Polyamine biosynthesis of apple callus under salt stress: importance of the arginine decarboxylase pathway in stress response.

To clarify the involvement of the arginine decarboxylase (ADC) pathway in the salt stress response, the polyamine titre, putrescine biosynthetic gene expression, and enzyme activities were investigated in apple [Malus sylvestris (L.) Mill. var. domestica (Borkh.) Mansf.] in vitro callus under salt stress, during recovery after stress, and when ADC was inhibited by D-arginine, an inhibitor of ADC. Salt stress (200 mM NaCl) caused an increase in thiobarbituric acid-reactive substances (TBARS) and electrolyte leakage (EL) of the callus, which was accompanied by an increase in free putrescine content, during 7 d of treatment. Conjugated putrescine was also increased, but this increase was limited to the early stage of salt stress. Accumulation of putrescine was in accordance with induction of ADC activity and expression of the apple ADC gene (MdADC). When callus that had been treated with 200 mM NaCl was transferred to fresh medium with (successive stress) or without (recovery) NaCl, TBARS and EL were significantly reduced in the recovery treatment, indicating promotion of formation of new callus cells, compared with the successive stress treatment. Meanwhile, MdADC expression and ADC activity were also decreased in the callus undergoing recovery, whereas those of the callus under successive stress were increased. Ornithine decarboxylase (ODC) activity showed a pattern opposite to that of ADC in these conditions. D-Arginine treatment led to more serious growth impairment than no treatment under salt stress. In addition, accumulation of putrescine, induction of MdADC, and activation of ADC in D-arginine-treated callus were not comparable with those of the untreated callus. Exogenous addition of putrescine could alleviate salt stress in terms of fresh weight increase and EL. All of these findings indicated that the ADC pathway was tightly involved in the salt stress response. Accumulation of putrescine under salt stress, the possible physiological role of putrescine in alleviating stress damage, and involvement of MdADC and ADC in response to salt stress are discussed.

Effects of tetrandrine on cytosolic free calcium concentration in corpus cavernosum smooth muscle cells of rabbits.

AIM: To study the relaxation mechanisms of tetrandrine (Tet) on the corpus cavernosum smooth muscle. METHODS: The corpus cavernosum smooth muscle cells from New Zealand white rabbits were cultured in vitro. [Ca(2+)](i) was measured by Fluorescence Ion Digital Imaging System, using Fluo-2/AM as a Ca(2+)-sensitive fluorescent indicator. RESULTS: Tet (1, 10 and 100 micromol/L) had no effect on the resting [Ca(2+)](i) (P>0.05). In the presence of extracellular Ca(2+) (2.5 mmol/L), Tet (1, 10 and 100 micromol/L) inhibited [Ca(2+)](i) elevation induced by high K(+) and phenylephrine (PE) in a concentration-dependent manner (P>0.05). In calcium free solution containing egtaic acid, Tet (1 and 10 micromol/L) had no inhibitory effects on [Ca(2+)](i) elevation induced by PE (P>0.05). However, Tet (100 micromol/L) inhibited [Ca(2+)](i) elevation induced by PE (P>0.05). CONCLUSION: Tet inhibited the Ca(2+) influx from the extracellular site via voltage-activated Ca(2+) channel and alpha(2)-adrenoceptor-operated Ca(2+) channel. At a high concentration, Tet might inhibit the cytosolic calcium pool release in cultured corpus cavernosum smooth muscle cells. This inhibitory action on [Ca(2+)](i) might be one of the relaxation mechanisms of Tet on the corpus cavernosum smooth muscle.

[Effects of different immunodepressants on the sperm parameters of kidney transplant recipients].

OBJECTIVE: To study the effects of different immunodepressants on the sperm parameters of kidney transplant recipients. METHODS: In 15 healthy fertile men and 37 kidney transplant recipients, ejaculates were aseptically obtained by masturbation. Thirty-seven patients were divided into two groups, 20 patients were treated with Prograf (FK506) combination with mycophenolate mofetil (MMF) and prednisone; 17 patients were treated with cyclosporine (CsA) combination with azathioprine with prednisone. The sperm viability, mobility parameters such as prorsad percentage motility, straight line velocity (VSL), curve line velocity (VCL), velocity of average path (VAP) and morph were estimated with a computer-assisted sperm analyzer (CASA) provided with a multiple-exposure photography system. RESULTS: There were no significant difference in sperm viability rate [(81.7 +/- 5.7)%, (79.4 +/- 6.8)% and (83.8 +/- 6.0)%], VCL [(24.1 +/- 8.6)%, (23.9 +/- 4.4)%, (24.8 +/- 4.2)% ] and VAP [(19.7 +/- 6.6)%, (18.6 +/- 2.9)%, (21.0 +/- 4.0)%] among groups of FK506, CsA and control, respectively (P > 0.05). The rate of anomaly [(67.8 +/- 5.7)%], the prorsad percentage motility [(46.4 +/- 8.1)%] and VSL [(15.4 +/- 4.6)%] in the group of FK506 were respectively significantly lower and higher than those in the group of CsA [(80.1 +/- 5.6%, (33.3 +/- 6.4)%, (10.2 +/- 2.4)%] (P < 0.05). CONCLUSION: The application of FK506 combined with MMF could help recover the mobility and morphology of the sperm in kidney transplantation recipients.

[Influence of captopril on human sperm motility parameters in vitro].

OBJECTIVE: To determine the effects of captopril on human sperm motility in vitro. METHODS: Sperm specimens were aseptically obtained by masturbation and prepared by Percoll gradient-centrifugation technique to produce a spermatozoa suspension of high motility from 12 healthy fertile men. The spermatozoa suspension were incubated with captopril at 1:9 ratio of volume at 37 degrees C in vitro, which was administrated at three different concentrations. Measurements were carried out within 5 minutes for all specimens including VAP, VSL, VCL, STR, ALH and progressive motility. RESULTS: Within 5 minutes, three different concentrations of captopril could change the human spermatozoa motility and forward motile sperm percentage, but captopril did not decrease the other motility parameters, including VSL, VCL and VAP etc. The high concentration group could decrease the VCL more than those of the other two groups and it seemed that the velocity parameters of the middle concentration group were higher than those of the other two groups, but there were no significant difference statistically. CONCLUSION: It suggests that captopril can decrease the human spermatozoa motility and forward motile sperm percentage, but it can not directly affect the spermatozoa velocity in vitro.

[Molecular identification of Ca2+ channels in human ejaculated sperm].

OBJECTIVE: To identify the types of voltage-dependent Ca2+ channels (VDCCs) in human ejaculated sperm using molecular technology. METHODS: Based on the WHO criteria, the donated semen was screened by computer-aided sperm analysis (CASA), the sperm optimized by swim-up method, and the VDCCs' alpha1 subunits examined by reverse transcription polymerase chain reaction (RT-PCR). RESULTS: Such mRNA messages as alpha1H, alpha1G, alpha1E, alpha1B and alpha1C were detected, but alpha1A and alpha1D were not. CONCLUSION: Human motile sperms express multiple VDCC RNAs, among which T-type and non-L-type channel messages are likely to be predominantly expressed. Not only T-type but also non-L-type calcium channels may be major gates for the external calcium influx, which helps regulate sperm motility and acrosome reaction.

[Chromosomal imbalance in primary lung squamous cell carcinoma and their relationship with smoking].

BACKGROUND & OBJECTIVE: Chromosomal imbalance plays an important role in tumorigenesis of lung cancer, and may be influenced by different carcinogens. This study was to examine chromosomal imbalance in primary lung squamous cell carcinoma (LSCC), and their association with smoking. METHODS: Chromosomal gains and losses in 39 specimens of LSCC were identified by comparative genomic hybridization (CGH), the association between chromosomal imbalances in LSCC and smoking was analyzed. RESULTS: The most frequent chromosomal gains of LSCC were detected on chromosomal arms 3q (74.4%, 29/39), 5p (66.7%, 26/39), 1q (43.6%, 17/39), 8q (41.0%, 16/39), 12p (42.6%, 18/39), 2p (38.5%, 15/39), and 18p (33.3%, 13/39), with minimal amplified regions (MAR) at 3q26.2-29 (74.4%, 29/39), 5p14.3-15.3 (66.7%, 26/39), 1q41-44(41.0%, 16/39), 8q23 (41.0%, 16/39), 12p13 (41.0%, 16/39), and 18p11.2 (33.3%, 13/39)u high-copy-number amplification at chromosomal arms 3q, and 5p were found in 15 (38.5%), and 6 (15.4%) patients. Chromosomal losses mainly involved chromosomal arms 3p (56.4%, 22/39), 5q (53.8%, 21/39), 13q (51.3%, 20/39), 8p (46.1%, 18/39), 4p (43.6%, 17/39), 4q (43.6%, 17/39), 1p (41.0%, 16/39), 2q (38.5%, 15/39), 9q (35.9%, 14/39), 13p (35.9%, 14/39), 16p (35.9%, 14/39) ,6p (33.3%, 13/39), and 6q (30.7%, 12/39), with minimal deleted regions (MDR) at 3p14.2-21.2 (51.3%, 20/39), 5q15-22 (51.3%, 20/39), 13q14.2-21.2 (48.7%, 19/39), 8p21.1-22 (43.6%, 17/39), 2q32 (35.9%, 14/39), and 16p12-13.1 (33.3%, 13/39). Amplification rates of chromosomal arms 3q, and 8q in smoking LSCC patients were significantly higher than those in non-smoking LSCC patients (P=0.002,P=0.031). While high incidences of gains at chromosomal arms 5p and 12p, and of losses at chromosomal arms 3p, 4q, and 5q were the common feature of chromosomal changes of smoking and non-smoking LSCC patients. CONCLUSION: 3q, 5p, 1q, 8q, 12p, 2p, 18p gains and 3p, 5q, 13q, 8p, 4p, 4q, 1p, 2q, 9q, 13p,16p, 6p, 6q loses might be involved in tumorigenesis and/or progression of LSCC, smoking-induced lung cancer may be associated with 3q, 8q gains.

[Microcirculation changes in the ventral prostates of rats after castration].

OBJECTIVE: To study the microcirculation changes in the ventral prostates of rats after castration and the role of microcirculation during the apoptosis of prostatic cells. METHODS: Thirty-six male adult rats were randomized to 6 groups: one was taken as the control, while the other 5 underwent measurement of the microcirculation in vivo by a D95 physiological signal acquisition system 12 h, 24 h, 72 h, 7 d and 14 d respectively after castration, and then were perfused with Chinese ink to trace the microvessels of the prostates. RESULTS: The microcirculation of the rats'prostates changed dramatically following castration. The diameter and density of the microvessels, especially in the distant and mediate ducts of the prostates, decreased dramatically, and so did the bloodflow velocity. CONCLUSION: The microcirculation plays a role during the process of apoptosis of prostatic cells, and might be the mechanism of " apoptosis shift".

[Effect of photodynamic therapy with metalloporphyrin compound on human prostate cancer PC-3 cells in vitro].

OBJECTIVE: To investigate the effect of the photodynamic therapy (PDT) with the new water-soluble metalloporphyrin compound on human prostate cancer PC-3 cells in vitro and the anticancer mechanism of PDT. METHODS: The new water-soluble manganese, 5,10,15, 20-tetra (N-methyl4-pyridyl) porphinato (2-) tetraiodide salt, was synthesized. The PC-3 cells were treated with the compound of serial concentrations(0, 0.1, 1, 1.0 micromol/L) followed by irradiation of different dosages of visible light. The techniques of MTT and Annexin-V/propidium iodide double-labeled flow cytometry (FCM) were applied to measuring the inhibitory effect of the compound on the growth activity and apoptosis of the cells. RESULTS: When the metalloporphyrin compound concentration was within 10 micromol/L and the irradiation time was within 30 min, the water-soluble metalloporphyrin compound had a significant inhibitory effect on the proliferation of PC-3 cells and induced PC-3 cell apoptosis, and the effects depended greatly on metalloporphyrin concentration and illumination dosages. Higher concentrations and dosages induced the death of the majority of PC-3 cells. CONCLUSION: The PDT of the water-soluble metalloporphyrin compound followed by light irradiation has a distinctive killing effect on PC-3 cells in vitro, and the rates of proliferation inhibition and cell apoptosis are correlated with metalloporphyrin concentration and the dosages of light irradiation. The results suggest that the mechanism of metalloporphyrin PDT may be involved with the induction of apoptosis in human prostate cancer cells.

[Recurrence risk factors of platinum-sensitive epithelial ovarian cancer].

BACKGROUND & OBJECTIVE: The prognosis of platinum-sensitive ovarian cancer patients was better than that of chemoresistant ones. However, platinum-sensitive ovarian cancer patients still have a high recurrence rate, which affects their prognosis. This study was designed to analyze clinical features and recurrence risk factors of platinum-sensitive epithelial ovarian cancer. METHODS: Factors that might relate to recurrence of 90 platinum-sensitive epithelial ovarian cancer patients, admitted in Cancer Center of Sun Yat-sen University from 1993 to 1999, with complete remission of more than 6 months, were assessed. Univariate analysis was performed using Chi(2) test; while multivariate analysis was carried out using Cox proportional hazard model. RESULTS: Among the 90 patients, 36(40.0%) relapsed with the median recurrence-free interval of 20 months. Pelvic cavity (18/36, 50.0%) was the most frequently involved. The 3-and 5-year survival rates of all patients were 79.6% and 69.5%; while those of the recurrent ones were 62.3% and 39.6%. Univariate analysis showed that the early FIGO stage group, mucinous type group, and no neoadjuvant chemotherapy group had lower recurrence rates than advanced FIGO stage group, non-mucinous type group, and neoadjuvant chemotherapy group, respectively (P=0.001, P=0.002, and P=0.025). Cox multivariate analysis showed that only FIGO stage was the independent risk factor of recurrence of ovarian cancer (risk ratio=1.771, P=0.003). There was no significant difference in recurrence rate between CBP and other postoperative chemotherapy regimen groups. More cycles of chemotherapy could not reduce the recurrence rate. CONCLUSION: Since FIGO stage is an independent recurrence risk factor of platinum-sensitive epithelial ovarian cancer patients, early diagnosis is the key point to decrease the recurrence rate.

[1H-MRS of the hippocampus of psychogenic erectile dysfunction].

OBJECTIVE: To study the changes of the hippocampus metabolites with MRS to provide some clues for exploring the possible underlying unrecognised factors and pathophysiological mechanisms of psychogenic erectile dysfunction (ED). METHODS: Fifteen cases of psychogenic erectile dysfunction and 15 normal volunteers (the control) were studied by a clinical 1. 5T MRI/MRS system. Proton multi-voxel spectroscopy imaging (1H-MRSI) was obtained from both sides of the hippocampus region. N-acetylaspartate (NAA), creatine and phosphocreatine (Cr) and choline-containing compounds (Cho) were determined and the ratios of NAA/Cr and Cho/Cr were calculated respectively. RESULTS: The NAA/Cr ratio was significantly lower in the ED patients than in the control (P < 0.05). There was no significant difference in the Cho/Cr ratio between the two groups (P > 0.05). CONCLUSION: Psychogenic erectile dysfunction may not be simply a functional disease. The hippocampus may be involved in the pathophysiology of psychogenic ED. The disease may have some previously unrecognised underlying aetiological factors and pathophysiological mechanisms.

[Instructions for the combination of PDE5 inhibitors with alpha-blockers].

The incidences of both benign prostatic hyperplasia and erectile dysfunction are increasing with aging, so the coexist of the two disease in the same patient is not rare. However, it is stumble for the clinicians to treat the patients. The problem of the combination of phosphodiesterase-5 (PDE5) inhibitors with alpha-blockers is getting more and more attraction. The article reviews the drug information of the combination of sildenafil, vardenafil and tadalafil with alpha-blockers respectively, and reports the results of the latest studies regarding it. The purpose is to help clinicians understand the problem across the board, so as to make the application of this class of drug correctly and the treatment of the disease suitably.

Polysaccharide extract isolated from ganoderma lucidum protects rat cerebral cortical neurons from hypoxia/reoxygenation injury.

The effect of polysaccharide extract isolated from Ganoderma lucidum (Gl-PS) on rat cortical neuronal cultures exposed to hypoxia/reoxygenation (H/R) was studied in vitro. Gl-PS (1, 10, 100 microg/ml) increased neuron viability following H/R as measured by the inhibition of MTT reduction. Gl-PS also significantly reduced malondialdehyde content and reactive oxygen species production and increased the manganese superoxide dismutase (Mn-SOD) activity; furthermore, the translocation of nuclear factor-kappa B induced by H/R was blocked. These findings suggest that Gl-PS might be useful in treating H/R-induced oxidative stress and Mn-SOD might play a critical role in the neuroprotective effect of Gl-PS against H/R injury.

[Sentinel lymph node identification with methylene blue in cervical cancer].

BACKGROUND & OBJECTIVE: Study of sentinel lymph node(SLN) in cervical cancer has been initiated since recent years, and there are still a lot of unknown factors about SLN identification in cervical cancer. This study was to investigate influential factors of identifying SLN with methylene blue in cervical cancer. METHODS: For 41 patients with cervical cancer enrolled from Jun. 2002 to May 2003, 2-4 ml of methylene blue was injected into cervix at 4-6 sites around the tumor about 90-400 min before operation. The blue-dyed lymph node (BDLN) was considered as SLN. HE staining in step sections, and immunohistochemistry were applied to detect SLN. The influential factors of using methylene blue to detect SLN in cervical cancer were assessed based on the identification rate of SLN, and its false negative rate. RESULTS: SLNs were detected in 31 of 41(75.6%) patients with cervical cancer of stage Ib1-IIb. A total of 85 SLNs were identified,and most frequently located in obturator fossa. SLNs were successfully detected in 20 of 23 (87.0%) patients who had no preoperative radiotherapy or chemotherapy, while in only 11 of 18 (61.1%) patients who had preoperative radiotherapy and/or chemotherapy. SLNs were detected in only 17 of 27(63.0%) patients who were injected with 2-3 ml of methylene blue, while in all of 14 patients whose dose was 3.4-4 ml. Eight patients were confirmed of lymph node metastases by pathology. CONCLUSIONS: The dose of methylene blue recommended to detect SLN in cervical cancer is 3-4 ml. SLN varies in different sites, but most frequently located in obturator fossa.

[Preparation and feasibility of superparamagnetic dextran iron oxide nanoparticles as gene carrier].

BACKGROUND & OBJECTIVE: Application of magnetic nano- particles as gene carrier in gene therapy of tumor has developed quickly. To obtain a new type non-viral gene introduction and therapy system,which is convenient,and can drive target gene to express highly and stably,this study was designed to explore the preparation of superparamagnetic dextran iron oxide nanoparticles(SDION),and the feasibility of SDION used as gene carrier in vitro. METHODS: SDION were prepared by chemical co-precipitation,separated by gel filtration chromatography on Sephacryl S-300HR,and centrifugation techniques,characterized by transmission electron microscopy,laser scattering system,and vibrating sample magnetometer signal processor. The green fluorescent protein-C2 (GFP-C2) plasmid was used as target gene. SDION-GFP- C2 compounds were synthesized by oxidation-reduction reaction. The connection rate of SDION and GFP-C2 was analyzed by agarose electrophoresis,and evaluated by measuring concentration of GFP in the supernatant after centrifugation. Liposome transfection was used as control,the efficiencies of SDION and liposome in transferring GFP gene into human bladder cancer BIU-87 cells were evaluated under fluorescence microscope in vitro. RESULTS: The diameter of SDION ranged from 3 nm to 8 nm, the effective diameter was 59.2 nm, and the saturation magnetization was 0.23 emu/g. After oxidized by sodium periodate of 10 mmol/L,and deoxidized by sodium hydride boron of 0.5 mol/L, SDION could connect with GFP in maximum degree,the transfection efficiency of SDION as gene carrier was about 45%, even higher than that of liposome (about 30%). CONCLUSION: SDION could connect with GFP plasmid by oxidation- reduction reaction,and success to transfer GFP gene into human bladder cancer BIU-87 cells in vitro.

[Correlation between P53 codon 72 polymorphism and tumorigenesis of cervical cancer].

BACKGROUND & OBJECTIVE: The p53 codon 72 polymorphism affects human papillomavirus (HPV) E6-mediated degradation of p53. This study was to investigate distribution of p53 polymorphism in Guangdong women, and relationship between p53 polymorphism and tumorigenesis of cervical cancer. METHODS: Cervical smears of 46 patients with cervical cancer (case group), and 84 patients with benign gynecologic tumor (control group) treated in our hospital from Sept. 2002 to May 2003 were collected. DNA, extracted from cervical smears,were examined by polymerase chain reaction (PCR) for detection of HPV DNA and p53 codon 72 polymorphism. RESULTS: Positive rate of HPV DNA in case group was 47.8%, in control group was 20.2%. Proportions of genotypes Arg/Arg, Pro/Pro, and Arg/Pro in case group were 56.5%, 21.7%,and 21.7%, respectively; in control group were 71.4%,20.2%, and 8.3%,respectively. There were no significant differences in proportions of Arg/Arg (OR,0.520; 95% CI,0.245-1.102), and Arg/Pro (OR, 1.095; 95% CI,0.454-2.639) between 2 groups; proportion of Pro/Pro in case group was significantly higher than control group (OR, 3.056; 95% CI,1.076-8.678), but no significant difference was found in women with HPV infection. CONCLUSION: Arg/Arg genotype is not a high-risk factor for cervical cancer in Chinese population, and individuals with Pro/Pro genotype are likely to develop cervical cancer.

[Advance in calcium channel blockers relaxing corpus cavernosum smooth muscle].

Calcium channels exist extensively in the membrane of cardiac, skeletal, smooth muscle cell and neuron. Calcium channel blockers (CCB) were widely used for the treatment of cardiovascular diseases because they could relax vascular smooth muscle. Experimental researches on calcium channel blockers relaxing corpus cavernosum smooth muscle have been reported recently. Whether the blockers can be used for the clinical diagnosis and treatment of erectile dysfunction still need to be further investigated.

[Genetic variation of Citrus calli revealed by the ploidy analyser].

The cell DNA content of forty-eight Citrus calli of different genotype were measured by using the Ploidy Analyser. The results showed that 93.8% out of forty-eight Citrus calli had double DNA content except that Ruby. Weizhang and Kinnow had little varied cells and did not show second peak. Moreover, Pineapple, Meiwa kumquat, Changsha, Russ navel, Guoqing No. 4 and Carter had DNA varied cells including triploids and aneuploids based on the DPAC software analyse. Among the forty-eight Citrus calli, Pineapple had the highest of the DNA varied cells, which was up to 18.57%, Anliucheng had the lowest, 4.07%. There existed significant difference in the variation among genotypes by Duncan Analyse. At the same subcultured medium and at the same subcultured period, the effect of cultural duration on the variation of calli was not significant.

Influences of dibutyryl cyclic adenosine monophosphate and forskolin on human sperm motility in vitro.

AIM: To study the influences of dibutyryl cyclic adenosine monophosphate (dbcAMP) and forskolin on human sperm motility in vitro. METHODS: Semen samples, aseptically obtained by masturbation and prepared by swim-up technique from 20 fertile men, were incubated with different concentrations of dbcAMP and forskolin at 37 deg. Measurements were carried out after 10 min, 20 min, 30 min and 60 min incubation. Motility parameters were estimated by using an automatic analyzing system. RESULTS: Treatment with dbcAMP or forskolin resulted in a significant increase in sperm motility and progressive motility. The larger the concentrations of dbcAMP or forskolin, the greater the effect appeared. The straight linear velocity and curvilinear velocity were not affected by both agents. CONCLUSION: dbcAMP and forskolin increase the motility and progressive motility of human sperm in vitro.
