[The effects of interleukin-12 inhalation on airway inflammation and T helper cell subsets in mouse asthma models].

OBJECTIVE: To observe the effects of aerosol interleukin-12 (IL-12) on the antigen-induced airway inflammation and T helper cell subsets in mouse asthma models. METHODS: Thirty BALB/c mice were divided into 3 groups, ten in each group. The mouse asthma models (asthma group) were established by sensitization and challenge with ovalbumin (OVA). One part of sensitized BALB/c mice (IL-12 group) were interfered with aerosol IL-12 from the beginning of sensitization to the end of challenge. The healthy control mice were (control group) treated with normal saline instead of OVA. Broncho-alveolar lavage fluid (BALF) and spleen mononuclear cells (SMNCs) were collected 24h after the last challenge. The total cell numbers and eosinophils (Eos) of BALF were counted. The levels of interferon-gamma (IFN-gamma) and IL-4 of BALF supernatant were measured by ELISA. The mRNA expressions of IFN-gamma and IL-4 in SMNCs were examined by reverse transcript-polymerase chain reaction. RESULTS: (1) The total cell counts and Eos counts in asthma group [(12.10 +/- 2.23) x 10(9)/L and (2.02 +/- 0.85) x 10(9)/L, respectively] were significantly increased than those in control group [(6.17 +/- 1.34) x 10(9)/L and (0.05 +/- 0.01) x 10(9)/L, respectively] (P < 0.01), while in IL-12 group the above counts [(8.41 +/- 1.71) x 10(9)/L and (0.15 +/- 0.08) x 10(9)/L, respectively] were significantly decreased than those of asthma group (P < 0.05). (2) The IFN-gamma and IL-4 levels of BALF supernatant in asthma group [(43.60 +/- 15.60) ng/L and (27.15 +/- 10.42) ng/L] had significant differences from those in control group [(83.76 +/- 19.02) ng/L and (0.84 +/- 0.69) ng/L, respectively] (P < 0.01) and from those in IL-12 group [(62.30 +/- 12.50) ng/L and (6.22 +/- 2.28) ng/L, respectively](P < 0.01). (3) The IFN-gamma and IL-4 mRNA expression of SMNCs in asthma group (0.64 +/- 0.14 and 0.89 +/- 0.31), respectively) had significant differences from those in control group (1.18 +/- 0.34 and 0.71 +/- 0.16, respectively) (P < 0.05) and those in IL-12 group (0.89 +/- 0.26 and 0.79 +/- 0.25, respectively) (P < 0.05) as well. CONCLUSION: The experiment data suggested that IL-12 aerosol inhalation might inhibit antigen-induced airway inflammation and partly reverse the deviation of Th cell subsets in mouse asthma models.

Autoantibodies against the myocardial beta1-adrenergic and M2-muscarinic receptors in patients with congestive heart failure.

OBJECTIVE: To determine whether autoantibodies against beta(1)-adrenergic and M(2)-muscarinic receptors are related to patients with congestive heart failure (CHF). METHODS: Both synthetic peptides corresponding to amino acids sequence 197-222 and 169-173 of the second extracellular loops of the beta(1) and M(2) receptors were used as antigens to screen sera from 265 patients.188 were congestive heart failure ( CHF) patients with different heart diseases, among them 42 were ischemic cardiomyopathy (ICD) and 52 were idiopathic dilated cardiomyopathy (IDCM) 44 were hypertensive heart disease (HHD) 50 were rheumatic valvular heart disease (RVHD); 77 were controls, among them 36 were simple hypertension and 41 were healthy donors (NC). RESULTS: Positive sera for beta(1)-adrenergic receptor was found in 45.73% (86/188) of CHF patients, while in the controls it was 10.4% (8/77) (P < 0.01); positive sera for M(2)-muscarinic receptor in CHF patients was found in 49.5% (99/188), while in the control it was 11.7% (9/77) (P < 0.01). The positive ratio of autoantibodies against beta(1)-adrenergic and M(2)-muscarinic receptors in CHF patients with cardiac function class II-III (NYHA) were significantly higher than cardiac function class IV. The average titer of autoantibodies against beta(1)-adrenergic and M(2)-muscarinic receptors of the former was significantly higher than the latter; 56.1% of patients with autoantibodies against beta(1)-adrenergic receptor had autoantibodies against M(2)-muscarinic receptor. CONCLUSIONS: Autoantibodies against beta(1)-adrenergic receptor and M(2)-muscarinic receptor were found in sera from heart failure patients with different cardiac diseases. We propose that autoantibodies against beta(1) and M(2) receptors are not only related to the IDCM, but also to cardiac structural and functional changes.

[Correlation of expression of vascular endothelial growth factor and matrix matalloproteinase-2 to invasion of ovarian tumor cells in vitro].

BACKGROUND & OBJECTIVE: Vascular endothelial growth factor(VEGF) and matrix metalloproteinase-2 (MMP-2) are the important factors in tumor metastasis. To the author's knowledge, their relationship has not been addressed to date. This study was designed to investigate the in vitro invasion and the expression level of VEGF and MMP-2 in ovarian tumor cells, and to evaluate the correlation of expression of VEGF and MMP-2 in the ovarian tumor metastasis. METHOD: Boyden chamber in vitro invasion assay was used to detect the invasive capacity in vitro in two ovarian tumor cell lines CaOV-3 and COC1. The expression levels of VEGF and MMP-2 in CaOV-3 and COC1 were evaluated by semiquantitative RT-PCR and Western blot. The activity of MMP-2 in two cell lines was detected by substrate zymography. RESULTS: Boyden chamber in vitro invasion assay indicated that the mean invasion percentage of CaOV-3 cells (21.9 +/- 1.5) was significantly higher than that of COC1 cells (8.8 +/- 0.9) (P < 0.05). Contrast to COC1 cells, CaOV-3 cells expressed significantly higher levels of mRNA of VEGF and MMP-2 as well as protein by RT-PCR and Western blot. Substrate zymography showed that the MMP-2 activity of CaOV3 cells was two times than that of COC1 cells. CONCLUSION: The in vitro invasive ability of ovarian tumor cells appeared to be positive correlated to high expression of VEGF and MMP-2. There may be relationship between VEGF and MMP-2 in the process of metastasis of ovarian carcinoma.

Molecular inhibition of angiogenesis and metastatic potential in human squamous cell carcinomas after epidermal growth factor receptor blockade.

Tumor metastasis represents a complex multistep process that requires migration, invasion, and angiogenesis. In this study, we examined the impact of molecular blockade of the epidermal growth factor receptor on the invasive and metastatic capacity of human squamous cell carcinoma (SCC) of the head and neck using in vitro and in vivo model systems. Treatment with the anti-epidermal growth factor receptor antibody C225 attenuated the migration of SCC-1 tumor cells through a chemotaxis chamber in a dose-dependent manner. Incubation of SCC cells with 10-100 nM C225 for 4 h resulted in 40-60% inhibition of cell migration. Furthermore, in the presence of C225, the capacity of SCC-1 to invade across a layer of extracellular matrix (Matrigel) was significantly inhibited. Using an in vivo orthotopic floor-of-mouth xenograft model, locoregional tumor invasion of SCC-1 into muscle, vessel, bone, and perineural tissues was inhibited in C225-treated mice. This inhibition was additionally characterized by down-regulation in the expression of matrix metalloproteinase-9. These data suggest that inhibition of metastatic potential by C225 may be mediated via decreased migration and invasion of SCC cells. Regarding angiogenesis in vitro, we first studied human umbilical vascular endothelial cells, which established a capillary-like network structure (tube formation) in the presence of reconstituted Matrigel. Treatment with C225 reduced cell-to-cell interaction of human umbilical vascular endothelial cells, resulting in disruption of tube formation. The effect of C225 was additionally examined using an in vivo tumor xenograft neovascularization model of angiogenesis. Systemic treatment with C225 not only reduced tumor growth and the number of blood capillaries but also hindered the growth of established vessels toward the tumor. Taken together, these results provide evidence that C225 can suppress tumor-induced neovascularization and metastasis in SCC of the head and neck.

[The clinical and pathologic features of Charcot-Marie-Tooth disease].

OBJECTIVE: To study the clinical and pathological features of Charcot-Marie-Tooth disease (CMT). METHODS: The general clinical data, the changes of neuroelectrophysiology and the pathological features of neural and muscular biopsy of 20 CMT patients were comprehensively analyzed. RESULTS: The clinical manifestations in the two types of CMT were nearly the same, but the changes of neuroelectrophysiology and the pathological characteristics of the neural and muscular biopsy in the two types were obviously different. 16 cases of CMT type 1 were characterized by decreased sensory nerve conduction velocity (SNCV < 30 m/s) and are associated with demyelinating hypertrophic neuropathy. 4 cases of CMT tylpe 2 were characterized by normal SNCV and associated with axonopathy. CONCLUSION: Neuroelectrophysiology and neural and muscular biopsy are important for the diagnosis of CMT.

[Effect of traditional Chinese medicine on expression of extracellular signal-regulating kinase-1 and mitogen activated protein kinase phosphatase-1 in brain and liver of fetal rats with fetal growth restriction].

OBJECTIVE: To investigate the effect of reinforcing kidney, replenishing Qi and blood activating prescription (RKRQBAP) on extracellular signal regulating kinase-1 (ERK-1) and mitogen activated protein kinase phosphatase-1 (MKP-1) of fetal rats with fetal growth restriction (FGR). METHODS: Passive smoking was used to establish animal model of FGR. All pregnant rats were divided into four groups: FGR group, L-arg group: rats with FGR treated with L-arginine, Chinese medicine group (CHM group): rats with FGR treated by traditional Chinese medicine and normal control group. The expression of ERK-1 and MKP-1 in brain and liver were measured by western-blot. RESULTS: (1) According to FGR group, L-arg group, CHM group and normal group, the expression of ERK-1 in brain was 7.63 +/- 0.22, 10.03 +/- 0.41, 11.03 +/- 0.30, 11.44 +/- 0.09 and MKP-1 was 7.41 +/- 0.38, 10.35 +/- 0.60, 10.60 +/- 0.14, 11.60 +/- 0.62. (2) The expression of ERK-1 in liver was 4.73 +/- 0.54, 5.83 +/- 0.17, 11.37 +/- 0.12, 12.34 +/- 0.14 accordingly and MKP-1 was 9.07 +/- 0.61, 10.66 +/- 0.08, 14.27 +/- 0.73, 14.92 +/- 0.17. (3) The expression of ERK-1 and MKP-1 in brain and liver was much lower in FGR group than in normal group (P < 0.01), and that of CHM group was much higher than in FGR group (P < 0.01) and closed to normal group (P > 0.05). The expression of ERK-1 in brain was much higher in CHM group than in L-arg group (P < 0.05), the expression of MKP-1 was almost the same (P > 0.05); Both of them in liver were higher in CHM group than in L-arg group (P < 0.01). CONCLUSION: The possible mechanism which the RKRQBAP cure FGR is to affect the expression of ERK-1 and MKP-1 and to promote growth- related genes expression and restrain apoptosis. In addition, RKRQBAP is superior to L-arginine in treatment of FGR.

[Monitoring impact of measurement of urinary beta-human chorionic gonadotropin on orthotopic implantation model of human ovarian carcinoma in athymic nude mice].

BACKGROUND & OBJECTIVES: Tumor orthotopic transplantation model has become the main carrier for tumor animal experiment. However, there was no convenient and reliable method to monitor tumor growth in animal body. This study was designed to evaluate the monitoring impact of urinary beta-human chorionic gonadotropin (beta-HCG) on tumor growth in orthotopic implantation model. METHODS: Pclone-beta-HCG stably transfected human ovarian carcinoma cell line A2780-CG was made as the orthotopic implantation model. The urinary beta-HCG/creatine ratio was continuously determined and plotted into curve, so as to indirectly know the rule of tumor growth in nude mice and the monitoring value of the ratio for intraperitoneal chemotherapy by cisplatin. RESULTS: The tumorigenesis time in beta-HCG system transfected tumor cell A2780-CG was similar to that in the original cell line. The content of urinary beta-HCG/creatine had a positive correlation to tumor weight in animal body (r = 0.98); After chemotherapy with cisplatin, the beta-HCG/creatine ratio showed progressively decreased. CONCLUSION: beta-HCG system could be used to sensitively and non-invasively monitor the tumor growth in orthotopic implantation model of human ovarian carcinoma in nude mice. Meanwhile, it provides a new convenient monitoring method for tumor treatment in the model.

[Vasodilators for sudden sensorineural hearing loss: a systematic review of randomized controlled trials].

OBJECTIVE: To assess the effects and safety of vasodilators on sudden sensorineural hearing loss(SSHL). SEARCH STRATEGY: Electronic databases: MEDLINE from 1966, EMBASE from 1974, the Cochrane Controlled Trails Register, Chinese Bio-medicine Database from 1989. Hand search: Five kinds of Chinese otolaryngology journals were selected. Literature references were checked intensively. SELECTION CRITERIA: Randomized controlled trials comparing vasodilators with placebo or other drugs in patients with SSHL. DATA COLLECTION AND ANALYSIS: Three reviewers independently accessed the quality of trials and extracted the data. RESULTS: Thirteen trials with 1,155 patients were eligible and covered in the systematic review. Ten of the trials were conducted in developed countries, and three in China. None of the four trials showed the effects of vasodilators on SSHL were better than that of placebo. None of the seven trials comparing different drugs showed the effects of one kind of vasodilators were better than that of the other vasodilators. Two trials showed that some other drugs, such as batroxobin and hypaque were probably better than the vasodilators (dextran, papaverine, 654-2, danshen). Eight trials showed side effects of vasodilators, such as pruritus, allergy, etc. CONCLUSIONS: Based on the systematic review of current eligible randomized controlled clinical trials, there is no evidence to prove that vasodilator therapy is more effective than placebo or other therapies for SSHL, or the effects of one kind of vasodilators to be better than that of the other vasodilators. So far we can't draw a reliable conclusion about the effects of vasodilators for SSHL. In addition, we must pay attention to their potential adverse reactions.
