Computer program for connection and data management of an automated coagulation system: the KC 10.

In Europe, the KC 10, manufactured by Amelung Germany, is one of the instruments most commonly found in coagulation laboratories. For facilitating the work of technical validation, we wrote a software adapted to any IBM or compatible PC running under MS-DOS, to manage the analyser performance. Data are automatically collected via the BCD interface from the analyser or keyed in for the other techniques. The software deals with 64 different analyses entirely 'user defined'. An 'electronic worksheet' presents the results, by page of ten patients. This enables the laboratory technician to assess the coherence of the various data and to perform verifications or complementary tests if necessary. As an option, a blinking asterisk can signal all results outside predetermined range. By moving the cursor through the table, a test result can be deleted, modified or added. A function displays the patient's previous files in a window because the data are recorded in long-term archives at the end of the day. This long-term recording allows a search of previous files to decide additional tests if the patient is unknown. A daily archive function classifies and prints the whole day's work in alphabetical order. A protocol of communication allows connection to a mainframe Bayer-Technicon computer. Blood Coagulation Tests

A data management program for the Electra 800 automatic analyser.

The Electra 800 automatic coagulation analyser rapidly performs most chronometric coagulation tests with high precision. To facilitate data handling, software, adaptable to any PC running under MS-DOS, was written to manage the analyser. Data are automatically collected via the RS232 interface or can be manually input. The software can handle 64 different analyses, all entirely 'user defined'. An 'electronic worksheet' presents the results in pages of ten patients. This enables the operator to assess the data and to perform verifications or complementary tests if necessary. All results outside a predetermined range can be flagged and results can be deleted, modified or added. A patient's previous files can be recalled as the data are archived at the end of the day. A 120 Mb disk can store approximately 130,000 patient files. A daily archive function can print the day's work in alphabetical order. A communication protocol allows connection to a mainframe computer. This program and the user's manual are available on request, free of charge, from the authors.

Differences in the metabolism of epicainide in rats and man.

1. Epicainide is a new potent antiarrhythmic agent, metabolized differently in rats and man. 2. In rats, the aromatic part of the molecule (the two phenyl rings linked to the quaternary carbon atom) undergoes metabolic attack by the mono-oxygenases. The hydroxy and methoxy-hydroxy metabolites are predominant and are excreted in urine and bile both as free and conjugated forms. 3. In contrast, the aromatic moiety of epicainide remains unchanged in man. It is the ethylpyrrolidine ring which is mainly attacked, resulting in the expected N-deethylation, subsequent oxidation of the pyrrolidine moiety, and reduction or hydrolysis of the amide function. 4. A preliminary kinetic analysis of epicainide in man reveals a linear open 2-compartment model. The radioactivity recoveries confirm the absence of any accumulation of the drug in the organism.

[Cutaneous apoprotein B and coronary atherosclerosis].

Cutaneous and plasma lipids (cholesterol and Apoprotein B) were studied in 2 populations (average age 57.5 years), one with pathological and the other with normal coronary angiography. Skin biopsy was performed during the incision of thoracotomy. The concentrations of Apo B and cholesterol in the skin were compared to those of plasma lipids, lipoproteins and apoprotein B for the diagnosis of atherosclerosis. This study showed that skin Apo B was the best marker of coronary atherosclerosis in patients with plasma Apo B concentrations of less than 1.3 g/l. The skin Apo B concentration was closely correlated to the presence but not to the severity of this arterial pathology. The cardiovascular risk factors of this population, studied separately and in a cumulative manner, confirmed the results of previously published reports.

Determination of apoprotein B in the skin.

To determine the origin of cutaneous cholesterol a method has been developed for extracting and determining by electro-immuno-assay apoprotein B in the skin as already performed by Hoff at the arterial level. This study demonstrated the presence of apoprotein B in the skin and its quantification. The easy and reliable technique employed required only conventional laboratory instruments. Repeatability and reproducibility studies give encouraging results comparable with those obtained on arterial tissue. They show that at least one part of skin cholesterol originates in low density lipoproteins.

Skin cholesterol and skin apoprotein B in atherosclerosis.

In order to quantify the accumulation of apoprotein B in skin we determined the amount of apo B in the skin of atherosclerotic patients using a modified Hoff's method with extraction and electroimmunoassay. Skin biopsies were taken from the lower limbs of nineteen male patients with arteriosclerosis obliterans during revascularization and from the thoracic region of thirty male patients with ischemic heart disease during coronary bypass graft surgery and thirty one male patients with abnormalities of cardiac valves during valvuloplasty. A significant positive correlation between skin apo B and skin cholesterol was found in three groups. Our data support the hypothesis that cholesterol deposit in the skin of patients with atherosclerosis is derived from plasma lipoprotein B and that the content of skin lipids parallels with the development of atherosclerosis in man.

Cytochrome P-450 monooxygenase activities in human and rat liver microsomes.

Microsomes were prepared from human livers obtained from renal donors of various ages and both sexes. Their drug-metabolizing capacity was measured and compared to that of rat liver microsomes. The following parameters were investigated: cytochrome P-450, cytochrome B5, NADPH-cytochrome c reductase, epoxide hydrolase, aryl hydrocarbon hydroxylase, benzphetamine N-demethylase, p-nitroanisole-O-demethylase, ethoxycoumarin-O-deethylase, steroid-16 alpha-hydroxylase. In addition, the metabolism of benzo(a)pyrene, progesterone, pregnenolone, testosterone, dehydroepiandrosterone and estradiol was studied in detail in vitro. The inhibitory effect of metyrapone and alpha-naphthoflavone on 7-ethoxycoumarin-O-deethylase was measured. The microsomal proteins of both species were separated by polyacrylamide gel electrophoresis in the presence of dodecyl sulfate. The following conclusions were drawn from the results obtained. Human liver microsomes can be stored under optimal conditions for the measurement of a large variety of enzymic activities. Human liver microsomes are able to metabolize the various xenobiotics used as substrates with a rate similar to that of female rat liver microsomes. No sex-linked difference in enzymic activity was observed in human microsomes. Significant differences in benzo(a)pyrene and steroid metabolism were registered when human and rat liver microsomes were compared. The monooxygenase activities, the sensitivity to in vitro alpha-naphthoflavone and metyrapone, the results of steroid metabolism, and slab gel electrophoresis are strong indications for multiplicity of human liver cytochrome P-450.

Cutaneous cholesterol and plasma lipoproteins in elderly active and bedridden patients compared with young adults.

This report concerns two groups of elderly patients, one active and the other inactive with cardiovascular histories. Cutaneous and plasma cholesterol (Ch) and the lipoprotein fractions were analyzed in each group and compared with those of a young group. The cutaneous histological ageing type was studied in the three groups. No histological difference was seen to exist between the elderly populations (skin type II), the young group had normal type O skin. A significant difference was seen to exist between the levels of cutaneous Ch of the two elderly groups and that of the young group. Changes in the cutaneous Ch with respect to the ratio (total Ch/HDL Ch) were found to differ in the two elderly groups: the active elderly patients presented a positive correlation as did the young subjects but the inactive patients showed a negative correlation.

Chronic lathyrism. Plasma, aorta and rat skin treated for five months with beta-aminopropionitrile with or without high fat diet.

Previous experiments proved that white Wistar rats, resistant to spontaneous or experimental atheroma, present arteriosclerosis (parietal aorta lesions) and atheroma lesions (fat deposits) after a 9 week-treatment by Beta-aminopropionitrile (BAPN) followed by a high fat diet. The present work shows that BAPN alone, administered for 5 months, does not induced atheroma; its formation needs both lipid humoral disorders and parietal aortic lesions. It confirms certain facts noted in the aging of man and in the formation of atheroma.

[Effect of pyridinol carbamate on long-term lathyrism and experimental atheroma in the rat. Histological and biochemical study].

Three-week-old rats were fed with Beta-aminopropionitrile (BAPN) with a normal or hyperlipidic diet. After a few weeks BAPN induced aortic elastic fibres disruption which as not worsened when the treatment was continued up the seven months. Adding a hyperlipidic diet produced atherosclerotic-like lesions. Pretreatment with Pyridinol Carbamate (PDC) whichever the diet used did not modify serum of aortic cholesterol levels. It decreased the severity of aortic lesions and delayed the onset of less severe atheromatous lesions. It seems that PDC increased the free/esterified aortic cholesterol ratio resulting in a better elimination of the aortic cholesterol.

Aminopropionitrile

Cutaneous cholesterol and plasma lipoproteins in young subjects.

The present report concerns the biochemical and anatomo-pathological study of the skin and the plasma lipoprotein fractions in a young population (average age : 22 +/- 1.5 years). The levels of cutaneous cholesterol and the ratio HDL cholesterol/total cholesterol are closely linked even through the histological structure of the skin is still normal; the level of cutaneous cholesterol giving a good indication of the degree of vascular ageing. The observations confirm the usefulness of measuring these two parameters in the detection of early atherosclerosis.
