of enzyme-linked immunosorbent assay (ELISA). Immunohistochemical staining for HSP27 was performed in 49 rats (98 eyes) for studying the distribution of the target gene products in retinal ganglion cells and retinal nerve fiber layer. RESULTS: Electrocoagulation treatment caused a significant increase of IOP and was maintained for 8 weeks (P < 0.01), which was greater than that in the contralateral eyes, sham control eyes and normal eyes. The baseline IOP was (15.69 +/- 3.20) mm Hg (1 mm Hg = 0.133 kPa). IOP was increased to (27.52 +/- 6.63) mm Hg (3 days after the operation), (31.42 +/- 6.18) mm Hg (1 week after the operation) and stabilized thereafter for 8 weeks. Serum ELISA titers of HSP27 antibody were higher in ocular hypertension group as compared to the control groups in both sham and normal group at 2 weeks after operation (P < 0.01) and 3 weeks after operation (P < 0.05). The expression of HSP27 was significantly enhanced in RGCs with IOP elevated and sustained. It was also significantly increased in retinal nerve fiber layers (RNFL). CONCLUSION: Enhanced expression of endogenous HSP27 might play an important role in the development of glaucomatous optic neuropathy.

[A qualitative study of in vivo pulsed electric field distribution model in rabbit liver tissues].

Pulsed electric fields (PEFs) with fixed frequency, width and gradually increased peak value of voltage was applied to 30 healthy rabbit liver tissues. The specific aims were to explore the feasibility of establishing a model of in vivo PEFs distribution in healthy rabbit liver tissues and to provide important references for clinical electrochemotherapy and for electrotransfer. Repeated experiment and self-comparison statistics design were implemented. The rabbit underwent the experiment under intravenous anesthesia and their liver tissues, after exposure to PEFs, were sent for HE staining. Necrotic borderline was visible 3 days after PEFs application, the necrotic shape of concentric circle was evident around the electrodes under optical microscope at lower voltage, as voltage increasing, two necroses in the shape of concentric circle gradually enlarged; nuclei with chromatin condensation, fragmentation and lysis alterations were seen in the middle region between the needles; concentric circles changed into ellipse fusiform and finally overlaped each other forming irregular necrosis contours. Cell cavitation and tissues ischemia were also observed within electric field. The shape of tissue necrosis from the experiment was noted to correlate with theoretic simulation of electric field distribution. Therefore, rabbit liver tissues can be a good carrier for in vivo modeling of electric field distribution when the lethal effects of PEFs in tissues are investigated. PEFs also show safety for the surrounding normal tissue while causing damage or injury to the target area therapeutically.

[Anatomy of the sphenoidal sinus in transsphenoidal approach through a single nostril].

OBJECTIVE: To study the anatomy of the sphenoid sinus for safe pituitary adenoma resection via transsphenoidal approach through a single nostril. METHODS: Eight fresh adult cadavers and the magnetic resonance images (MRI) of the sphenoidal sinus of 5 adults were observed. In anteroposterior axes, the root of the nasal columella was defined as the point O, the inferior border of the sphenoidal sinus anterior wall as point B, the superior border of the spheniodal sinus anterior wall as point C, and the midpoint of the sellar floor as point D. Line OA ran through the point O in parallel with the maxillary alveolar process. Angle AOB', angle AOC' and angle AOD' were the supplementary angles of angle AOB, angle AOC and angle AOD respectively, and angle AOB', angle AOC', angle AOD', OB, and BC were measured. RESULTS: In the 8 fresh adult cadavers, angle AOB', angle AOC', angle AOD', OB, and BC were (43.2+/-4.3) degrees, (22.9+/-3.0) degrees, (35.4+/-4.1) degrees, 66.3+/-3.6 mm, and 20.9+/-1.5 mm, respectively, with 2 cases having ethmoid cell superior to the sphenoid bone; in the MRI of 50 adults, the measurements were (44.1+/-5.5) degrees, (25.7+/-6.4) degrees, (34.2+/-5.9) degrees, 68.7+/-4.9 mm, and 23.3+/-3.1 mm, respectively, with 15 cases having ethmoid cell superior to the sphenoid bone. Two independent-sample t test and Chi-square test revealed no significant differences in the measurements between the two groups (P>0.05). CONCLUSION: Preoperative MRI facilitates safe opening of the anterior wall of the spheniodal sinus and the sellar floor, and the best angle between the axis of the speculum and line OA was 39 degree.

[Over one hundred year sedimentary record of polycyclic aromatic hydrocarbons in the Pearl River Estuary, South China].

The western shoal of the Lingding Bay is known to be an important deposition zone of terrestrial substance in the Pearl River Estuary (PRE). In this study, the vertical variations of concentration and fluxes of polycyclic aromatic hydrocarbon (PAH) are investigated in a core from the west shoal of Lingding Bay. In combining with 210Pb-dating, the PAHs sedimentary record in the last 100 years is reconstructed. The sigmaPAH concentration ranged from 59 ng x g(-1) to 330 ng x g(-1) throughout the core with two distinct peaks. An initial increase in sigmaPAH concentration was found around the 1860s, followed with the first maximum in the 1950s. There was a decrease in total PAHs concentration and flux in 1960s and 1970s. A sharp increase in PAHs levels was observed from 1980s and a maximum was found in the 1990s. PAHs diagnostic ratios indicate that PAH in the sediment core are mainly of pyrolytic origin. PAHs concentrations are found to correlate positively with the Gross Domestic Production, vehicle numbers and power generation in the surrounding regions, indicating that the PAHs in the sediment core are mainly anthropogenic. Atmospheric deposition and land runoff may serve as the important pathway of PAHs input to the sediment.

[Axial split osteotomy of free fibular flap for mandibular angle reconstruction: a clinical study].

PURPOSE: To introduce an axial split osteotomy of free fibular flap for mandible angle reconstruction and discuss its indications and surgical technique. METHODS: 16 patients underwent axial split osteotomy of free fibular flap(12 males and 4 females, aged from 23 to 62). 11 patients with benign tumors (8 ameloblastomas and 3 odontogenic keratocyst), 3 patients with osteoradionecrosis, and 2 patients with secondary mandibular deformity were included. The length of free fibular flap ranged from 10-17 cm (mean 14.6 cm). The length of the vascular pedicel ranged from 5-11 cm (mean 8.2 cm). The fibular flap harvested was performed in routine manner. After elevating the periosteum and performing minimal dissection at osteotomy site only, axial splitting was performed using an oscillating saw. The first osteotomy was made on the external surface of the fibula, perpendicular to the main axis of the bone down to the level of the medullary canal. The second osteotomy was made along the main axis of the bone. The length of this osteotomy was between 2 and 3 cm. The last osteotomy was made perpendicular to the internal surface at one junction of the anterolateral and posteromedial surfaces down to the medullary canal, as was done for the first osteotomy. Then the harvested flap was contoured, which was achieved by raising the two bone segments and fitting them together. The angulation was 120 degrees. RESULTS: 16 flaps were monitored by Doppler ultrasound postoperatively and examined by colored ultrasound 3 months, 6 months after operation. The results revealed that the recipient sites healed primarily without complications. CONCLUSIONS: Axial split osteotomy is a reliable technique with maximum contact surface for bone union, and less bone loss, which can lengthen the transplanted bone to a certain extent. It is suitable especially for reconstruction of the defect of the angle of the mandible.

[Matrix metalloproteinase-1 expression in the circulation of patients with Kawasaki disease and its role in the pathogenesis of coronary artery lesions].

OBJECTIVE: Kawasaki disease (KD) is an acute and self-limited systemic vasculitis syndrome of unknown origin that mainly affects small and medium-sized arteries, particularly the coronary arteries, which is followed by aneurysm formation. Increased levels of matrix metalloproteinase-1 (MMP-1) have been detected in aortic aneurysms in adults, suggesting an important role of MMP-1 in arterial wall destruction and resultant aneurysm formation. The aim of this study was to investigate the potential role of MMP-1 in the pathogenesis of coronary artery lesions in patients with KD. METHODS: Forty patients with KD, including 23 patients without coronary artery lesions (CAL) and 17 patients with CAL, as well as age-matched 10 febrile and 10 healthy afebrile controls were studied. The duration of KD was divided into three phases: the acute phase, the subacute phase and the convalescent phase. Enzyme-linked immunosorbent assay was used to detect the protein levels of MMP-1 in the sera. MMP-1 mRNA expression in the circulating leucocytes was studied using reverse transcription-polymerase chain reaction. RESULTS: Levels of MMP-1 protein in serum and MMP-1 mRNA expression in the leucocytes were significantly elevated at the acute phase in the two groups of KD patients (CAL group: 14.91 +/- 3.88 ng/ml and 0.89 +/- 0.15 ng/ml; NO-CAL group: 11.27 +/- 3.28 ng/ml and 0.77 +/- 0.14, respectively), compared with febrile (7.05 +/- 1.98 ng/ml and 0.45 +/- 0.12 ng/ml, respectively) and afebrile (5.13 +/- 1.20 ng/ml and 0.29 +/- 0.12 ng/ml, respectively) controls (P < 0.01). Furthermore, MMP-1 protein and MMP-1 mRNA levels were significantly higher in KD patients with CAL than in KD patients without CAL (P < 0.05). There was a significantly positive correlation between the serum protein level of MMP-1 at the acute phase of KD and the circulating leucocytes counts (r = 0.750, P < 0.01). The MMP-1 serum protein level and mRNA expression in the leucocytes at the acute phase of the two KD groups decreased obviously from the subacute through the convalescent phases (P < 0.05 or P < 0.01). CONCLUSION: The expression of MMP-1 at the acute phase of KD was significantly elevated, especially in KD patients with CAL. MMP-1 might be involved in the formation of coronary artery lesions and pathogenesis of KD.
