[Assessment of toxicity of heavy metal contaminated soils by toxicity characteristic leaching procedure].

The contents of heavy metals (Cu, Zn, Pb and Cd) in soils from a lead-zinc mine in Shangyu, Zhejiang Province, China, were analyzed and their toxicity was assessed by toxicity characteristic leaching procedure (TCLP), which was developed by US EPA. The TCLP method is a currently recognized international method for evaluation of heavy metal pollution in soils. The available levels of Cu, Zn, Pb and Cd were 8.2-36 mg x kg(-1), 23-143 mg x kg(-1), 6.4-1367 mg x kg(-1) and 0.41-2.2 mg x kg(-1), respectively, while the international standard were 15 mg x kg(-1), 25 mg x kg(-1), 5 mg x kg(-1) and 0.5 mg x kg(-1), respectively. The results show that soils around the mine are polluted with heavy metals Cu, Zn, Pb and Cd, especially polluted by Zn and Pb, followed by Cd and Cu. Moreover, the heavy metals in the soils extracted by TCLP indicate that fluid 2 is more effective than fluid 1 in extracting the heavy metals from the polluted soils and there is a positive correlation between fluid 1 and fluid 2. The contents of available heavy metals by TCLP are correlated with contents of total heavy metals.

A field experimental study of lignin sand stabilizing material (LSSM) extracted from spent-liquor of straw pulping paper mills.

A new technique was introduced for sand stabilization and re-vegetation by use of lignin sand stabilizing material (LSSM). LSSM is a reconstructed organic compound with lignin as the most dominant component from the extracts of black-liquor issued by straw pulp paper mills. Unlike the polyvinyl acetate or foamed asphalt commonly used for dune stabilization, the new material is plant-friendly and can be used with virescence actions simultaneously. The field experimental study was conducted since 2001 in China's Northwest Ningxia Hui Autonomous Region and has been proved that LSSM is effective in stabilizing the fugitive dunes, making the arenaceous plants survive and the bare dune vegetative. The advisable solution concentration is 2% and the optimal field spraying quantity is 2.5 L/m2. The soil nutrients of the stabilized and greened dune, such as organic matter, available phosphorous and total nitrogen are all increased compared with the control treatment, which is certainly helpful to the growth of arenaceous plants. The technique is worthwhile to be popularized because it is provided not only a new method for desertification control but also an outlet for cleaning contaminants issued from the straw paper mills.

[Chronic prostatitis related sexual dysfunction and its psychologic treatment].

OBJECTIVE: To explore the relationship between sexual dysfunction and chronic prostatitis and investigate the effect of psychologic treatment. METHODS: A total number of 346 patients with chronic prostatitis complicated by sexual dysfunction were randomized into two groups, one treated with routine medication (antibiotic, indomethacin, alpha1 receptor blocker and Chinese traditional medicine), and the other with both routine medication and psychologic treatment. Results were analyzed by comparing NIH-CPSI and IIEF-5 between pre- and post-treatment groups. RESULTS: The prostatitis symptoms and sexual dysfunction were improved obviously after treatment in the two groups, but compared with the control, more marked improvement was observed in the combined psychologic treatment group (P <0.01). The degree of chronic prostatitis related sexual dysfunction was not correlated with that of prostatitis symptoms. CONCLUSION: Psychology plays a very important role in chronic prostatitis related sexual dysfunction. Psychologic treatment not only works on the sexual dysfunction, but also improves prostatitis symptoms.

[Analysis and application of SNP and haplotype in the human genome].

Single nucleotide polymorphism (SNP) is the most common type of genetic variant in human genome. Haplotype, defined as a specific set of alleles observed on a single chromosome, or a part of a chromosome, has been an integral part of human genetics for decades. The goal of the international HapMap project is to determine the common patterns of DNA sequence variation and find the Tag SNPs representing all SNPs in the human genome. Some studies demonstrated that the analyses of haplotype defined by the grouping and interaction of several variants rather than any individual SNP correlated with complex phenotypes. Here, we describe the definitions of SNPs, genotype, haplotype and some information of the HapMap project. In this review, we summarize the current three haplotype-inference methods, including Clark' method, EM algorithm and Byes approach, and the different defining methods for haplotype block, as well as the methods for choosing tag SNPs and association studies of complex diseases using haplotype. The major public SNP databases and applications of SNPs and haplotype in common complex diseases and drug response are also introduced in the paper.

[Clinical observation on effect of modified huanglian wendan decoction in treating diabetic asymptomatic myocardial ischemia].

OBJECTIVE: To investigate the clinical efficacy of modified huanglian wendan decoction (MHWD) on diabetic asymptomatic myocardial ischemia. METHODS: Ninety patients were randomly divided into two groups. The control group (n=30) was given Xinkang tablet (XKT) at a dose of 20 mg, twice a day. The treated group (n=35) was given MHWD besides XKT as that given to the control group. The treatment course for both groups was 1 month. Indexes, including blood glucose, glycosylated hemoglobin, blood lipids, myocardial zymogram, hemorheologic parameters, urinary albumin, routine examination of blood and urine, function of liver and kidney, as well as 24h dynamic electrocardiogram and electrocardiogram exercise test were measured in the two groups before and after treatment. RESULTS: The total clinical effective rate in the treated group and the control group was 88.33% and 56.67% respectively (P < 0.05), showing significant difference between them. The frequency of ischemia attacking, paroxysmal cumulative time, motion related incidence were lower in the treated group after treatment than those in the control group. Besides, in the treated group after treatment, the level of blood lipids and hemorheologic parameters were significantly improved (P < 0.05 or P < 0.01), hematocrit was unchanged and triglyceride, red blood cell agglutination index and erythrocyte deformability index were obviously different to those in the control group (P < 0.05). While in the control group after treatment, except the improving of whole blood viscosity (P < 0.05), no significant change was found in the other indices. CONCLUSION: MHWD has effects in improving myocardial ischemia, bettering hemorheologic condition and reducing blood lipids.

[Evaluation of the psychosocial status of fluorosis patients in fluorosis epidemic areas].

OBJECTIVE: To explore the psychosocial effect of fluorosis on patients in fluorosis epidemic areas. METHODS: Multistage, stratified, clustered and random sampling method was used in this survey. Totally 416 inhabitants of 12, 15 and 35-44 years of age from rural fluorosis epidemic areas in Hebei province were selected. Dental fluorosis was classified by Dean's index (DI). A total of 178 patients with different types of fluorosis were involved in a psychosocial questionnaire investigation which included six five-point subscales, including attitude to teeth, subjective well-being, life satisfaction, interpersonal or social anxiety, effect of self-esteem, impact on behavior. RESULTS: Fluorosis prevalence in fluorosis epidemic areas of Xingtai in Hebei province was 71.20%, and significant difference existed among three age groups (Pearson Chi-square value = 167.51, P < 0.01). With the increase of DI score, the average value of subscale "attitude to teeth" rising, ranging from 2.58 to 3.51, and value of "subjective well-being" was also increasing, ranging from 2.35 to 2.9. Statistical difference of subscale average value was found only in these two subscales ("attitude to teeth": F = 4.787, P < 0.01; "subjective well-being": F = 2.538, P < 0.05). "Attitude to teeth" was significantly different between control group and moderate group (F = 0.52, P < 0.01) and between control group and severe group (F = 0.72, P < 0.01). While significant difference of "subjective well-being" could only be found between control group and severe group (F = 0.56, P < 0.01). According to the bivariate correlation analysis, strong correlation could be found between the degree of fluorosis and the average value of sub-scales. CONCLUSIONS: The psychosocial impact of fluorosis on patients in fluorosis epidemic areas mainly appeared in two aspects, i.e. attitude to teeth and the perception of well-being, especially in severe group.

[Effect of bite force lost on the expression of iNOS in the rat periodontium].

PURPOSE: To investigate the molecular mechanism of the effect of bite force loss on periodontium remodeling. METHODS: For establishing a rat model of different bite forces for the study, the left maxillary molars of Wistar rats were extracted and the left mandibular molar area was used as the model of the bite force lost. The animals were sacrificed after 6 hours,1,2,3 days and 1,2,3,4weeks. The prepared tissue specimens were processed for the study of the change in the histologic morphology and the expression of iNOS protein in PDLC and osteoblasts with HE staining, immunohistochemistry and RT-PCR techniques, and the results were analyzed by one-way ANOVA,Dunnet t test and paired samples test. RESULTS: In the group of lost biting force, the derangement of periodontal ligament and resorption of alveolar bone were observed in histologic study: the structure of periodontal ligament deranged ,fibers and cells arrayed abnormally, there were many holes in the alveolar bone, even osteoclasts were present; the expression of iNOS was observed dramatically changed: the expression of iNOS in periodontal ligament fibroblasts and osteoblasts was higher than normal, especially at 2 days and 3 weeks,so as the results of RT-PCR.There was a significant correlation between bite force lost and the increased expression of iNOSmRNA. CONCLUSIONS: Bite force lost significantly increased the expression of iNOSmRNA in PDLC and osteoblasts, which suggests that iNOS may play an important role in the process of periodontium remodeling.

Purinoceptors in renal microvessels: adenosine-activated and cytochrome P450 monooxygenase-derived arachidonate metabolites.

Cytochrome P450 (CYP)-dependent epoxyeicosatrienoic acids (EETs) dilate rat preglomerular microvessels (PGMVs) when adenosine 2A receptors (A(2A)R) are stimulated. As high salt intake increases epoxygenase activity and adenosine levels, we hypothesized that renal adenosine responses would be greater in high salt-fed rats. We have obtained evidence supporting this hypothesis in rats fed a high salt diet for 7 days. Stimulation of adenosine receptors with 2-chloroadenosine in kidneys obtained from rats on high salt (4%) intake produced an increase in EET release that was several-fold greater than in kidneys of rats on normal salt (0.4% NaCl) diets, which was associated with a sharp decline in renovascular resistance. Under conditions of high salt intake, an associated upregulation of A(2A)R and 2C23 protein expression was observed. As EETs are renal vasodilator and natriuretic eicosanoids, the antipressor response to salt loading may operate through an A(2A)R - EET mechanism. These findings expand the role of adenosine-related mechanisms in protecting renal function.

[Potential of human adipose tissue derived adult stem cells differentiate into endothelial cells].

OBJECTIVE: To investigate whether human adipose derived adult stem (hADAS) cells can differentiate into endothelial cells. METHODS: Stem cells were isolated and expanded from adipose tissue and then induced to differentiate into cells of osteogenic, adipogenic and neurogenic lineages in vitro. hADAS cells were induced with vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) to endothelial cells differentiation. hADAS cells were intravenously injected into mouse hindlimb ischemic models to test their ability to differentiate endothelial cells in vivo. RESULTS: hADAS cells were easily isolated and expanded in vitro. They had the ability to differentiate into osteogenic, adipogenic and neurogenic lineages. The cells expressed vascular endothelial growth factor receptor-2 (VEGFR-2, Flk1), and expressed endothelial markers when cultured with VEGF and bFGF. In response to local cues, hADAS cells in vivo differentiate into endothelial cells that contributed to neoangiogenesis in hindlimb ischemia models. CONCLUSIONS: Flk1+ hADAS cells have multipotential not only similar to bone marrow mesenchymal stem cells, but also exhibiting characteristics of endothelial progenitor cells. They may be a potential source of endothelial cells for cellular pro-angiogenic therapies.

Risk assessment of heavy metal contaminated soil in the vicinity of a lead/zinc mine.

Heavy metal contamination of soils through anthropogenic activities is a widespread and serious problem confronting scientists and regulators throughout the world. In this study we investigated the distribution, chemical species and availability of lead, zinc, cadmium and copper in nine surface (0 to 20 cm) soils from near an abandoned lead/zinc mine tailings located in Shaoxing, Zhejiang, China. Total heavy metal contents ranged from 5271 to 16369 mg/kg for Pb, 387 to 1221 mg/kg for Zn, 3.0 to 9.3 mg/kg for Cd and 65 to 206 mg/kg for Cu. In general, all heavy metals exceeded China National Standards for Soil Environmental Quality of Heavy Metals by a factor of 3-65 times. Comparison of the heavy metal concentrations (Pb, Zn, Cd and Cu) with clay content revealed a strongly significant relationship while significant relationship (P < 0.001) was also obtained between Cd + Zn and Pb + Cu. Solid phase speciation of the soils using Tessier procedure showed that the heavy metals were distributed in the order: residual > organically complexed-Fe-Mn oxides occluded > carbonate bound > exchangeable > water soluble. In the organic matter fraction, the ratio of Pb (29.1%) to its total concentration in the soils was higher than those of Zn (4.70%), Cd (3.16%) and Cu (9.50%). The percentages of the water soluble and the exchangeable fractions of Pb (1.80%) and Cd (2.74%) were markedly greater than those of Zn (0.10%) and Cu (0.15%), suggesting that Pb and Cd are relatively more mobile and hence more toxic in the contaminated soils. Strongly significant relationships between H2O-Pb, H2O-Zn and H2O-Cu, strong positive correlations between H2O-Pb, H2O-Zn, H2O-Cu and organic matter in soil were found. The content of H2O-Pb, H2O-Zn, H2O-Cu was negatively correlated with pH values. The similar negative relationships between pH values and exchangeable heavy metals were also recorded. It is suggested that increasing soil pH or liming the soil could decrease bioavailability of heavy metals in the soil.

[Chemical forms of heavy metals in the soils and plants of copper tailings yard].

The study on the chemical forms of heavy metals in the soils and plants at the copper tailings yard in Tongling City indicated that the soils were extremely poor, with organic mater content being 2.6-5.8 g x kg(-1), only 1/15 of the control, while their Cu, Cd, Pb and Zn contents were higher than the control, with Cu 809.30-1395.54 mg x kg(-1) and Cd 3.25-6.35 mg x kg(-1), as 30-60 times as those of the control. The heavy metals contents in Zoysia japonica and Trifolium pratense had a significantly positive correlation with the contents of their exchangeable and organic forms in soils, a significantly or very significantly negative correlation with the forms of Fe-Mn oxides and carbonate, and no correlation with residual form. The main forms of Cu, Zn and Pb in Z. japonica and T. pratense were HAC-and HCl-extractable, and residual. The NaCl-extractable Zn content in the roots, stems and leaves of Z. japonica and T. pratense occupied a higher proportion, while NaCl-extractable Cd was the main form of Cd in both of the plants, not only in their roots but also in their leaves.

[In vitro anti-myeloma effects induced by myeloma idiotype-protein pulsed dendritic cell vaccine].

OBJECTIVE: To investigate the specific antitumor immune response induced by idiotype protein (Id)-pulsed dendritic cells (DC) in vitro. METHODS: DC was generated from peripheral blood monocytes of the multiple myeloma (MM) patients using GM-CSF, IL-4, and TNF-alpha. The DCs were pulsed with idiotypic fragment, the F(ab')2 fragment of M protein from MM patient at the immature stage. The morphologic characteristics of the cells were observed with light and electron microscopes. The phenotypic features were analyzed with FACS, MTT assay was employed to evaluate the proliferation of autologous T cells and the inhibition rate of MM cells. RESULTS: DC precursors in peripheral blood could be induced to typical mature DC in medium containing GM-CSF, IL-4 and TNF-alpha. Mature DC with Id could increase the proliferation of the autologous T cells and activate naive T cells to become tumor specialized cytotoxic T lymphocytes (CTL). The CTL at different doses showed significant inhibition on or killing ability to autologous MM cells in vitro. CONCLUSIONS: In a suitable cytokine environment, the DC precursors from peripheral blood of MM patients could be induced to functional DC, and vaccination of Id-pulsed DC could induce active antitumor immune response.

[Influence factors analysis for laser-induced plasma spectroscopy measurements].

An experimental equipment has been set up for laser-induced plasma spectroscopic (LIPS) measurement based on a Nd: YAG Q-switched solid laser (average single pulse energy is 38 mJ) as exciting light source and the copper alloy as sample. The instrument time distribution of Cu radiation was obtained, and the software and hardware influence factors on the measured spectra were analyzed. The experiments proved that the fluctuation of laser output energy, average number of each datum, the position between the target surface and the lens focus, the position between the optical fiber bundle for receiving the radiation and the normal of the target surface, and the effective receiving area of the optical fiber bundle all influence the intensity of spectra and the reproducibility of measurement.
