[Genescan analysis of the loss of heterozygosity on the long arm of chromosome 6 in non-small cell lung cancer].

To investigate if there are microsatellite loci in the long arm of chromosome 6 that have close relationship with non-small cell lung cancer, Multiple PCR approach was used to analyze the 36 loci in the long arm of chromosome 6. The PCR products were analyzed in PAGE and then the electrophoresis maps were analyzed with Genescan and Genotyper. There is different LOH frequency in different loci. The total frequency of LOH in 41 lung cancers was 78%(32/41), with the highest frequency of LOH was detected on the locus D6S302(75%). There are 14 loci which have LOH frequency more than 20% and the loci are mainly located in 2 regions: 6q16.3-q21 [6 loci D6S458 (21.43%), D6S1694 (26.92%), D6S1717 (35.71%), D6S1565 (40%), D6S302 (75%), D6S1706 (36.36%) and 6q26-q27 (5 loci D6S1550 (38.46%), D6S264 (20%), D6S1585 (25%), D6S446 (33.33%), D6S281 (30.77%)], There may be tumor suppressor genes located in the two regions, which have a close relationship with non-small cell lung cancer.

[Inhibition of glucose transporter 1 overexpression in mesangial cells by rhein].

OBJECTIVE: To evaluate the effect of rhein on the overexpression of glucose transporter 1(GLUT 1) in mesangial cells transfected with GLUT 1 gene in vitro. METHODS: Rat mesangial cells were transduced with the human GLUT 1 gene(MCGT 1) by retrovirus vector. Mesangial cells transduced with bacterial beta-galactosidase(MCLacZ) were used as control. Glucose uptake was detected by 2-deoxyglucose(2-DG). Cell volume, RNA/DNA ratio and protein/DNA ratio were evaluated by flow cytometry analysis. The synthesis of collagen IV and fibronectin were measured by 3H-proline incorporation and flow cytometry. The activity of glutamine: fructose-6-phosphate aminotransferase (GFAT) was assayed by spectrophotometry method. The expression of collagen IV, fibronectin and GFAT were analyzed by RT-PCR. RESULTS: MCGT 1 exhibited a higher 2-DG uptake with increased Vmax value as compared to MCLaZ(741 +/- 60.5) dpm.microgram prot-1 vs (92.2 +/- 9) dpm.microgram prot-1, (P < 0.01), Even cultured in normal glucose concentration, MCGT1 showed cell hypertrophy, including increased cell volume, RNA/DNA and protein/DNA ratios; increased synthesis of extracellular matrix. 3H-proline incorporation significantly enhanced in MCGT1 compared with that in the control(7.0 +/- 0.4) dpm.cell-1 vs (4.6 +/- 0.6) dpm.cell-1, (P < 0.01); and GFAT activity increased to 1.8 fold. Rhein could inhibit 2-DG uptake of MCGT1(560 +/- 64) dpm.microgram prot-1 vs (741 +/- 60.5) dpm.microgram prot-1, (P < 0.05) in a dose-dependent manner, and reverse the cell hypertrophy of MCGT 1. In addition, rhein could diminish the enhanced GFAT activity of MCGT 1 with no influence in mRNA expression. CONCLUSION: It is suggested that glucose transport activity was an important modulator of cellular glucose metabolism in mesangial cells. Rhein could ameliorate the metabolic derangement of MCGT 1 by decreasing glucose uptake. These findings have led us closer to the identification of therapeutic approaches to abort GLUT 1 overexpression in diabetic nephropathy.

[Relationship between water supply and consumption of main planting tree species of protection forests in loess area of western Shanxi Province].

Based on three years point-fixed observation, this paper analyzed the relationship between water supply and consumption of black locust(Robinia pseudoscacia L.) and Chinese pine(Pinus tabulaeformis Carr.), the main tree species of protection forests in the loess area of the western Shanxi Province. In April, May and June, the water supply was less than water consumption, while in raining season, the water supply was abundant, and the water storage in soil was increased. During dry seasons and years, more water consumption and less available water supply occurred in high-density stand, showing a serious water deficiency. The water deficiency was in order of south-facing slope > semi-south-facing slope > north-facing slope. The study shows that water consumption characters coefficient could be used as an index to indicate the growth pattern of trees, and the relationship between water supply and consumption.

[Runoff loss ways of nutrients in non-irrigated farmland in Hefei outskirts].

Taking non-irrigated flatlands in Hefei outskirts as experiment observation areas, yellow brown soil as selected soil type, and < 3 degrees as ground slope, the time of runoff development, runoff quantities, and silt carrying capacity under different rainfall intensity were observed on the non-irrigated farmlands with different kinds of crops such as grain, oil, cotton and vegetables, and with different coverage for continuous 3 years. Based on the observation, water and soil samples were collected and analyzed. The results showed that the loss way of the surface soil and nutrients in non-irrigated flatlands was obviously different from that on the sloping fields and on the upland. Over 98% of available N, P, and K nutrient loss were run off by chemical erosion. Physical erosion was the main way of the surface soil and organic matter loss, and 96.14% of total organic loss was caused by physical erosion.

[Analysis on the genotype of measles virus epidemic strains in Dongguan city, Guangdong province in 2000].

OBJECTIVE: To analyze the genotype of the measles virus epidemic strains in Dongguan according to the patients suffered measles in 2000. METHODS: The saliva of 55 patients with acute measles symptoms were collected and detected with RT-PCR. The PCR products of the 5 samples were sequenced and analyzed with the software DNASTAR. RESULTS: The positive detection rate was 87.3%, whereas the positive rate of anti MV-IgM in serum was only 70.9%. 68.8% of the saliva samples of patients with anti MV-IgM negative results showing MV RNA positive. The 293 bp fragment of N gene and 194 bp fragment of M gene were amplified from the vaccine virus and the specimens. The sequences of the epidemic strains were identical in both M gene and N gene. CONCLUSION: These results suggested that the outbreak came from a single MV strain. The epidemic strain was obviously different from the standard strains discovered in other countries of the world (the deviation were 1.4%-11.7%) that might belong to H genotype.

WAIS-III reliability data for clinical groups.

Reliability estimates for psychological tests are almost always reported for nonclinical populations (e.g., the normative samples). Such practice will no longer be sufficient as the new standards for testing call for an adequate assessment of psychometric properties within the specific population being tested. The purpose of this study was to provide internal consistency reliability estimates for clinical groups on the Wechsler Adult Intelligence Scale-Third Edition. The study included data from 403 clinical participants composed of 10 groups of adults recruited as part of the WAIS-III clinical validity studies. Split-half reliability coefficients were obtained for these groups replicating the procedure used in the WAIS-III. With 8 of the clinical groups, the split-half reliability coefficients were comparable to, or even higher than, those reported for the WAIS-III standardization sample. In general, the split-half coefficients for the Verbal subtests tended to be higher than the coefficients for the Performance subtests. The high magnitude and general pattern of these coefficients demonstrate that the WAIS-III scales do not include additional error variance above and beyond what is reported in the WAIS-III-WMS-III Technical Manual when it was used to assess certain clinical groups. For the ADHD/ADD and learning disabilities groups, however, the internal consistencies coefficients of some subtests were relatively lower, although not statistically significant, than the normative sample. These findings may reflect more heterogeneity within the groups. The implications for assessment and for using alternate methods of determining the psychometric properties in these populations are discussed.

Criterion validity of new WAIS-II subtest scores after traumatic brain injury.

The criterion validity of the new subtests from the Wechsler Adult Intelligence Scale-Third Edition (WAIS-III; Wechsler, 1997) was evaluated in a sample of 100 patients with traumatic brain injury (TBI). Letter-Number Sequencing and Symbol Search, but not Matrix Reasoning, yielded statistically significant differences in performance between patients with moderate-severe TBI, patients with mild TBI, and demographically matched controls. Level of education accounted for a statistically significant amount of variance in the performance of patients with TBI, in addition to that explained by injury severity variables. It is concluded that Letter-Number Sequencing and Symbol Search have satisfactory criterion validity, but that they need to be supplemented with other measures in the context of neuropsychological evaluations. Matrix Reasoning, on the other hand, is not sensitive to the sequelae of TBI and more studies are needed to determine how it can be used for neuropsychological assessment purposes.

Effect of all-trans retinoic acid and arsenic trioxide on tissue factor expression in acute promyelocytic leukemia cells.

OBJECTIVE: To study the effect of all-trans retinoic acid (ATRA) and arsenic troxide (As2O3) on tissue factor (TF) expression and procoagulant activity (PCA) of acute promyelocytic leukemia (APL) cells in vivo and in vitro. METHODS: PCA from freshly isolated APL blasts from APL patients treated with ATRA or As2O3 was detected using a one-stage clotting assay. TF antigen was detected by ELISA and TF mRNA by RT-PCR. The maturation sensitive (NB4) or resistant subclones (NB4-R1) of the promyelocytic NB4 cell line, as well as U937 cells infected with pMSCV-PML-RARa treated with or without ATRA or As2O3, were also examined. RESULTS: Both ATRA and As2O3 can down-regulate the TF antigen, its mRNA transcription and membrane PCA of APL cells in vivo and in vitro, in a time-dependent manner. The TF antigen level in PML-RARa + U937 cells was significantly higher than that in U937 cells infected with retrovirus vector. Both ATRA and As2O3 can also down-regulate the TF antigen in U937 cells transfected with or without PML-RARa. CONCLUSION: Tissue factor expression and PCA in APL cells may be down-regulated by ATRA and As2O3. By down-regulating TF expression, As2O3 might also be used to improve the DIC-related hemorrhage in APL. Our data indicate that elevated TF antigen in PML-RARa + U937 may be related to the fusion protein PML-RARa. The down-regulating effect of ATRA and As2O3 on TF expression in U937 cells might not involve this fusion protein.

Monoclonal anti-idiotype antibody bearing the internal image of nasopharyngeal carcinoma associated antigen.

OBJECTIVE: To generate and characterize anti-idiotypic monoclonal antibody (Ab2) that bears the internal image of nasopharyngeal carcinoma (NPC) associated antigen. METHODS: Using NPC monoclonal antibody (Ab1) as immunogen, hybridoma cells were obtained by fusion of SP2/0 myeloma cells with immunized murine spleen cells. Positive clones were screened by Sandwich ELISA and a binding inhibition test. To determine whether Ab2 possess the internal image of the original antigen or not, mice were immunized with Ab2. ELISA and the competitive inhibition assay tested anti-anti-idiotypic antibodies (Ab3) in anti-sera. Cell-mediated immunity to tumors induced by Ab2 was investigated by a delayed-type hypersensitivity response and the mouse T-cell proliferation assay. RESULTS: Anti-idiotypic monoclonal antibodies against the monoclonal anti-NPC antibodies FC2 and HNL5 were generated that recognize NPC associated antigens. These Ab2, which were designated 2H4 and 5D3, could inhibit the binding of FC2 or HNL5 to NPC cell lines. Anti-sera from the immunized mice, which contained Ab3, could compete with FC2 or HNL5 for binding with NPC cell by a competitive inhibition assay. Mice immunized with 2H4 or 5D3 coupled with keyhole limpet hemocyanin (KLH), showed a positive and specific delayed-type hypersensitivity (DTH) reaction after stimulation by NPC cells. The mouse T cell proliferative assay indicated that there was a significantly higher proliferative response of the splenocytes in the experimental groups than that in control groups. CONCLUSIONS: Anti-idiotypic antibodies 2H4 and 5D3 are Ab2 beta bearing the internal image of the epitope of NPC associated antigen. Either 2H4 or 5D3 expressing three-dimensional shapes that resemble the structure of natural antigens could induce humoral and cellular immune response.

[Nonlinear behavior of nerve fibers with constant current stimulation].

There are abundant nonlinear phenomenons in life systems. Using the Hodgkin-Huxley model, we studied the excitability of single nerve fiber under constant current stimulation by means of computer simulation. It was found that stimulated with different current intensities, the membrane potentials of the nerve fiber are at equilibrium points or periodic oscillations. And the frequencies of the periodic oscillations are obtained.

Contrast enhanced MR angiography in pulmonary sequestration.

OBJECTIVE: To evaluate the feasibility of contrast enhanced three-dimensional (3D) magnetic resonance angiography (MRA) in identifying the systemic blood supply in pulmonary sequestration. METHODS: Three patients who were suspected of pulmonary sequestration were examined by contrast enhanced 3D MRA. MR images were compared with surgical findings. RESULTS: Contrast enhanced 3D MRA clearly demonstrated systemic arteries from the descending thoracic aorta supplying the basilar segments of the lower lobe in each case. Pulmonary veins from these segments, which drained into the left atrium in two cases, were also detected. MRA images were consistent with those observed in surgery. CONCLUSION: Contrast enhanced 3D MRA allows accurate diagnosis of pulmonary sequestration and may obviate the need for more invasive investigations.

[Morphological observation of Mahaim's fibers in 7 cases].

OBJECTIVE: To investigate the Mahaim's fibers of the cardiac conduction system (CCS) of human hearts and study their morphological features. METHODS: The CCS from 165 cases of sudden death without extracardiac causes and those from 760 cases of non-cardiac death were sampled using a method established by the authors. Serial sections were prepared for histological observation. RESULTS: Mahaim's fibers were discovered in 7 cases, 5 cases were in the sudden death group, accounting for 3% in that group, 2 cases were in the non-cardiac death group, accounting for 0.26% in that group. All cases belonged to the bundle-ventricular branch type. The characteristics of Mahaim's fibers were found to be as follows. Their transverse diameter was 10 - 35 microns. The bodies of the Mahaim's fibers that originated from A-V bundle were slender with nucleus in the middle of the cell. While the shape of cells from the left bundle branch varied according to the origin of the Mahaim's fiber with size enlarging from up downward. Both types of cells showed abundant lightly stained cytoplasm, similar to A-V bundle and left bundle branch cells. The cells were not bifurcated and some of them were surrounded by a thin layer of fibrous tissue. CONCLUSION: Mahaim's fibers are not a rarity and it is an abnormal development of an A-V by-path.

[The germination and preliminary formation of science of TCM formula in the pre Qin dynasty].

Shennong tasted hundreds of grasses and found herbs during the legend period. The appearance of prescription should be so early if single drug was regarded as single -- item recipe. However, the explicit records in literature appeared at the end of the Spring and Autumn period and the beginning of the Warring States at the latest, evidenced by the custom of "presenting a recipe" of the Qi states. The medical books of Mawangdui, mainly Wu shi er bing fang (Prescriptions for 52 Kinds of Diseases), recorded the classification of prescriptions based on the sort of diseases, composing, preparation, method of decocting medicinal herbs, contraindication of prescriptions, and the therapeutic methods, compatibility of complex prescriptions and determination of treatment based on the differentiation of syndromes (Zheng), etc. reflected in the recipes, which could represent the primary level of science of TCM formula of the Warring States in China. The classic medical book Nei jing (Internal Canon of Medicine), including denominations, composition, principles of compatibility and rules of clinical application of concrete prescriptions, especially therapies and theories of composed prescription, have great actions on the development of science of TCM formula.

[Effect of PCNA antisense oligooxynucleotides and VEGF antisense oligoxynucleotides on growth of hepatocellular carcinoma transplanted in nude mice].

OBJECTIVE: To study the inhibitory effect of PCNA antisense oligonucleotides or/and VEGF antisense oligonucleotides on growth of human hepatocellular carcinoma in nude mice. METHODS: Human hepatocellular carcinoma transplanted subcutaneously in nude mice model was established, and subsequently was divided into four groups: PCNA-ASODN group, VEGF-ASODN group, combined group and control group. Different treatments were given respectively. The weigh and volume of subcutaneous tumors was measured, and the morphological changes of tumor cells was observed under microscope. RESULTS: The growth inhibitory rate in the PCNA-ASODN group was 72.8%, 44.9% in the VEGF-ASODN group, and 87.2% in the combined group. The combined group which was treated with PCNA-ASODN and VEGF-ASODN showed more effective inhibitory than the PCNA-ASODN group or the VEGF-ASODN group respectively. CONCLUSIONS: The growth of human hepatocellular carcinoma in nude mice can be inhibited by PCNA antisense oligonucleotides and VEGF antisense oligonucleotides respectively. Better inhibitory effect can be obtained if PCNA antisense oligonucleotides and VEGF antisense oligonucleotides are combined in use.

Secretory Expression of Different C-terminal Truncated HCV E1 Proteins in Mammalian Cells and Characterization of the Expressed Products.

Three fragments of the HCV envelope 1 (E1) with different C-terminal truncation at aa310, aa325, aa340 were cloned into the mammalian expression vector pSecTagB. An epitope in the hepatitis B surface antigen, preS1(21--47), were genetically engineered onto the N-terminus of the recombinant protein and used as an affinity tag for detection and purification. The resulting pSec-preS1-E1t310, pSec-preS1-E1t325 and pSec-preS1-E1t340 were transiently expressed in the HeLa cells and the antigenicity, secretory efficiency and glycosylation type of the recombinant E1 proteins were compared. All of the three recombinant proteins could be detected by both preS1 monoclonal antibody and E1 polyclonal antiserum. The expression products were secreted and highly mannose-type glycosylated, with S1E1t325 being secreted, indicating the influence of the hydrophobic regions on the secretion of the E1 protein. Three CHO cell lines expressing the proteins, S1E1t310, S1E1t325 and S1E1t340, were established and the CHO/pSecS1E1t325 was chosen for further study. The secreted S1E1t325 could be enriched from cell culture medium by the preS1 antibody-coupled Sepharose. The glycosylation analysis indicated the lack of complex glycogen even after the E1 was secreted via Golgi complexes. The established stable cell lines and anti-preS1 affinity method could be utilized to enrich and purify the HCV E1 expressed in mammalian cells, and may be used for further characterization of this protein.
