Polymorphisms of mouse apolipoprotein A-II: seven alleles found among 41 inbred strains of mice.

In mice, apolipoprotein A-II (apoA-II) associates to form amyloid fibrils in an age-associated manner. We determined the complete nucleotide sequences of the apoA-II gene (Apoa2) cDNA in 41 inbred strains of mice including Mus musculus domesticus (laboratory mouse), Mus musculus castaneus, Mus musculus molossinus, Mus musculus musculus and Mus spretus. Among these strains we identified 7 alleles (Apoa2a1, Apoa2a2, Apoa2b, Apoa2c, Apoa2d, Apoa2e and Apoa2f). Polymorphisms of nucleotides at 15 positions were detected and amino acid substitutions were found at 8 positions. Apoa2a1 was found in all mouse subspecies, but Apoa2b and Apoa2c were found only in Mus musculus domesticus. Two strains of Mus spretus have the unique alleles Apoa2e and Apoa2f which resemble Apoa2c. We confirmed that VICS in which we found severe amyloidosis here and other amyloidoneic strains in published reports have Apoa2c allele. We determined the plasma concentrations of total and HDL cholesterol in the strains of Mus musculus domesticus with the Apoa2a1, Apoa2b and Apoa2c alleles. Significantly higher concentrations of plasma cholesterol were observed in mouse strains with the Apoa2b allele. These findings provide fundamental data on mouse Apoa2 alleles. Furthermore, differences in these alleles likely have considerable influence on traits related to amyloidosis and lipid metabolism.

Incidence and treatment of hepatic artery complications after orthotopic liver transplantation.

AIM: To investigate the incidence and treatment of hepatic artery complications after orthotopic liver transplantation. METHODS: From February 1999 to May 2002, orthotopic liver transplantations (OLT) were performed in 72 patients with end-stage liver diseases with an average age of 40.2 +/- 13.6 years (ranged from 11 to 68 years), 56 were males and 16 females. The preoperative evaluation for the 72 patients was performed using duplex sonography, abdominal CT scan, and angiography of the hepatic artery. All donor grafts were perfused and preserved in University of Wisconsin solution at 4 degrees. OLT was performed with standard techniques with or without a veno-venous bypass. Reconstructions of hepatic artery were performed between the branch patches of gastroduodenal/hepatic or splenic/common hepatic artery confluence of the donors and recipients, and an end-to-end anastomosis between other arterial vessels of the donors and recipients was done. Arterial anastomosis was performed with interrupted 7-0/8-0 monofilament polypropylene suture under 3.5 x loupe magnification. Diagnosis of the complications of hepatic artery after OLT was based on the clinical presentations, ultrasound findings and arterial angiography. All patients were followed up regularly for duplex ultrasound scan after discharge. RESULTS: The overall incidence of arterial complications in 72 patients after OLTs was 1.4% (1/72). One 3 cm pseudoaneurysm at the side of anastomotic site of hepatic artery was found by urgent arteriogram due to hemoperitoneum secondary to bile leakage after OLT. Subsequently the pseudoaneurysm was successfully embolized and the blood flow toward the donor liver in hepatic artery remained. The overall postoperative 30-day mortality rate was 8.33%. The one-year survival rate was 83.72% in 50 patients with benign diseases and was 71.43% in 22 patients with malignant diseases following OLT. No death associated with complications of hepatic artery occurred. CONCLUSION: Careful preoperative evaluations and intraoperative microsurgical technique for hepatic artery reconstructions are the keys in prevention of hepatic artery complications after OLT.

[Blue-on-yellow perimetry in patients with primary open-angle glaucoma].

OBJECTIVE: To investigate the early changes of the blue-on-yellow (B/Y) perimetry in patients with early primary open-angle glaucoma(POAG). METHODS: Thirty-one cases (45 eyes) of POAG underwent the central 30 degrees field examination of B/Y as well as routine white-on-white (W/W). RESULT: No significant difference of mean index deficiency (MD) between B/Y and W/W perimetry was detected (P>0.05). However, there were marked changes in index GHT (glaucoma hemisphere test) and more defect points in B/Y visual field than those in W/W (P<0.01). CONCLUSION: B/Y perimetry might be more sensitive than W/W perimetry a potentially more and valuable method in detection of early POAG lesion.

Transfection of B7-1 cDNA empowers antigen presentation of blood malignant cells for activation of anti-tumor T cells.

OBJECTIVE: To define roles of B7-1 co-stimulation factor expressed in human malignant cell lines in mediating anti-tumor T cell immune responses. METHODS: Examining human leucocyte antigen (HLA) and B7 expressions on 8 human blood malignancies cell lines by flow cytometry. Transfecting B7-1 gene to B7-1 negative (B7(-)) Raji and B7(-) Jurkat cell lines by liposome, and comparing the potencies of blood malignant cell lines in the induction of T cell activation by examination of T cell cytokine mRNAs before and after transfection using semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). RESULTS: High level of HLA I and II molecules were expressed in most human blood malignant cell lines examined, and the co-stimulatory factor B7-2 was also highly expressed. In contrast, another member of B7 family: B7-1 was either not expressed or very limitedly expressed in most of these hematopoietic malignant cell lines. Most importantly, transfection of B7-1 gene to B7(-). Raji and B7(-). Jurkat cell lines made these cell lines better antigen presenting cells for stimulation of anti-tumor T cell activation, which was demonstrated by up regulation of expression of T cell cytokines IL-2, IL-4 and INF-gamma mRNAs after incubation of these tumor cells with T cells for 24 h. CONCLUSIONS: B7 co-stimulation plays an important role in anti-tumor immunity. Transfection of B7-1 gene to the human hematopoietic malignant cell lines that are deficient in the B7-1 expression empowers their antigen presentation potency for activation of anti-tumor T cells. Our results suggested that repairing the deficiency of B7-1 co-stimulatory pathway in tumor cells might be a novel immunotherapeutic approach for human hematopoietic malignancies.

[Characteristics and environmental significance of soil dissolved organic matter].

Soil is a complex ecosystem with multi-interface. A numerous studies on soil dissolved organic matter (DOM) were carried out, and proved that DOM was one of the most active chemical components in the environment. Increasing attention has been paid on the study of soil DOM, especially in recent years, and the study has become an interdisciplinary focus in the fields of soil science, ecology, and environmental science due to the important roles of DOM in the biogeochemical cycles of carbon, nitrogen, phosphorus, sulfur, etc. In addition, DOM has significant effects on pedogenesis, growth and metabolism of soil microorganisms, decomposition and transformation of soil organic matter, and transport of pollutants in soils. The recent literatures about extraction methods, origin, composition, contents and controlling factors, bioavailability, and environmental significance of DOM were therefore reviewed, and future research aspects on this topic were also proposed.

[Detection of bovine-derived materials in import animal feeds and food by PCR assay].

A PCR assay for detection and identification of bovine--derived materials in import animal feeds and food is developed. A 271 bp sequence from bovine specific mitochondrial DNA is amplified. The amplified specific sequence of the bovine mtDNA from meat and feed samples is demonstrated by both direct sequencing and restriction endonuclease digestion yield analysis. This method can detect bovine mtDNA in those less than 0.125% of bovine meat. Owing that this method is specific, rapid and sensitive, it can be utilized as a routine control assay in prevention of spreading of bovine spongiform encephalopathy in China caused by imported food and animal feeds especially meat and bone meals.

[Diagnosis and surgical treatment of primary duodenal carcinoma].

OBJECTIVE: To investigate the early diagnosis of primary duodenal carcinoma and its outcome after surgical procedure. METHODS: Twenty-two patients with primary duodenal carcinoma treated operatively between 1983 and 1997 were analyzed retrospectively. Eleven patients complained of epigastric pain and discomfort, 5 jaundice, 4 nausea and vomiting and 2 epigastric fullness. The correct diagnosis rate for endoscopy was 90.0% (9/10), for duodenography 86.7% (13/15), for ultrasound examination 33.3% (4/12) and for computerized tomography (CT) scanning 58.3% (7/12), respectively. All of the 22 patients received surgery including pancreaticoduodenectomy (12 patients), segmental resection (4) and bypass operation (gastrojejunostomy and cholecystojejunostomy or cholangiojejunostomy) (6). RESULTS: Primary duodenal carcinoma was characterized nonspecifically, and the correct diagnosis was based on endoscopy and duodenography. The follow-up rate of this group was 86.4% (19/22). The 1-, 3-, 5-year survival rates of patients receiving radical resection (n = 16, 1 patient lost follow-up) were 86.7% (13/15), 46.7% (7/15), and 26.7% (4/15), respectively. The 5-year survival rate of patients receiving pancreaticoduodenectomy was 27.3% (3/11), and the 5-year survival rate of patients having segmental resection was 1/4. No patient (n = 6, 2 lost follow-up) with primary duodenal carcinoma treated by bypass procedure survived more than one year. There was a significant difference between patients receiving radical procedure and bypass operation (chi(2) = 6.84, P < 0.01). CONCLUSION: Radical resection might improve the survival of patients with primary duodenal carcinoma.

Highly efficient expression, purification of recombinant LTB protein and its activity against mucosal immunoadjuvant by nasal immunization.

OBJECTIVE: To develop an efficient expression, purification system of recombinant Escherichia coli heat-labile enterotoxin B subunit (rLTB) and study its activity against mucosal immunoadjuvant by nasal immunization. METHODS: A recombinant, pMMB68-LTB was generated by cloning the LTB cDNA fragment into an expression vector (pMMB68) and transformed it into the host strain marine vibrio VSP60. The relevant target protein was identified using SDS-polyacrylamide gel electrophoresis (SDS-PAGE) and Western blot. Sephacryl S-100 gel filtration chromatography was carried out for purification of rLTB in engineering bacteria VSP60. BALB/c mice received hen egg lysozyme (HEL) alone or combined with rLTB by nasal administration. After three times immunization, IgG and IgA antibody levels in serum or small intestine wash samples were determined using ELISA. RESULTS: rLTB protein was highly expressed in VSP60. After gel filtration with Sephacryl S-100, the purity of rLTB reached 98.1%, the yield rate was about 52%. After immunization, IgG and IgA antibody responses specific to HEL in system and mucosa of HEL + rLTB groups were significantly increased, compared with the HEL alone group (P < 0.001). CONCLUSIONS: A set of protocols for large-scale rLTB preparation has been established, which is simple, efficient and applicable. The rLTB protein we prepared was proved to be a powerful mucocal adjuvant, which could greatly enhance systemic and mucosal immune responses to nasally co-administered antigen.

[Effect of the different diameter of posterior capsulotomy on visual field].

OBJECTIVE: The purpose of this study is to determine the influence of two different diameter of posterior capsulotomy on visual field. METHODS: Forty patients with posterior capsule opacification (PCO) of grade II after phacoemulsification and intraocular lens implantation were randomly divided into two groups. 20 cases received laser capsulotomy with 2 mm diameter. Other 20 cases received laser capsulotomy with 4 mm diameter. One and half months after laser capsulotomy, vision field of every patient was examined using the automated perimetry for central 30 degrees and peripheral 60 degrees. Visual field in 21 patients with a transparent posterior capsule after cataract surgery was also evaluated by the same automated perimetry. RESULTS: There were significant reductions in the total visual field sensitivity, the mean deviation (MD) and the pattern standard deviation (PSD) in 2 mm diameter capsulotomy group compared with 4 mm diameter capsulotomy group (P < 0.001). There were no significant difference in visual field between 4 mm capsulotomy group and the transparent posterior capsule group (P > 0.05). CONCLUSIONS: Better visual acuity could be obtained after laser capsulotomy with 2 mm capsulotomy, however the generalized sensitivity reduction and irregular localized depression of visual island might occurred. 4 mm diameter capsulotomy may be a better choice in laser capsulotomy in patients with PCO.

[Overexpression of COX-2 and its clinical significance in non-small cell lung cancer].

OBJECTIVE: To study the relations among COX-2 expression in non-small cell lung cancer (NSCLC), its clinical characteristics and brognosis. METHODS: Immunostaining was performed with COX-2 antibody to the surgically resected tissue samples from 79 patients with NSCLC. Vessel epithelium cell COX-2 expression was taken as positive control. RESULTS: The positive rate of adenocarcinoma and squamous cell carcinoma was 85% and 57%, respectively (P = 0.013). COX-2 expression was associated with the extent of adenocarcinoma differentiation, tumor size, and TNM period, but not with the extent of squamous cell carcinoma differentiation. In the COX-2 positive group, the 5-year survival rate and a median survival time were 27.1% and 53 months; however in the negative group they were 52.0% and 61 months (P = 0.029). CONCLUSION: Invasive development of NSCLC is related to inceased expression of COX-2. COX-2 overexpression may be one of the risky factors for the prognosis of NSCLC.

[Research on PAHs fingerprints of vehicle discharges].

Air samples of 14 PAHs were collected from vehicle discharges of different types, oils, and mileages and analyzed by HPLC. It showed that the total PAHs discharged from the vehicle in 30 minutes were 41.53-121.1 micrograms/m3, among which, BaA was the most predominant, about 33.3%, followed by Naphthalene, Benzo (ghi) pyrene and In (1,2,3-cd) pyrene, about 16.8%, 12.9% and 12.1%, respectively. Diesel vehicle discharged more total PAHs compared to gasoline vehicle. The two vehicles all exhausted much BaA and Naph, however diesel vehicle mainly emitted Ac, Fluor, Bghip, In, while gasoline discharged mainly Bghip, In, BkF. PAHs with five or more rings in gasoline discharges were relatively higher than those in diesel discharges, however to 3-ring PAHs, diesel vehicle produced more than gasoline vehicle. As for vehicle of different mileage, there was a clear increase for almost all PAHs, especially Flur, Py, BaP, Bghip.

[Influence of initial chirp on the evolution of spectra].

The influence of dispersion and nonlinearity on the spectra was analysed. The nonlinear Schrödinger equation was solved numerically in the frequency domain. The effect of the initial chirp on the spectra of different formats was simulated. It was found that the influence of the initial chirp on the spectra changed with different formats. The positive initial chirps make spectra spreading and negative initial chirps have little influence upon spectra for Gauss pulses. For super Gauss pulses, the spectra have some side tails, which cause energy loss, but the initial chirp has very little effect on the spectra. The initial chirps have a great influence on the spectra for soliton pulse, especially for high order soliton, which suggests that initial chirp must be decreased for stable pulse transmission.

[Properties of the stimulated emission in the ZnO thin film].

ZnO thin film is a promising material for short-wave laser and LED etc, due to its high excition binding energy, intense stimulated emission, low lasing threshold, and high working temperature. ZnO thin films were prepared by laser molecular beam epitaxy (L-MBE) in our work. At room-temperature we reported the measurements of absorption spectra and emission spectra of ZnO thin films excited by various optical pumping intensities. High structural perfection of our sample was shown in these figures. We studied the properties and mechanism of stimulated emission in ZnO thin films. The relation between emission intensity and pumping intensity was obtained. Time behaviors of the stimulated emission under relatively high pumping intensity, spontaneous emission, and laser pulses were compared, and hence the stimulated emission of ZnO thin films was proved.

Posterior chamber phakic intraocular lens implantation for high myopia.

OBJECTIVE: To evaluate the efficacy, safety and stability of posterior chamber phakic intraocular lens (PIOLs) implantation for the correction of high myopia. METHODS: Thirty-nine eyes of twenty patients with high myopia (between -11.75 and -25.75 diopters) had a posterior chamber PIOL (Staar ICL) implanted. During 6 - 48 months' follow-up, visual acuity, refraction, intraocular pressure (IOP), corneal reaction and space between crystal lens and intraocular lens (IOLs) were tested. RESULTS: Successful implantation was achieved in all patients. Visual acuity without correction greater than 0.5 was found in 34 eyes at 1 day and 3 months postoperatively. Thirty-five eyes maintained a low negative power of refraction (-1.42 +/- 1.32 doipters), which did not prevent the patients from most of their daily activities. During 3 - 48 months' follow-up, refraction was stable and no cornea edema and glaucoma was found. Two eyes of one patient had corticosteroid glaucoma and another eye showed cataractogenesis under anterior capsular membrane. CONCLUSION: Posterior chamber PIOL implantation is predictable, safe, and effective in the correction of high myopia, and its indications should be carefully selected.
