Hydrogen-rich water combined with traditional Chinese medicine compound in the treatment of kidney stones: a randomized controlled prospective clinical trial.

JOURNAL/mgres/04.03/01612956-202701000-00009/figure1/v/2026-09-13T085902Z/r/image-tiff The formation and development of kidney stones are related to abnormal urine metabolism, oxidative stress and inflammation. Hydrogen-rich water has clear efficacy in antioxidant and improving inflammatory status, while Ye-Shi-Shi-Lin-Formula is a clinically effective traditional Chinese medicine compound preparation for treating kidney stones. This randomized controlled prospective clinical trial from July 2025 to March 2026 at the Seventh People's Hospital Affiliated to Shanghai University of Traditional Chinese Medicine examined the effect of hydrogen-rich water and Ye-Shi-Shi-Lin-Formula on kidney stones. The included 100 patients with kidney stones were randomly divided into blank, hydrogen-rich water, hydrogen-rich water and Chinese medicine, and Chinese medicines groups. The patients received drinking hydrogen-rich water and/or Ye-Shi-Shi-Lin-Formula daily for 12 weeks. All subjects received basic treatment following the guidelines. Imaging examination, urine metabolism testing, renal function, inflammation and oxidative stress index, blood routine and liver function are used to detect the efficacy and safety of hydrogen-rich water and Ye-Shi-Shi-Lin-Formula. Results showed that the total effective rate of kidney stone treatment in blank group, Chinese medicines group, hydrogen-rich water and Chinese medicine group and hydrogen-rich water group was 28%, 76%, 80% and 32%. The Ye-Shi-Shi-Lin-Formula can partially alleviate the oxidative stress, uric acid metabolism and urinary magnesium levels of patients with kidney stone and improve the function of renal tubules. The hydrogen-rich water therapy showed only efficacy in improving glutathione reductase but the combination of hydrogen-rich water and Ye-Shi-Shi-Lin-Formula has shown superior efficacy in improving oxidative stress and related metabolic factors of blood uric acid and urine stones, as well as in renal tubular function. The results indicate that the addition of hydrogen-rich water can improve urinary metabolism and oxidative stress status in the treatment of kidney stones with Ye-Shi-Shi-Lin-Formula. The study was registered at the International Traditional Medicine Clinical Trial Registry (Registration No. ITMCTR2025001446).

soFusion: facilitating tissue structure identification via spatial multi-omics data fusion.

The rapid advancement of spatial multi-omics technologies has opened new avenues for dissecting tissue architecture with unprecedented resolution. However, inherent disparities across omics modalities, such as differences in biological hierarchy and resolution, pose significant challenges for integrative analysis. To address this, we present soFusion, a method for representation learning on spatial multi-omics data that enables automated identification of tissue compartmentalization. soFusion employs a graph convolutional network (GCN) to extract latent embeddings from spatial omics profiles. To simultaneously capture both cross-modality relationships and modality-specific features, we introduce a novel strategy for intra- and inter-omics feature learning. Moreover, modality-specific decoders are designed to preserve the unique information embedded in each omics type. We evaluated soFusion on multiple datasets including gene expression, protein expression, and epigenetic features. Across all benchmarks, soFusion consistently outperformed existing methods in delineating anatomical structures and identifying spatial domains with improved continuity and reduced noise. Collectively, soFusion offers an effective solution for spatial multi-omics integration, substantially enhancing the robustness of spatial domain identification.

TRIM63 Overexpression in FISH-Negative MiTF Family Altered Renal Cell Carcinoma (MiTF RCC).

TFE3 and TFEB break-apart fluorescent in situ hybridization (FISH) assays are the "gold standard" for diagnostic confirmation of microphthalmia-associated transcription factor (MiTF) family-altered renal cell carcinoma (MiTF RCC), which includes TFE3-rearranged RCC and TFEB-altered RCC. However, FISH assays, for multiple reasons, may lead to equivocal or false-negative results, especially in cryptic fusions resulting from intrachromosomal inversions involving 5' partner genes, such as non-POU domain-containing octamer-binding protein (NONO); GRIPI-associated protein 1 (GRIPAP1); RNA-binding motif protein, X chromosome (RBMX); and RNA-binding motif protein 10 (RBM10). When FISH results are negative in cases with strong morphological suspicion of the listed tumor entities, pathologists may recommend targeted RT-PCR or panel-based RNA fusion sequencing for diagnostic confirmation. Our recent RNA in situ hybridization (RNA ISH)-based study demonstrated RNA expression of the tripartite motif containing 63 (TRIM63) to be highly enriched in TFE3-rearranged RCC and TFEB-altered RCC, including 2 FISH false-negative RCC cases harboring RBM10::TFE3 fusion. Based on these observations, we hypothesized that TRIM63 positivity could aid in diagnosing cases that are negative by conventional FISH assay but remain morphologically suspicious, representing an unmet clinical need in this area. We collected 20 RCC cases with morphological suspicion (with equivocal/indeterminate immunohistochemistry panel) of MiTF RCC, which were TRIM63 positive, negative/equivocal for TFE3/TFEB gene rearrangement by FISH, and underwent next-generation sequencing (NGS). On NGS correlation, 14 of 20 (70%) FISH-negative TRIM63-positive tumors harbored an MiTF gene rearrangement. In the remaining 6 cases, we were unable to fully ascertain the MiTF rearrangement status due to the inherent limitation of the NGS panel utilized. The cases with MiTF gene rearrangement include TFE3 rearrangement in 60% (12/20) and TFEB low-level copy gains (with an additional missense mutation in 1 case) in 10% (2/20) of samples. RBM10:TFE3 fusion was seen in 67% (8/12) of TFE3-rearranged RCC in this cohort. TRIM63 RNA ISH assay could aid in identifying cases that harbor TFE3 or TFEB rearrangement associated with false-negative or equivocal TFE3/TFEB FISH results, especially those involving gene fusions with a paracentric Xp11 inversion. Overall, employment of TRIM63 RNA ISH coupled with TFE3/TFEB FISH assays and follow-up genomic interrogation enhanced diagnostic accuracy for patients with MiTF RCC.

MitoTracer facilitates the identification of informative mitochondrial mutations for precise lineage reconstruction.

Mitochondrial (MT) mutations serve as natural genetic markers for inferring clonal relationships using single cell sequencing data. However, the fundamental challenge of MT mutation-based lineage tracing is automated identification of informative MT mutations. Here, we introduced an open-source computational algorithm called "MitoTracer", which accurately identified clonally informative MT mutations and inferred evolutionary lineage from scRNA-seq or scATAC-seq samples. We benchmarked MitoTracer using the ground-truth experimental lineage sequencing data and demonstrated its superior performance over the existing methods measured by high sensitivity and specificity. MitoTracer is compatible with multiple single cell sequencing platforms. Its application to a cancer evolution dataset revealed the genes related to primary BRAF-inhibitor resistance from scRNA-seq data of BRAF-mutated cancer cells. Overall, our work provided a valuable tool for capturing real informative MT mutations and tracing the lineages among cells.

A stomatin-domain protein essential for touch sensation in the mouse.

Touch and mechanical pain are first detected at our largest sensory surface, the skin. The cell bodies of sensory neurons that detect such stimuli are located in the dorsal root ganglia, and subtypes of these neurons are specialized to detect specific modalities of mechanical stimuli. Molecules have been identified that are necessary for mechanosensation in invertebrates but so far not in mammals. In Caenorhabditis elegans, mec-2 is one of several genes identified in a screen for touch insensitivity and encodes an integral membrane protein with a stomatin homology domain. Here we show that about 35% of skin mechanoreceptors do not respond to mechanical stimuli in mice with a mutation in stomatin-like protein 3 (SLP3, also called Stoml3), a mammalian mec-2 homologue that is expressed in sensory neurons. In addition, mechanosensitive ion channels found in many sensory neurons do not function without SLP3. Tactile-driven behaviours are also impaired in SLP3 mutant mice, including touch-evoked pain caused by neuropathic injury. SLP3 is therefore indispensable for the function of a subset of cutaneous mechanoreceptors, and our data support the idea that this protein is an essential subunit of a mammalian mechanotransducer.

Simultaneously inhibition of HIV and HBV replication through a dual small interfering RNA expression system.

Human immunodeficiency virus (HIV) is often acquired in individuals already infected with hepatitis B virus (HBV) as a result of shared routes of transmission. Since current options for the treatment of HIV and HBV infections are limited, there is an essential need for the development of effective therapies against HIV/HBV co-infections. RNA interference (RNAi) has been used as a powerful tool to silence genes in cells and animals. In this study, we developed a small interfering RNA generation system that expressed two different siRNAs to target the HBs gene of HBV and the gp120 gene of HIV in Bel-7402 and HEK293T cells, respectively. Our results demonstrated that the two siRNA molecules could simultaneously inhibit the expression of HBs and gp120 by 81% and 89%, respectively. In addition, dual siRNA molecules significantly decreased the production of HBs, and simultaneously inhibited the replication of HBV and HIV. This dual siRNA generation system not only proved to be a novel approach for studying functions of multiple genes simultaneously, but also provides a potential approach for the treatment and prevention of HIV and HBV co-infection.

Mechanosensitive currents in the neurites of cultured mouse sensory neurones.

Almost all sensory neurones in the dorsal root ganglia have a mechanosensory function. The transduction of mechanical stimuli in vivo takes place exclusively at the sensory ending. For cutaneous sensory receptors it has so far proved impossible to directly record the mechanically gated receptor potential because of the small size and inaccessibility of the sensory ending. Here we investigate whether mechanosensitive currents are present in the neurites of freshly isolated adult mouse sensory neurones in culture. Almost all sensory neurone neurites possess currents gated by submicrometre displacement stimuli (92%). Three types of mechanically activated conductance were characterized based on different inactivation kinetics. A rapidly adapting conductance was found in larger sensory neurones with narrow action potentials characteristic of mechanoreceptors. Slowly and intermediate adapting conductances were found exclusively in putative nociceptive neurones. Mechanically activated currents with similar kinetics were found also after stimulating the cell soma. However, soma currents were only observed in around 60% of cells tested and the displacement threshold was several times larger than for the neurite (approximately 6 microm). The reversal potential of the rapidly adapting current indicated that this current is largely selective for sodium ions whereas the slowly adapting current is non-selective. It is likely that distinct ion channel entities underlie these two currents. In summary, our data suggest that the high sensitivity and robustness of mechanically gated currents in the sensory neurite make this a useful in vitro model for the mechanosensitive sensory endings in vivo.

Mandibular distraction osteogenesis enhanced by bone marrow mesenchymal stem cells in rats.

OBJECTIVE: This study was to observe the effects of bone marrow mesenchymal stem cell transplantation on new bone formation in a rat mandibular osteodistraction model. MATERIAL AND METHODS: Autologous bone marrow stem cells were obtained from tibiae of 40 male rats. Two weeks after cell harvest, the rats underwent right mandibular distraction and were then randomly divided into two groups (group A=20, group B=20). After distraction was complete, the stem cells were injected into the distracted gaps in group A, while the rats in group B only received physiological saline. Twenty rats (10 from each group) were sacrificed on postoperative days 27 and 55, respectively. The distracted mandibles were harvested and processed for radiographic, histological and histomorphometric analysis. RESULTS: The radiodensity of the distraction zone was higher in group A than in group B at both time points. Histologically callus was found in both groups but more bone was formed in group A. Histomorphometric analysis also demonstrated that both new bone volume and thickness of the new trabeculae were significantly greater in group A than in group B. CONCLUSION: The results of this study suggest that autologous bone marrow stem cell transplantation may be considered as a potential method to accelerate bone regeneration in the distraction gap, and enhance consolidation.

Association of RNA helicase a with human immunodeficiency virus type 1 particles.

RNA helicase A (RHA) belongs to the DEAH family of proteins that are capable of unwinding double-stranded RNA structure. In addition to its involvement in the metabolism of cellular RNA, RHA has been shown to stimulate RNA transcription from the long terminal repeat promoter of human immunodeficiency virus type 1 (HIV-1) as well as to enhance Rev/Rev response element-mediated gene expression. In this study, we provide evidence that RHA associates with HIV-1 Gag in an RNA-dependent manner. This interaction results in specific incorporation of RHA into HIV-1 particles. Knockdown of endogenous RHA in virus producer cells leads to generation of HIV-1 particles that are less infectious in part as a result of restricted reverse transcription. Therefore, RHA represents the first example of cellular RNA helicases that participate in HIV-1 particle production and promote viral reverse transcription.

Autogenous coronoid process as a new donor source for reconstruction of mandibular condyle: an experimental study on goats.

OBJECTIVE: To evaluate the feasibility and effectiveness of autogenous coronoid process grafts for reconstruction of the mandibular condyle. STUDY DESIGN: Two groups of 9 goats each were used. Each animal underwent unilateral total condyle and disk removal and was treated by grafting an autogeneous coronoid process. The animals in group A were sacrificed at 4, 12, and 24 weeks after surgical procedure, and their grafted coronoid processes were harvested for histological observation. All animals in group B received 3-dimensional CT scanning examination at different times and were sacrificed at 24 weeks after surgical procedure. The items of ramus height (RH), transverse dimension of the condyle (CT), and posteroanterior dimension of the condyle (CP) were measured for comparison with the operated and nonoperated sides. RESULTS: Despite the differences in the measurements of RH, CT, and CP between the operated and nonoperated sides, the grafted coronoid process had a similar shape and histological structures to the normal condyle at 24 weeks after surgical procedure. The head of the neocondyle was covered by a cap of fibrous tissue, which might play the role of the articular disc. CONCLUSIONS: Under the mechanical stimuli of the temporomandibular joint site, the grafted coronoid process could gradually remodel to a neocondyle with functional shape and structure in goats. This animal study suggests that autogenous coronoid process could be considered as a new donor source for reconstruction of the mandibular condyle.

Surgical repair of hard palate cleft with absorbable membrane: the new surgical technique and its clinical application.

This study evaluated a new surgical technique with absorbable membrane to repair hard palate cleft without extensive mobilisation of the mucoperiosteum. From 2001 to 2002, 32 selected patients with complete unilateral clefts underwent this surgical operation. The traditional flap surgical operation was performed at the soft palate, uvula and anterior alveolar cleft. The absorbable membrane was implanted to the hard palate cleft gap to guide the regeneration of the mucoperiosteum. The patients were followed up for 1-6 months after the operation. The speech assessment was carried out 12 months after the operation. Of 32 patients, 30 were successfully operated by this method and no obvious complications occurred. Primary healing on tissue defect of hard palate was obtained in 27 patients and secondary healing in 3 patients. Eighty percent of the 30 patients had good or excellent speech 12 months after the operation. The operation failed in 2 patients. The surgical technique with absorbable membrane to repair hard palate appears to have several valuable advantages including the decreased area of the hard palate involved and favourable outcome for speech in the majority of cases.

Assessment of long-range correlation in time series: how to avoid pitfalls.

Due to the ubiquity of time series with long-range correlation in many areas of science and engineering, analysis and modeling of such data is an important problem. While the field seems to be mature, three major issues have not been satisfactorily resolved. (i) Many methods have been proposed to assess long-range correlation in time series. Under what circumstances do they yield consistent results? (ii) The mathematical theory of long-range correlation concerns the behavior of the correlation of the time series for very large times. A measured time series is finite, however. How can we relate the fractal scaling break at a specific time scale to important parameters of the data? (iii) An important technique in assessing long-range correlation in a time series is to construct a random walk process from the data, under the assumption that the data are like a stationary noise process. Due to the difficulty in determining whether a time series is stationary or not, however, one cannot be 100% sure whether the data should be treated as a noise or a random walk process. Is there any penalty if the data are interpreted as a noise process while in fact they are a random walk process, and vice versa? In this paper, we seek to gain important insights into these issues by examining three model systems, the autoregressive process of order 1, on-off intermittency, and Lévy motions, and considering an important engineering problem, target detection within sea-clutter radar returns. We also provide a few rules of thumb to safeguard against misinterpretations of long-range correlation in a time series, and discuss relevance of this study to pattern recognition.

Evidence for histamine as a neurotransmitter in the cardiac sympathetic nervous system.

The colocalization of histamine (HA) and norepinephrine (NE) immunoreactivities was identified within the superior cervical ganglia neurons of the guinea pig. HA and NE immunoreactivity levels were significantly attenuated after chemical sympathectomy with 6-hydroxydopamine (6-OHDA). Coexistence of NE and HA was also visualized in the cardiac sympathetic axon and varicosities labeled with anterograde tracer biotinylated dextran amine. Depolarization of cardiac sympathetic nerve endings (synaptosomes) with 50 mM potassium stimulated endogenous HA release, which was significantly attenuated by 6-OHDA or a vesicular monoamine transporter 2 (VMAT2) inhibitor reserpine pretreatments. Compound 48/80, a mast cell releaser, did not affect cardiac synaptosome HA exocytosis. Furthermore, K+ -evoked HA release was abolished by the N-type Ca2+ -channel blocker omega-conotoxin but was not affected by the L-type Ca2+ -channel blocker lacidipine. Cardiac synaptosome HA exocytosis was augmented by the enhanced synthesis of HA or the inhibition of HA metabolism. HA H3-receptor activation by (R)-alpha-methylhistamine inhibited high K+ -evoked histamine release. The HA H3 receptor antagonist thioperamide enhanced K+ -evoked HA release and blocked the (R)-alpha-methylhistamine effect. The K+ -evoked endogenous NE release was attenuated by preloading the cardiac synaptosomes with L-histidine or quinacrine. These inhibitory effects were reversed by thioperamide or antagonized by alpha-fluoromethylhistidine. Our findings indicate that high K+ -evoked corelease of NE and HA may be inhibited by endogenous HA via activation of presynaptic HA H3-receptors. The H3-receptor may function as an autoreceptor, rather than a heteroreceptor, in the regulation of sympathetic neurotransmission and HA may be a novel sympathetic neurotransmitter.

Characterization of a rice (Oryza sativa L.) gene encoding a temperature-dependent chloroplast omega-3 fatty acid desaturase.

A cDNA, designated Osfad8, encoding a chloroplast omega-3 fatty acid desaturase responsible for trienoic fatty acid formation, was isolated from the leaves of Oryza sativa L. by RT-PCR. Southern blot hybridization indicated that a small gene family composed of two copies or closely linked genes exists. RNA in situ hybridization showed that the accumulation of Osfad8 mRNA was abundant in leaves but hardly detectable in roots. The Osfad8 transcript level in leaves was much higher at 15 degrees C than at normal temperature (25 degrees C). In situ hybridization also showed particularly prominent expression of Osfad8 in the palisade layer and spongy parenchyma cells of leaves when exposed to 15 degrees C conditions for 5 days and 10 days. Two transgenic lines (8S-52 and 8S-101) harboring the Osfad8 ORF in sense orientation under the control of the CaMV 35S promoter contained increased levels of hexadecatrienoic (16:3) and linolenic (18:3) fatty acids. When exposed to 2 degrees C for 7 days, the damage observed to the control plants was significantly alleviated in the 8S-52 and 8S-101 lines. The amounts of trienoic fatty acids in an Osfad8 antisense line (8A-35) declined 40.2% compared to the control plants. The 8A-35 plants survived after growth at 44 degrees C for 3 days while the control plants died. These data suggest that Osfad8 encodes a temperature-dependent chloroplast omega-3 fatty acid desaturase.

Reconstruction of mandibular condyle by transport distraction osteogenesis: Experimental study in rhesus monkey.

PURPOSE: This study investigated histomorphologic changes in the newly formed condyle reconstructed by transport distraction osteogenesis through a nonhuman primate model. MATERIALS AND METHODS: Six adult rhesus monkeys were used in this study. Bilateral condyles and articular discs were extirpated, and the technique of transport distraction osteogenesis was carried out to reconstruct the condyles. X-ray and 3-dimensional (3D) computed tomography (CT) scanning films were taken at various intervals. Two monkeys were killed respectively at 4, 12, and 24 weeks after completion of distraction; the transport segments and the distracted calluses were harvested and processed for histologic examination. The original condyles removed at surgery served as normal control. RESULTS: Open bite was seen in all animals postoperatively and diminished at the end of distraction. Bone regeneration was perfect in the distraction gap between the transport disc and the pre-existing mandible. The bony transport disc gradually remodeled to a neocondyle that was similar to the original condyle in the appearances and structures. The head of neocondyle was covered with a fibrous cap, which might play the role of an articular disc. CONCLUSION: A neocondyle with functional shape can be created by transport distraction osteogenesis. This suggests that this technique is an alternative method for reconstruction of the mandibular condyle.

Reproductive outcome of septate uterus after hysteroscopic treatment with neodymium:YAG laser.

OBJECTIVE: The aim of this study was to assess fertility outcome and obstetrical prognosis of 46 patients after hysteroscopic section of uterine septa with neodysmium:yttrium-aluminum-garnet (Nd:YAG) laser. BACKGROUND DATA: Approximately 15-25% of spontaneous miscarriages are related to Müllerian fusion defects, most of which are associated with uterine septa. Operative hysteroscopy is a safe and effective treatment for uterine septa. METHODS: Forty-six sterile patients with septate uterus received hysteroscopic metroplasty with Nd:YAG laser. Forty-one patients with incomplete septate uterus were under the guidance of B-ultrasonography, while the other 5 patients with complete septate uterus were treated by laparoscopy. RESULTS: The metroplasty was successfully conducted in 45 patients, with a success rate of 98% (45 /46). Postoperative follow-up examinations of 28 patients revealed pregnancy within 12 months in 20 patients, 16 of whom acquired term delivery. The pregnancy rate within 1 year was 71%. CONCLUSION: Hysteroscopic treatment of septate uterus with Nd:YAG laser is effective. The cervix may not be excessively dilated, the intraoperative bleeding can be thoroughly controlled, and the procedure under local anesthesia is relatively safe.

Analysis of biomedical signals by the lempel-Ziv complexity: the effect of finite data size.

The Lempel-Ziv (LZ) complexity and its variants are popular metrics for characterizing biological signals. Proper interpretation of such analyses, however, has not been thoroughly addressed. In this letter, we study the the effect of finite data size. We derive analytic expressions for the LZ complexity for regular and random sequences, and employ them to develop a normalization scheme. To gain further understanding, we compare the LZ complexity with the correlation entropy from chaos theory in the context of epileptic seizure detection from EEG data, and discuss advantages of the normalized LZ complexity over the correlation entropy.
