[Genetic diversity of F-ATPase subunits gene uncA amplified from Streptococcus mutans clinical isolates].

OBJECTIVE: To study the genetic diversity of F-ATPase alpha subunit gene uncA derived from Streptococcus mutans (S. mutans) clinical isolates and to investigate the relationship between the genetic diversity of acidurance factor and S. mutans aciduric ability, also and the cariogenicity. METHODS: Sixty-four S. mutans strains derived from 34 caries-active individuals and 30 caries-free individuals, including 18 strains displaying high acid tolerance and 20 strains displaying low acid tolerance. Gene uncA was amplified with specific primers from S. mutans genomic DNA, then the PCR products were analyzed by RFLP and sequenced. RESULTS: Two genotypes A and B of PCR-RFLP were revealed when digested with Hph I. Mbo II also produced two different pattern C and D. The distributions of A and B genotype strains with different caries-sensitivity groups were different (P < 0.05), and the proportion of A genotype strains from caries-activity group was higher than that from caries-free one. The distributions of C and D genotype strains with different acidurance strains were different (P < 0.05), and the proportion of C genotype strains from high acid tolerance group was higher than that from low acid tolerance group. These amplified uncA genes from different group were sequenced and there existed variation of Hph I and Mbo II recognized sites. CONCLUSIONS: This study indicates that uncA gene of S. mutans F-ATPase obviously displayed genetic diversity. The different Hph I-RFLP and Mbo II-RFLP genotypes could be related to the cariogenicity and acid tolerance of S. mutans strains.

[Passive FTIR remote sensing of gaseous pollutant in heated plume].

The principle and techniques of passive remote sensing of gaseous pollutant in heated plume was illustrated and discussed in this paper. The algorithm of radiance spectra and transmittance spectra in measured region was proposed, and the method of retrieving gas concentrations with NLLS fitting algorithm was also proposed. The remote senseing of actual gaseous pollutant of smokestack was done, and the quantitative analysis of SO2 and CO2 was completed.

[Concentration calibration method of ambient trace-gas monitoring with tunable diode laser absorption spectroscopy].

Tunable diode laser absorption spectroscopy (TDLAS) is a new method to detect trace-gas qualitatively or quantificationally based on the scan characteristic of the diode laser used to obtain the absorption spectroscopy in the characteristic absorption region It needs to be combined with a long absorption path in the ambient trace-gas measurements. TDLAS is a new trace gas detective method developed with the combination of a tunable diode laser source and a long absorption path; it has significant advantages not only in the sensitivity but also in rapidity of response. It has been widely used in many atmospheric trace-gases detection, ground trace-gas detection and, gas leakage detection. On-line calibrating is necessary to most trace gas monitor, and in the present paper the authors introduced a simple and accurate method, analyzed it in the theory, and proved it's feasibility in the experiment.

[Error analysis and calibration method of complex spectrum of Fourier transform infrared spectrometer].

The phase shift in complex spectrum of Fourier transform infrared (FTIR) spectrometer was discussed. A rigorous derivation showed the relation between the phase shift and complex spectrum. The reason for the radiation offset in FTIR measurement was analysed. The present article presents a calibration method of FTIR complex spectrum, which can adjust the radiation offset. The comparison between experiment data and theoretical computation showed that the method is precise.

[Study on the genetic diversity within the extended-V region of the surface protein of Streptococcus mutans].

PURPOSE: The purpose of this study was to investigate the genicity of the extended- variable region (V+) of surface proteins of Streptococcus mutans (S. mutans, serotype c,f) and provide some genetic informations on the molecular structure of S. mutans surface protein. METHODS: 7 standard strains and 110 clinical isolates of S.mutans were selected.The bacterial DNA was extracted and the extended-V region (srV+ 1384-2514 bp) was amplified using polymerase chain reaction (PCR). The genetic diversity of srV+ was assessed by using restriction fragment length polymorphism (RFLP) with restriction endonuclease DdeI. RESULTS: The results showed that five different patterns (A,B,C,D,E) of the PCR-RFLP among the 117 strains were reveled by DdeI. The ration of the genotype A and B were the most among the strains, C followed by genotype, the D and E were 1 and 3 strains per genotype, respectively.OMZ175 (serotype f) belonged to B genotype. CONCLUSIONS: The study supported that the extended-variable region (srV+) of the surface proteins of Streptococcus mutans have diversity. There are five patterns of genotypings. The most genotypings were p1-like,spaP /sr-like and pac-like.

[The fluorescence emission of water at upconversion of frequency by laser induced fluorescence].

The fluorescence spectra of several types of water samples in combination with laser-induced fluorescence (LIF) measurements using 532 nm wavelength excitation source were measured in the laboratory, and the fluorescence matters such as dissolved organic matter (DOM), chlorophyll a (Chl a) and carotenoids were pointed out with spectral fluorescent signature (SFS) technique. It is considered that the peak at 455 nm in the spectrum is contributed by carotenoid (PPC). Finally, the dynamic model of PPC's fluorescence emission is showed.

[The study of infrared background radiation of earth and atmosphere by airborne FTIR spectrometer].

The present article describes the experiment of airborne Fourier transform infrared (FTIR) spectrometer detecting the background radiation of earth and atmosphere. Considering the surface radiation and atmospheric absorbance and emission, the expression of radiations measured by airborne FTIR is given. It is indicated that the background radiations depend on four parameters: surface temperature, surface emissivity, atmospheric transmission and temperature. Based on theoretic analysis the measured spectra are compared and analyzed according to the differences in altitude and surface. Spectral characters of infrared background radiation are described.

[The influence of temperature change on determined concentrations of dissolved organic matter in water].

The influence of temperature change on the determined concentrations of dissolved organic matter (DOM) in water was investigated by laser induced fluorescence (LIF) technique in laboratory. The temperature was altered within the range of 20 to 75 degrees C, and the intensity of DOM fluorescence and water Raman scattering were found to decrease with rising temperature. A linear fit of the normalized fluorescence intensity versus temperature yielded a mean value of the temperature coefficient of -5.24 x 10(-4) x degrees C(-1), and a mean value of the temperature coefficient of -3.42 x 10(-3)(mg x L(-1)) x degrees C(-1) was obtained according to the relationship between normalized fluorescence intensity and concentration of DOM. The temperature change will cause relative changes of up to 8.45% in DOM concentration within the range of 20 to 75 degrees C if we assume that the normalized fluorescence intensity remains the same.

[A study of genetic diversity in lactate dehydrogenase of Streptococcus mutans from clinical isolates].

OBJECTIVES: Lactate dehydrogenase (LDH) is shown to be an important virulence factor resulting in acid production of Streptococcus mutans (S. mutans), on which the cariogenic potential of S. mutans depends. Differences in cariogenic abilities of S. mutans isolates may be determined by genetic heterogeneity from virulent factors. The relationship between LDH activity or genetic diversity and cariogenicity of S. mutans (serotype c) isolates was studied in this research. METHODS: The genome DNA of S. mutans isolates were isolated and LDH gene (ldh) were amplified with specific primers. These isolates came from 34 caries-active individuals and 36 caries-free ones, in which 24 strains showed the high LDH activity and 21 strains showed the low LDH activity. Then genetic diversity of PCR products were analyzed or assessed by restriction fragment-length polymorphism (RFLP). Some of amplified LDH genes from different group were sequenced and assayed. RESULTS: It is testified that two genotypes A and B of ldh-RFLP were revealed when LDH genes were digested with Mse I, but Hph I, Mnl I, Dde I, Nla III and Alu I digesting fragments of Idh gene did not show different pattern. Furthermore, Fisher Exact one-Tail Test showed that the proportion of genotype B among strains from caries-free individuals was higher than that from caries-active ones (P = 0.033), while the distribution of genotypes with different LDH activity was different between two groups (two-Tail Test P = 0.017). The sequencing DNA testified that the specific base mutation would lead to multiple kind of genotype resulted. CONCLUSION: This study indicated that LDH gene of S. mutans clinical strains from different individuals is conservative, while there still is the gene mutation in. The ldh genetic diversity may be related to the low caries sensitivity, and closely correlated with the differences in LDH enzyme activity of S. mutans strains.

[Impact of hydrothermal process on the dewaterability and degrease performance of restaurant garbage].

In order to ameliorate the dewaterability and degrease performance of restaurant garbage, and to improve the treatment effect, a complete trail series with 2 factors on 5 levels was implemented. The 2 factors were temperature and retention time respectively as the main influencing factors of hydrothermal process. By means of analyzing the variation of the resistance, dewatering rate and floatable oil content of the treated restaurant garbage, and constructing the solid grease extracting kinetics, the mechanism of impact of hydrothermal process on the dewaterability and degrease performance of restaurant garbage was studied. It showed that the dewaterability of the product decreases at the beginning, after heating for 40min, it begins to increase. Moreover, it increases more quickly as temperature increases. The optimal dewaterability of the treated garbage appears at 180 degrees C and heating for 100 minutes. As temperature rises and heating time increases, the degrease performance is improved. Furthermore, this trend becomes more remarkably as temperature increases. When the temperature and retention time reach 160 degrees C and 80 minutes respectively, most of the solid grease in the garbage is extracted out to become floatable oil which can be separated and recovered readily. Subsequently, the amount of floatable oil begins to decrease since chemical reactions such as the partial hydrolysis of the oil take place. Additionally, the extraction of solid grease from interior accords with first-order reaction dynamic model.

[Determination of the error of aerosol extinction coefficient measured by DOAS].

The method of defining the error of aerosol extinction coefficient measured by differential optical absorption spectroscopy (DOAS) is described. Some factors which could bring errors to result, such as variation of source, integral time, atmospheric turbulence, calibration of system parameter, displacement of system, and back scattering of particles, are analyzed. The error of aerosol extinction coefficient, 0.03 km(-1), is determined by theoretical analysis and practical measurement.

[Relationship of the genetic diversity within the V-region, P-region and C-terminus of surface protein of Streptococcus mutans to dental caries].

OBJECTIVE: To gain an understanding of the relationship between the genetic diversity within the V-region, P-region, C-terminus of surface protein of Streptococcus mutans (serotype c) and their adherent abilities. METHODS: The clinical isolates of S. mutans (serotype c) in two groups with different adherent abilities (cpm>2000, cpm<1000) had been prepared in our experimental laboratory. The genome DNA was extracted, and the spaP-pv (2060-3157 bp) and spaP-c (4003-4851 bp) were amplified by PCR, respectively. Genetic diversity was assessed by restriction fragment-length polymorphism (RFLP) with restriction endonucleases Alu I. RESULTS: (1) Two different patterns of spaP-pv RFLP among strains were revealed after the amplified product being digested with Alu I. The distributions of two genotypes (a and b) in the clinical strains with different adherent abilities differed significantly (P<0.05). The proportion of genotype b in the strains with higher adherent abilities was higher than that in the strains with lower adherent abilities. (2) Two different patterns (c and d) of spaP-c RFLP among strains were also revealed after the spaP-c being digested with Alu I. No statistically significant difference was observed in the distribution of the two genotypes in the clinical strains. CONCLUSION: The genetic diversity within spaP-pv of clinical isolates of S. mutans might be related to the differences of their adherent abilities.

[Passive remote sensing of VOC in atmosphere by FTIR spectrometry].

The paper expatiates the passive remote sensing experiment of VOC in atmosphere by FTIR spectrometry, a mode of simulated spectra for pollution gas in a complicated environment is proposed, and the detecting limit of passive FTIR remote sensing is discussed. Using this measurement technique and the data analysis method, the authors can obtain column density and effective radiation temperature of target-gas that does not require a previously measured background spectrum.

[Study on characteristics of fluorescence spectra of dissolved organic matter with different water quality].

The fluorescence spectra of dissolved organic matter (DOM) in several types of water samples in combination with laser-induced fluorescence (LIF) measurements were measured in the laboratory, and the spectral characteristics of DOM were analysed. The curve of normalized fluorescence intensity against corresponding concentration of humic acid is showed. The results demonstrate the possibilities of water quality monitoring based upon fluorescence spectral characteristics of DOM by means of LIF method.

[Study the characteristics of dissolved organic matter and turbidity of water using laser induced fluorescence and laser scattering].

In the experiment a frequency doubled radiation of Nd:YAG laser was used for excitation at 532 nm. The fluorescence spectra of dissolved organic matter (DOM) and chlorophyll a (Chl-a) in several types of water samples with laser-induced fluorescence (LIF) measurements using a 532 nm wavelength excitation source were measured in the laboratory, and the spectral characteristics of DOM and Chl-a were analysed. The characteristics of turbidity were investigated by measuring the amount of scattering light of suspended particles in the water volume. The curve of scattering intensity against corresponding turbidity of SiO2 is showed. The possibilities of water quality monitoring based upon the fluorescence spectral characteristics of contaminations and the turbidity by means of LIF method.

[Study on UV-visible DOAS system based on photodiode array (PDA)].

A long-path differential optical absorption spectroscopy (DOAS) system is introduced. A photodiode array is employed as the detector to replace the complicated SD detector which consists of a PMT and a slotted disk. The properties of the detector and the spectrometer unit such as offset, dark current, noise, linearity, resolution, and wavelength range were measured. This system was also tested to measure SO2 and NO2 in the atmosphere. The detection limits of this system for SO2, and NO2 over a 713 m light path were determined.

[Application of FTIR spectra fitting method in retrieving gas concentrations].

The method of retrieving gas concentrations by FTIR spectra measurement system is studied. NLLS fitting algorithm is used to complete the quantitative analysis of CO gas under WINDOWS operating system. The spectra of HITRAN database is employed as calibrating and training sets during the procedure of fitting FTIR spectrum. The measured IR transmittance spectra of CO are best fitted with the calculated spectra in HITRAN. The CO concentrations are acquired after the best fitting. The absolute accuracy of the results of retrieve achieves 1%-5%.

Frequent ventricular premature beats increase blood pressure variability in rats.

AIM: The present study was designed to test a hypothesis that nonfatal ventricular arrhythmia such as ventricular premature beats (VPB) is a contributing factor in the elevation of blood pressure variability (BPV). METHODS: Blood pressure (BP) and electrocardiogram were continuously recorded. The relation between VPB and BPV was observed under conscious state in chronic myocardial infarction (MI) rats one month after ligation of the left coronary artery, and further verified under anesthetized state in rat model of ventricular arrhythmia produced by acute intravenous infusion of aconitine. RESULTS: MI rats exhibited a big difference in the count and pattern of VPB, and were divided into no VPB, occasional VPB, and frequent VPB groups. Among the three groups, there were no differences in BP, heart period (HP), and MI size. However, BPV was markedly higher in frequent not occasional VPB rats, and HP variability (HPV) was larger in both frequent and occasional VPB rats, when compared with no VPB rats. In the whole population of MI rats, BPV was positively correlated with VPB and HPV, not with BP, HP and MI size. Infusion of aconitine had no effect on BP, HP, BPV, and HPV during the period without VPB. Frequent VPB after several minutes of aconitine infusion induced significant increase in BPV and HPV with no change in BP and HP. BPV was also positively correlated with VPB and HPV, not with BP and HP. Hemodynamics in aconitine-evoked ventricular tachycardia was characterized as lower BP, higher BPV, and higher HPV. CONCLUSION: High BPV can be caused by frequent not occasional VPB in rats.
